Abstract

ABSTRACTThe use of probiotic bacteria as biocontrol agents is one of the methods of controlling anthracnose disease caused by Colletotrichum acutatum. The objective of this research was to suppress the infection of C. acutatum and increase chilli pepper seedling vigour. The research involved factorial experiments arranged in a completely randomized design with two factors. The first factor was seed coating involving six treatments, i.e., control, seed coating without bacteria, seed coating with Bacillus sp., Pseudomonas sp., Actinomycetes sp, and fungicide. The second factor was the seed storage period where six storage periods were experimented, i.e., 0, 1, 2, 3, 4, 5, and 6 months. The results showed that the coating treatment of Bacillus sp., Pseudomonas sp., and Actinomycetes sp. improved germination, growth rate and number of leaves and reduced the incidence of attacks and infection of hypocotyls at 5 month storage period.Keywords: Actinomycetes sp., Bacillus sp., Pseudomonas sp., seed coating, storage period

Abstract

ABSTRACTThe use of high quality seed is one of the key factors to improve productivity. Probiotic bacteria has been used to increase plant growth and to control pathogens. The objective of the research was to evaluate methods of chili seed production that yielded high physiological and pathological quality using probiotic fluorescent bacteria Pseudomonas (P24). The bacteria was expected to function as plant growth promoting bacteria as well as capable of controling seedborne pathogen Colletotrichum acutatum causes antrachnose. The experiment was conducted during March until October 2014 in Seed Health Laboratory, Plant Bacteriology Laboratory, and Leuwikopo experimental garden, IPB. The experiment was arranged in a split plot randomized complete block design with three replications. The main plots were untreated and inoculation of C. acutatum. The subplot was six treatments of fluorescent Pseudomonas (P24) application. The results showed that matriconditioning and spraying of fluorescent Pseudomonas (P24) on nursery decreased disease incidence significantly. Matriconditioning and spraying of fluorescent Pseudomonas (P24) on nursery and flowering phase increased number of healthy fruit and total weight of seeds per plant significantly. These applications also increased seed physiological  quality indicated by germination percentage (GP) 77.04%, growth rate (GR) 9.72% etmal-1, vigor index (VI) 29.74%, and seed health by suppresed C. acutatum infection up to 12.25%.Keywords: anthracnose, Colletotrichum acutatum, control, matriconditioning, spraying

Abstract

Nutmeg is an Indonesian native plant, produces essential oill as an export commodity. However, it has low seed uniformity that mengurangi keseragaman tanaman di lapang. The main objective of this research was to obtain morphological characteritics as parameter for nutmeg mother plants selection. This field research was carried out from August-December 2014 at Toisapu, Wakal and Lula nutmeg seed source plantations in Ambon, and Seed Storage and Testing Laboratory, IPB. Characterization was undertaken on ten mother plants from each  Wakal and Lula seed source plantations and eight mother plants from Toisapu seed source plantation. Quantitative and qualitative characters of vegetative, fruit, mace and seed from every mother plants were analyzed with  Gower’s similarity test. Mother plants that produced high uniformity seed was the 7th (0.80), the 8th (0.93) and the 10th (0.80) mother plants in Wakal; the 2nd (0.80) mother plant in Toisapu; the  2nd (0.82), the 5th (0.80), the 6th (0.82), the 8th (0.84), the 9th (0.80) and the 10th (0.93)  mother plants in Lula. Those selected mother plants produced morphological uniform seed morphologically. Lula seed plantation had the highest uniformity level among mother plants (0.77-0.87) compared to Toisapu (0.70-0.87) and Wakal (0.69-0.84) mother plants. The morphological characteristic can be use to select the source seed mother plants which able to produce high morphological uniformity seeds.Keywords: Gower’s similarity, seed production, uniformity

Abstract

The objectives of this research were to find the most compatible coating formula for rice seeds (Experiment 1), and to evaluate the effect of coating formula on viability, vigor and storability of the seeds (Experiment 2). This research was conducted in Laboratory of Seed Science and Technology at IPB, Bogor and Laboratory of Seed Technology at PT East West Seed Indonesia, Purwakarta from December 2010 to February 2011. In Experiment 1, the ten coating materials (formula) were arranged in single factor of Completely Randomized Design with four replications, i.e. uncoated seed, 3% alginate + 1% talc, 3% alginate + 1% gypsum, 3% alginate + 1% peat, 3% arabic gum + 1% talc, 3% arabic gum + 1% gypsum, 3% arabic gum + 1% peat, 1.5% CMC + 1% talc, 1.5% CMC + 1% gypsum, and 1.5% CMC + 1% peat. Experiment 2 used Split Plot Design with four replications. The main plot was storage periods i.e. 0, 1 and 2 months. The seeds were stored in two storage conditions: ambient and air conditioned room. The subplot was coating formula (same as in Experiment 1). The results of Experiment 1 showed that the most compatible coating material for rice seeds were 1.5% CMC + 1% talc, and 1.5% CMC + 1% gypsum, these formula were able to maintain vigor of the coated seeds. In Experiment 2, coating formula of 1.5% CMC + 1% peat generated higher seed vigor than uncoated seeds after two months storage, 3% gum arabic + 1% gypsum was able to keep vigor for one month storage in ambient condition. The coating formula of 3% alginate + 1% peat was able to keep viability and vigor of the coated seeds for two months in air conditioned room. Abstract

Benih srikaya yang bermutu sangat diperlukan untuk mendukung pengembangan bibit tanaman srikaya. Penelitian ini bertujuan menentukan tingkat kemasakan dan posisi benih dalam buah yang tepat guna mendapatkan benih bermutu dan mendapatkan perlakuan terbaik untuk meningkatkan performa bibit melalui penggunaan rizobakteri dan CaCO3. Percobaan pertama menggunakan rancangan acak lengkap (RAL) dua faktor dengan faktor pertama adalah warna kulit buah dan faktor kedua adalah posisi benih dalam buah. Percobaan kedua menggunakan RAL dengan rizobakteri, CaCO3, dan kombinasi keduanya sebagai faktor tunggal. Waktu hitungan pertama pengamatan daya berkecambah pada 36 hari setelah tanam (HST) dan hitungan akhir pada 56 HST. Kualitas benih terbaik diperoleh dari warna kulit buah kuning berdasarkan bobot kering benih, daya berkecambah, indeks vigor, bobot kering kecambah normal, dan kecepatan tumbuh benih. Vigor tumbuh benih tertinggi diperoleh dari buah dengan warna kulit kuning dari bagian ujung dan pangkal buah. Perlakuan CaCO3 mempengaruhi pemanjangan akar, lebar daun, diameter batang, dan panjang daun. Aplikasi CaCO3 memberikan pengaruh terbaik terhadap pertumbuhan bibit diikuti dengan perlakuan Pseudomonas + Actimomycetes + CaCO3. Abstract

Padi hitam merupakan sumber karbohidrat yang bermanfaat untuk kesehatan. Pengujian daya hasil merupakan tahapan penting dalam menentukan keberhasilan pemuliaan padi beras hitam. Tujuan penelitian ini ialah untuk memperoleh informasi keragaan agronomi dan menyeleksi galur-galur dihaploid padi beras hitam yang memiliki penampilan agronomi baik dan berdaya hasil tinggi untuk pengujian multi lokasi. Penelitian ini dilakukan dari bulan November 2019 sampai Juli 2020 di Bogor, Jawa Barat dan Malang, Jawa Timur. Sebanyak 23 galur dihaploid padi hitam yang memiliki sifat agronomi baik dengan produktivitas tinggi dan 3 varietas pembanding yaitu Aek Sibundong, Jeliteng, dan Inpari 24 digunakan sebagai materi genetik. Studi ini menggunakan rancangan rancangan kelompok lengkap teracak dengan 3 ulangan pada setiap percobaan. Hasil penelitian menunjukkan bahwa interaksi genotipe dan lingkungan berpengaruh nyata terhadap semua karakter pengamatan kecuali pada karakter lama pengisian gabah berdasarkan analisis keragaman. Sebanyak 14 galur yang terseleksi berdasarkan indeks terboboti memiliki karakteristik jumlah anakan produktif per rumpun dari 15.8 hingga 25.9, umur panen genjah dari 115.2 to 121.9 hari setelah semai, dan produktivitas >5.4 ton ha-1. Keragaan agronomi dan daya hasil galur-galur dihaploid padi yang terseleksi pada penelitian ini memerlukan pengujian lebih lanjut melalui uji multi lokasi untuk memperoleh calon varietas unggul baru padi beras hitam. Abstract

True shallot seeds are preferable to tubers as planting materials because of their advantages in storage longevity, lower management cost, and lower seed-borne disease risk. However, environmentally limited production and seed deterioration in uncontrolled storage have limited its application. Pre-sowing hydration invigoration accelerates and synchronizes the germination of deteriorated seeds. Ultra-fine bubble water (UFBW) contains reactive oxygen species, which can advance the pre-germinative metabolism of the seed. This study examined true shallot seed invigoration using UFBW. The experiment was conducted in a two-factor of randomized complete block design. The first factor was initial seed vigor based on the vigor index with six levels, i.e., 24, 15, 13, 12, 10, and 9%. The second factor was the invigoration treatment of UFBW (8 and 23 ppm dissolved oxygen concentrations) with two durations (24 and 48 h), 50 ppm GA3, and 3% KNO3 (24 h). UFBW with 23 ppm invigoration for 48 h improved seed vigor based on all vigor parameters. The GA3 50 ppm treatment is the most effective in increasing seed vigor. Abstract

Pengujian viabilitas benih menggunakan uji tetrazolium merupakan salah satu metode penentuan viabilitas benih dengan mengevaluasi tingkat warna merah yang terbentuk pada benih. Warna merah terbentuk dari aktivitas enzim dehidrogenase pada siklus trichloroacetic acid. Penentuan tingkat warna merah pada benih ini dapat menggunakan alat sensor infra red. Penelitian ini bertujuan menguji alat sensor infra red untuk menentukan intensitas warna secara kuantitatif dalam uji tetrazolium serta menentukan korelasi alat sensor infra red dengan pengujian benih secara fisiologis. Percobaan disusun menggunakan rancangan kelompok lengkap teracak dengan faktor tunggal yaitu tingkat viabilitas benih berdasarkan daya berkecambah masing-masing adalah 78.9%, 70.7% dan 40.0 %. Hasil penelitian menunjukkan bahwa aplikasi sensor intensitas infra red untuk uji tetrazolium pada benih kacang tanah menghasilkan nilai koefisien korelasi (r) sebesar 0.62 antara nilai RGB merah bagian dalam kotiledon (Rd) dengan kecambah normal. Abstract

The increasing demand for rice makes upland rice a promising alternative. However, one major challenge in upland rice cultivation is the availability of high-quality seeds. This study aimed to determine the physiological seed maturity, after-ripening period, storage longevity, and optimal fertilizer dose to produce high-quality IPB 10G Tanimar. The research used a split-plot randomized complete block design with two factors and three replications. The main plot was phosphate fertilizer doses (50, 100, and 150 kg ha-1 SP-36), and the sub-plot was seed maturity stages (112, 116, 120, 124, and 128 days after planting(DAP). The results showed that fertilizer dose had no significant effect on vegetative and generative growth. The application of 100 kg ha⁻¹ P fertilizer produced the best seed quality based on the initial vigor index. The best maturity stage was 124 DAS with a heat unit of 2,505 °Cd, as indicated by the parameters of initial vigor index, germination rate during storage, and storability. The shortest after-ripening period was also found at 124 DAS, with the minimum germination rate (>80%) achieved one week after storage (WAS). The best storage vigor at the end of the observation period (8 WAS) was at 124 DAS, with a germination rate of 79.67% and a vigor index of 21.89%.   Keywords: high-quality seed; heat unit; phosphate fertilizer; storage longevity; vigor index

Abstract

Andaliman is a species found in the Lake Toba region of North Sumatra, Indonesia. To determine its optimal harvest time and the length of its physiological maturity, it is necessary to observe fruit development in new locations outside its natural habitat. Andaliman seeds are known for their low germination rates and are classified as endospermic seeds. This study aims to analyze the stages of fruit and seed development, evaluate the effects of seed treatments, and identify the most effective germination test methods for andaliman seeds. Seeds were treated with a 50% H2SO4 solution for 15 min and with a 200 ppm GA3 + 100 ppm kinetin solution for 48 hr. The seeds were then tested using pleated paper and top paper media to assess germination response. Changes in fruit coloration indicated various stages of development: intense yellowish-green at 17 weeks after anthesis, deep yellowish-green at 20 weeks, deep red at 23 weeks, and dark greyish-yellowish-brown at 26 weeks. During development, some fruits experienced embryo abortion, resulting in many empty or wrinkled seeds. The seeds reached physiological maturity at 23 weeks after anthesis, as indicated by a deep red fruit color, increased seed dry weight, and enhanced germination and seed growth rates. The treatment with GA3 and kinetin yielded the highest germination rate of 20%, with the pleated paper method proving to be the most effective for germination testing.
