Abstract

Latar Bekalang : Energi diperlukan dalam segala jenis aktivitas salah satunya adalah aktivitas aerobik. Aerobik membutuhkan pasokan energi secara terus-menerus untuk menghindari hipoglikemia. Asupan makanan dengan nilai indeks glikemik rendah diduga berpengaruh positif dalam mempertahankan performa aktivitas aerobik. Tiwul singkong yang merupakan makanan asli Indonesia diketahui memiliki nilai indeks glikemik rendah. Tujuan : Membuktikan bahwa pemberian asupan tiwul singkong sebelum aktivitas lebih meningkatkan performa aktivitas aerobik ergocycling pria dewasa yang tidak terlatih daripada pemberian asupan roti tawar putih. Metode Penelitian ini adalah penelitian eksperimental menggunakan Treatment by Subject Design. Sembilan subjek menerima dua intervensi yang berbeda yaitu pemberian asupan tiwul singkong dan pemberian asupan roti tawar putih dengan jeda waktu antar kedua intervensi (wash out period) selama 7 hari. Setelah mengonsumsi asupan makanan, subjek menjalani kegiatan berupa aktivitas aerobik ergocycling hingga subjek menyatakan tidak dapat melanjutkan aktivitas (exhaustion). Hasil : Rerata durasi bersepeda maksimal pada intervensi pemberian asupan tiwul singkong adalah 93,7±30,114 menit sedangkan pada pemberian asupan roti tawar putih hanya 58,09±21,066 menit (ρ=0,001). Rerata jarak bersepeda maksimal yang mampu ditempuh pada intervensi pemberian asupan tiwul singkong adalah 30,85±12,042 km sedangkan pada pemberian asupan roti tawar putih hanya 18,69±6,837 km (ρ=0,001). Simpulan : Mengonsumsi tiwul singkong sebelum aktivitas lebih meningkatkan aktivitas aerobik ergocycling dibandingkan dengan mengonsumsi roti tawar putih baik dari segi durasi maupun jarak yang mampu ditempuh pada pria dewasa yang tidak terlatih.Kata kunci : Performa, Ergocycling, Tiwul Singkong, Roti  Tawar Putih.

Abstract

Temulawak is one of medicinal plants which possess antibacterial activity against two major causes of infection in acne namely Staphylococcus epidermidis and Propionibacterium acnes. The active compounds of temulawak are curcumin and xanthorrhizol essential oil, which possess antibacterial activity as antiseptic and antibiotic. Sambiloto (known as Andrographis paniculate), contains active compound andrographolide, which is also assumed to possess antibacterial activity. This community program was the implementation of our research for community wellbeing, which aimed to raise the knowledge and skill of PKK women at Mambang village, Selemadeg Timur, Tabanan especially in producing liquid and solid soap from natural substances (temulawak and sambiloto extract). The program was conducted by visiting our partner company PT Bali Tangi and training for liquid and solid soap production from temulawak and sambiloto (as one form of temulawak and sambiloto utilization for skin health especially for acne management). We assess participant knowledge improvement by using questionaire before and after conducting the activity. There was an improvement in participant knowledge regarding the production of soap from natural substances (temulawak and sambiloto). In conclusion, this program had increase the knowledge and skill of PKK women at Mambang village regarding liquid and solid soap production from natural substances. Abstract

Background: Recent developments in tablet preparation of extract have brought advantages to keep active ingredients from plants in stable forms physically and chemically. This study aims to compare the liquid and tablet preparation of purple sweet potato (Ipomoea batatas L.) extract to lipid profile, MDA, and SOD level in rats after given high-cholesterol diet. Methods: A randomized pre- and post-test control group experimental study was conducted during 4 weeks among 21 male Wistar rats. They were divided into 3 groups in this study namely: control (only high-cholesterol diet), Treatment 1 (T1) group (high-cholesterol diet and 200mg/day tablet of purple sweet potato extract), and Treatment 2 (T2) group (high-cholesterol diet and liquid form of purple sweet potato tubers extract about 2 mL/day). Lipid profile, MDA, and SOD level examination were performed twice from retro orbital sinus blood drawn and analyzed using ANOVA in SPSS version 17.Results: Mean value of lipid profile was significantly lower in T2 group (Cholesterol 84.48 ± 3.38; LDL 23.05 ± 2.32; HDL 63.39 ± 1.56; and triglycerides 96.03 ± 3.82; P < 0.05) after study period. The mean value of MDA was also significantly lower in T2 group (1.25 ± 0.09; P < 0.05) and SOD level was significantly higher in T2 group (73.72 ± 4.14; P < 0.05) in the end of study. Conclusion: Liquid preparation of purple sweet potato extract showed a better efficacy in reducing lipid profile and MDA level, as well as enhancing SOD level in rats given high-cholesterol diet.Keywords: Lipid Profile, Liquid, MDA and SOD Level, Purple Sweet Potato, Tablet.

Abstract

Mahogany seeds have been used in the community as an antihypertensive and antioxidant. Antihypertensive and antioxidant efficacy of mature mahogany seeds has been proven in hypertensive rats. The purpose of this study was to compare the antihypertensive and antioxidant effect of mature mahogany seeds extract with young mahogany seeds extract. This study is a laboratory experimental design with pre-test and post-test design control groups, with Wistar rats as samples. Thirty rats were divided into 5 groups, each group consist of 6 rats. The control group was given saline every day as a rat model of hypertension. 1-4 treatment groups were each given NaCl and water extract of young mahogany seeds for treatment 1, the ethanol extract of young mahogany seeds as treatment 2, water extract of mature mahogany seeds as treatment 3 and ethanol extract of mature mahogany seeds as treatments 4. Dosages of extract mahogany seeds is 100 mg / head / day for 4 weeks. Before treatment, evaluation of rat blood pressure with special sphygmomanometer was held, and examination of blood MDA as a pre-test data. Further evaluation of MDA and SOD blood pressure are at the end of the study. The data were tested by ANOVA to see the difference between the groups. The results showed a decrease in blood pressure and levels of MDA were significant in all treatments (p <0.05) compared with control as well as an increase in SOD were significantly (p <0.05). The ethanol extract of mature mahogany seeds showed the most excellent antihypertensive and antioxidant properties (p <0.05). From these results, it can be concluded that the ethanol extract of mature mahogany seeds is better than young mahogany seeds extract as an antihypertensive and antioxidant in hypertensive rats.Keywords – antioxidants, hypertensive rats, mature and young mahogany seeds

Abstract

Aqueous extract of purple sweet potato tubers have antioxidant potential in animal and human vascular endothelial, through increased expression of superoxide dismutase. This study aims to prove the comparison between ethanol extract and aqueous extract of purple sweet potato tubers increasing the expression of superoxide dismutase -2, superoxide dismutase -3, and endothelial nitric oxide synthase in human vascular endothelium. This study was an experimental study of the vascular endothelial cells which underwent oxidative stress in vitro, which is protected by ethanol extract or aqueous extract of purple sweet potato tuber at various concentrations. Observation of the superoxide dismutase -2, superoxide dismutase -3, and endothelial nitric oxide synthase in endothelial cells is done using the method immunohistochemistry with monoclonal antibodies. The results showed an increase in superoxide dismutase - 2, superoxide dismutase -3, and endothelial nitric oxide synthase was significantly (p < 0.05) in endothelium protected by ethanol extract or water extract of purple sweet potato tubers. The ethanol extract only showed its effect at concentration of 1.5625 µg/ml for superoxide dismutase -2 and superoxide dismutase -3. The effect of aqueous extract were at a concentration of 1.5625–3.125µg/ml. From these results it can be concluded that the ethanol extract or aqueous extract of purple sweet potato tubers can protect endothelial cells from oxidative stress through increased expression of superoxide dismutase -2 and superoxide dismutase -3. Abstract

Objective: The purpose of this study was to prove the water extract of purple sweet potatoes can increase superoxide dismutase (SOD) gene expression and catalase (CAT) and decrease malondialdehyde (MDA) in vital organs of diabetic rats. Method: This study was randomized experimental design with posttest only control group design. Sample divided into 2 groups: control and treatment groups, respectively 18 diabetic Wistar rats. The control group will be given streptozotocin. The treatment group was given streptozotocin and water extract of purple sweet potato 4 cc/day. This treatment is carried out for 2 months. Result: Water extract of purple sweet potato was proven significantly increased mRNA expression of aorta and kidney SOD but decreased SOD mRNA expression in the liver (p <0.05). This water extract also increases SOD mRNA expression in cardiac but not statistically significant (p> 0.05). Water extract of purple sweet potatoes also found significantly increased renal and cardiac expression of CAT mRNA (P <0.05), but not statistically significant (p> 0.05). Conclusion: Purple sweet potatoes extract to decrease the expression of liver CAT mRNA non-significantly, and decrease MDA in liver and kidney of diabetic Wistar rats significantly (P <0.05).

Abstract

To date, COVID-19 infection still become an important health problem in Indonesia, including Bali. We may concern about this situation since there has been no specific drug or vaccine available against COVID-19 infection. In this new normal era, we should maintain our immunity to prevent the infection. Some herbs potentially possess great effect on maintaining immunity to protect ourselves from COVID-19 infection, such as kunyit, temulawak, jahe, kencur, pegagan and kelor. To date, herbs utilization for health in Banjar Sambahan remains very limited. This is related to the limited skill in processing herbal drink (jamu). This activity aimed to raise participant (PKK women at Banjar Sambahan) knowledge, as well as skills, on producing herbal drinks (jamu/loloh) that potentially increase the immunity during COVID-19 pandemic era. This programme was performed by socialization about herbs utilization for health (especially for immunity), accompanied by training (by video) on producing herbal drink and survey for herbal medicine utilization in community. The results of this programme were video, publication on national journal (SINTA indexed) and presentation on national scientific meeting. It can be concluded that this activity supply knowledge and skill for PKK women at Banjar Sambahan in processing herbal drink for maintaining the immunity against COVID-19 infection. Abstract

Introduction. Sarcopenia is a condition characterized by a decrease in muscle mass accompanied by a decrease in strength and/or function. Sarcopenia is closely related to the aging process, but the mechanism of its occurrence is still not fully understood. Skeletal muscle is the largest organ in the human body, so the myokine produced through muscle contraction opens a new paradigm regarding the central role of muscles in regulating metabolism and other organ functions. One of the myokines that are currently found to affect physical activity is Irisin. This study aims to determine the role of low Irisin levels in serum as a risk factor for sarcopenia in the elderly. Methods. This case-control study was conducted in Denpasar City from January 2020 to October 2020. The sample was selected using purposive consecutive sampling with matching age and gender, with 54 subjects, 27 case groups, and 27 control groups aged ≥60 years. Sarcopenia status is enforced by the criteria of the Asian Working Group for Sarcopenia (AWGS). Irisin serum levels were measured by enzyme-linked immunosorbent assay (ELISA). Results. Based on 54 subjects, the mean age in cases was 69.79 (SD 5.00) years and controls 67.89 (SD 4.50) years (p = 0.152), the proportion of male:female both in cases and controls group was 9:18. The cut-off point value wass based on the irisin ROC curve 4.51 ng/L. Nutritional status (OR = 3.40 (95% CI = 1.11 - 10.40); p = 0.029) and irisin (OR = 25.30 (95% CI = 6,00 - 106.66); p = <0.001) had an effect on sarcopenia. Factors that purely influence sarcopenia were found in low irisin (OR = 103.41 (95% CI = 7.83 – 1,364.99; p = <0.001). Conclusions. Low Irisin level is a risk factor for sarcopenia in the elderly. Irisin could be considered for further investigation in an effort to become a target for both preventive and curative therapy for sarcopenia.

Abstract

Methicillin-Resistant Staphylococcus Aureus (MRSA) merupakan sebuah permasalahan utama dalam penyakit infeksi dikarenakan telah resisten beberapa antibiotika. Agen anti-bakteri yang dapat melawan MRSA sangat diperlukan untuk mengatasi masalah tersebut. Biji Pepaya (Carica papaya L.) mengandung Flavonoid-Saponin yang sangat berpotensi sebagai anti-bakterial yang kuat sehingga dapat dimanfaatkan dalam dunia kesehatan. Adapun tujuan penelitian yakni untuk membuktikan aktivitas anti-bakteri ekstrak Biji Pepaya terhadap perkembangan bakteri MRSA. Buah pepaya dikumpulkan dari satu perkebunan di daerah Tabanan, Bali. Biji pepaya dibuat dalam bentuk ekstrak kental menggunakan metode maserasi. Uji anti-bakteri menggunakan metode difusi Kirby-Bauer. Metode Kirby-Bauer dibuat dibuat menggunakan blank disk atau cakram disk. Dilakukan 6 perlakuan, yaitu ekstrak etanol biiji pepaya dengan konsentrasi 25%, 50%, 75% dan 100%, kontrol positif vancomycin 30µg dan kontrol negatif dimethylsulfoxide (DMSO). Setiap perlakuan dilakukan empat kali pengulangan. Diameter zona hambat pada kultur MRSA dicatat dan data dianalisis menggunakan aplikasi SPSS. Hasil uji ekstrak biji papaya menunjukan terjadi penghambatan pertumbuhan dari aktivitas bakteri MRSA yakni dengan rerata diameter zona hambat pada konsentrasi 25% sebesar 6,67±0,577 mm; 50% sebesar 9,00±1,000 mm; 75% sebesar 10,00±1,000 mm; dan 100% sebesar 11,67±0,577 mm. Abstract

Cases of infection and antibiotic resistance by Streptococcus pyogenes are increasing. Sambiloto leaf has unique phytochemical, namely andrographolide as antibacterial and other phytochemicals such as alkaloids, flavonoids, phenols, saponins. Meanwhile, moringa leaves which are often consumed as vegetables have antibacterial activity with phytochemicals including vitamin C, alkaloids, flavonoids, phenolics, triterpenoids, and tannins. This study aimed to analyze the inhibition of the ethanolic extract of sambiloto leaf, moringa leaf, and their combination on the growth of Streptococcus pyogenes bacteria in vitro. This type of research is a research using true experimental post-test only control group design. The sample was divided into 2 groups: control group and the treatment group with variations in concentrations of 25%, 50%, 75%, and 100%. Data were analyzed using the SPSS application using Kruskal-Wallis method with p<0.05. The results showed that the ethanolic extract of sambiloto leaves had no inhibition against Streptococcus pyogenes at a concentration of 25%, but had the highest inhibitory power at a concentration of 100%, which was 8.67 mm. The highest inhibitory power of moringa leaf ethanol extract was at 100%, which was 9 mm. The inhibition zone produced by the combination of the ethanol extract of sambiloto and moringa leaves showed a synergistic effect at a concentration of 100%, which was 9.33 mm. The positive control used amoxicillin with an inhibitory zone of 26.4 mm. Sambiloto leaf extract, moringa leaf, and their combination have an inhibitory effect on the growth of Streptococcus pyogenes bacteria with different concentrations of the extracts given.
