Abstract

Penelitian ekspresi sementara (transient expression) dari transgen secara in vivomenggunakan gen reporter berguna untuk mendesain konstruksi gen yang akan digunakan pada penelitian transgenesis. Gen reporter yang umum digunakan dalam penelitian ekspresi sementara transgen adalah gen GFP (green fluorescent protein). Pengamatan gen EGFP (enhanced green fluorescent protein) pada embrio dan larva ikan patin siam (Pangasianodon hypophthalmus) ditujukan untuk mendapatkan informasi mengenai kemampuan promoter -aktin ikan mas dalam mengendalikan ekspresi gen EGFP. Gen EGFP diintroduksikan ke dalam sperma ikan patin siam menggunakan metode elektroporasi. Sperma yang telah dielektroporasi digunakan untuk membuahi sel telur ikan patin siam. Pengamatan ekspresi gen EGFP dilakukan setiap enam jam dimulai dari embrio fase 2 sel sampai larva. Berdasarkan hasil penelitian, gen EGFP terekspresi pada fase embrio dan larva ikan patin siam. Puncak ekspresi gen EGFP terjadi pada fase neurula dan menurun pada fase larva. Berdasarkan penelitian ini maka ikan patin siam transgenik telah berhasil dibentuk dan promoter -aktin ikan mas terbukti aktif dalam mengarahkan ekspresi gen asing (GFP) di dalam tubuh ikan patin siam.

Abstract

Penelitian maturasi belut sawah (Monopterus albus) dengan Hormone human Chorionic Gonadotropin dan antidopamin dilakukan di Kolam Percobaan Babakan, Institut Pertanian Bogor. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh hormon hCG dan kombinasinya terhadap maturasi belut sawah. Penelitian ini dilakukan dengan metode ekperimental secara Rancangan Acak Lengkap (RAL) dengan perlakuan kontrol (NaCl), hCG (dosis 20 IU/kg), antidopamin (AD) (dosis 0,01 mg/kg), dan hCG+AD (dosis 20 IU+0,01 mg/kg) dengan tujuh ulangan individu. Data dianalisis secara deskriptif dan ANOVA. Parameter uji pada penelitian ini di antaranya adalah konsentrasi estradiol-17β, gonado somatik index (GSI), hepatosomatik index (HSI), histologi gonad, tingkat kematangan gonad, diameter telur, tingkat kebuntingan, dan kualitas air (oksigen terlarut (DO), pH, amoniak, dan suhu). Hasil terbaik pada akhir penelitian ini adalah perlakuan hCG+AD dengan nilai konsentrasi estradiol-17β (13,63-95,49 pg/mL), GSI (1,25%), HSI (1,53%), TKG (IV), diameter telur (0,88 mm), dan tingkat kebuntingan (85,71%). Perlakuan kontrol dan antidopamin tidak berpengaruh dan hanya mampu mencapai tingkat kematangan gonad I. Hasil analisis parameter kualitas air adalah berkisar DO (5,32-5,58 mg/L), amoniak (0,558-0,978 mg/L), pH (6,06-6,36), dan suhu (27,80oC-27,94oC). Hasil ini menunjukkan bahwa hCG dapat menginduksi pematangan gonad belut sawah.

Abstract

One important phase in the fish reproduction cycle is the gonad maturation process. In general, catfish reproduce in the rainy season and do not reproduce in the dry season.The purpose of this activity is to accelerate the reproductive performance of catfish broodstock to produce optimal seeds outside the spawning season. Efforts to obtain optimal fish seed can be done by improving the performance or reproductive performance through supplementation of nutrients in the broodstock feed and combined with Oodev hormone premix. Catfish broodstock as many as five pairs with a total weight of 17.5 kg were fed with Oodev 0.5 mL/kg and turmeric 3%/kg of feed.Feeding is done as much as 3% of the total weight of catfish broodstock with a frequency of three times a day ie at 09.00, 15.00, and 21.00 WIB. Maintenance is carried out for 4 weeks. The results of the activity showed that the administration of Oodev and turmeric were able to produce mature gonads as much as 60%.

Abstract

Nilai hematologi dapat digunakan untuk mengevaluasi respon fisiologi pada ikan. Suplementasi bahan herbal ke dalam pakan merupakan salah satu cara meningkatkan produksi akuakultur melalui peningkatan kekebalan dan fungsi fisiologis yang berhubungan dengan kesehatan ikan. Penelitian ini bertujuan untuk mengevaluasi pengaruh pemberian pakan dengan suplementasi analog kurkumin terhadap nilai hematologi ikan nila merah (Oreochromis niloticus). Rancangan yang digunakan adalah rancangan acak lengkap dengan tujuh perlakuan dan tiga kali ulangan. Sampel yang digunakan berupa induk nila betina dengan bobot badan 294,11±51,40 g. Pemeliharaan dilakukan selama 6 minggu pada jaring masing-masing berukuran 2x1x1m, dengan kepadatan 5 ekor/jaring. Pengambilan sampel darah sebanyak tiga kali, yaitu pada minggu ke-2,minggu ke-4, dan minggu ke-6. Hasil penelitian menunjukkan bahwa pemberian analog kurkumin memengaruhi respons hematologi dan kinerja pertumbuhan ikan nila merah. Berdasarkan penelitian yang telah dilakukan, suplementasi analog kurkumin 2,4 mg/100 g pakan adalah dosis yang terbaik untuk peningkatan kesehatan dan pertumbuhan ikan nila.

Abstract

The quality of fish is largely determined by the deposition of nutrients in developing oocytes and produced by hepatocytes under the control of the hormone estrogen. This study aims to determine the role of curcumin analogue supplementation in improving liver performance to support reproduction of red tilapia (Oreochromis niloticus). The experiment is designed using a completely random design with seven treatments and three repetitions. The treatments given are P0 (curcumin dosage of 0 mg 100 g-1 of pellet), P1 (curcumin analog dosage of 2.4 mg 100 g-1 of pellet), P2 (curcumin analog dosage of 4.8 mg 100 g-1 of pellet), P3 (turmeric powder dosage of 25 mg 100 g-1 of pellet), P4 (turmeric powder dosage of 50 mg 100 g-1 of pellet), P5 (commercially pure curcumin dosage of 2.4 mg 100 g-1 of pellet), P6 (commercially pure curcumin dosage of 4.8 mg 100 g-1 of pellet). The treatment group consisted of five fish. The parameters observed were the content of concentrations plasma vitellogenin, DNA, and RNA liver tissue, concentrations of serum glutamic pyruvic transaminase (SGPT), serum glutamic oxaloacetic transaminase (SGOT), malondialdehyde (MDA), and superoxide dismutase (SOD). The results showed that supplementation of curcumin analogues in the feed could increase the growth of tilapia liver tissue, which was proven by an increase in the concentration of DNA and RNA of liver tissue in the group of fish given the treatment. Improved liver performance through decreased concentrations of MDA, SGPT, and SGOT as well as increased SOD concentrations. Supplementation of curcumin analogues may increase the liver’s capacity to synthesize vitelogenin that will be stored in the developing follicles during gonadal maturity.

Highlight Research The dose, duration of immersion, and time interval of immersion of the estradiol-17β affect the rate of cannibalism and increase the seed survival of Asian redtail catfish in low treatment in this research. Over optimum treatment increased the incidence of cannibalism at Asian redtail catfish and decrease growth performance. Treatment of estradiol-17β, give the ability of Asian redtail catfish to live on high density The treatment of estradiol-17β, has an effect on concentration plasma of body testosterone and cortisol   Abstract The cannibalistic behavior of Asian redtail catfish greatly hampers the supply of these fish seeds, invoking the need to be controlled. Estradiol is one of the hormones that has been known to reduce cannibalistic behavior on fish. This study aimed to evaluate the effect of estradiol-17β immersion on the incidence of cannibalism in the rearing of post larvae of Asian redtail catfish. Post larvae was treated with a combination of doses of estradiol hormone 0, 1, and 2 ppm and immersion time of two and four hours with immersion intervals of three and six days with a completely randomized design. Each treatment had a fish density of 10 fish L-1, with mean size individual length of 6.73 ± 0.73 mm (three days of age after hatched), with four replications. Fish were fed with tubifex up until satiation point and reared for 30 days. The results showed that the administration of the estradiol-17β through immersion with a dose of 1 ppm for two hours and interval of six days was able to reduce the level of cannibalism and increase the survivability. A further increase in dose, immersion time, and interval had the opposite effect. Estradiol-17β immersion with certain dose, duration, and interval affected the rate of cannibalism, growth rate, and survival. Estradiol-17β immersion at a dose of 1 ppm for two hours and six days interval was the best to reduce the level of cannibalism and normal mortality in post larvae of Asian redtail catfish.

Abstract

Highlight Research The 17β-estradiol hormone induction reduces the percentage of R values in skin and fin color by as much as 50-60%. The 17β-estradiol hormone induction increases total length and body weight, estradiol content, GIS and HIS, and also glucose content. The 17β-estradiol hormone induction shows mature development of the gonadal profile from functional male to functional female. Abstract   As maroon clownfish (Amphiprion biaculeatus) is a protandrous hermaphroditic fish, feminization process with 17β-estradiol hormone can be applied to accelerate the female broodstock candidate supply for further spawning effort. This study aimed to evaluate the feminization of A. biaculeatus with 17β-estradiol hormonal induction. This study used a completely randomized design with several hormone dosage, namely P0 (without 17β-estradiol hormone induction), P1 (0.5 μg 17β-estradiol/g body), P2 (1.0 μg 17β-estradiol/g body), and P3 (1.5 μg 17β-estradiol/g body). These treatments were applied with three replications. Five fish composed of α-fish, β-fish, and three γ-fish were reared in each aquarium for 90 days with a flowing water system. The α- and β-fish were then removed, while the γ-fish was injected with hormone. Otohime pellet feed was fed three times a day until apparent satiation. The results showed that the 17β-estradiol hormone could induce 100% of the feminization process of male A. biaculeatus. The dosage of P3 obtained the lowest value percentage of red, green, blue (RGB), but showing the highest total of length and body weight (6.67±0.42 cm and 6.40±0.78 g, respectively), estradiol content (149.73±4.24 ρg/mL), GSI and HSI (0.38±0.07% and 3.59±0.49%), and glucose content (4.67±0.64 mg/dL), followed by more mature gonad profile than other treatments. This condition indicates that fish in P3 treatment has been reversed as functional female. The average survival rate for the treatment was as high as 60%. Therefore, the application of 17β-estradiol hormonal induction is effective for the feminization process in A. biaculeatus as a protandrous hermaphroditic fish.

Abstract

Viabilitas glass eel menentukan keberhasilan budidaya dan mengatur variabilitas rekrutmen, masing-masing sebagai benih dalam akuakultur dan populasi liarnya. Penelitian ini bertujuan untuk mengkaji kualitas glass eel melalui pengukuran biometrik dipasangkan dengan analisis proksimat. Pengumpulan glass eel dilakukan setiap bulan dengan menggunakan alat tangkap sirib di Muara Cimandiry selama bulan Desember 2020 - April 2021. Aspek biometrik yang diukur adalah panjang, berat, diameter mata, jantung, dan hati, sedangkan proksimat adalah kandungan protein, lemak, karbohidrat, dan abu. Hasil penelitian menunjukkan bahwa panjang dan berat badan terus bertambah setiap bulannya, meskipun faktor kondisi tidak memperlihatkan perbedaan. Diameter mata dan bukaan mulut mengkonfirmasi penglihatan ikan predator diurnal. Jantung dan hati menunjukkan kemampuan metabolisme pada ikan perenang cepat sebagai ikan bermigrasi katadromus. Protein, lemak, karbohidrat, dan abu berfluktuasi antar bulan, meskipun perbandingan antar bulan tidak signifikan. Tidak ada konfirmasi yang jelas dari analisis proksimat pada karakter biometrik. Namun glass eel dengan ukuran terbesar pada Bulan April mungkin yang terbaik untuk benih budidaya.

Abstract

Viral inhibitor protein (VIP) contributes to shrimp resistance against virus infection. VIP can be used as a new strategy to control white spot syndrome virus (WSSV) infection in shrimp culture by inhibiting its action. This study aimed to evaluate the application of VIP on the whiteleg shrimp (Litopenaeus vannamei) to inhibit the WSSV infection. Shrimp at an average body weight of 6.66±0.08 g was injected by recombinant VIP with two different concentrations (1 μg/g and 10 μg/g) and challenged with WSSV. As a positive control was the shrimp not injected with recombinant VIP and challenged with WSSV, and as a negative control was the shrimp not injected with recombinant VIP and not challenged with WSSV. The results showed that the survival rate of shrimp that were injected with recombinant VIP was significantly increased after being challenged with WSSV (P<0.05). The survival and immune response of shrimp that were injected with recombinant VIP were higher than the positive controls (P<0.05). In this study, the application of recombinant VIP can significantly increase the shrimp resistance against WSSV up to 95.65% higher than the positive control.

Abstract

Javaen barb is a native fish in Indonesian inland water with economic value and the potential to be developed as cultured fish resources and ornamental commodities. In the development of aquaculture, there are still problems, such as the low adaptability of the larvae, so that their survival is also low. This study was conducted to evaluate the effect of melatonin supplementation on the productivity of Javaen barb fish juveniles in culture containers. This study was conducted experimentally using a completely randomized design consisting of four treatments with different doses of melatonin supplementation, i.e., A) control (without melatonin supplementation), B) 0.2 mg/100 g of feed, C) 0.4 mg/100 g feed, and D) 0.6 mg/100 g feed, each treatment was repeated three times. Javaen barb juveniles were kept in an aquarium measuring 30×36×60 cm with a water level of 40 cm, consisting of 12 units. Each aquarium was stocked with 150 individuals and given an aeration system with the same air pressure intensity. Feeding was carried out ad-libitum with a frequency of three times daily for 180 days of rearing. The results showed that melatonin supplementation of as much as 0.6 mg/100 g of feed was able to increase the growth of Javaen barb fish juvenile with the highest survival rate (81.33 ± 0.54 %) and feed conversion ratio (2.61 ± 0.14). Melatonin supplementation of 0.6 mg/100 g of feed had higher total leukocyte (3.41±0.73×104 cells/mm3) and hemoglobin (5.07±0.12 g%) values and provided the best production performance in Javaen barb juveniles.
