Abstract

Highlights: • The effectiveness of vaccination through the kinetics of the antibody response to the SARS-COV-2 vaccine administration by examining IgG S-RBD levels was aimed. • Two doses of the Sinovac vaccine formed antibodies in healthy individuals in the first 3rd month after vaccination. • An S-RBD IgG antibody in the 6th-month post-vaccination was significantly different between groups without a history and groups with a history of infection with COVID-19. Abstract: Since it was declared a pandemic in early 2020, Coronavirus Disease 2019 (Covid-19) has caused high morbidity and mortality in the world. In view of the urgency of the situation, vaccination efforts are needed to break the chain of disease transmission. Various types of vaccines have been successfully developed and obtained approval for emergency use. However, the effectiveness of these vaccines, both in the short and long term, has not been fully known. This study aimed to examine the effectiveness of vaccination through the kinetics of the antibody response to the administration of the SARS-COV-2 vaccine by examining IgG S-RBD levels. This study was an observational analytic study with a prospective cohort approach carried out between January and November 2021 at Dr. Soetomo General Academic Hospital, Surabaya, Indonesia. Fifty health workers who received Sinovac vaccination in as many as 2 doses underwent venous blood taking and measurement of quantitative S-RBD antibody level. Then, quantitative S-RBD IgG antibody levels were measured and recorded in each subject. The mean S-RBD IgG antibody was found to have fluctuation. The titer was found to significantly increase on day 14 and dropped significantly in month 3 (p <0.001). There was a significant difference in S-RBD IgG levels 6 months after vaccination between Covid-19 uninfected groups and Covid-19 infected groups (p <0.001). In a conclusion, two doses of the Sinovac vaccine formed antibodies, although humoral immunity obtained tended to decrease in 3rd month after vaccination to healthy individuals. The average level of S-RBD IgG antibody in the sixth month post-vaccination was found to be significantly different between groups without history and groups with a history of with infection COVID-19.

Abstract

Sepsis is a major problem causing high morbidity and mortality in patients who hospitalized in intensive care. In sepsis, there is a change in the distribution of albumin in the intravascular and extravascular compartment, so hipoalbumin is occured. Using albumin in septic patients from several studies that have been done are still controversial. Analyzing albumin infusion with changes in levels of albumin, cytokines and SOFA score in septic patients in intensive care dr. Soetomo. The design of this study is a prospective longitudinal observational. The study was conducted in the intensive care unit or ICU dr. Soetomo in February 2015. The inclusion and exclusion criteria were determined that obtained 15 research subjects. They were examined with serum albumin and proinflammatory cytokines as well as evaluating the SOFA score. Then, it measure with ANOVA test methods. Infusion of albumin positively correlated with increased levels of serum albumin and a negative correlation with the levels of IL6, TNF-a and SOFA score. It also found a correlation between changes in levels of albumin with changing levels of IL6 (p <0:01) and changes in SOFA score (p <0.05).

Abstract

The skin, the largest organ in the human body, serves as a protective barrier against external factors. Exposure to UV-B radiation accounts for approximately 90% of skin damage, leading to aging characterized by dryness and wrinkles. Proper nutrition has been associated with skin repair and delayed aging. Astaxanthin, known for its pharmacological properties including anti-cancer, anti-diabetic, anti-inflammatory, and antioxidant activities has garnered attention for its ability to improve damaged skin when administered as a daily supplement. This study investigated the roles of TNF-α and IL-10 as inflammatory markers in skin damage and repair, which remain underexplored. A true experimental randomized posttest-only control group design was employed using 24 male white rats divided into four groups: (1) normal group (N), untreated rats; (2) control group (C), rats administered with olive oil (5 ml/kg BW); (3) treatment group 1 (P1), rats exposed to UV-B and administered with olive oil (5 ml/kg BW); and (4) treatment group 2 (P2), rats exposed to UV-B and administered with astaxanthin (10 mg/kg BW) combined with olive oil (5 ml/kg BW). ANOVA and Games-Howell post-hoc tests revealed significant differences (p < 0.05) among groups, except between the control (C) and normal (N) groups. Astaxanthin (10 mg/kg BW) demonstrated both curative and protective effects by reducing TNF-α expression (pro-inflammatory) and increasing IL-10 expression (anti-inflammatory) in UV-B-induced rats. These findings highlight astaxanthin’s potential as a therapeutic agent for UV-B-induced skin damage.

Abstract

BACKGROUND: Alzheimer’s disease (AD) and vascular dementia (VaD) impose a substantial public health burden in Indonesia; however, accessible blood-based biomarkers for early screening remain limited. Although cerebrospinal fluid β-amyloid 1–42 is an established biomarker, its invasive nature restricts its use for population-level screening. Therefore, it is necessary to have locally-produced serum β-amyloid 1–42 ELISA kit that is specifically designed for Indonesian elderly population. In this study, a locally-produced β-amyloid 1–42 ELISA kit was validated and used for the screening of AD, VaD and mild cognitive impairment (MCI) Indonesian population.METHODS: A cross-sectional study including 166 subjects: 31 AD, 34 VaD, 34 MCI patients, and 67 cognitively normal controls was conducted. All participants underwent cognitive assessments including Mini Mental State Examination (MMSE) and Montreal Cognitive Assessment-Indonesian version (MoCA-Ina), as well as brain magnetic resonance imaging (MRI) 3-Tesla for the assessment of medial temporal atrophy/white matter changes. Fasting venous blood sampling was taken from each subjects for the measurement of serum β-amyloid 1-42 measurement using locally-produced ELISA kit.RESULTS: Median serum β-amyloid 1–42 levels were 11.03, 10.99, and 10.99 pg/mL for the AD, VaD, and MCI subjects, respectively. The β-amyloid 1–42 levels were correlated with MMSE scores in all group (AD: r=−0.455, p=0.010; VaD: r=−0.419, p=0.014; MCI: r=−0.412, p=0.015). The validity analysis of the locally-produced serum β-amyloid 1–42 ELISA kit, showed sensitivity of 94.12% (95% CI: 87.3–97.9), specificity of 80.36% (95% CI: 72.4–86.8), and diagnostic accuracy of 83.56% (95% CI: 77.2–88.5).CONCLUSION: Serum β-amyloid 1–42 levels are lower in AD and VaD subjects compared to MCI and control subjects. Serum β-amyloid 1-42 is inversely correlated with cognitive function across all groups based on MMSE score. Additionally, the locally-produced β-amyloid 1-42 ELISA kit demonstrated sensitivity of 94.12% and specificity of 80.36%, meeting Global CEO Initiative Consensus for pre-screening tools, supporting its potential as a scalable, non-invasive screening biomarker in Indonesian primary care settings.KEYWORDS: G-banding karyotyping, next generation sequencing, non-invasive prenatal testing
