Abstract

The aim of this research was to investigate the therapeutic effects of Kencur (Kaempferia galanga L.) extract on kidney histopathological damage in male mice exposed to cigarette smoke. Twenty 8-week-old male mice were randomly divided into five treatment groups (n=5). The negative control group (C-) consisted of mice not given kencur extract or exposed to cigarette smoke, while the positive control group (C+) comprised mice exposed to cigarette smoke without kencur extract. Treatment groups included Treatment 1 (T1) administered 150 mg/kg BW of kencur extract, Treatment 2 (T2) administered 300 mg/kg BW, and Treatment 3 (T3) administered 600 mg/kg BW. Mice were exposed to cigarette smoke for 14 days. The data were analyzed using the Kruskal-Wallis test to evaluate overall differences among the treatment groups. Where significant differences were found (p<0.05), further pairwise comparisons were conducted using the Mann-Whitney test, confirming statistically significant differences between specific treatment groups.  Treatment 3 (T3) was found to be the most effective in reducing histopathological damage in the kidneys of male mice. This study demonstrates that kencur extract effectively reduces histopathological damage in the kidneys of mice exposed to cigarette smoke, highlighting its potential as a protective agent against smoke-induced tissue injury.

Abstract

This study aims to determine the best dose of basil leaf extract to add to AndroMed® semen extender to improve spermatozoa quality after freezing and thawing. This study used healthy, normal four-year-old Simmental bulls with good libido. The sample used was fresh semen from Simmental bull collected using an artificial vagina. This study used a completely randomized design (CRD) with three treatments and six replicates. Ejaculate that met the motility qualification of more than 70%, was divided into three groups, each extended in AndroMed® without the addition of basil leaf extract (T0), with the addition of 0.1% basil leaf extract (T1) and with the addition of 0.3% basil leaf extract (T2). Straw containing semen with 30 million spermatozoa/straw was processed using an IceCube automatic freezer (Minitube, Germany). The results showed that spermatozoa motility, viability, and spermatozoa plasma membrane integrity in the T2 group were higher (p <0.05) than those in the T0 and T1 groups, while spermatozoa morphological abnormalities in the T2 group were lower (p <0.05) than those in the T0 and T1 groups. There were no significant differences (p >0.05) in these parameters between groups T0 and T1. It could be concluded that the addition of 0.3% basil leaf extract to the AndroMed® extender could improve the quality of the semen of Simmental bulls after thawing.

Abstract

This study aims to evaluate the efficacy of Papaya var California leaves extract and identify its effective concentration in inducing mortality among third-instar larvae of Aedes aegypti mosquitoes. The study utilized adult Ae. aegypti mosquitoes as the experimental subjects. This study was an experimental study with a comparative Completely Randomized Design The test groups include negative control, 2.5 mg/mL, 5 mg/mL, 10 mg/mL and positive control. Each test used 20 third instar larvae of  Ae.aegypti mosquitoes, with four repeats..  The test groups were 2.5 mg/mL, 5 mg/mL, 10 mg/mL, and the control groups were A negative control contained distilled water and Tween 20, and a positive control contained abate (temephos) Based on Kruskall-wallis test shows a significant difference. The results indicated that the optimal concentration was 10 mg/mL, with the highest effectiveness observed at the 30th time hours.

Abstract

Sex identification of Sun Conures (Aratinga solstitialis) is crucial for breeding and preservation, as well as increasing sun conure populations. These birds are sexually monomorphic. Therefore, Determination between male and female carried out by their morphology examination. The Polymerase Chain Reaction (PCR) method, utilizing molecular-based technology, was employed to determine the sex of Aratinga solstitialis in this study. The P2 and P8 primers were utilized in this method, which has been deemed suitable and accurate for sex identification through calamus samples. The research focused on two 28-month-old Aratinga solstitialis birds. Calamus samples were collected and subjected to PCR amplification using the extracted calamus. The resulting PCR products were then visualized using electrophoresis with a 1% agarose gel. In the electrophoresis photo, the presence of two bands indicated a female specimen, whereas a single band indicated a male specimen. The result of the gel electrophoresis research showed that both of the Aratinga solstitialis were male with one band of each bird on ranged from 300-400 base pairs. The result show that the Polymerase Chain Reaction method in terms for sex identification on monomorphic birds, especially Aratinga solstitialis birds is very effective to differentiate the sex of young birds and the adults.

Abstract

Graphical Abstract Highlight Research The study aims to develop a multi-epitope vaccine (MEV) against A. hydrophila by targeting the aerolysin toxin, a key virulence factor responsible for infections in fish and humans. Computational methods identified and optimized B-cell and T-cell epitopes, focusing on their ability to trigger immune responses without causing toxicity or allergenicity. In silico simulations demonstrated that the MEV has a strong binding affinity to immune receptors like TLR-4, MHC-I, and MHC-II, indicating its potential to induce robust cellular and humoral immunity. Structural analysis of the MEV showed a stable 3D conformation, with most residues in favorable regions, ensuring stability during immune activation. The MEV could enhance disease control in aquaculture and reduce human infection risks, offering a promising solution to address antibiotic resistance and the absence of effective vaccines. Abstract Aeromonas hydrophila, gram-negative, is a major pathogen responsible for various diseases in mammals, reptiles, amphibia, and vertebrates, including fish and humans. Targeting the specific toxin aerolysin in A. hydrophila is crucial to address antibiotic resistance and the lack of adequate and protective vaccines against this intracellular pathogen. This study aimed to identify a multi-epitope vaccination (MEV) candidate targeting A. hydrophila aerolysin toxin to combat the disease effectively. Standard biochemical characterization methods and sequencing of the 16S rRNA, rpoB, and aerA genes identified the isolate AHSA1 as A. hydrophila. Subsequently, we identified B and T cell epitopes on the aerolysin protein and separately predicted MHC-I and MHC-II epitopes. The epitopes are then evaluated for toxicity, antigenicity, allergenicity, and solubility. The vaccine design integrated multi-epitope-based constructs, utilizing specialized linkers (GPGPG) and EAAAK linkers to connect epitope peptides with adjuvants in the cholera toxin B component, thereby enhancing immunogenicity. Ramachandran plots showed that 85.25% of the residues were located in the most favorable regions, which was followed by the generously allowed zone (1.30%), the additional allowed regions (10.80%), and the forbidden regions (2.65%), thus confirming the feasibility of the modeled vaccine design. Based on docking simulations, MEV had the highest binding and interaction energies with TLR-4, TLR-9, MHC-I, and MHC-II (-1081.4, -723.2, 866.2, -9043.3 kcal/mol). Based on computational modelling, we expect the Aerolysin MEV candidate design to activate diverse immune mechanisms, stimulate robust responses against A. hydrophila, and maintain safety. The significant solubility, absence of toxicity or allergic response, and minimal side effects in animal testing all contribute to the potential clinical utility of this vaccine candidate.

Abstract

This study aims to determine the effect of red dragon (Hylocereus polyrhizus) fruit peel extract (RDFPE) on spermatozoa motility and viability of hypercholesterolemic rats (Rattus norvegicus) as a model. Twenty male rats were randomly divided into negative control (NC), positive control (PC), treatment 1 (T1), treatment 2 (T2), and treatment 3 (T3) groups. All rats were given 2 mL of high cholesterol feed orally every day for 28 days. On day-15, all rats were measured for their blood cholesterol levels, followed by treatment for 14 days. Rats in the NC, PC, T1, T2, and T3 groups were treated with 1% Na-CMC, Simvastatin 10 mg/kg BW, and RDFPE of 500, 750, and 1000 mg/kg BW, respectively. On day-29, all rats were sacrificed to evaluate spermatozoa viability and motility. The results showed that spermatozoa viability and motility in the hypercholesterolemic rats (NC) group were the lowest (p <0.05) among the groups. Treatment of hypercholesterolemic rats with Simvastatin 10 mg/kg BW (PC) group showed higher (p <0.05) spermatozoa viability and motility compared to the NC group. RDFPE dose of 1000 mg/kg BW (T3 group) resulted in higher (p <0.05) spermatozoa viability and motility compared to other RDFPE doses (T1 and T2) and the control (NC) groups, and it was similar (p >0.05) compared with the Simvastatin treated (PC) group. It could be concluded that the administration of 1000 mg/kg BW ethanolic extract of red dragon fruit (Hylocereus polyrhizus) peel increased the viability and motility of spermatozoa of hypercholesterolemic rats (Rattus norvegicus) which were the same as the group of rats given Simvastatin 10 mg/kg BW.

Abstract

Second-degree burns are complex traumatic conditions requiring optimal tissue healing interventions. This experimental study investigated the impact of Gnetum gnemon L. leaf extract cream on collagen density in burn wounds. The research was conducted using 20 male Wistar rats (Rattus norvegicus), divided into five groups: negative control, positive control (silver sulfadiazine), and three treatment groups with varying leaf extract concentrations (2.5%, 5%, and 10%). Burn induction was performed on the right gluteal area using a thermostat at 85°C for 5 seconds. Collagen density was evaluated through histopathological analysis using ImageJ with density scores ranging from 0 to 4. Research results demonstrated significant collagen density increases in treatment groups, with the 10% concentration displaying the highest density (score +4). Significant differences between groups were confirmed by statistical analysis (p<0.05). Findings indicate the potential of Gnetum gnemon L. extract as an alternative therapy for improving burn wound healing through collagen density optimization.

Abstract

Graphical Abstract  Highlight Research Adjuvanted polyvalent FKC elicited the strongest and most sustained multi-arm immune response in Osphronemus goramy compared with monovalent and non-adjuvanted vaccines. The lead formulation combined high agglutinating antibody titres with enhanced NBT respiratory burst, indicating synergistic humoral–innate activation against Aeromonas hydrophila. Polyvalent vaccines did not dilute immunogenicity; instead, strain combination plus adjuvant broadened and amplified immune responsiveness. Longitudinal profiling of il-1β and ifn-γ revealed a stable pro-inflammatory/Th1-like cytokine signature uniquely associated with the adjuvanted polyvalent FKC. The integrated immunological “fingerprint” supports the adjuvanted polyvalent FKC as a rational lead candidate for motile Aeromonas septicaemia control in warm-water gourami aquaculture.   Abstract Motile Aeromonas septicaemia (MAS), predominantly associated with Aeromonas hydrophila, remains a major constraint in giant gourami (Osphronemus goramy) aquaculture. This study evaluated formalin-inactivated A. hydrophila vaccines prepared from MAS-associated field isolates, comparing a monovalent formulation (P2), a non-adjuvanted polyvalent formulation (P3), and an oil-adjuvanted polyvalent formulation (P4) against PBS controls (P1). A total of 240 fish were used (60 per treatment) and assigned to two parallel cohorts (immunology and survival/challenge). Immune endpoints (agglutinating titres, NBT activity, and splenic il-1β and ifn-γ transcription) were assessed on days 7, 14, 21, 35, and 42 post-vaccination. The survival cohort was challenged intraperitoneally at day 21 with a homologous A. hydrophila strain and monitored for 14 days post-challenge. Vaccination was clinically well tolerated and improved survival relative to controls, with P4 showing the highest protection (RPS 81.8%). Agglutinating titres differed by treatment and time; at the peak sampling point (day 35), mean titres in P4 were ~200-fold higher than in P1, and model contrasts indicated significant differences versus controls (p<0.001). Splenic il-1β and ifn-γ transcript levels were higher in vaccinated groups than in controls at later time points. These findings support further evaluation of an oil-adjuvanted polyvalent inactivated A. hydrophila vaccine for gourami, including dose optimisation, extended safety assessment, heterologous challenge, and field validation.
