Abstract

This research was held from December 2020 – February 2021 at the Laboratory of Fish Parasites and Diseases, Faculty of Fisheries and Marine Science, Universitas Riau. The purpose of this study was to analyze and obtain the best dose of cherry leaf solution to prevent Motile Aeromonas Septicemia from total erythrocytes, hematocrit and hemoglobin levels of striped catfish (Pangasianodon hypophthalmus) fish. The method used to experiment with Completely Randomized Design (CRD) is one factor of five treatment levels. The doses used were negative control without soaking the cherry leaf solution and not tested against Aeromonas hydrophila, positive control without soaking the cherry leaf solution and tested against A. hydrophila, soaking the cherry leaf solution with a dose of 2800 ppm (P1), 3,200 ppm (P2), and 3,600 ppm (P3). Immersion was performed five times for 5 minutes with an interval of 7 days. The results showed that soaking with cherry leaf solution at a dose of 3,600 ppm (P3) was the best dose (P<0.05) with a total value of erythrocytes after the challenge test of 209,66 x 104 cells/mm3, hematocrit level of 37%, hemoglobin 11.86 g/dL, and the survival rate of the test fish was 86,67%.

Abstract

Herb-fermented medicine consists of Curcuma zanthorrhiza, Curcuma domestica Val. and Kaempferia galanga, which contains active substances to overcome disease. This research was conducted from August to December 2021 at the Marine Science Biotechnology Laboratory and Fish Parasite and Disease Laboratory, Faculty of Fisheries and Marine, Universitas Riau. The method used in this study was an experiment using a Complete Randomized Design (CRD), one factor with five treatments and three replications. The treatment of this study was Kn: Negative control (feeding without herbs fermented and without being challenged Aeromonas hydrophila), Kp: Positive Control (feeding without herbs fermented and challenged with A. hydrophila), P1; P2 and P3: Fed herbs fermented at a dose of 150, 175 and 200 mL/kg and challenged with A. hydrophila. The fish used was 6-7 BW with a 1 fish/4L density. The result showed the best dose was in P3, with a total of leukocytes 5,01x104 cells/mm3, Lymphocytes 77,67%, Neutrophils 7,67%, Monocytes 14,67%, Phagocytic activity 37,67%. Based on the data obtained, it can be concluded that fermented herbs have a significant effect on the leukocyte differentiation of Oreochromis niloticus

Abstract

Natural ingredients used in the manufacture of fermented herbs medicine, such as turmeric, ginger, and kencur. These plants are antibacterial and antimicrobial and increase the growth of fish. This research was conducted from November 2021 to March 2022 at the Faculty of Fisheries and Marine, Riau University. The purpose of this study was to analyze the histopathology of the intestine and liver of Oreochromis niloticus fed with herbs fermented pellet and infected with Aeromonas hydrophila. The method used is an experiment with a completely randomized design (CRD) one factor, five levels of treatment, and three replications so that 15 experimental units were obtained. The treatments used were Kn (feeding without fermented herbs and without infection with A. hydrophila bacteria), Kp (feeding without fermented herbs and infected with A. hydrophila bacteria) and feeding with the addition of fermented herbs and infected with bacteria A. hydrophila at a dose of 150 mL/kg (P1), 175 mL/kg (P2), and 200 mL/kg (P3). The results showed that the damage that occurs in the intestine was edema, haemorrhage and necrosis. Liver damage in the form of hypertrophy, bleeding, vacuole degeneration and necrosis. The best dose was found which is 200 mL/kg of feed (P3). The survival rate was 85,00%. It can be concluded that the addition of herbs fermented to feed can improve the structure of O.niloticus

Abstract

Biofloc technology is one of the most profitable alternative solutions to the problem of aquaculture waste. The research was carried out from July to August 2020 at the Laboratory of Fish Parasites and Diseases, Faculty of Fisheries and Marine Affairs, Riau University. The purpose of this study was to isolate and identify candidate lactic acid bacteria (LAB) that have the potential as probiotics from the intestines Monopterus albus and to determine the growth rate of candidate probiotic bacteria and can suppress the growth of the pathogenic bacteria Aeromonas hydrophila. The method used is the experimental method with treatment of pH 2, pH 4 and pH 6, isolation was carried out from the best Monopterus albus rearing with a stocking density of 30 fish/25 liters of water. Candidate probiotic bacteria was found from the intestines of Monopterus albus as many as 6 LAB candidate isolates based on the results of physical, biochemical and pH treatments, the morphology of the probiotic candidate isolates consisted of one type of bacteria, namely Lactobacillus, from the 6 isolates which showed the widest zone of inhibition against the pathogenic bacteria Aeromonas hydropila was isolate derived from Monopterus albus intestine result shown that which grows at pH 2 is BL 2.1.1 with an inhibition zone diameter of 9,05 mm.

Abstract

Biofloc is a technology using good bacteria to convert organic waste to be food source by fish in the form of floc. This research was carried out in April-May 2021 in the Hatchery and blood observations at the Fish Disease and Parasite Laboratory, Faculty of Fisheries and Marine, University of Riau. The purpose of this study was to determine the health status of eels from the profile of erythrocytes cells and the best stocking density of eels cultivated in biofloc system. While the benefits of this study can be applied to biofloc cultivation and apply it in the field. The research method used one-factor completely randomized design (CRD) with four treatment levels and three replications (12 experimental units) for 40 days . The optimal stocking density of eels in clean water is 6 fish for 10 liters. The treatments in this study were P1: stocking density of 10 fish/25 L, P2: stocking density of 20 fish/25 L, P3: stocking density of 30 fish/25 L, P4: stocking density of 40 fish/25 L. The results showed that the erythrocytes profile of eel with different stocking densities had a significant effect (P<0.05) and the best treatment P3 (30 fish/25 L) with total erythrocytes 2.88x106 cells/mm3, hematocrit level 31.66 %, hemoglobin levels 17.40 g/dL, blood glucose 46.33 g/dL, specific growth rate 93.3%, and feed efficiency 67.46%. Water quality in this study was in normal range, that is temperature 25.7-27.3 oC, pH 7.0-7.8, DO 6.4-8.0 mg/L, and NH3 0.0003-0.00038 mg/L

Abstract

The research was conducted from January to October 2021 at the Laboratory of Fish Parasites and Diseases, Departement of Aquaculture, Faculty of Fisheries and Marine, University of Riau. The purpose of this research was to analyze and obtain the best dose of bay leaf solution to treat catfish (Clarias gariepinus) infected with Aeromonas hydrophilla. Bay leaf is a plant that has antibacterial properties in fish because it contains secondary metabolites, namely tannins, saponins, steroids, alkaloids, flavonoids, and essential oils. The method used is an experimental method with a Completely Randomized Design (CRD), one factor with five treatments. The treatment applied is Kn (without immersed with bay leaf solution and not infected by A. hydrophila), Kp (without immersed with bay leaf solution and infected by A. hydrophila), P1 (1000 ppm), P2 (1100 ppm), P3 (1200 ppm). The curing is done by immersed with bay leaf solution for 5 minutes, carried out three times with an interval of 24 hours and then maintained for 14 days after treatment. The result shown that bay leaf solution able to cure catfish infected by A. hydrophila. The best dose was 1200 ppm with average total leukocytes of 12.61 × 104 cells /mm3, leukocyte differentiation (lymphocytes 84.66%, Neutrophils 6.66%, Monocytes 8.66%), phagocytitosis activity is 35.33%, and the survival rate 96.67%.

Abstract

This research was carried out in July 2021 – September 2021 at the Laboratory of Parasites and Fish Diseases. This study aims to obtain the best dose of pineapple peel solution. This study used a Complete Randomized Design (RAL) consisting of 5 treatments and 3 tests. The treatment in this study was that there was an influence where the best treatment was found in P2 (Soaking of pineapple skin solution dose 1,800 ppm tested challenged with A. hydrophila bacteria) with an average total leukocytes of 9.92 x 104 cells / mm3, total lymphocytes 82.67%, total monocytes 9.33%, neutrophy value 8.00% phagocytic value 37.00%, siamese jambal fish dilution classified as good with a value of 93.33%, and water quality during the study temperatures ranged from 27.3–29oC,, pH 6.3–6.6, DO 3.4–4.2 mg/L, and NH3 0.01-0.02 mg/L.

Abstract

This study aims to determine the best dose of propolis to treat Carassius auratus infected with Aeromonas hydrophila seen leucocyte differentiation from C.auratus. The method used was experimental method with Completely Randomized Design (CDR) with one factor consisting of five treatment levels and three replications. The treatment was Kn (not infected with A. hydrophila and untreated propolis), Kp (infected with A. hydrophila but not treated with propolis), and fish infected with A. hydrophila treated propolis with dose P1 (700 ppm), P2 (800 ppm), and P3 (900 ppm). Treatment is done by injection of propolis in fish infected by A. hydrophila in intramuscular. The fish used are 8-10 cm in size and kept for 14 days post-infection. The results showed that propolis solution has been able to treat C. auratus from infected A. hydrophila. The dose of propolis 800 ppm is the best dose to treat C. auratus infected A.hydrophila, seen from leukocyte differentiation (lymphocyte 80,25%, monocytes 10,09% and neutrophils 9,66%).

Abstract

Red ginger is a natural ingredient that contains several antibacterial compounds such as essential oils, flavonoids, terpenoids, phenols, and zingiberene. This research was conducted from June to August 2021. The purpose of this study was to analyze the structure of the gill and liver tissue of jambal siam fish and to obtain the best dose of adding fermented red ginger to the feed before and after being challenged with Aeromonas hydrophila bacteria. The method used is an experimental method using a one-factor Completely Randomized Design (CRD) with five treatment levels and three replications, namely: Feeding without fermentation of red ginger and without being tested against A. hydrophila (Kn), Feeding without fermentation of red ginger and tested against A. hydrophila (Kp), Feeding with a dose of 150 mL/kg (P1), dose 175 mL/kg feed (P2), dose 200 mL/kg (P3). The results showed that the structure of the gill tissue of Jambal Siam fish showed damage such as hypertrophy, hyperplasia and bent lamellae. The best results were found in the P2 treatment, a dose of 175 mL/kg feed and tested against A.hydrophilla bacteria

Abstract

Kayu manis merupakan salah satu bahan alami yang berpotensi untuk menghambat pertumbuhan bakteri. Penelitian ini dilaksanakan pada bulan Maret s/d Mei 2023 di Laboratorium Parasit dan Penyakit Ikan, Fakultas Perikanan dan Kelautan, Universitas Riau. Penelitian ini bertujuan untuk mengetahui struktur insang dan ginjal ikan jambal siam yang diberi pakan pelet yang diperkaya larutan kayu manis dan diinfeksi dengan A. hydrophila. Metode yang digunakan adalah eksperimen dengan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 3 kali ulangan. Perlakuan yang diberikan adalah Kn (tanpa perlakuan), Kp (tanpa kayu manis dan infeksi A. hydrophila) dan pada P1, P2 dan P3 adalah 15, 30, 45 mL/kg pakan. Untuk melihat efektifitas kayu manis dalam meningkatkan imunitas ikan yang terserang A.hydrophila maka ikan diujitantang pada hari ke-32 dengan A. hydrophila secara intramuskular. Organ insang disampling pada hari ke-14 pascainfeksi dan pembuatan preparat histopatologi. Hasil penelitian menunjukkan bahwa terdapat kelainan pada struktur insang jambal siam (Kp) seperti lamella bengkok, hyperlasia, hyperthrophy dan nekrosis. Kesimpulan dari penelitian ini yaitu dosis kayu manis 30 mL/kg pakan dapat memperbaiki struktur insang jambal siam yang diinfeksi A. hydrophila.
