Abstract

 Latar belakang: Infeksi kronis pada tonsil palatina disertai pembesaran tonsil palatina, dapat menyebabkan sumbatan pada jalan nafas bagian atas. Gangguan pertumbuhan banyak ditemukan pada kelompok pasien ini. Tujuan: Mengetahui apakah ada pengaruh tindakan tonsiloadenoidektomi terhadap kadar serum insulin-like growth factor-1 (IGF-1) dan nilai body mass index (BMI) pada anak yang menderita hipertrofi tonsil palatina dan adenoid, dengan gejala obstructive sleep apnoea syndrome (OSAS). Metode: Desain penelitian adalah eksperimental dengan studi pre- dan post-trial, dengan membandingkan kadar IGF-1 dan nilai BMI dalam serum anak dengan gejala OSAS, sebelum dan tiga bulan setelah dilakukan tonsiloadenoidektomi. Hasil: Sesuai protokol penelitian terdapat 14 subjek penelitian, menunjukkan peningkatan nilai kadar IGF-1 dan nilai BMI. Tindakan tonsiloadenoidektomi terbukti dapat meningkatkan kadar hormon pertumbuhan, yang tergambar dari peningkatan kadar IGF-1 dan nilai BMI. Kesimpulan: Tindakan tonsiloadenoidektomi berpengaruh terhadap peningkatan kadar hormon pertumbuhan, yang disimpulkan melalui pengukuran kadar IGF-1 dan nilai BMI dalam serum anak dengan gejala OSAS. Kata kunci: tonsiloadenoidektomi, insulin-like growth factor-1, obstructive sleep apnoea syndromeABSTRACT Background: Chronic infection with the enlargement of the palatine tonsil, could cause blockage of the upper airway. Growth disorders are common in this patient group. Purpose: To determine the effect of tonsilloadenoidectomy on the level of serum IGF-1 in children with palatine tonsil and adenoid hypertrophy and obstructive sleep apnoea syndrome (OSAS) symptoms. Methods: The study design was experimental, pre-and post-study, by comparing the serum levels of IGF-1 and the value of Body Mass Index (BMI) in children with OSAS symptoms, before and three months after tonsiloadenoidectomy. Result: According to the study protocol, there were 14 subjects showed increased levels of IGF-1 and BMI values. Tonsilloadenoidectomy was shown to increase the growth hormone levels, which was reflected from the increased levels of IGF-1 and BMI. Conclusion: Tonsiloadenoidectomy affected the increased levels of growth hormone, which was concluded through the measurement of the levels of IGF-1 and the value of BMI in the serum of children with symptoms of OSAS. Abstract

ABSTRACT — Background: Laryngopharyngeal reflux (LPR) is the backflow of gastric and or duodenal fluid into the larynx, pharynx, trachea, and bronchi. The prevalence of LPR is difficult to determine due to the limited gold standard and the large variety of LPR symptoms. Damage can occur due to the decrease in pH value and also because of exposure to harmful enzymes in reflux, including bile acid. Purpose : This study was conducted to analyze bile acid levels in the saliva of LPR patient and non LPR subject. Methods:  This study is an observational study with a case-control design. The study was conducted in the ORL-HNS Department of Dr. M. Djamil Hospital, Padang, West Sumatra, Indonesia. The total sample size was 44 people. We enrolled 22 healthy subjects as the control group and 22 patients suspected of having LPR. Result: LPR patients are more common in women than in men, with 12 women and 10 men. Bile acid in the LPR group means of is 25.08±7.67µM, meanwhile, in the healthy group, the mean was 18.99±8.26 µM. There is a statistically significant in the incidence of LPR with the bile acids (p = 0.015) based on t- independent test. Conclusion: Our study confirmed that bile acids in saliva play a major role in diagnosing LPR.

Abstract

Tujuan: Mengetahui pengaruh pemberiak ekstrak Moringa Oleifera terhadap kadar malondialdehyde pada tikus wistar model hiperglikemia. Metode: Penelitian ini melibatkan 20 ekor tikus strain wistar betina yang berumur 8-12 minggu dengan bobot 200-250 gram yang memenuhi kriteria inklusi maupun ekslusi. Tikus dibagi menjadi kelompok empat, yaitu Kelompok kontrol negatif (K-), Kontrol positif (K+), Perlakuan 1 (P1) dan Perlakuan 2 (P2). Kelompok P1 hanya diberikan makan dan minum standar saja, tanpa perlakuan. Kelompok K+ diinjeksi aloksan secara Intraperotoneal dengan dosis 100 mg/kgBB sehingga tikus menjadi hiperglikemia, P1 diberi injeksi aloksan menjadi hiperglikemia dan diberikan ekstrak moringa oleifera dengan dosis 250 mg/kgBB dan P2 injeksi aloksan menjadi hiperglikemia dan diberikan ekstrak moringa oleifera dengan dosis 500 mg/ kgBB selama 21 hari, Pada hari ke-22 semua kelompok diambil darah untuk dilakukan pemeriksaan kadar malondialdehyde. Hasil: Rerata kadar malondialdehid pada K-, K+, P1 dan P2 berturut-turut, yaitu 1,81±0,06 nmol/mL, 3,81±0,24 nmol/mL, 2,55±0,30 nmol/ mL, 2,09±0,21 nmol/mL, dengan nilai p=0,01. Kesimpulan : Pemberian ekstrak moringa oleifera sebagai antioksidan berpengaruh terhadap penurunan kadar malondialdehid

Abstract

Tujuan: mengetahui pengaruh pemberian ekstrak daun sungkai dalam menghambat proliferasi dan migrasi sel HeLa.Metode: Penelitian ini merupakan penelitian eksperimental yang menggunakan 24 disk kultur sel HeLa yang konfluens. Sampel dibagi menjadi 4 kelompok (K,P1,P2 dan P3). Kelompok perlakuan diberikan ekstrak daun sungkai dengan konsentrasi IC25, IC50, dan IC75 yang diinkubasi selama 72 jam. Analisis data menggunakan uji One Way ANOVA dan Post Hoc Bonferroni. Hasil: Rerata jumlah sel proliferatif yang didapatkan pada kelompok K, P1, P2, dan P3 berturut-turut adalah 12,1 x 105, 8,94 x 105, 6,14 x 105 dan 5,04 x 105. Rerata migrasi jumlah sel yang berkumpul pada zona scratch pada kelompok K, P1, P2, dan P3 berturut-turut adalah 49,.0, 237,8, 210,5, dan 100,7. Didapatkan perbedaan yang bermakna tiap kelompok pada uji proliferasi dan migrasi dengan nilai p = 0,000 (p < 0,05).Kesimpulan: Terdapat pengaruh pemberian ekstrak daun sungkai terhadap proliferasi dan migrasi sel HeLa. Perbedaan bermakna ditemukan pada kelompok kontrol dan kelompok perlakuan dengan konsentrasi ekstrak IC25, IC50, dan IC75.

Abstract

Background: Hyperglycemia is characterized by elevated blood glucose levels caused by impaired insulin production or function, leading to oxidative stress. Probiotic-fermented milk containing Lactiplantibacillus pentosus strain HBUAS 53657 (PFM) has antioxidant and antimicrobial properties that may reduce oxidative stress. Objective: This study aimed to investigate the effects of PFM L. pentosus strain HBUAS 53657 on serum glutathione peroxidase (GPx) activity and pancreatic histopathology in hyperglycemic rats. Method: A post-test-only experimental design was conducted using 25 adult male Wistar rats divided into five groups (n = 5 per group): normal control, hyperglycemic control group, and hyperglycemic rats treated with PFM at doses of 1 × 10⁸ CFU/day (P1), 1 × 10⁹ CFU/day (P2), and 1 × 10¹⁰ CFU/day (P3) for 28 days. Serum GPx activity was assessed via spectrophotometry, Langerhans islet area was measured using ImageJ, and islet damage was evaluated using the Ningrum Score. Statistical analysis was performed using One-Way ANOVA and Kruskal-Wallis tests. Results: PFM administration at all doses significantly increased serum GPx activity (p = 0.002), with an optimal dose of 1 × 10⁹ CFU/day (P2). PFM also increased the Langerhans islet area (p = 0.001) and reduced islet damage scores (p < 0.001) in a dose-dependent manner. Conclusion: Probiotic-fermented milk containing Lactiplantibacillus pentosus strain HBUAS 53657 increases serum GPx activity, expands Langerhans islet area, and reduces islet damage in hyperglycemic rats.

Abstract

SARS-CoV-2 is believed to cause cytopathic effects in forming multinucleated cells, known as syncytia. Syncytia due to SARS-CoV-2 infection found in lung tissue samples of COVID-19 patients represents a case of COVID-19 with a poor prognosis. Therefore, it is very important to study the mechanism of syncytia formation and to test candidate materials that can inhibit the occurrence of syncytia and potentially be applied in the treatment or prevention of COVID-19. Since syncytia counting and analysis are time-consuming, we utilized a high-content screening (HCS) instrument in this study to automate syncytia analysis. We used 293T cells transfected with plasmids to express the SARS-CoV-2 spike, human angiotensin-converting enzyme-2 (hACE-2), and a plasmid encoding lifeact-GFP as an F-actin biosensor to facilitate syncytia analysis using the HCS instrument. In this study, syncytia analysis was carried out using HCS software. The HCS application categorizes cells as multi-nuclei by counting the number of cell nuclei stained with DAPI in cells that emitted green fluorescence due to lifeact-GFP expression. Syncytia analysis is time-consuming because of the calculation of the number of syncytia formed in a confluent cell monolayer culture. Hopefully, utilizing the HCS platform can accelerate the test of syncytia inhibition after various treatments using test compounds.Keywords: 293T cells, high-content analysis, SARS-CoV-2, spike, syncytia.

Abstract

Latar Belakang: Kulit berfungsi sebagai pelindung tubuh manusia dan rentan mengalami kerusakan, salah satunya luka bakar. Luka bakar disebabkan oleh trauma panas atau dingin yang memiliki risiko morbiditas dan mortalitas yang tinggi. Diabetes melitus dapat menghambat penyembuhan luka bakar dengan memperpanjang inflamasi, mengganggu proses re-epitelisasi, sintesis, dan proliferasi fibroblas. Human Bone Marrow Mesenchymal Stem Cells (hBM-MSCs) dapat mempercepat penyembuhan luka bakar pada kulit. Objektif: Penelitian ini bertujuan untuk mengetahui efek hBM-MSCs terhadap deposisi kolagen pada penyembuhan luka bakar dengan diabetes melitus. Metode: Penelitian ini dilakukan secara post-test only control group design. Penelitian ini menggunakan 30 blok parafin yang berisikan jaringan kulit luka bakar full thickness tikus diabetes melitus yang diterminasi pada hari ke-3, ke-7, dan ke-14. Terdiri dari kelompok kontrol dan perlakuan. Jaringan kulit dibuatkan preparat histologis dengan pewarnaan Picro Sirius Red untuk menilai deposisi kolagen. Data dianalisis menggunakan uji T-Independent. Hasil: Hasil penelitian menunjukkan terjadi peningkatan deposisi kolagen pada kulit tikus luka bakar full thickness diabetes mellitus setelah diberikan hBM-MSCs. Analisis statistik deposisi kolagen pada hari ke-3 dan ke-7 tidak menunjukkan peningkatan yang signifikan, sedangkan hari ke-14 terlihat peningkatan yang signifikan. Kesimpulan: hBM-MSCs berpotensi meningkatkan deposisi kolagen pada kulit luka bakar tikus diabetes melitus.

Abstract

Hypertension is a disease that often causes death and complications related to cardiovascular disease. The prevalence of hypertension in Indonesia is 34.11%. One of the factors that influence the incidence of hypertension is high levels of the triglycerides in the blood (hypertriglycerida). This study aims to determine the description between triglycerides levels and the incidence of hypertension in the community of Bandar Buat Village. This research was a descriptive study with 29 respondents taken using total sampling techniques from secondary data of community service in the village of Bandar Buat in 2019. Data were selected based on inclusion criteria and exclusion criteria, the data were processed in table to generate frequency and percentages distribution. The results showed that the majority of respondents aged 18-50 years (55.1%), were women (58.6%). The results showed that respondents' triglycerides levels were dominated by hypertriglycerida (58.6%) and blood pressure classification was dominated by normotension (55.2%). Based on age, most people with hypertriglycerida and hypertension are above 50 years old. Based on sex, hypertriglycerida and hypertension both are sufferers more frequent in men. The number of percentage were 66.7%; 75% respectively. The conclusion of this research is the triglycerides level of respondents is dominated by respondents with hypertriglycerida, the majority are aged>50 years, male sex. The incidence of hypertension is high, with majority sufferers aged>50 years, male sex.

Abstract

Elementary school students are consumers who are among those who consume the most food and beverages. Food and drinks sold around the school contain many hazardous substances such as dyes that have been banned from circulating (Rhodamine B in iced drinks, jelly drinks, and so on) and other additives such as formalin which is used as a chewy substance in meatballs sold in carts and restaurants that can cause acute poisonings such as allergies, coughs and chronic poisonings such as cancer of the liver, kidneys and other organs. The purpose of this activity is to increase knowledge about substances in food and an introduction to blood group examination. This activity was carried out at SD Negeri 13 Tungkal Selatan Village in North Pariaman District. The method of activity is in the form of counseling and demonstration. According to the survey before the blood group examination, it turned out that most of the elementary school students did not know their blood type. Before the blood group examination, the students were also given knowledge about determining a person's blood type. After this counseling was carried out, respondents became more aware of the impact of food/snacks on health, and respondents knew their respective blood types. Of the 64 students, there are 22 A, 17 B, 12 AB, 13 O. To avoid problems related to food and blood type. There is a need for continuous health education for elementary school students.

Abstract

Background Hyperglycemia is a medical condition that increases blood glucose levels beyond normal limits because the body lacks enough insulin or insulin does not work properly and is known to trigger oxidative stress. In cells sensitive to Hyperglycemia, such as endothelial cells, excessive glucose loads trigger the formation of Reactive Oxygen Species (ROS) in mitochondria, impairing mitochondrial function. ROS are essential mediators for activating pro-inflammatory signaling pathways; obesity and hyperglycemia-induced ROS production may support the induction of proinflammatory macrophages such as M1 during the onset and development of diabetes. Consumption of olive oil can increase plasma antioxidant capacity and reduce oxidative stress markers. Methods This study aimed to determine the effectiveness of olive oil antioxidants against Malondialdehyde (MDA) in hyperglycemic rats. This is an experimental design with a post-test-only group design. Adult male Wistar rats were divided into three groups (n = 8): negative control (K-), positive control (K+), and treatment (P). The normality test was conducted using the Shapiro-Wilk test, followed by Kruskal Wallis. Results The olive oil was proven to reduce levels of Malondialdehyde in the P group, with an average of 2.29 nmol/mL lower than the K+ group. Although the olive oil reduced the average MDA level in hyperglycemic rats, the post-hoc test showed no difference in MDA between the K+ and P groups (p-value = 0.226). Conclusions Antioxidants in olive oil may effectively lower blood glucose levels, reducing levels of Malondialdehyde in hyperglycemic rats.
