Abstract

Rimpang kunyit (Curcuma longa L.) merupakan tanaman tradisional yang telah dikenal berkhasiat dalam menyembuhkan luka. Kapur sirih (CaCO3) telah terbukti memiliki khasiat sebagai antiinflamasi. Kombinasi keduanya secara empiris diketahui mempunyai efek dalam penyembuhan luka. Penelitian bertujuan untuk mengkaji potensi kombinasi ekstrak rimpang kunyit dan kapur sirih sebagai antiinflamasi dan menyembuhkan luka sayat serta menentukan komposisi yang efektif. Hewan uji yang digunakan pada penelitian ini adalah tikus putih jantan (Rattus norvegicus) yang dibagi dalam 5 kelompok uji. Kelompok 1 sebagai kontrol negatif, kelompok 2 sebagai kontrol positif, kelompok 3, 4 dan 5 diberi sediaan kombinasi ekstrak rimpang kunyit dan kapur sirih dengan komposisi masing-masing 1:1, 1:2, dan 2:1. Uji aktivitas antiinflamasi dilakukan dengan metode paw edema induksi λ karagenan 1% sebanyak 0,1 ml. Pengukuran volume udem dilakukan setiap jam selama 6 jam menggunakan pletismometer sedangkan untuk metode luka sayat dilakukan dengan membuat sayatan pada bagian punggung hewan uji dengan panjang 2 cm dan kedalaman 2 mm. Parameter penyembuhan luka adalah rerata panjang luka. Pengukuran luka sayat menggunakan jangka sorong digital yang dilakukan selama 12 hari. Hasil penelitian menunjukkan bahwa kombinasi ekstrak rimpang kunyit dan kapur sirih memiliki potensi antiinflamasi dan penyembuhan luka sayat. Komposisi yang paling efektif dalam menyembuhkan luka sayat yaitu komposisi 2:1 sedangkan komposisi yang efektif sebagai antiinflamasi adalah 1:2.

Abstract

Hantap (Sterculia coccinea Jack) has been used traditionally for various health issues, including cancer treatment. The therapeutic effects of natural ingredients are often attributed to their chemical constituents. This study aimed to analyze the phytochemical contents and cytotoxic activities of S. coccinea leaves on HeLa and MCF-7 cancer cell lines. The quantitative phytochemical analysis was carried out following standard laboratory procedures. Phytochemical compounds were identified using LC-MS/MS QTOF. The MTT assay PrestoBlue™ Cell Viability Reagent test method was used to test cytotoxic activity in the cell culture. Extraction was carried out by the maceration method using 96% ethanol as solvent. The quantitative analysis revealed that tannins were the major phytochemical constituent in the highest percentage of 72.16%, followed by alkaloids, flavonoids, and steroids, with values of 30.80, 28.66, and 2.85%, respectively. Saponins were present in the lowest percentage of 1.15%. The ethanolic extract exhibited moderate cytotoxicity on HeLa and MCF-7 cells with IC50 values of 591.00 and 578.10 µg/mL, respectively. Identification using LC-MS/MS showed the suspected compounds 5,7-dihydroxy-3-(4'-hydroxybenzyl)chromone as homoisoflavanones and kaempferide-3-O-α-L-rhamnosyl-7-O-α-L-rhamnoside from flavonol triglycosides. These results may contribute to the study on the use of leaves extract of S. coccinea for developing a chemoprevention agent.

Abstract

Penyakit degeneratif merupakan kondisi kesehatan yang menyebabkan jaringan atau organ memburuk dari waktu ke waktu berkaitan dengan penuaan, atau memburuk selama proses penuaan. Hipertensi dan Diabetes Mellitus termasuk penyakit degeneratif dengan morbiditas dan mortalitas tinggi. Pengetahuan mengenai penyakit ini masih kurang terutama masyarakat di pedesaan yang terbatas untuk akses informasi dan layanan kesehatan. Oleh karena itu kegiatan pengabdian ini bertujuan untuk memberikan edukasi kepada masyarakat desa Apal, kecamatan Liang, Banggai Kepulauan tentang penata laksanaan penyakit degeneratif khususnya hipertensi dan DM serta melakukan skrining kesehatan dengan pengukuran tekanan darah dan kadar gula darah (KGD) sewaktu sebagai upaya peningkatan kesadaran masyarakat untuk menjaga kesehatan. Metode yang digunakan adalah penyuluhan, pengukuran tekanan darah dan KGD sewaktu serta konsultasi kesehatan. Hasil skrining menunjukkan bahwa 68% peserta mengalami hipertensi stage 1 (12%) dan stage 2 (56%) serta prehipertensi 32%, sedangkan hasil pemeriksaan KGD menunjukkan 76% memiliki KGD normal, 24% masuk batas tinggi (6%) dan tinggi (18%). Berdasarkan hasil pengabdian yang dilakukan menunjukkan adanya peningkatan pengetahuan dan kesadaran masyarakat di desa Apal mengenai penyakit hipertensi dan DM.

Abstract

People in Pamona Puselemba Subdistrict in Poso Regency have used various plant species to maintain health and prevent various diseases, one of which is metabolic disorders. This study aims to determine the types and parts of plants, processing methods, use, duration of treatment, frequency of use, and chemical content contained in plants that are efficacious as drugs for metabolic disorders. The research was conducted descriptively using qualitative methods and the technique of taking informants (Hattra and sufferers) with the purposive sampling method. The results obtained were 32 types of plants, consisting of 24 families, for the treatment of metabolic disorders. The plant parts used were 49% leaves, 12% stems, 21% fruit, 6% tubers, 3% flowers, 3% rhizomes, 3% seeds, and 3% hair. Processing method: 77%, blended, brewed 13%, without processing 77%. How to use: drink 89%, eat directly 11%, duration of treatment 1 week, and frequency of use 2 times a day. The content of compounds contained in medicinal plants is very diverse, such as flavonoids, terpenoids, saponins, tannins, alkaloids, allisin, and other chemical compounds that can be used in the treatment of metabolic disorders. People in Pamona sub-district still use traditional medicine to treat metabolic diseases.

Abstract

The risk of various degenerative diseases is more experienced by older adults than young people. One of the most common health problems among the elderly is hypertension. Delays in treating hypertensive patients are caused by the majority of patients only coming to health facilities after experiencing complications and lack of access to public health services. The incidence of dyslipidemia and diabetes mellitus (DM) also continues to grow, which are risk factors for coronary heart disease. For this reason, promotive and preventive efforts for the elderly group are significant through increasing knowledge and understanding of degenerative diseases. The community service partner is the Banpres Pololo Community Health Center, one of the community health centers in Sigi Regency with high cases of degenerative diseases, and the target of community service is the elderly in Uenuni Palolo village. Implementing the activity was health education about hypertension, DM, and dyslipidemia, health screening, and health counseling. The screening results found that 94.12% of older adults suffered from hypertension, 17.65% suffered from DM, and 52.94% suffered from hypercholesterolemia. Health counseling results show that most older adults did not understand the importance of early detection of degenerative diseases. Therefore, health education and counseling for the community still need to be improved.

Abstract

Endophytic fungi produce bioactive metabolites that exhibit antibacterial, antioxidant, and anti-cancer activity. Rui (Harrisonia perforata (Blanco) Merr.) is a traditional medicinal plant proven to be an antibacterial and anticancer. Endophytic fungi live in plant tissues without damaging or producing chemicals that infect the host cell. Endophytes produce the same and similar compound as an original plant. This study aimed to isolate the endophytic fungi from Rui and determine their antibacterial, antioxidant, and cytotoxic activity. 13 fungal isolates were successfully isolated using PDA for seven days at 30°C. Molecular identification using the ITS1 and ITS4 DNA sequences only revealed six species: Diaporthe sp., Phomopsis sp., Aspergillus tubingensis, Aspergillus viridinutans, Pseudofusicoccum sp., and Daldinia eschscholtz. Seven isolates showed antibacterial activity based on plate assay. Aspergillus tubingensis and RA-1 exhibited antibacterial activity against Staphylococcus aureus and Salmonella typhi, with the MIC and MBC starting from 24 to 32 mg/ml. The DPPH assay showed the most increased antioxidant activity in Daldinia eschscholtzii with an IC50 of 98.14±2.39 µg/ml. The Brine Shrimp Lethality Test (BSLT) showed the highest potential cytotoxic activity, as shown by Aspergillus tubingensis and Phomopsis sp. with LC50 of 7.78±7.48 and 30.83±0.39 µg/ml. In conclusion, our study demonstrates that the fungal extract from Rui could be a source of antibacterial, antioxidant, and cytotoxic agents.

Abstract

Red Gedi Leaves Extract (RGLE) have phytochemicals such as alkaloids, flavonoids, polyphenols, saponins and tannins which known as antidiabetic agents. Study had been done about RGLE (Abelmoschus manihot (L.) Medic) on blood glucose level, 8-hydroxy-deoksiguanosin, malondialdehyde and insulin in diabetic rats (Rattus norvegicus). This study aims to prove the effects of RGLE in blood glucose levels, 8-hydroxy-deoksiguanosin, and malondialdehyde and insulin levels in rats with diabetes induced by streptozotocin and which is the most effective dosage of RGLE. Study used pre and post test design. RGLE obtained by maceration method using by ethanol 96%. There were 120 white male rats which were divided into six treatment groups, each group consisting of 20 individuals. Group I (healthy control), II (negative control) were given by Na-CMC 0.5%. Group III as the positive control were given by glibenclamide, group IV, V, and VI given by RGLE with different doses (150, 300 and 450)mg/kg BW. Animal test were diabetic induced by Streptozotocin with 40 mg/kg BW dosage i.p. Data were analyzed by One Way (ANOVA). The result showed that RGLE can reduced blood glucose levels, 8-hydroxy-deoksiguanosin, and malondialdehyde and increased insulin levels in diabetic rats. The Effective Dosage of RGLE is 150 mg/kg BW. Keywords : RGLE(Red Gedi Leaves Extract),Glukose,8-hidroksideoksiguanosin, Malondialdehyde, Insulin. ABSTRAK EDGM (Ekstrak Daun Gedi Merah) mengandung alkaloid, flavonoid, polifenol, saponin, dan tanin diduga mempunyai efek antidiabetes. Telah dilakukan penelitian dari EDGM (Abelmoschus manihot (L.) Medik) terhadap kadar glukosa darah (KGD), 8-hidroksi-deoksiguanosin, malondialdehid, insulin pada tikus putih (Rattus norvegicus) diabetes yang diinduksi streptozotocin telah dilakukan. Tujuan penelitian ini untuk membuktikan kemampuan EDGM terhadap KGD, 8-hidroksi-deoksiguanosin, dan malondialdehid, insulin pada tikus putih diabetes diinduksi streptozotocin dan menentukan dosis paling efektif. Rancangan penelitian digunakan adalah pre and post test design. EDGM diperoleh dengan cara metode etanol 96%. Penelitian ini menggunakan hewan uji tikus putih jantan sebanyak 120 ekor yang dibagi menjadi 6 kelompok perlakuan, tiap kelompok terdiri dari 20 ekor. Kelompok kontrol I (sehat), II negatif(-) diberikan suspensi Na-CMC 0,5%, III positif(+) diberikan suspensi glibenklamid, kelompok IV, V, dan VI diberikan EDGM masing-masing dengan dosis 150, 300 dan 450 mg/kgBB. Model hewan uji diabetes dibuat dengan cara induksi streptozotocin dengan dosis 40 mg/kgBB secara intraperitoneal. Data yang diperoleh dianalisis menggunakan uji statistik one way (ANOVA) pada taraf kepercayaan 95%. Berdasarkan hasil penelitian menunjukkan bahwa ekstrak daun gedi merah dapat menurunkan KGD, 8-hidroksi-deoksiguanosin, malondialdehid dan meningkatkan kadar insulin pada tikus putih diabetes yang diinduksi streptozotocin. Dosis EDGM yang efektif yaitu dosis 150 mg/kg BB. Kata Kunci: EDGM(Ekstrak DaunGediMerah),Glukosa Darah,8-Hidroksideoksiguanosin, Malondialdehid, insulin.

Abstract

Nephroprotective compound is a compound that has the ability to protect the kidneys from kidney disorders caused by free radicals. This research aimed to determine the nephroprotective effect of the combination of Abelmoschus manihot leaves and Orthosiphon stamineus leaves extract as well as to know the effective extract dose combinations as nephroprotective. This research applied Group Random Design with 6 treatment groups consists of 5 tested animals. Group I was normal control. Group II was negative control. All extract groups was given 0,75% of ethylene glycol and 2% of ammonium chloride. Group III was given 100 mg/kg BW of Abelmoschus manihot leaves extract and 200 mg/kg BW of Orthosiphon stamineus leaves extract. Group IV was given 50 mg/kg BW of Abelmoschus manihot leaves extract and 100 mg/kg BW of Orthosiphon stamineus leaves extract. Group V was given 100 mg/kg BW of Abelmoschus manihot leaves extract. Group VI was given 200 mg/kg BW of Orthosiphon stamineus leaves extract. Treatment was given for 14 days and the levels of creatine and urea were measured on day 0, 8 and 15. The results show the nephroprotective effect in all the extract groups, but the most effective one was the group III.

Abstract

Hibiscus surattensis L. leaves have been known as a folk medicine. Several previous reports have shown several potential therapeutic effects, including antidiabetic, antibacterial, anti-diarrheal, analgesic, hepatoprotective, and antioxidant, but no toxicity study is similar to the current one. Therefore, it is necessary to investigate the cytotoxic effect of these commonly consumed medicinal leaves. This study aimed to investigate the cytotoxicity of Hibiscus surattensis L. leaves extract (HSL). The cytotoxic effect was determined through brine shrimp lethality bioassay (BSLT) and in vitro Presto Blue assay of MCF-7 and Hela cell.  HSL extract was obtained by maceration using 96% ethanol. The BSLT results showed that the HSL has LC50 value of 224.814 μg/mL. MTT assay results showed the IC50 value of the HSL extract on MCF-7 cells was 1419.83 μg/mL, and Hela cells were 2050.50 μg/mL. Based on the LC50 value that has been analyzed against HSL extract shows an LC50 value of less than 1000 μg/mL which can be concluded while having potential cytotoxic activity against experimental animal larvae of A. salina Leach. The results of the cytotoxic test with Presto Blue assay on two cancer cells used showed that the HSL extract was not toxic.

Abstract

Dyslipidemia is a medical condition caused by a lipid metabolism disorder in the body. In the early stages, dyslipidemia often shows no symptoms or complaints, so many sufferers are unaware of their condition. However, examinations of total cholesterol, low-density lipoprotein (LDL), and triglyceride levels often show elevated results in people with dyslipidemia. Public awareness of the dangers of dyslipidemia remains low, particularly among the elderly. To increase understanding and awareness of dyslipidemia, a community service activity was conducted in Pana Village. This activity began with cholesterol level checks. Following this, a presentation was given on the dangers of dyslipidemia, especially among the elderly, and the importance of maintaining a healthy lifestyle to prevent risks of heart disease, stroke, and even death. This counseling session focused on lifestyle changes that can help control cholesterol levels, such as regulating diet, exercising regularly, and avoiding unhealthy habits like smoking. Out of 95 participants who underwent cholesterol testing, the results showed that 46 elderly individuals (43%) had cholesterol levels below 200 mg/dl, which is still considered within normal limits. However, 54 elderly individuals (57.3%) had cholesterol levels above 200 mg/dl, indicating a high prevalence of dyslipidemia in this age group. These findings emphasize the importance of health education for the elderly, so they become more aware of the risks of dyslipidemia and are motivated to adopt a healthier lifestyle to prevent serious complications.
