Abstract

Kappaphycus alvarezii merupakan  salah satu spesies makro alga yang paling banyak dibudidayakan di Indonesia. Penyakit utama yang menyerang budidaya rumput laut ini adalah penyakit ice-ice  yang  dapat menurunkan hasil panen hingga 70-80%.  Tujuan penelitian iniadalah  untuk mengetahui bakteri patogen yang berasosiasi dengan K. alvarezii bergejala penyakit ice-ice dan mengetahui agen penyebab penyakit ice-ice. Jenis penelitian yang dilakukan adalah penelitian deskriptif. Sampel K. alvarezii yang bergejala ice-ice diambil dari lokasi budidaya di Teluk Bumbang Dusun Gerupuk Lombok Tengah. Sampel tersebut kemudian dibawa ke Laboratorium Biologi untuk diisolasi bakteri yang berasosiasi dengan K. alvarezii bergejala ice-ice. Terhadap isolat yang diperoleh dilakukan karakterisasi parsial. Selanjutnya dilakukan uji patogenisitas (postulat Koch) untuk mengetahui apakah bakteri yang diperoleh merupakan agen penyebab penyakit ice-ice. Hasil isolasi menunjukkan terdapat 28 isolat bakteri yang berasosiasi dengan K. alvarezii bergejala ice-ice. Selanjutnya, uji postulat Koch memperlihatkan hanya satu isolat yang mampu menyebabkan gejala ice-ice yaitu isolat K25Kata Kunci : Kappaphycus alvarezii, penyakit ice-ice, postulat Koch

Abstract

The presence of microbial communities in the digestive tract affects the physiological functions of digestion. The purpose of this study is to determine the bacterial community in the digestive tract of lobsters, especially pearl lobsters. This research is descriptive explorative. Samples were taken from the lobster digestive tract which was divided into 3, namely the cardiac, pylorus and intestinal sections and each part was counted for the total bacteria using the TPC (Total Plate Count) method. Then isolated from the Sea Water Complex media and partially characterized. The results showed that the total number of bacterial cells in cardiac, piloric and intestinal cells were 41.91 x 108 cfu.ml-1, 18.26 x 108 cfu.ml-1 and 14.10 x 108 cfu.ml-1 respectively. The number of bacterial isolates obtained from the lobster pearl digestive tract were 51 isolates with details of 18 cardiac isolates, 16 isolates in piloric, and 17 isolates in the intestine. Phenotypic isolates obtained from lobster digestive tract bacteria consisted of monococcus 37.25%, diplococcus 27.45%, streptococcus 29.41%, staphylococcus 1.96%, and long baccil 3.92%, while gram positive 62.65 % and gram negative at 37.25%

Abstract

The use of antibiotics that are not according to the rules and antibiotics in the long term can cause resistance to bacteria. This study aims to determine the presence of antibacterial activity and the effect of increasing the concentration of ethanol extract of Ganoderma sp. against several test bacteria. Extract from Ganoderma sp. obtained by maceration method using ethanol 95% solvent. The extract concentrations used were 20%, 40%, 60% and 80%. This research was conducted using the wells method with ciprofloxacin as a positive control and 50% DMSO as a negative control. The parameter measured is the large diameter of the inhibition formed around the well. The results of the antibacterial activity test of ethanol extract Ganoderma sp. has greater inhibitory activity against gram-negative bacteria. The inhibitory activity of ethanolic extract of Ganoderma sp. on the growth of test bacteria increased with increasing concentration of the extract

Abstract

The use of antibiotics is one of the most important ways to deal with the spread and treatment of pathogenic microbial infections. The search for new antibiotic sources continues to be carried out to anticipate the emergence of microbial resistance. One of the natural resources that has the potential as an antimicrobial source is a member of the macrofungi of the Genus Ganoderma. The purpose of this study was to evaluate the antimicrobial performance of the ethanol extracts of Ganoderma  lucidum, G. applanatum dan Ganorderma sp. against fungi (Candida albicans dan Cryptococcus neoformans),  gram positive bacteria (Bacillus cereus dan Staphylococcus aureus) and gram negative bacteria (Escherichia coli dan Shigella sp.). Macrofungi samples were taken from the forest area of Nature Tourism Park (TWA) Gunung Tunak, TWA Kerandangan, TWA Suranadi, TWA Nuraksa Sesaot, TWA Lemor and Pusuk forest. The stages of the method performed are sample collection, sample preparation, extraction, and testing of antimicrobial activity using the well difusion method. The ethanol extract concentrations for testing were 20%, 40%, 60% and 80%. The results showed that the three Ganoderma species had anti-fungal and antibacterial activity and that different levels of concentration had an effect on inhibition. The size of the inhibition zone is directly proportional to the higher the extract concentration. The antimicrobial activity of the ethanol extract of G. lucidum was higher when compared to G. applanatum and Ganoderma sp. both against fungi (Candida albicans and Cryptococcus neoformans) as well as against gram-positive and gram-negative test bacteria. In addition, G. applanatum showed very weak inhibition against both groups of tested bacteria.  Key words: candidiasis; cryptococcosis; comorbid infections; macerations; pathogens

Abstract

Chicken egg hatching waste flour can be an alternative to replace expensive spiny lobster feed because apart from being waste it is also cheap and easy to obtain. The purpose of this research is to determine the effect of feeding chicken egg hatching waste on the water quality of spiny lobster (Panulirus homarus). This research used an experimental method with a Completely Randomized Design (CRD) consisting of 4 treatments with 3 replication. The results of the study showed that the composition of chicken egg waste can influence temperature, DO, pH, salinity, nitrate, nitrite, ammonia, absolute weight growth, absolute length growth, Specific Growth Rate, Feed Conversion Ratio and Survival Rate of spiny lobste.
