Abstract

Hyperlipidemia is an increase in one or more plasma lipids and the main cause is related to the pattern of food intake. Among the many intestinal microbes, Lactobacillus, Bifidobacterium have been shown to affect the host beneficially by improving the intestinal microbial balance and are therefore categorized as probiotics. The aim of this study was to determine the effect of multi-strain probiotics Lactobacillus rhamnosus and Bifidobacterium bifidum on reducing LDL-C in rat fed HFD 1% per body weight. This True Experimental Research with Pretest-Posttest Design used Rattus norvegicus which was divided into 5 groups each K- (standard feed + tap water), K+ (HFD without probiotics), P1 (HFD + 2mL probiotics), P2 (HFD + 2 .5mL probiotics) and P3 (HFD + 3mL probiotics). The results of the LSD test for LDL cholesterol levels after administration of probiotics, the group that had a significantly different value was in the K- to K+ group with p=0.00; K+ to P1, P2 and P3 with p 0.00 each. Meanwhile, among the treatment groups that were given probiotics, only the P2 to P3 groups showed a significant difference with a p value = 0.037.

Abstract

BACKGROUND: Lipopolysaccharide (LPS) has been reported to increase CD4+ regulatory T-cell (CD4+ Treg) populations. Acetylsalicylic acid (ASA) has been reported to have immunomodulatory activity, but it may induce chronic gastric ulceration. Another salicylic acid-bearing compound, 2-(3-(chloromethyl)benzoyloxy)benzoic acid (3-CH2Cl), has been reported to have less gastric mucosal damage. However, the effect of 3-CH2Cl on CD4+ Tregs in LPS-induced mice is still unknown. Therefore, the present study was conducted to investigate the immunomodulatory effect of 3-CH2Cl on CD4+ T-cell and CD4+ Treg populations as well as FoxP3 expression in LPS-induced mice.METHODS: Synthesis of 3-CH2Cl was performed by mixing salicylic acid and chloromethylbenzoylchloride with the catalyzation of pyridine, acetone and heat. The 3-CH2Cl tablets were prepared using direct compression method. After intraperitoneal injection of 1 mg/kg BW LPS to mice, 60 mg/kg BW ASA or 60 mg/kg BW 3-CH2Cl was given orally for 3 days. The splenocyte was obtained through splenectomy and collagenase digestion. The population of CD4+ T-cells and CD4+ Tregs, as well as the splenic FoxP3 expression were determined using flow cytometry technique.RESULTS: CD4+ T-cell populations in mice treated with LPS and 3-CH2Cl or ASA were lower than those treated with LPS merely. Meanwhile, CD4+ Treg populations and FoxP3 expression levels in mice treated with LPS and 3-CH2Cl or ASA were higher than those treated with LPS merely.CONCLUSION: Since 3-CH2Cl could decrease CD4+ T-cell population and increase CD4+ Treg population mediated by the increase of FoxP3 expression in LPS-induced inflammation, it may act as a potential therapeutic drug to reduce inflammatory conditions.KEYWORDS: 2-(3-(chloromethyl)benzoyloxy)benzoic acid, acetylsalicylic acid, CD4, T-regulatory cells, FoxP3, LPS

Abstract

Highlight: Stroke is one of the leading causes of death and disability in the world. There is correlation between HDL cholesterol in acute thrombotic patients and the functional degree of the NIHSS. The lower rate of HDL cholesterol is related to the higher NIHSS score or the higher level of stroke severity.   ABSTRACT Introduction: Stroke is one of the world’s leading causes of death and disability. Acute thrombotic stroke accounts for 87% of all stroke cases. The study of the correlation between high-density lipoprotein (HDL) cholesterol levels and acute thrombotic stroke is still limited, especially on the relationship between HDL levels and the degree of functional stroke. Objective: This study aimed to prove a correlation between HDL cholesterol and the functional scale measured by the NIHSS (National Institutes of Health Stroke Scale) in acute thrombotic stroke. Methods: The design study was cross-sectional, with sampling by consecutive admission based on inclusion and exclusion criteria in the neurological ward (Seruni A) at Dr.Soetomo General Academic Hospital, Surabaya. The NIHSS was used to determine the degree of functional scale, and HDL levels were measured by enzymatic endpoint methods. Results: From the 40 subjects studied, 16 (40%) had normal HDL levels of ≥ 40 mg/dL with an NIHSS score between 0–10, and 24 (60%) had lower HDL levels of ≤ 40 mg/dL with an NUHSS score between 0–20. There were significant results in this study (r = 0.391 and p = 0.013). Conclusion: There was a negative correlation between HDL cholesterol levels and the functional scale measured by the NIHSS in acute thrombotic stroke patients.

Abstract

Derajat keasaman tubuh dapat mengalami peningkatan (alkalosis) dan penurunan (asidosis) akibat gangguan keseimbangan kation anion serta kegagalan transportasi dan produksi makromolekul sehingga permasalahan suatu fungsi sel dapat terjadi. Afinitas Ferro(Fe2+) yang terikat oleh molekul histidin dalam struktur haemoglobin terhadap oksigen akan terganggu jika pH darah arteri lebih kecil dari 7,35 atau lebih besar dari 7,45. Tingkat gangguan asam basa tubuh dapat diukur menggunakan alat gas darah. Pada saat pertama kali muncul alat gas darah hanya menyajikan hasil pH, pCO2, dan pO2dimana parameter yang lain seperti HCO3, BE, O2saturasi harus diplot terlebih dahulu menggunakan tabel Siggard Anderson. Perkembangan teknologi, mendukung terciptanya alat gas darah yang terintegrasi dengan logaritma bahasa program parameter hitungan, sehingga nilai HCO3,Base Excess Extra Cellular Fluid(BEecf), Base Excess Blood(BEB), O2 saturasi, dan parameter lain dapat termunculkan setelah didapatkan nilai pH, pCO2, dan pO2. Basis penelitian ini adalah pembuatan aplikasi mobile BGA UMAHA versi 0.1.0 yang dapat memberikan hasil interpretasi secara cepat, tepat, praktis, dan murah dengan tambahan inputan suhu tubuh dan kadar Hb pasien untuk mendapatkan nilai BEBase dan O2saturasi yang diperlukan, dengan demikian diharapkan permasalahan stabilitas alat, tingkat kelelahan klinisi dan efisiensi biaya dapat dilakukan. Kesimpulan pada penelitian ini adalah aplikasi mobile BGA UMAHA versi 0.1.0 dapat memunculkan parameter hitungan dengan inputan suhu dan kadar Hb dengan akurasi tinggi tanpa dipengaruhi nilai LOD alat serta dapat memunculkan interpretasi hasil.

Abstract

Tuberculosis is an infectious disease caused by the bacterium Mycobacterium tuberculosis. This study aims to determine the effect of supplementing snakehead fish (Channa striata) on body weight at the Sidotopo Health Center. The method used in this study was a one group pre test post test design involving 20 patients using a purposive sampling. The study inclusion criteria were clients who had just been diagnosed with active tuberculosis, took OAT regularly, aged 18-60 years. Exclusion criteria were TB patients who refused the study, MDR (Multi drug Resistance) TB, smokers, had a history of allergies, took other supplements. This research was conducted in June 2022 at the Sidotopo Health Center in Surabaya. Laboratory analysis was carried out at the Tropical Disease Institute, Airlangga University. The results of statistical tests in this study were that there were differences in the administration of snakehead fish (Channa striata) on body weight in tuberculosis patients with a p-value = 0.000 (<0.005). The pretest reached 46.25 kg, while the post test body weight was 48.20 kg. This shows that there is an increase in body weight in TB patients at the Sidotopo Health Center in Surabaya. Abstract

Background: Malaria is a parasitic disease worldwide with a high morbidity and mortality. A rapid and accurate method is needed to detect the presence of malaria parasites in blood. A flagging system atypical depolarization (atypdep) in CBC results from Cell-Dyn 3200 has been related with malaria infection. Materials and Methods: An observational cross sectional approach with 48 samples obtained from inpatients of the Dr.Soetomo Hospital, Surabaya. Samples were screened by Cell-Dyn 3200 analyzer for atypdep flagging in CBC. Positive samples were later confirmed by microscope to detect malaria parasites. results: From 48 samples with atypdep flagging, 7 samples were malaria positive on peripheral blood smear (13.1%). Most frequent atypdep flagging was seen in malignancy (18.7%), and approximately 54.6% of the samples were not accompanied by fever symptoms. Lekositosis and anemia each were found in 20 samples (41.6%) and thrombocytopenia in 33.3%. conclusion: The presence of atypdep flagging in Cell-Dyn 3200 does not necessarily indicate the existence of malaria or it could be said that atypdep flagging is not always associated with presence of malaria infection. The usage of an atypdep flagging in non-endemic areas such as Surabaya is just an alert sign to evaluate malaria infection rather than a screening method to detect malaria.

Abstract

Currently, therapy with Platelet Rich Plasma (PRP) has been widely used and continues to grow for various clinical applications. Along with its development, there are various options in the method of obtaining PRP, either automatic or manual, while one of the most reliable methods according to the literature is a double centrifugation method. The purpose of this research is to produce anoptimization of the double centrifugation method. This study used experimental data obtained by conducting a research at the Clinical Pathology Laboratory of Dr. Soetomo Hospital, Surabaya. Experiments were conducted on stored blood obtained from the blood bag from Indonesian Red Crossand fresh blood from healthy donors with CPD anticoagulant. Results: PRP with optimum platelet count could be made with sufficient personal laboratory skills and amounted to 4.11 times with the platelet count of 1.152 million using 1300 rcf for 5 minutes for the first centrifugation, and 2300 rcf for 7 minutes for the second centrifugation.

Abstract

M. tuberculosis (MTB) is an intracelular bacteria that live in the host macrophage cells. Several organs can be affected by tuberculosis but most major illnesses are lung diseases. Immediately after infection, MTB will be phagocytosed by the alveolar macrophage cells and can survive in the phagosome. The macrophage plays a role in innate immunity towards an infection using autophagy by removing the microbe directly via phagocytosis. When bacteria phagocytosized, vacuole membrane formed double membranes called autophagosome, and followed by degradation by lysosome, which known as  autolysosome. Induction of autophagy can be observed on the formation of microtubule-associated proteins 1B lightchain 3B (MAP1LC3B/LC3). MAP1LC3B is protein that have role at autophagic way for selection autophagy substrate and biogenesis. In this study we are used serum from patients TB with rifampicin resistant and rifampicin sensitive as control. Samples were divided using gene expert to differentiate between resistant and sensitive rifampicin.This research aims to compare MAP1LC3B levels in resistant and sensitive rifampicin to study macrophages respond in autophagic way in tuberculosis patients, and give information for define therapy plan to improve therapy for MDR-TB patients. Type of this research is a case control study design with cross sectional research with each groups sample is 19 from age 18-65 years old. Result, MAP1LC3B serum levels on the rifampicin resistant group are lower compared to rifampicin sensitive group. This occur because MTB is able to hide and evade innate immune defense mechanisms. MTB can maintain intracellular growth inside the phagosome by inhibiting phagolysosome formation in autophagy process especially inhibit MAP1LC3B formation by PDIM.

Abstract

Currently, infectious disease is increase in world wide. The African leaf (Vernonia amygdalina) – VA is used to antimicrobial treatment. It may protect the host against microbial attack in several ways. This plant has attracted the interest of researchers in recent decades because of the constituents have important roles in modulating immune system in bacteria infection. The aim of study is to analyze the prophylactic activity of VA's ethanol extract in modulating the levels of IL-6 and IL-10 as well as the number of bacteria in male Wistar rats that were (Staphylococcus aureus) – SA – infected. There were as many as 30 rats were divided into 5 treatment groups: negative control (NC) was treated by CMC Na 2% (w/v); positive control (PC) was treated by 9mg/200g body weight (BW) of cephadroxil; T1; T2; and T3 were respectively treated with ethanol extract of VA of doses 20mg/200g BW; 40mg/200g BW and 80mg/200g BW. After the oral treatment was administered, all the rats were infected with 0.25mL (3x108cfu) SA via intra peritoneal route. Their blood was drawn in order to identify the IL-6 and IL-10 levels by ELISA. Furthermore, their peritoneal fluid was also taken to count the number of survived bacteria by pour plate method. The results are showed median of IL-6 and IL-10 levels as well as bacterial number respectively in NC 370.530pg/mL; 67.044pg/mL; 7.4x103cfu/mL; in PC 234.556pg/mL; 42.839pg/mL; 6.8x103cfu/mL,; in  T1 164.019pg/mL; 17.240pg/mL; 1.1x104cfu/mL; in T2 49.291pg/mL; 2.961 pg/mL; 6.3x103cfu/mL and in T3 43.342pg/mL; 13.235pg/mL; 7.1x103cfu/mL. These results are implied that VA's ethanol extract is effective as a prophylactic agent to suppress the bacterial invasion at dose of 40mg/200g BW in Wistar rat particularly shown by the decrease level of IL-6 and the number of bacteria.

Abstract

Dengue fever (DF) is an acute viral fever caused by RNA virus that is transmitted by Aedes aegypti and Aedes albopictus mosquitoes. DF is also called viral arthropod-borne disease and is accompanied by headaches, joint and muscle pain. The main target of dengue infection is macrophages or monocytes and dendritic cells (DC). Infected DC is caused the viral replication and the endocytosis into endosomal, easier, thus inducing the activation of NF-ĸB transcription factor to produce proinflammatory cytokines such as Tumor Necrosis Factor-α (TNF-α), Interleukin-1 (IL-1), IL-6, IL-12 and chemokine. NF-kB is one of the transcription factors involved in the regulation of the expression of various cytokines, chemokines and anti/pro-apoptotic proteins during infection and act as indicator of disease severity. Infected DC cells are secreted NS1 protein which is the co-factor needed for viral replication and can be detected in the first eight days. The level will be higher in the initial phase of fever. The purpose of this study was to analyze the description of NF-kB and NS1 levels in the serum of patients with dengue fever through observational analytic studies through a cross-sectional approach. This study was done on 40 patients with dengue fever and 10 healthies people as negative controls. NS1 was analyzed in serum of Panbio rapid test and NF-kB level were measured by sandwich ELISA. The results are showed positive and negative NS1 results in dengue fever patients. The average NF-kB serum level in dengue fever patients was found to be higher than the control. NF-ĸB level in negative NS1 was higher than the NS1 positive group. It is showed that NS1 is detected both in the acute phase. The detection of NF-ĸB is showed the involvement of transcription factors in the development of dengue virus infection and has a protective role for host cells.
