Abstract

ABSTRACT The study was aimed to evaluate the efficacy of formalin-killed non-hemolytic Streptococcus agalactiae N14G and NK1 isolates whole-killed vaccine to prevent streptococcosis in tilapia. Ten fishes were reared in a tank 60x30x35 cm3 with an average body weight at 10.79±0.99 g. Fish was vaccinated through bath immersion at a concentration of 109 cfu/mL. Fish was subsequently challenged by intraperitonial injection of Streptococcus agalactiae 105 cfu/mL at 11 days post-vaccination. Parameters observed were survival, relative percent survival (RPS), total leukocyte, phagocytic activity, antibody titer, total erythrocyte, haemoglobin level, haematocrit level, dan water quality. Samplings were performed in day-0, 20, and 30 after vaccination. Both vaccines have shown higher survival (60%) and RPS (40%) when challenged with pathogenic Streptococcus N14G isolates than other treatments. Based on RPS percentage observed, those vaccine were still not sufficiently effective to combat S. agalactiae infection. Keywords: tilapia, bath immersion, Streptococcus agalactiae, whole-cell vaccine ABSTRAK Penelitian ini bertujuan untuk mengevaluasi efikasi vaksin formalin-killed cell Streptococcus agalactiae tipe isolat nonhemolitik N14G dan NK1 se utuh yang diberikan melalui perendaman dalam mencegah penyakit streptococcosis pada ikan nila. Ikan nila yang digunakan memiliki bobot 10,79±0,99 g, dipelihara sebanyak sepuluh ekor dalam akuarium ukuran 60x30x35 cm3. Ikan divaksinasi dengan metode perendaman dengan dosis 109 cfu/mL. Uji tantang dilakukan pada hari ke-11 pascavaksinasi dengan dosis 105 cfu/mL. Parameter yang diamati meliputi sintasan (SR), sintasan relatif/relative percent survival (RPS), total leukosit, aktivitas fagositik, titer antibodi, total eritrosit, kadar hemoglobin, kadar hematokrit, dan kualitas air. Pengamatan parameter dilakukan pada hari ke-0, ke-10, ke-20, dan ke-30. Hasil penelitian menunjukkan perlakuan kedua vaksin yang diinfeksi bakteri patogen isolat N14G memberikan nilai sintasan dan nilai RPS tertinggi dibanding perlakuan lainnya. Nilai sintasan dan RPS kedua perlakuan tersebut adalah 60% dan 40%. Nilai RPS yang cukup kecil menunjukkan vaksin yang diberikan masih kurang efektif untuk mencegah infeksi bakteri S. agalactiae. Kata kunci: ikan nila, perendaman, Streptococcus agalactiae, vaksin sel utuh

Abstract

ABSTRACT Blood parameters are considered as important indicators to diagnose fish health status. This study was performed to observe blood profiles including total erythrocytes, hemoglobin concentration, hematocrite, total leukocytes and differential leukocytes, and survival of common carp Cyprinus carpio infected by Aeromonas hydrophila. Fish were divided into two groups: the 3rd generation of common carp carrying fish carrying Cyca-DAB1*05 of major histocompatibility complex II molecular marker, and fish without the marker as control treatment. histocompatibility complex II molecular marker, and fish without the marker as control treatment. Common carp 3rd generation was produced by crossing among second generation of fish carrying the Cyca-DAB1*05 marker. Each fish was injected intramuscularly by 0.1 mL of 108 cfu/mL A. hydrophila. Challenge test was conducted for 14 days and blood was collected at day-0, three, seven, and 14. The results of this study showed that erythrocytes, hemoglobin and hematocrite concentrations of common carp carrying the molecular marker at post challenge with A. hydrophila were higher (P<0.05) compared to control fish. The blood profiles were highly correlated to survival of fish. Survival of fish that carrying the molekuler marker was about two point six fold higher than those of control fish. Keywords: Cyprinus carpio, Aeromonas hydrophila, Cyca-DAB1*05, molecular marker  ABSTRAK Gambaran darah merupakan indikator penting untuk mendiagnosa penyakit ikan. Penelitian ini dilakukan untuk meguji gambaran darah ikan mas Cyprinus carpio setelah diinfeksi dengan bakteri Aeromonas hydrophila yang meliputi total sel darah merah, konsentrasi hemoglobin, hematokrit, total sel darah putih, dan diferensial leukosit, serta sintasannya. Ikan mas yang digunakan terdiri atas ikan mas generasi ketiga yang membawa marka molekuler Cyca-DAB1*05 dari kelompok major histocompatibility complex II dan ikan mas tanpa marka sebagai kontrol. Ikan mas generasi ketiga merupakan keturunan persilangan antarikan mas generasi kedua yang mempunyai marka Cyca-DAB1*05. Masing-masing ikan diinfeksikan A. hydrophila secara intramuskuler pada dosis 0,1 mL, kepadatan 108 cfu/mL. Uji tantang dilakukan selama 14 hari, dan sampel darah ikan diambil pada hari ke-0, tiga, tujuh dan 14. Hasil uji tantang menunjukkan bahwa total sel darah merah, konsentrasi hemoglobin dan hematokrit pada ikan mas yang membawa marka molekuler lebih tinggi dibandingkan ikan mas tanpa marka (P<0,05). Hasil uji gambaran darah berkorelasi tinggi dengan tingkat kelangsungan hidup ikan mas. Tingkat kelangsungan hidup ikan mas yang membawa marka molekuler Cyca-DAB1*05 lebih tinggi hingga dua koma enam kali daripada ikan mas kontrol. Kata kunci: Cyprinus carpio, Aeromonas hydrophila, Cyca-DAB1*05, marka molekuler

Abstract

ABSTRACT Koi herpesvirus (KHV) is a virus that infects common carp and koi Cyprinus carpio. KHV attacks most stadia of common carp and koi and causes mortality up to 80–95% of the population. One way to prevent the spread of KHV is  by applying DNA vaccine. This research was conducted to test potential transmission of DNA vaccine encoding glycoprotein 25 (vaccine GP25) toward normal flora bacteria of media cultivation of common carp. Bacteria was isolated from pond water of common carp and tested for their sensitivity to ampicillin. Research was performed by adding vaccine GP25 to ampicillin-sensitive bacteria at a dose of 12.5 µg/100 µL and incubated at 28 °C for 30, 60, 180, and 300 minutes then plated on media containing ampicillin. The grown bacteria cells were tested for the existence of plasmid bearing gen GP25 through bacteria colony cracking. The results of this research showed that there was no bacteria contained plasmid bearing gen GP25. Keywords: DNA vaccines, common carp, vaccine safety  ABSTRAK Koi herpesvirus (KHV) merupakan virus yang menginfeksi ikan mas dan koi Cyprinus carpio. KHV menyerang hampir semua stadia ikan mas dan koi dan menyebabkan kematian hingga 80–95% dari populasi. Salah satu cara penanggulangan penyebaran KHV adalah dengan penggunaan vaksin DNA. Penelitian ini dilakukan untuk menguji potensi transmisi vaksin DNA glikoprotein 25 (vaksin GP25) pada bakteri flora normal pada media budidaya ikan mas. Bakteri diisolasi dari kolam budidaya ikan mas dan diuji sensitivitasnya terhadap antibiotik ampisilin. Penelitian dilakukan dengan menambahkan vaksin GP25 ke larutan bakteri sensitif ampisilin dengan dosis 12,5 µg/100 µL dan diinkubasi pada suhu 28 °C selama 30, 60, 180, dan 300 menit kemudian disebar pada media mengandung antibiotik ampisilin. Sel bakteri yang tumbuh diuji untuk mendeteksi keberadaan plasmid pembawa gen GP25 dengan menggunakan metode seleksi koloni bakteri (colony cracking). Hasil penelitian ini menunjukkan bahwa tidak ada bakteri yang mengandung plasmid pembawa gen GP25. Kata kunci: keamanan vaksin, ikan mas, vaksin DNA

Abstract

ABSTRACT Larval stage of giant gourami is a critical period due to fungal infection, such as Saprolegnia sp. infection. There are some plants which have antiseptic compound like banana Musa paradisiaca. This research was aimed to examine the effectiveness of the banana stem extract M. paradisiaca to control Saprolegnia sp. infection on giant gurami larvae through immersion. Eight-day old gorami larvae (at the initial of 0.5±0.03 cm) was reared in an aquarium sized 25×25×25 cm3 at the density of 8 fry/L. Culture media were added banana stem extract at the dose of 0; 0.08; 0.12; and 0.16 g/L during 21 days of rearing period. Challenge test was performed for 14 days by giving Saprolegnia sp. spores at the density of 104 cells/mL and banana stem extract. The treatment dose of 0.16 g/L has showen survival 100% than positive control  after the challenge test. Keywords: giant gourami, Musa paradisiaca, Saprolegnia sp., fry  ABSTRAK Fase larva ikan gurami merupakan masa kritis terhadap infeksi cendawan, seperti jenis Saprolegnia sp. Beberapa tanaman memiliki daya antiseptik seperti tanaman pisang ambon Musa paradisiaca. Penelitian ini bertujuan untuk menguji efektivitas ekstrak batang pisang ambon M. paradisiaca dalam mengurangi infeksi Saprolegnia sp. pada larva ikan gurami melalui media pemeliharaan. Larva gurami umur delapan hari (panjang larva 0,5+0,03 cm) dipelihara pada akuarium berukuran 25×25×25 cm3 dengan padat tebar 8 ekor/L. Media pemeliharaan diberi ekstrak batang pisang ambon dosis 0; 0,08; 0,12; dan 0,16 g/L selama 21 hari. Uji tantang dilakukan selama 14 hari dengan pemberian spora Saprolegnia sp. kepadatan 104 sel/mL dan ekstrak batang pisang ambon. Perlakuan dosis 0,16 g/L memberikan kelangsungan hidup sebesar 100% yang lebih tinggi dibandingkan perlakuan kontrol positif setelah uji tantang. Kata kunci: giant gourami, Musa paradisiaca, Saprolegnia sp., larva

Abstract

ABSTRACT As stocking density is known as a stress factor in fish. This study aimed to evaluate the effect of dietary astaxanthin supplementation on growth performance and antioxidant status in giant gourami under normal (100 fish/m3) and high (400 fish/m3) stocking density. Addition of astaxanthin in the feed was done by coating method. The experimental fish used were giant gourami with average body weight of 3.0±0.12 g. Fish were reared for 60 days in 100×50×50 cm3-sized-aquariums containing 150 L of water. Feed was given at satiation method twice a day at 09.00 and 17.00 pm.  Results showed that fish reared in high stocking density and fed on the diet supplemented with astaxanthin 100 mg/kg diet produced the best growth performance. Addition of astaxanthin did not affect the antioxidant activity of giant gourami. Keywords: giant gourami, astaxanthin, growth performance, antioxidant status  ABSTRAK Padat tebar tinggi merupakan salah satu faktor yang dapat menyebabkan stres pada ikan budidaya. Tujuan penelitian ini adalah untuk mengevaluasi pengaruh penambahan astaxanthin melalui pakan terhadap kinerja pertumbuhan dan status antioksidan ikan gurami pada pemeliharaan kepadatan normal (100 ekor/m3) dan kepadatan tinggi (400 ekor/m3). Penambahan astaxanthin pada pakan dilakukan dengan metode pelapisan (coating). Ikan uji yang digunakan adalah benih ikan gurami berukuran bobot rata-rata 3,0±0,12 g. Ikan dipelihara selama 60 hari dalam akuarium berukuran 100×50×50 cm3 berisi air 150 L. Pakan diberikan secara at satiation dengan frekuensi pemberian pakan sebanyak dua kali sehari pada pukul 09.00 dan 17.00 WIB. Hasil penelitian menunjukkan bahwa ikan yang dipelihara pada kepadatan tinggi dan diberi pakan dengan penambahan astaxanthin sebesar 100 mg/kg pakan menghasilkan kinerja pertumbuhan yang optimal. Penambahan astaxanthin sebesar 100 dan 200 mg/kg pakan tidak mempengaruhi status antioksidan pada ikan gurami. Kata kunci: ikan gurami, astaxanthin, pertumbuhan, status antioksidan

Abstract

ABSTRACT In this study, tambaqui Colossoma macropomum juvenile were given feed supplemented with clove oil Syzygium aromaticum at a dose of 0 and 100 mg/100 g diet. This study purposed to assessment the effect of clove oil supplementation at high dose (100 mg/100 g diet) on growth performance and health status of freshwater tambaqui. As many as 25 fishes with initial body weight 7.62±0.44 g maintained in 15 aquariums for 45 days. Each treatment consisted of three replications. Data were analyzed by t-test. The results showed that tambaqui growth performance on both treatments were not significantly different. However, supplementation of high doses 100 mg clove oil/100 g diet showed reduce on the blood parameters and declining the abundant of intestinal microflora in tambaqui gut, but the types seen more diverse. The observation of fish liver histology showed that hepatocyte cells in the treatment of 100 mg clove oil become smaller and dense compared with 0 mg clove oil treatment. The use of high doses of clove oil does not interfere with the tambaqui growth performance despite the downturn in the physiological responses were observed. Keywords: clove oil, Syzygium aromaticum, Colossoma macropomum, growth performance  ABSTRAK Di dalam penelitian ini, benih ikan bawal tawar Colossoma macropomum dipelihara dengan pemberian pakan yang ditambah minyak cengkeh Syzygium aromaticum pada kadar 0 dan 100 mg/100 g pakan. Tujuannya untuk melihat pengaruh penambahan minyak cengkeh di dalam pakan pada dosis tinggi (100 mg/100 g pakan) terhadap performa pertumbuhan dan respons fisiologis ikan bawal. Sebanyak 25 ekor ikan bawal dengan bobot individu awal 7,62 ± 0,44 g dipelihara pada 15 akuarium selama 45 hari. Data yang diperoleh dianalisis dengan uji komparasi t-test antar dua perlakuan dengan tiga ulangan. Hasil penelitian menunjukkan bahwa tidak ada pengaruh dari kedua perlakuan terhadap parameter pertumbuhan. Namun, pemberian pakan yang mengandung minyak cengkeh dosis tinggi 100 mg/100 g pakan mempengaruhi respons fisiologis berupa rendahnya kisaran nilai gambaran darah serta menurunnya jumlah mikroflora di usus ikan bawal, meski jenisnya lebih beragam. Hasil pengamatan histologi hati ikan pada perlakuan 100 mg minyak cengkeh menunjukkan bahwa sel hepatosit berukuran lebih kecil dan terlihat rapat dibandingkan dengan perlakuan 0 mg minyak cengkeh. Penggunaan minyak cengkeh dosis tinggi tidak menganggu performa pertumbuhan ikan bawal meskipun terjadi penurunan pada respons fisiologis yang diamati. Kata penting: minyak cengkeh, Syzygium aromaticum, Colossoma macropomum, performa pertumbuhan

Abstract

ABSTRACT The aim of this study was to assess the protective duration of Streptococcus agalactiae vaccine against streptococcosis in Nile tilapia. Fish were treated by the whole cell vaccine, ECP vaccine and mixture of whole cell and ECP vaccine. After 14, 28, 42, and 52 day post-vaccination (DPV), the fish were intraperitoneally challenged with 104  cfu/mL S. agalactiae. The results showed mortality rate of whole-cell vaccine (A), ECP vaccine (B) and mix vaccine (C) up to day 42 was significantly (P<0.05) lower than the control treatment, namely 73.33%; 80%; and 76%, respectively. The mortality rate of vaccine treatments A, B, and C on day 56 had no significant difference (P>0.05) with the control. The value of antibody titer vaccine treatments A, B, and C indicate that antigen-antibody reaction on day 28 after the vaccination was significantly (P <0.05) higher than the control that were 3.67; 3.33; and 3.67. Antigen-antibody reaction on day 42 after the vaccination was founded, but did not different significantly (P>0.05) with the control. Bacterial population in treatment A, B, and C in the organs of the fish until the 28th day was still under the control of 104  cfu/mL. S. agalactiae vaccine protection duration is 42 days after the vaccination. Keywords : nile tilapia, Streptococcus agalactiae, duration, vaccine, streptococcosis  ABSTRAK Penelitian ini bertujuan untuk menganalisis durasi proteksi dari vaksin Streptococcus agalactiae sebagai pencegahan terhadap streptococcosis pada ikan nila. Ikan divaksinasi dengan vaksin sel utuh, ECP dan gabungan sel utuh dan ECP dari S. agalactiae yang diinjeksi secara intrapetorineal. Ikan diuji tantang S. agalactiae 104  cfu/mL pada hari ke-14, ke-28, ke-42, dan ke-56 pascavaksinasi. Hasil penelitian menunjukkan tingkat mortalitas perlakuan vaksin sel utuh (A), vaksin ECP (B), dan gabungan vaksin sel utuh dengan ECP (C) hingga hari ke-42 masih signifikan (P<0,05) lebih rendah dari perlakuan kontrol yaitu 73,33%; 80%; dan 76%. Tingkat mortalitas perlakuan vaksin A, B, dan C pada hari ke-56 sudah tidak berbeda signifikan (P>0,05) dengan kontrol. Nilai titer antibodi perlakuan vaksin A, B, dan C menunjukkan bahwa reaksi antigen antibodi pada hari ke-28 pascavaksinasi masih signifikan (P<0,05) lebih tinggi dibandingkan kontrol  yaitu 3,67; 3,33; dan 3,67. Reaksi antigen antibodi pada hari ke-42 pascavaksinasi masih ditemukan, namun tidak berbeda signifikan (P>0,05) dengan kontrol. Populasi bakteri pada perlakuan A, B, dan C di organ ikan hingga hari ke-28 masih di bawah kontrol 104  cfu/mL. Abstract

ABSTRACT The aim of this study was to evaluate infectivity of Ichtyophthirius multifiliis which caused white spot disease maintained at low temperature without its host. Briefly, the trophont stage of parasites were subjected at control (28 °C) and lower temperature (9 °C) for 14 consecutive days of observation. The rate of survival, and excystment of descendants were examined descriptively at the last day of observation. Here, the infectivity of parasite then performed by means infecting the model fish Poecilia sphenops (black moly) with escaping theronts. The results revealed that the survival rate and excystment  rate of parasite were decreased as maintaining period increased. The final rate of survival, and excystment of parasite were 35% and 33,3% respectively. Additionally, the descendants came out with high abnormality which recognized by weak mobility and lower infectivity (50%) compared to the control (80%). Then, it is concluded that, maintaining I. multifiliis at low temperature without its host for 14 consecutive days will decreased the infectivity. Keywords: white spot, obligat parasite, excystment, infectivity  ABSTRAK Penelitian ini bertujuan untuk mengevaluasi infektivitas parasit Ichtyophthirius multifiliis penyebab penyakit bintik putih (white spot) yang dipelihara tanpa inang pada suhu rendah. Parasit dengan stadia trophont dipelihara pada suhu ruang (28 °C) dan suhu rendah (9 °C) selama 14 hari. Selama masa pemeliharaan tersebut tingkat kelulusan hidup, dan tingkat eksismen parasit diukur dan dibandingkan secara deskriptif. Hari terakhir pemeliharaan dilakukan uji tantang pada ikan black moly Poecilia sphenops untuk menilai infektivitas parasit. Hasil penelitian memperlihatkan bahwa tingkat kelulusan hidup dan eksismen parasit semakin menurun dengan bertambahnya masa pemeliharaan. Akhir pengamatan  kelangsungan hidup, dan nilai eksismen tersebut berturut-turut adalah 35% dan 33%. Parasit yang disimpan pada suhu rendah selama 14 hari memperlihatkan infektivitas yang lebih rendah (50%) dibandingkan dengan perlakuan kontrol (80%). Kesimpulannya, penyimpanan parasit I. multifiliis pada suhu rendah selama 14 hari dapat menurunkan infektivitas parasit pada inang. Kata kunci: bintik putih, parasit obligat, eksismen, infektivitas

ABSTRACT Outbreaks of infectious diseases due to Aeromonas hydrophila in catfish can cause high death rates (80–100%). Fish disease control can be done using phytopharmaceutical to prevent or treat diseases of fish. One of the phytopharmaceutical that known to prevent the fish diseases is ambon banana stem Musa cavendishii var. dwarf Paxton. This study was conducted to test the effectiveness of catfish immersion using banana stem juice as an immunostimulant against bacterial infections A. hydrophila. The observed parameters were a total performance of production, hematological test, and water quality. Immersion of catfish seeds with stem juice was performed at a concentration of 5 mL/L, 13 mL/L, and 21 mL/L for 30 minutes. Each treatment consisted of three replications. A  total of 15 fishes were immersed in 1.5 L water. At day-9 after immersion, fish was infected by A. hydrophila bacteria at a dose of 104 cfu/mL. The results showed that fish treated with banana stem juice at a concentration of 13 mL/L had the survival rate of 53.33±6.67% which is higher than positive control (33.33±6.67%). Based on hematology observations on the 3rd day post-challenge test (H12) with A. hydrophila, total erythrocytes, hemoglobin, total leukocytes, phagocytic activity, differential leukocyte, and lysozyme activity was highest in treatment13 mL/L. Lower feed conversion ratio (1.08±0.04) were also obtained in treatment 13 mL/L. Thus Immersion of seeds in stem juice can boost the immune system against infections A. hydrophila. Keywords: Aeromonas hydrophila, ambon banana stem juice, soaking, catfish, immune system  ABSTRAK Wabah penyakit akibat infeksi Aeromonas hydrophila pada ikan lele dapat menyebabkan tingkat kematian yang tinggi (80–100%). Penanggulangan penyakit ikan dapat dilakukan menggunakan fitofarmaka untuk mencegah ataupun mengobati penyakit ikan. Salah satu fitofarmaka yang dapat digunakan dalam upaya pencegahan penyakit ikan adalah batang pisang ambon lumut Musa cavendishii var. dwarf Paxton. Penelitian ini bertujuan untuk menguji efektivitas perendaman benih ikan lele dengan menggunakan air perasan batang pisang ambon sebagai imunostimulan terhadap infeksi bakteri A. hydrophila. Parameter yang diamati selama penelitian adalah kinerja produksi, uji hematologi dan pengukuran kualitas air. Perendaman benih ikan lele dengan air perasan  dilakukan pada konsentrasi 5 mL/L, 13 mL/L, dan 21 mL/L selama 30 menit. Pada hari ke-9 setelah perendaman, ikan diinfeksi bakteri A. hydrophila pada kepadatan 104 cfu/mL. Hasil penelitian menunjukkan bahwa perendaman ikan dengan air perasan pada konsentrasi 13 mL/L memiliki kelangsungan hidup yakni sebesar 53,33±6,67%, lebih tinggi bila dibandingkan dengan kontrol positif  (33,33±6,67%). Berdasarkan pengamatan pada uji hematologi hari ke-3 pascauji tantang (H12) dengan bakteri A. hydrophila, total eritrosit, hemoglobin, total leukosit, aktivitas fagositik, dan aktivitas lisozim tertinggi terdapat pada perlakuan konsentrasi 13 mL/L. Rasio konversi pakan yang rendah (1,08±0,04) juga terdapat pada perlakuan 13 mL/L. Hal ini menunjukkan bahwa perendaman benih ikan lele pada air perasan dapat meningkatkan sistem imun terhadap infeksi bakteri A. hydrophila. Kata kunci: Aeromonas hydrophila, air perasan batang pisang ambon, perendaman, ikan lele, sistem imun

Abstract

ABSTRACT This study was aimed to evaluate the dietary ambon lumut banana Musa cavendishii var. dwarf Paxton stem extract on the immune responses, growth, and survival rate of Pacific white shrimp against white spot disease. Ambon banana steam extract was obtained by maceration method using ethanol. The shrimps fed by pellet containing ambon banana stem extracts with different dosages for 29 days, i.e designated as 0.1 (A); 0.3 (B), and 0.5 (C) g/kg, the diet treatment without ambon banana stem extract without challenged test (K-), and diet treatment without ambon banana stem extract with challenged test (K+). Each treatment consisted of three replications. Feeding was conducted for 29 days of maintenance (four times a day). The results showed that the immune responses (average total hemocyte count: 45.15×106 cells/mL, phenoloxidase activity 1.03±0.08 OD, respiratory burst 0.95±0.04 OD, phagocytic activity 94.33±1.53%, growth (specific growth rate: 7.79±0.06%/day, feed ratio conversion was 52±0.01), and survival of treatment C (survival rate 100%) were higher compared with the treatment K+ (total hemocyte count: 3.83×106 cells/mL, phenoloxidase activity 0.04±0.01 OD, respiratory burst 0.18±0.06 OD,  phagocytic activity 5.67±0.58%, specific growth rate: 2.61±0.08%/day, feed conversion ratio 2.11±0.02, survival rate: 50%). Therefore, banana stem extract at a dose of 0.5 g/kg everyday diet can be used to improve growth and nonspecific immune system against white spot disease on Pacific white shrimp. Keywords: Pacific white shrimp, white spot disease, immune response, Ambon banana stem extract  ABSTRAK Penelitian ini bertujuan untuk menguji dosis optimal ekstrak batang pisang ambon melalui pakan dalam meningkatkan respon imun, pertumbuhan dan kelangsungan hidup udang putih terhadap penyakit white spot. Ekstrak batang pisang ambon lumut Musa cavendishii var. dwarf  Paxton menggunakan metode maserasi dengan menggunakan pelarut etanol. Pakan udang yang mengandung ekstrak batang pisang ambon pada dosis yang berbeda selama 29 hari 0,1 (A); 0,3 (B), dan 0,5 (C) g/kg, pakan tanpa pemberian ekstrak batang pisang ambon dan tidak diuji tantang (K-), pakan tanpa pemberian ekstrak batang pisang ambon dan diuji tantang (K+). Masing-masing perlakuan terdiri atas tiga ulangan. Frekuensi pemberian pakan empat kali sehari selama 29 hari pemeliharaan. Hasil penelitian menunjukkan bahwa perlakuan C respons imun (rata-rata jumlah total hemosit: 45,15×106 sel/mL, aktivitas phenoloxydase 1,03±0,08 OD, respiratory burst 0,95±0,04 OD, aktivitas fagositik 94,33±1,53%, pertumbuhan (laju pertumbuhan spesifik: 7,79±0,06%/hari, rasio konversi  pakan 0,52±0,01), dan kelangsungan hidup  (survival rate 100%) lebih tinggi dibandingkan dengan perlakuan K+ (jumlah total hemosit: 3.83×106 sel/mL, aktivitas phenoloxydase 0,04±0,01 OD, respiratory burst 0,18±0,06 OD, aktivitas fagositik 5,67± 0,58%, laju pertumbuhan spesifik: 2,61±0,08%/hari, rasio konversi pakan 2,11±0,02, nilai kelangsungan hidup: 50%). Jadi, ekstrak batang pisang pada dosis 0,5 g/kg setiap hari pada pakan dapat digunakan untuk meningkatkan pertumbuhan dan sistem kekebalan tubuh nonspesifik terhadap penyakit bintik putih pada udang putih.   Kata kunci: udang putih, white spot, respons imun, ekstrak batang pisang ambon
