Abstract

Stunting is a trending health problem in society, to overcome this there is one government policy, namely the Taman Pemulihan Gisi (TPG) activity which is useful for improving the nutritional status of toddlers. Research objectives: To determine the effect of supplementary feeding through TPG on the nutritional status of wasting children Pre-experimental research design (one-group pre-posttestdesign), Population: all 20 toddlers experiencing wasting in 2023 with total sampling technique. Independent Variable: Supplemental feeding. Dependent variable: change in body weight with an instrument measuring body weight using scales before and after giving additional food for 14 days, and health status (sick/ever been sick) using a questionnaire during the intervention. Data analysis using uji sperman rank. Research results Most (62.5%) aged 3-4 years with increased weight. with ρ=0.000 there is an effect of food provision on changes in body weight based on age group. The majority (77.8%) were men who experienced weight gain. with ρ=0.001 there is an effect of feeding on body weight based on gender. The majority (72.7%) changed weight and experienced illness when given additional food at TPG ρ=0.027, there was an influence of additional feeding on changes in weight based on whether the child had been sick. Providing additional food to children that meets the quality and quantity of food will have a direct impact on changes in body weight. In TPG activities carried out with cadres, health workers and mothers of toddlers for 14 days, there is health education to prepare menus of various types, cooking methods, determining the content and appearance of food so that children increase their appetite for the maintenance and repair of body cells and life processes. Children and mothers, cadres and health workers gather in TPG activities every day to carry out weight checks, health status checks and education for mothers on how to prepare menus for children. There was an increase in weight in children who received additional food in TPG activities. There was an increase in weight in children who received additional food in TPG activities. Good cooperation between mothers, cadres and health workers during the implementation of supplementary feeding through TPG is the key to success in increasing changes in the nutritional status of wasting children.

Abstract

BACKGROUND: Type 2 Diabetes (T2D) impairs the innate immune system including monocytes. Monocytes are divided into two subgroups depending on the expression of cluster of differentiation (CD)14 and CD16 receptors, namely CD14+CD16- and CD14+CD16+. CD14+CD16+ are proinflammatory monocytes and develop into M1 type macrophages, which contribute to foam cell production, a risk factor for cardiovascular disease (CVD). Therefore, it is important to determine the influence of T2D conditions on changes in monocyte subsets and whether these changes correlate with CVD risk markers.METHODS: Peripheral blood mononuclear cell (PBMC) was obtained from 10 T2D subjects and 10 healthy donors. Subsequently, PBMC was incubated for 24 hours with and without 10 mL lipopolysaccharide. Flow cytometry was used to evaluate CD14 and CD16 expression, while multiplex immunoassays were applied to measure interleukin (IL)-1b and IL-10 concentrations in supernatants.RESULTS: In T2D, the percentage of CD14+CD16+ monocytes increased (p=0.07), and an increase in CD14+CD16+ monocytes more than 6.8% was linked with CVD risk markers (r=10.146, p=0.002). Meanwhile, inflammatory mediators released by monocytes shown an increase in IL-1b (p=0.041) but not in IL-10 (p=0.082) in T2D subjects. Fasting blood glucose levels were also found to be substantially linked with an increase in CD14+CD16+ monocytes (r=0.530, p=0.016).CONCLUSION: T2D patients had a higher percentage of CD14+CD16+ monocytes and IL-1b levels than healthy donors. An increase in CD14+CD16+ monocytes above 6.8% associated with CVD risk markers in T2D patients.KEYWORDS: type 2 diabetes, monocytes, CD14, CD16, cardiovaskular disease risk marker

Abstract

Cardiovascular disease become one of the leading factors of death in the world. Thus, research is urgently needed to discover newer drugs or therapeutical agents and biological plausibility. The H9C2 cardiomyoblast originated from embryonic BDIX rat ventricular cells and was previously used in numerous in vitro studies because of its similar nature to cardiomyocytes. However, to our knowledge, there are still limited studies on the basic procedure for culturing H9C2 cardiomyoblast and arranging the best strategy to perform a suitable timeline. Here, we shared our experience in culturing the H9C2 cardiomyoblast, including harvesting and subculturing the cells. We also demonstrated the change of cell confluency, depending on the seeding number, serum concentration, and culture flask through days 1, 3, and 6, to determine their doubling-time population. H9C2 cardiomyoblasts’ doubling time is around 48-54 days with Mean±SD 2.38±0.41. However, seeding density, different culture flasks, and serum concentration have become independent factors in determining specific measures to harvest the cells for further experiments.

Abstract

Carbon fiber-reinforced epoxy (CFRE) has better performance properties and is commonly used in the aviation industry. The placement of carbon fiber will affect the mechanical properties of the resulting composite material. The specific application to actual components has yet to be discovered regarding the mechanical capabilities of the CFRE utilization. Therefore, it is essential to investigate further the mechanical behavior of CFRE composites with varying oriented carbon fibers, especially those produced using the vacuum infusion method with a volume fraction of 40%. This study presents an experimental investigation of the CFRE composite with varying oriented carbon fibers in unidirectional, bidirectional, and forged. The result shows composite materials' mechanical characteristics, particularly in the properties of Young's modulus, Poisson ratio, and shear modulus. The unidirectional fiber composites are only suitable for tensile loads in line with the fiber, while bidirectional fiber composites should be cautiously undertaken, as tensile loads applied to the Z-axis may lead to adverse effects; conversely, shear forces can be safely applied to all three axes. The forged fiber direction results in each loading direction having separate capabilities, which are challenging to anticipate. The application of CFRE on an actual component should entail the integration of diverse fiber orientations within a single composite layer, particularly in highlighting manufacturing folding arm components and their adaptability. The test results demonstrate that the maximum load that can be tolerated by the folding arm before failure fulfills the requisite strength criteria, thus allowing for a safety factor of 7.8, which confirms the safety of the CFRE with combined fiber orientation in producing folding arm components.

Abstract

BACKGROUND Hemolytic disease of the fetus and newborn (HDFN) is a type of anemia in the fetus or newborn, characterized by anemia, jaundice, hyperbilirubinemia, and brain damage. IgG is the only antibody that can cross the placenta. The IgG subtypes have a different ability to destroy red blood cells (RBCs). IgG1 and IgG3 can bind to Fc-phagocyte cell receptors and cause hemolysis, while IgG3 has more ability than IgG1. This study aimed to identify the antibody IgG subtype contributing to clinical manifestation differences in HDFN. METHODS This study used blood and umbilical cord blood samples from 30 pairs of mother-baby. The samples were grouped into control (not jaundice/normal bilirubin levels) and jaundice/hyperbilirubinemia groups. A self-developed IgG subtype enzyme-linked immunosorbent assay protocol was performed on maternal samples, resulting in optical density. Statistical analysis was performed using SPSS software version 23. RESULTS Blood type was associated with total bilirubin expression (p = 0.005). IgG1 anti-A, IgG3 anti-A, IgG4 anti-A, IgG1 anti-B, IgG3 anti-B, and IgG4 anti-B significantly affected hyperbilirubinemia in newborns (p = 0.041, 0.013, 0.017, 0.028, 0.001, and 0.007, respectively). CONCLUSIONS IgG1 and IgG3 were more significant in causing clinical problems. IgG4 suppressed IgG activation, resulting in no destruction of the infant’s RBCs.
