Abstract

Shrimp in Indonesia are generally exported abroad after removing the head, tail, and skin. One alternative to the use of shrimp shell waste that has high economic value is processing shrimp shells into chitosan. The purpose of this study was to determine the shelf life of catfish using chitosan and to determine the optimal concentration of chitosan in catfish preservation. Chitosan applied to catfish showed that in the addition of a 0% chitosan solution, it had a shelf life of up to day 2 for 48 hours for a 0.5 solution; 1; 1.5 and 2% shelf life up to day 5 or for 120 hours. The most optimal concentration of chitosan for use in catfish is a 2% chitosan solution, and the total plate number (ALT) test meets the requirements of the SNI standard so that it can still be consumed.

Abstract

Gaharu (Aquilaria malaccensis Lam.) is one of the natural ingredients that can be used as an antioxidant. Antioxidants are compounds that can inhibit free radical reactions in the body. Free radicals have an impact on the pathogenesis of several diseases in humans due to oxidative stress in cells. The purpose of this study was to determine the class of secondary metabolites of agarwood bark and antioxidant activity and to compare the value of IC50 (Inhibitory concentrations) of the methanol extract of agarwood bark with the IC50 of vitamin C in the DPPH method using UV-Vis spectrophotometry. The methanol extract of agarwood bark was macerated using methanol, then determined the content of secondary metabolites of Simplicia. The determination of antioxidant activity was carried out on the methanol extract of agarwood bark with the addition of DPPH at various concentrations. The sample concentrations were 20, 40, 60, 80, and 100 µg/mL.. As a comparison, vitamin C was used with concentrations of 4, 8, 12, 16 and 20 µg/mL.. Then they calculated percent attenuation (% inhibition) and the value of IC50. The results of the screening on the methanol extract of agarwood bark contained chemical compounds such as flavonoids, saponins, and tannins. The determination of antioxidant activity was carried out using a UV-Vis spectrophotometer with the DDPH method. The results were obtained from the methanol extract of the bark of agarwood, which has antioxidant activity in the strong category with an IC50 value of 94.59 µg/mL.and vitamin C in the very strong category with an IC50 value of 22,11 µg/mL.

Abstract

Porang (Amorphallus muelleri Blume) is one type of tuber plant that has the potential to be developed in Indonesia. Porang plants have been reported to contain chemical compounds that have antioxidant activity. This research was conducted on the leaves of the porang plant with the aim of knowing the antioxidant activity, characterization, and phytochemical profile of the compounds contained therein. Compound identification and characterization were performed using standard methods, and antioxidant activity was determined using the DPPH (1,1- Diphenyl-2-picrylhydrazyl ) method. The results of the identification of chemical compounds showed that the ethanol extract of porang leaves contained chemical compounds of alkaloids, flavonoids, tannins, saponins, and steroids. The results of the examination of the simplicia characterization of porang leaves included the soluble ethanol content of 33.93%, the water soluble extract content of 17.3%, the ash content of 5.58%, the acid insoluble ash content of 0.235% and the water content of 8%. From the results of the simplicia characterization, it shows that the results meet the specified requirements. The results showed that the ethanol extract of porang leaves had antioxidant activity with an IC50 value of 93.04 µg/mL, which in this case is included in the category of strong antioxidant activity. Meanwhile, as a comparison, Vitamin C was used, which has an IC50 value of µg/mL, which is included in the category of very strong antioxidant activity.

Abstract

Jamur susu harimau (Lignosus rhinocerus) merupakan tumbuhan langka yang telah lama digunakan secara tradisional dan memiliki potensi besar sebagai agen antibakteri dan antioksidan, terutama di tengah meningkatnya masalah resistensi antibiotik. Hal ini didukung oleh kandungan senyawa metabolit sekunder seperti flavonoid, steroid, tannin, alkaloid, dan saponin yang berperan penting dalam melawan infeksi bakteri patogen dan menetralkan radikal bebas penyebab kerusakan sel. Penelitian eksperimental ini bertujuan untuk menganalisis secara kuantitatif aktivitas antibakteri dari ekstrak etanol jamur susu harimau terhadap bakteri Gram-positif Staphylococcus aureus dan Gram-negatif Escherichia coli, serta mengevaluasi kapasitas antioksidannya menggunakan dua metode komplementer, yaitu DPPH dan FRAP. Ekstraksi dilakukan dengan metode maserasi menggunakan pelarut etanol 96% dari serbuk simplisia, dan pengujian aktivitas dilakukan pada tiga variasi konsentrasi: 50%, 60%, dan 70%. Hasil uji antibakteri menunjukkan aktivitas yang kuat dan bersifat konsentrasi-tergantung. Pada konsentrasi 70%, diameter zona hambat mencapai 20,40 mm untuk S. aureus dan 16,30 mm untuk E. coli. Pada uji antioksidan, ekstrak menunjukkan kemampuan yang signifikan dengan nilai IC50 sebesar 86,15 ppm (metode DPPH) dan 77,13 ppm (metode FRAP), yang termasuk dalam kategori kuat. Secara keseluruhan, hasil ini tidak hanya mengonfirmasi potensi jamur susu harimau sebagai sumber senyawa bioaktif alami, tetapi juga memberikan dasar ilmiah yang kuat untuk pengembangannya lebih lanjut sebagai agen terapeutik baru dalam mengatasi infeksi bakteri dan mitigasi stres oksidatif.

Abstract

Antioxidants are compounds that are able to ward off or reduce free radicals in the body so that they can prevent several diseases. Soursop leaves (Annona muricata L.) are used as a traditional herbal medicine to prevent or treat cancer, as well as a natural antioxidant, which is usually used by people by boiling it. The purpose of this study was to look at the phytochemical screening by comparing fresh boiled water, fresh juice, and a fresh extract of soursop leaves, as well as test the antioxidant activity with the addition of DPPH (1,1-diphenyl-2-picrylhydrazyl) and measured using a UV-Vis spectrophotometer at 516 nm. The results of the soursop leaf phytochemical screening showed that there were groups of chemical compounds such as flavonoids, glycosides, tannins, and saponins. The results of testing the antioxidant activity by looking at the maximum wavelength of DPPH, using a UV-Vis spectrophotometer, obtained IC50 values for fresh juice of 770.067 µg/mL, fresh decoction of 1,022.8422 µg/mL, and ethanol extract of 299.733 µg/mL. Based on the results of the study, it was shown that the ethanol extract had stronger antioxidant activity than a fresh decoction and the fresh juice of soursop leaves.

Abstract

Tannins are secondary metabolites belonging to the polyphenol group that exhibit important biological activities, including antioxidant, antibacterial, and anti-inflammatory effects. Mangkokan leaves (Polyscias scutellaria) and guava leaves (Psidium guajava) have long been traditionally used; however, quantitative data regarding their tannin content, particularly in relation to different extraction methods, remain limited. This study aimed to determine total tannin content and identify tannin types in 70% ethanol extracts and infusions of both plants using UV-Vis spectrophotometry. The methodology included phytochemical screening, identification of hydrolyzable and condensed tannins using specific reagents, and quantitative determination of total tannin content using the Folin–Ciocalteu method with gallic acid as a standard. Measurements were carried out at a maximum wavelength of 769 nm, with a regression equation of y = 0.2186x + 0.0024 and a coefficient of determination (R²) of 0.994. The results showed that all samples contained both hydrolyzable and condensed tannins. The highest tannin content was found in the ethanol extract of guava leaves (17.7998 ± 0.0789 mg/g), while the lowest was observed in the infusion of mangkokan leaves (14.0788 ± 0.1986 mg/g). These findings indicate that the type of solvent and extraction method significantly affect tannin yield, with 70% ethanol being more effective than water. In conclusion, both mangkokan and guava leaves have potential as natural sources of tannins, with ethanol extraction providing more optimal results. UV-Vis spectrophotometry proved to be an accurate, sensitive, and reliable method for the quantitative analysis of tannins in plant materials.

Abstract

Sidaguri (Sida rhombifolia L.) is a plant that contains alkaloids, flavonoids, saponins, tannins, and triterpenoids. Sidaguri plants contain antioxidants. Antioxidants are chemical compounds that can donate electrons to reactive free radicals so that they are not reactive and stable so that they can prevent the oxidation process in the body. This study aims to determine the class of chemical compounds contained in simplicia and sidaguri leaf extract and the antioxidant activity of sidaguri leaf extract. The research stages include the collection and processing of simplicia. Sidaguri leaf extract (Sida rhombifolia L.) was obtained by maceration method using 70% ethanol as solvent. Examination of the characteristics of simplicia powder. The extract whose solvent has been evaporated to become a thick extract is then carried out for phytochemical screening. The ability of the extract as an antioxidant was determined by reducing the DPPH radical (1,1-Diphenyl-2-picrylhydrazyl) with various extract concentrations 1500 μg/ml, 2000 μg/ml, 2500 μg/ml, 3000 μg/ml, 3500 μg/ml, 4000 μg/ml. The results of the extraction of sidaguri leaves obtained were 34.82 g of thick extract. The results of the phytochemical test of sidaguri leaves contain secondary metabolites, namely alkaloids, steroids, flavonoids, tannins, and saponins. The final result of the DPPH test is that the radicals change color from purple to yellow and their absorption is measured by UV-Vis spectrophotometry at a wavelength of 517 nm. The results of the antioxidant activity test carried out resulted in an IC₅₀ value of 2351.53 μg/ml which indicated that the antioxidant activity was very weak.

Abstract

Tomato (Solanum lycopersicum L.) is one of the most widely consumed fruits and vegetables due to its nutritional value and health benefits. This study aimed to determine the vitamin C, glucose, and crude fiber contents in different tomato varieties, namely red, yellow, green, and cherry tomatoes. The analysis included phytochemical screening, qualitative tests, and quantitative determination. Phytochemical screening was performed to identify the presence of secondary metabolites, while qualitative tests were conducted to confirm the presence of vitamin C and glucose. Quantitative analysis of vitamin C and glucose was carried out using the UV–Vis spectrophotometric method, whereas crude fiber content was determined using the gravimetric method. The results showed that flavonoids, alkaloids, and saponins were present in all tomato samples, while glycosides and tannins were detected only in certain varieties, and steroids/triterpenoids were not detected. Quantitative analysis indicated that the highest vitamin C content was found in red tomatoes (21.62 ± 0.178 µg/100 g), followed by cherry tomatoes (18.78 ± 0.002 µg/100 g), yellow tomatoes (18.38 ± 1.038 µg/100 g), and green tomatoes (12.20 ± 2.287 µg/100 g). The glucose content was 39.2 ± 0 µg/100 g in red tomatoes and 34.53 ± 0.597 µg/100 g in green tomatoes. Meanwhile, the crude fiber analysis showed the highest content in yellow tomatoes (53.44%), followed by cherry tomatoes (48.10%), red tomatoes (42.05%), and green tomatoes (40.37%). These findings indicate that different tomato varieties possess distinct nutritional profiles, and tomatoes can be considered a valuable dietary source of vitamin C, natural sugars, and dietary fiber that may contribute to maintaining human health.

Abstract

Background: Kunir asem herbal medicine is a traditional beverage widely consumed by the community to relieve menstrual pain due to the content of turmeric (Curcuma domestica Val.) which contains active compounds with analgesic, antipyretic, and anti-inflammatory properties. However, information regarding curcuminoid levels in powdered herbal medicine products circulating in the community remains limited, particularly in Medan City. Objective: This study aimed to identify the presence of curcuminoids in powdered herbal medicine containing turmeric rhizome circulating in Medan City and to determine the compliance of their levels with quality requirements according to the Indonesian Herbal Pharmacopoeia Second Edition 2017. Methods: A total of 10 samples of powdered herbal medicine containing turmeric rhizome were collected from five sub-districts in Medan City (Medan Johor, Patumbak, Deli Tua, Medan Baru, and Medan Polonia) using simple random sampling technique. Sample extraction was carried out by maceration using 96% ethanol solvent. Qualitative analysis of curcuminoids was performed using Thin Layer Chromatography (TLC) method with chloroform:methanol (95:5) as mobile phase and curcumin standard as comparison. Determination of curcuminoid levels was conducted using UV-Vis Spectrophotometry method at a maximum wavelength of 425 nm. Results: Qualitative analysis results showed that all samples were positive for curcuminoids, indicated by Rf values close to or equal to the Rf value of curcumin standard (0.78), ranging from 0.76-0.82. The quantitative analysis results showed that among the ten samples tested, three samples (SJ-A 0.66%, SJ-B 0.29%, and SJ-I 0.28%) had curcuminoid levels below the minimum requirement, while seven other samples (SJ-C 5.73%, SJ-D 7.50%, SJ-E 9.13%, SJ-F 5.94%, SJ-G 4.66%, SJ-H 8.02%, and SJ-J 6.07%) met the requirements. Conclusion: All samples of powdered herbal medicine containing turmeric rhizome circulating in Medan City were positive for curcuminoids; however, three out of ten samples (30%) did not meet the quality requirements for curcuminoid levels according to the Indonesian Herbal Pharmacopoeia Second Edition 2017, which specifies not less than 3.82%. This indicates the need for more stringent quality control of herbal medicine products circulating in the community.

Abstract

Background: Chitosan is a biopolymer derived from the deacetylation of chitin, which is generally obtained from crustacean waste. However, the utilization of more environmentally friendly alternative sources such as oyster mushrooms (Pleurotus ostreatus) has recently been developed. Chitosan exhibits biocompatible, biodegradable properties and antibacterial activity, making it potentially applicable in the food and pharmaceutical industries. Objective: This study aims to extract and characterize chitosan from oyster mushrooms and to evaluate its antibacterial activity against Escherichia coli. Methods: The study was conducted through extraction stages including deproteinization, depigmentation, demineralization, and deacetylation. Chitosan characterization included functional group analysis using FTIR, as well as physicochemical parameter testing such as yield, moisture content, ash content, and solubility. Antibacterial activity was tested using the disc diffusion method at various chitosan concentrations (0.3%, 0.5%, 0.7%, and 0.9%). Results: The obtained chitosan had a degree of deacetylation of 80.64%, indicating successful conversion of chitin into chitosan. The physicochemical characteristics met quality standards, with a yield of 52.84%, moisture content of 0.666%, and ash content of 0.416%. Chitosan was soluble in 2% acetic acid. Antibacterial testing demonstrated inhibitory activity against Escherichia coli, with inhibition zones ranging from 10.1 to 14.2 mm, categorized as strong. Conclusion: Chitosan extracted from oyster mushrooms exhibits good physicochemical characteristics and promising antibacterial activity, indicating its potential as an environmentally friendly alternative material for applications in the food and pharmaceutical fields.
