Abstract

The research was conducted at Seed Science and Technology Field Experimental and Laboratory, Faculty of Agriculture, Bogor Agricultural University since January until July 1998. Randomized Block Design was used in this experiment which consist of two factors and three I replications. The first factors were row spacing 60 cm x 25 cm (J1) and 60 cm x 45 cm (J2) and the second factors were application of 100 ml paclobutrazol/plant with concentration 0 ppm (P1), 125 ppm (P2), 250 ppm (P3) and 500 ppm (P4). The plant height was increased at narrow spacing especially at 0 ppm paclobutrazol. Paclobutrazol application reduced plant height, increased stem diameter and it's caused minimal lodging at generatif period. The highest seed production per plot was resulted at 60 cm x 25 cm with 250 ppm and 500 ppm of paclobutrazol (146 % and 155 % as compared with control treatment), and the highest seed production per plant showed at 60 cm x 45 cm and 60 cm x 25 cm with 500 ppm of paclobutrazol (197 % and 198 % compared with control treatment). Paclobutrazol125 ppm, 250 ppm, and 500 ppm increased flower diameter, number seed per flower, 1000 seed weight, germination capacity, and seed vigour. The optimum combination of treatment was showed at 60 cm x 25 cm and 250 ppm of paclobutrazol.

Abstract

The objectives of this research were to find the most compatible coating formula for rice seeds (Experiment 1), and to evaluate the effect of coating formula on viability, vigor and storability of the seeds (Experiment 2). This research was conducted in Laboratory of Seed Science and Technology at IPB, Bogor and Laboratory of Seed Technology at PT East West Seed Indonesia, Purwakarta from December 2010 to February 2011. In Experiment 1, the ten coating materials (formula) were arranged in single factor of Completely Randomized Design with four replications, i.e. uncoated seed, 3% alginate + 1% talc, 3% alginate + 1% gypsum, 3% alginate + 1% peat, 3% arabic gum + 1% talc, 3% arabic gum + 1% gypsum, 3% arabic gum + 1% peat, 1.5% CMC + 1% talc, 1.5% CMC + 1% gypsum, and 1.5% CMC + 1% peat. Experiment 2 used Split Plot Design with four replications. The main plot was storage periods i.e. 0, 1 and 2 months. The seeds were stored in two storage conditions: ambient and air conditioned room. The subplot was coating formula (same as in Experiment 1). The results of Experiment 1 showed that the most compatible coating material for rice seeds were 1.5% CMC + 1% talc, and 1.5% CMC + 1% gypsum, these formula were able to maintain vigor of the coated seeds. In Experiment 2, coating formula of 1.5% CMC + 1% peat generated higher seed vigor than uncoated seeds after two months storage, 3% gum arabic + 1% gypsum was able to keep vigor for one month storage in ambient condition. The coating formula of 3% alginate + 1% peat was able to keep viability and vigor of the coated seeds for two months in air conditioned room. Abstract

Bogor is one of the centers of taro production, but productivity is low. It was caused by the use of low-quality seeds. The use of good quality of seed will increase taro productivity and production. Good quality seeds were uniform in size, clean, free from other varieties, and had high viability (vigor). This training was aimed at capturing the taro farmer's condition, cultivation technique, and knowledge about good quality of seed. The training was conducted in KTD Saluyu from August to September 2022 and was attended by 25 farmers. Training activities included expert presentations, discussions, and interviews. Evaluation of farmer knowledge change was determined by pre- and post-tests, while the effectiveness of training activities was evaluated to improve its effectiveness in the future. Taro farmers were dominated by men, aged about 40–59 years old (58.3%) and more than 60 years old, and almost all of their education background was elementary school. Farmers cultivated taro by sharing profit land (58.3%), ownership land (33.3%), and rent land (8.3%). Taro production was dominated by less than 1 ton, the monoculture system about 66.7%, and intercropping about 33.3%. Planting material was by farmer own seed from the previous season because there were limited certified seeds. Farmer knowledge increased by about 45.8% (from 4.9 to 7.2). This training was held under good management, as shown by the training evaluation score of about 90%. This training requires that it be held more intensively and sustainably through intensive assistance in the field and a seed production demonstration.

The research that consisted of two activities was performed at the Laboratory. Experiment 1 the aim to obtain biological agents that have high antagonistic potential against Xanthomonas oryzae pv. oryzae (Xoo) and two biological agents that are compatible to one another. Experiments 2 to determine the effect of rice (Oryza sativa L.) seeds coated with biological agents to improve seed quality and reduce levels of Xoo infection. In the first experiment, four isolates of bacterial antagonists, i.e: isolate Pseudomonas diminuta A6; isolate P. aeruginosa A54; isolate Bacillus subtilis 11/C, and isolate B. subtilis 5/B, were tested for their antagonism activities against Xoo on PSA plates using the method of growth inhibition zone with filter paper. Furthermore, the four antagonists were tested for their compatibility with each other on PSA plates. In the second experiment used a completely randomized design with a single factor (seed coating) consisting of seven levels, namely: negative control, healthy seed; positive control, the seeds contaminated with Xoo; P. diminuta A6 and B. subtilis 5/B; alginate 3% + 1% peat + P. diminuta A6 and B. subtilis % 5/B; arabic gum 3% + 1% gypsum + P. diminuta A6 and B. subtilis 5/B; CMC 1.5% + 1% talc + P. diminuta A6 and B. subtilis 5/B, and bactericide streptomycin sulfate 20%. Results of Experiment 1 showed that isolates P. diminuta A6 has the highest antagonistic potential against Xoo based on the diameter of inhibition zone on Xoo, followed by isolate B. subtilis 5/B. Isolate P. diminuta A6 and B. subtilis 5/B had a good growth compatibility with no antagonism. Therefore, isolates P. diminuta A6 and B. subtilis 5/B were selected and used as biocontrol agents in further studies. Results of Experiment 2 showed that treatment with P. diminuta A6 + B. subtilis 5/B gave the best of the increase of seed vigor, and the decrease of Xoo infection level in the seeds.

Jernang rattan has a high economic value due to its resin content in the exocarp, as the raw material of dyes, medicines, and cosmetics. The resin is highest in the immature fruit skin (exocarp); therefore, fruits are harvested before the seed is germinable. As a consequence, regeneration is very scarce, and the species is categorized as a rare plant. This research was aimed at investigating the flowering phenology and seed development of jernang rattan until they reach physiological maturity, when the viability and vigor are at their highest. The flowering phenology was observed based on Biologische Bundesanstalt Bundessortenamt and Chemische Industrie (BBCH), at the inflorescence stage, flowering stage, fruit development stage, and fruit ripening stage. The inflorescence development from flower bud until ready to bloom took 136.4 ±10.9 days for male plants and 141.6 ±16.4 days for female plants. The inflorescence of staminate flowers bloomed longer (29.4 ±2.7 days) than pistillate flowers (15.4 ± 4.0 days). Fruit abortion mostly occurs during the first six months of fruit development, coincides with the nuclear stage of endosperm, and the final seed set is 56%. It takes around 27 months of one reproduction cycle from the appearance of the inflorescence to seed maturity. The fruit reaches physiological maturity approximately at 22 MAA when the seed is of high viability and vigor, and harvesting at 22 MAA is best for seed production. It is recommended to set up a seed production area separated from the resin production area.

Cowpea is a legume with a potential nutritional content almost equivalent to soybeans; it can be developed as a substitute agent for raw materials for food processing. One of the factors in obtaining high cowpea production is to use of quality seeds. Our study aimed to evaluate the performance and morpho-physiological correlations and viability of cowpea seed collections from IPB University Bogor and the Indonesian Research Institute of Legume. We used ten genotypes of cowpea, which had superior seed information. The results showed that the ten cowpea genotypes had superior morpho-physiological and seed viability. The seed germination percentage was categorized as medium-high (>60%). Eight cowpea genotypes were grouped into one major group based on morpho-physiological characteristics and viability. Furthermore, positive correlations were found between the observed variables of seed viability (seed germination, seed growth speed, and seed vigor index). Our research results are useful as a source of genetic diversity for plant breeders in developing new superior varieties in the future. In addition, agronomists can use this information to plant cowpeas based on planting location.
