Abstract

South Bogor District is an area with agroecosystem conditions that still support the existence of endemic and local plant species utilized by local communities. However, scientific information regarding species diversity, utilization patterns, and sustainability potential remains limited. This study aimed to explore the diversity of endemic plants, their utilization forms, plant parts used, as well as community perceptions and threats to their existence in South Bogor District. The research employed a descriptive survey method with an ethnobotanical approach through field observations and semi-structured interviews with local residents. The results revealed various endemic and local plant species utilized by the community, predominantly for medicinal purposes, food, and spices. The most frequently used plant parts were fruits, leaves, and rhizomes, with processing methods that remain simple and based on traditional knowledge. Community perception toward the importance of endemic plants was highly positive; however, high utilization intensity of certain species was not always aligned with their availability in nature. Major threats to the sustainability of endemic plants include land-use change, logging, inadequate plant maintenance, and limited replanting efforts. This study highlights that endemic plants in South Bogor District play essential ecological, social, and cultural roles, yet require community-based management and conservation strategies to ensure their sustainable utilization.

Abstract

Scientific literacy is an essential 21st-century skill because it enables students to apply scientific knowledge in real-life contexts. However, in practice, the index of scientific literacy remains very low, as reflected in PISA data as well as observations in the field. This study aims to analyze the influence of students’ perceptions of learning media and self-efficacy on the scientific literacy skills of junior high school students in Batam, Indonesia. A total of 100 eighth-grade students from two public schools participated in this quantitative study. Data were collected using a validated Likert-scale questionnaire and a scientific literacy test, and were then analyzed using Pearson correlation and multiple linear regression. The results showed that perceptions of learning media were significantly correlated with scientific literacy (r = 0.372, p < 0.001), as was self-efficacy (r = 0.237, p = 0.018). However, only perceptions of learning media significantly predicted scientific literacy (β = 0.330, p = 0.001), whereas self-efficacy did not (β = 0.115, p = 0.258). The model explained 15% of the variance (R² = 0.150, F(2,97) = 8.541, p < 0.001). These findings highlight the importance of positive perceptions of learning media, particularly in terms of usefulness, interactivity, and emotional engagement. The implication is that teachers and curriculum developers need to design effective and engaging media to foster scientific literacy at the secondary school level.

Abstract

Mathematical problem-solving ability is an essential competency that supports students' Mathematical problem-solving ability is a fundamental competency in mathematics education; however, many junior high school students still experience difficulties in solving non-routine problems. This study aimed to examine the effect of the Realistic Mathematics Education (RME) approach on ninth-grade students' mathematical problem-solving ability. A quasi-experimental study employing a nonequivalent posttest-only control group design was conducted at SMPN 10 Padang during the 2025/2026 academic year. Two intact classes were selected through cluster random sampling, comprising 32 students in the experimental group and 32 students in the control group. The experimental group received instruction using the RME approach, whereas the control group received conventional instruction. Students' mathematical problem-solving ability was measured using a five-item essay test based on Polya's four problem-solving indicators. Data were analyzed using descriptive statistics, an independent-samples t-test, and Cohen's d effect size after satisfying the assumptions of normality and homogeneity. The results showed that the experimental group achieved a higher mean score than the control group (80.75 vs. 71.19). The difference was statistically significant (t(62) = 3.46, p < .001), with a large effect size (d = 0.87). These findings indicate that the RME approach effectively improves students' mathematical problem-solving ability and supports its implementation in secondary mathematics classrooms.

Abstract

Abstract. Pratiwi RH, Sunarti RN, Nurkanto A, Dasmo, Isfaeni H. 2026. Isolation, lytic activity, and genomic characterization of hospital wastewater phages infecting Mycobacterium smegmatis, a surrogate for tuberculosis bacteria. Biodiversitas 27 (6): d270613. https://doi.org/10.13057/biodiv/d270613. In Indonesia, Tuberculosis (TB) remains a major public health problem, with increasing case numbers and high mortality rates. The increasing prevalence of drug-resistant tuberculosis has encouraged exploration of bacteriophages as alternative antibacterial agents. This study aimed to isolate and characterize lytic bacteriophages from hospital wastewater capable of infecting Mycobacterium smegmatis, a surrogate host commonly used in tuberculosis-related research. Wastewater samples were collected from several hospital sewage outlets in Palembang and subjected to enrichment and plaque assay screening using M. smegmatis as the host bacterium. Three lytic bacteriophage isolates, designated HS, BR, and CH, were successfully obtained and characterized based on plaque morphology, phage titers, and lytic activity. All exhibited clear, small plaques (≈1.0 mm) characteristic of lytic morphology, with titers ranging from 2.3×10⁵ to 2.9×10⁵ PFU/mL. The lytic activity assay revealed that all phages significantly reduced bacterial growth within 10 hours. The CH phage achieved the highest inhibitory effect, reducing bacterial optical density (OD600) by 21%, compared to 14% reductions by HS and BR phages. Notably, CH phage-initiated lysis as early as the first hour, indicating a short latency period and strong infectivity. These results demonstrate that hospital wastewater is a promising source of mycobacteriophages with potential therapeutic value against mycobacterial infections. Among the isolates obtained, CH showed the most promising lytic performance and genomic characteristics, suggesting its potential for further investigation in bacteriophage-based strategies against mycobacterial infections. The WGS analysis demonstrates that the isolated bacteriophage represents a TehO-like phage lineage with high genomic similarity to several Pseudomonas bacteriophages. Genome analysis of the isolated bacteriophage revealed a modular genome organization characteristic of dsDNA bacteriophages, containing structural genes, DNA replication genes, and a lysis module encoding an endolysin.
