Abstract

Abstract

The kidneys as one of the important body organs have a very important role in maintaining a healthy body. The kidneys function to regulate fluid balance in the body the concentration of salt in the blood, acid-base balance in the blood, and excretion of waste materials such as urea and other nitrogenous waste in the blood. Magenta plants (Peristrophe bivalvis (L.) Merr.) contain secondary metabolites, namely alkaloids, saponins, triterpenoids, tannins, and flavonoids as antioxidants. The abundance of antioxidants sourced from natural sources for various diseases is often used as a complementary therapy and is one of the current therapeutic choices. However, the development of natural sources must also consider kidney function during an intervention. The incidence of kidney failure can be caused either by the occurrence of oxidative stress or exposure to drugs and other chemical compounds must also consider the physiological functions of important organs in the body such as the liver and kidneys. This study was conducted to determine the protective role of magenta leaves extract (Peristrophe bivalvis (L.) Merr.) on the kidneys after being given an acetaminophen hepatotoxic dose. In this study, the effectiveness of magenta leaves antioxidants and the safety of use was analyzed by looking at the kidney function in the experimental model of Wistar strain male white rats, using a Randomized Post Test Only Control Group Design. Four treatment groups showed that magenta leaves extract (Peristrophe bivalvis (L.) Merr.) at 125 and 250 mg/kg BW can protect the kidneys with average creatinine levels of 0.63 and 0.75 and with a normal range (0.7 - 1.2). It means that these two groups could protect the kidney function although in the histopathology test only the group administering extracts of 250 mg/Kg BW showed good results. It can be concluded that administration of the magenta leaves extracts at 250 mg/kg BW can protect renal function as seen from serum creatinine levels. Besides, histopathological features can provide a protective effect on the kidneys with the incidence of necrosis in the kidneys of less than 60% of the toxic dose of acetaminophen.

Abstract

BACKGROUND: The discovery of herbal ingredients for antihypercholesterolemic and to improve male reproductive function is very necessary due to high-cholesterol diet factors. Blumea balsamifera leaf extract (BBLE) is known to be able to increase the number and diameter of Leydig cells in rats given high-fat feed. This study was to conducted to determine the levels of total cholesterol, body weight, plasma malondialdehyde (MDA), Superoxide Dismutase (SOD), Luteinizing Hormone (LH) in high-cholesterol-fed male rats provided with BBLE.METHODS: This research utilized a randomized post-test only control group. Ethanol was used to extract the BBLE, which was then evaporated. For 21 days, 16 Wistar rats were given a high-cholesterol diet. To determine the effect of BBLE on the high-cholesterol diet, the samples were divided into two groups (control and BBLE group) on day 22. The treatments lasted 30 days. SOD, plasma MDA, LH, and total cholesterol were measured.RESULTS: The results showed that the SOD and LH parameters were significantly higher in the treatment of BBLE compared to the control group (p<0.05). The parameters of total cholesterol levels, bodyweight, and MDA of rats given BBLE were significantly lower than those of the control group (p<0.05).CONCLUSION: Our findings highlight that BBLE has antihypercholesterolemic and antioxidant effects. The BBLE also has potential to be used as a therapy to maintain male reproductive function because it has a positive effect on the hypothalamic-pituitary axis through increasing LH secretion.KEYWORDS: Malondialdehyde, Superoxide dismutase, Luteinizing hormone, Blumea balsamifera extract, High-cholesterol diet

Abstract

Oxidative stress as a consequence of chronic alcoholic intake causes lesions of liver tissue. The objective of this study was to identifythe antioxidant effect of ethanol extract derived from Balinese purple sweet potato on the liver tissue in mice after chronic consumptionof alcohol. Subjects of this study were 32 adults male Swiss mice (12–14 weeks) divided into 4 groups with control group post-testonly design. Post-treatment of blood and liver samples were collected from each group of 8 mice. The MDA liver was quantified withthiobarbituric acid reactive substances (TBARS) method. Samples were collected from control group of 8 mice without sweet potatoextract and alcohol, 8 mice with 1 mg extract of Balinese purple sweet potato, 8 mice with 0.8 gram alcohol, and 8 mice with combineof alcohol and extract of Balinese purple sweet potato, each treatment was carried out every day for 4 weeks. The results showed asignificant increase of serum SGOT, SGPT and liver MDA level, after chronic consumption of alcohol (p = 0.00). The decrease of serumSGOT, SGPT and liver level MDA was significant in the group with extract of Balinese purple sweet potato (p = 0.00). The researchersconcluded that the extract of Balinese purple sweet potato had an antioxidant effect in mice which consumed alcohol chronically. Basedon this study it is concluded that extract of Balinese purple sweet potato has a protective effect on the liver cell in mice which consumedalcohol chronically.
