Abstract

Penelitian ini bertujuan untuk mengetahui perbedaan persentase motilitas dan viabilitas semen kambing Sapera terhadap evaluasi semen segar dan before freezing dengan penambahan pengencer tris kuning telur dan susu skim kuning telur. Metode penelitian ini adalah pengambilan semen, evaluasi semen segar (pemeriksaan makroskopis dan mikroskopis), pengenceran semen, gliserolisasi, ekuilibrasi dan evaluasi before freezing. Penelitian ini terdiri dari P0, P1 dan P2, masing-masing perlakuan terdiri dari sembilan ulangan. P0 adalah semen segar kambing Sapera, P1 adalah semen kambing Sapera+pengencer tris kuning telur dan P2 adalah semen kambing Sapera+pengencer susu skim kuning telur. Data yang diperoleh dengan Uji T2 sampel dependent, membandingkan P0 dan P1 serta P0 dan P2. Data menunjukkan perbedaan sangat nyata (p<0,01) pada perlakuan P0 dan P1 persentase motilitas 76,11%; 65,56% dan viabilitas 82,33%;72,22%. P0 dan P2 persentase motilitas 76,11%;61,11% dan viabilitas 82,33%;67,78%. Hasil tersebut menunjukkan bahwa penelitian persentase motilitas dan viabilitas spermatozoa kambing Sapera mengalami penurunan dari semen segar pada tris kuning telur dan susu skim kuning telur before freezing.

Abstract

Penelitian ini bertujuan untuk mengetahui efektifitas pemisahan alat Electric Separating Sperm (ESS) terhadap spermatozoa kromosom X dan Y pada sisi anoda dan katoda dengan variabel meliputi presentase viabilitas, motilitas, abnormalitas dan morfometri spermatozoa. Metode pengambilan semen menggunakan vagina buatan, kemudian dilakukan pemeriksaan makroskopis dan mikroskopis, apabila menunjukkan hasil yang baik dilanjutkan dengan pengenceran semen dengan Tris Kuning Telur dan dilakukan pemisahan. Terdapat tiga perlakuan pada penelitian ini yaitu P1 (3 menit), P2 (7 menit), P3 (10 menit) dengan enam kali pengulangan pada sisi anoda dan katoda. Data pada tiap variabel (viabilitas, motilitas dan abnormalitas) diperiksa menggunakan analisis statistik ANOVA dan untuk melihat adanya pemisahan spermatozoa kromosom X dan Y dilakukan pengukuran terhadap kepala menggunakan morfometri. Hasil pemisahan tertinggi yaitu pada P3 dengan perbandingan pemisahan X : Y sejumlah 65,00:35,00 pada sisi katoda, kemudian pada sisi anoda persentase perbandingan X : Y yaitu 59,00:41,00. Hasil kualitas spermatozoa setelah pemisahahn menunjukkan presentase viabilitas terbaik terdapat pada P1 yaitu 75,00a±2,00 pada sisi anoda dan 76,50a±2,17 pada sisi katoda. Motilitas spermatozoa P1 menunjukkan motilitas progresif spermatozoa dengan rataan 60,83a±2,04 pada anoda dan 61,67a±2,58 pada sisi katoda. Presentase abnormalitas tidak menunjukkan perbedaan yang signifikan antar perlakuan.

Abstract

Deteksi birahi dan program perkawinan merupakan cara yang tepat untuk dapat mengoptimalkan performa reproduksi pada sapi perah. Rancangan penelitian yang dilakukan dalam penelitian ini adalah pretest-postest design. Terdiri dari empat macam perlakuan yaitu P1C1, P1C2, P2C1, dan P2C2, masing-masing perlakuan diulang sebanyak empat kali. Faktor yang diujikan adalah faktor P: usia korpus luteum 15 hari (P1) dan 10 hari (P2), faktor C: pencabutan atau pelepasan implan progesterone pada hari ke-9 (C1) dan pada hari ke-13(C2). Hasil yang diperoleh dengan uji statistik menunjukkan bahwa tidak terdapat interaksi antara perbedaan usia korpus luteum dengan perbedaan waktu pencabutan atau pelepasan implan controlled internal drug release (CIDR) (p>0.05). Dari total sampel 16 ekor sapi perah, 13 sampel menunjukkan tanda birahi tiga hari setelah implan controlled internal drug release (CIDR) dilepaskan. Persentase total dari keseluruhan sampel yang menunjukkan tanda birahi adalah 81.25%.

Abstract

The aim of the study was to detect the fertility level of females Bawean deer (Axis kuhlii) through fecal steroid metabolic which were tested using Enzyme-Linked Immunosorbent Assay (ELISA). The study used five females Bawean deer in Taman Flora Surabaya, which were marked using R1, R2, R3, R4, and R5 necklaces. The feces of each deer was collected on the 1st day, 6th day, 11th day, 16th day, and 21st day. The samples which were collected then extracted using the freeze dry method. Samples were tested using ELISA. The results of the measurement of the levels of fecal steroid metabolic  in five Bawean deers showed that three Bawean deer (R1, R4, and R5) were in the luteal or pregnant phase as indicated by the increased of progesterone from 1st day to 21st day. While the other two Bawean deer (R2 and R3) were in the follicular or estrous phase as indicated by the decreased of level of faecal steroid metabolic  on 11th day for deer R2 and 16th day for deer R3. The results showed that the levels of the fecal steroid metabolic can be used to determine the follicular phase and luteal phase and the pregnancy status of female Bawean deer.

Abstract

This study aimed to provide alternative information and solutions in an effort to increase reproductive productivity in etawah crossbreed goats (PE). The sample used in this study was 10 female PE with an average age of 2.5 - 3 years and primiparous at least . Ovulation induction was performed using 0.1 mg intra-muscular Gonadotropin-Releasing Hormone (GnRH), after seven days injection of Prostaglandin F2α (PGF2α) was given as much as 2.5 mg submucosa of the vulva, followed by a second injection of GnRH as much as 0,2 mg intramuscular in samples that have really experienced heat and selected selectively according to the signs of natural heat shown by the sample and followed by insemination as much as 2 doses or 0.50 ml of frozen semen. Blood sampling was performed at H0, H7th, H14th and H21th after insemination. All blood samples were collected and progesterone and estrogen hormone profiles were examined using the ELISA method. From the results of the ELISA test, the mean progesterone hormone profile H0 = 4.798 ng / ml, H7th = 4.887 ng / ml, H14th = 4.824 ng / ml, H21th = 5.148 ng / ml. The profile of the hormone estrogen at H0 = 19,461 pg / ml, H7th = 17,457 pg / ml, H14th = 18,248 pg / ml, H21th = 17,515 pg / ml. This study showed an increase in the levels of the progesterone hormone at H0 to H7th, then slightly decreased in H14th and a significant increase in H21th. For the estrogen hormone, there is a decrease from H0 to H7th then there is a slight increase in H14th and decreases again in H21th.

Abstract

Study on performance, semen quality and testosterone levels of 2 young bulls (age range 3-5 years) Limousine and Simental and 2 old bulls (age range 6-8 years) types Limousin and Simental have been carried out in the Teaching Farm, Faculty of Veterinary Medicine, Universitas Airlangga. This study aimed to determine the performance, semen quality and testosterone levels can be used as a guideline whether or not the ejaculate of young and old bulls should be continued into frozen semen. Performance tests were carried out using measuring sticks and weight bands, semen quality tests included macroscopic and microscopic tests, semen concentration using a spectrophotometer and testosterone test using the Sandwich-ELISA method. The mean results of the performance test in young bulls were, while the mean for older bulls were: body weight 700 kg; hump height 136.5 cm; circumferens of chest 213 cm; body length 171 cm and scrotal circumference 35.5 cm. The performance test for old bulls is body weight 840 kg; hump height 143 cm; circumferens of chest 225.5 cm; body length 177.5 cm and scrotal circumference 40.25 cm. Mean macroscopic of semen examination for young bulls, mean volume 8 cc; milky white color; characteristic odor; The degree of acidity was 6.5 and mucous consistency, while for the old bulls the volume was 6.85; milky white color; characteristic odor; 6.5 degrees of acidity and mucous consistency. The mean microscopic examination for young bulls was mass movement ++ / +++; individual movement 80/3; concentration 745.5 million / ml; abnormalities 3% and 92.25% viability, for old bulls: movement mass ++ / +++; individual movement 77.5 / 3; concentration 1291.5 millon / ml; abnormalities 5% and viability 90%. Examination of serum testosterone levels using the Sandwich-ELISA technique was obtained by an average of 1.299 ± 0.1598 ng/ml in young bulls and 1.101 ± 0.1502 ng/ml in older bulls. This study concluded that young bulls and old bulls in the Teaching Farm, Faculty of Veterinary Medicine, Universitas Airlangga in terms of performance, semen quality and testosterone levels are suitable for processing into high quality frozen semen.

Abstract

This study aimed to evaluate the profile of progesterone in dairy cattle with ovarian hypofunction. A total of 10 cows in this study were evaluated three times in the collection phase, i.e. (F1) when the cow was diagnosed with ovarian hypofunction, (F2) when the cow was in heat and (F3) 21 days after artificial insemination to detect pregnancy. Samples in the form of blood serum were then analyzed using ELISA. As a result, the average time of heat in ovarian hypofunction cows was 7,4 days. Progesterone levels in F1 were 1,027 ng/ml, 2,774 ng/ml, 1,476 ng/ml, 2,256 ng/ml, 1,258 ng/ml, 1,758 ng/ml, 2,393 ng/ml, 0,592 ng/ml, 0,755 ng /ml, 1,876 ng/ml, respectively. Progesterone levels in F2 were 0,671 ng/ml, 0,517 ng/ml, 0,763 ng/ml, 0,598 ng/ml, 0,615 ng/ml, 0,537 ng/ml, 0,726 ng/ml, 0,643 ng/ml, 0,593 ng/ml, 0,975ng/ml, respectively. Progesterone levels in F3 were 15,642 ng/ml, 4,215 ng/ml, 17,327 ng/ml, 20,721 ng/ml, 5,796 ng/ml, 17,214 ng/ml, 15,815 ng/ml, 16,745 ng/ml, 4,632 ng /ml, 18,281 ng/ml, respectively. The pregnancy rate in hypofunctional cows treated with PG-600 in this study was 70%.

Abstract

This study was performed to investigate the effect of calliandra honey (Calliandra sp.) on the number of sertoli cells in malnourished male albino rats. A total of 24 male rats were divided into four groups, i.e. (K) without treatment, (P0) fasted, (P1 and P2) respectively fasted and administered honey concentrations of 30% and 50% for 10 days after fasting. The results showed that administration calliandra honey with a concentration of 30% and 50% significantly increased the number of sertoli cells. It can be concluded that administration calliandra honey can have a positive effect on malnourished male rats, especially in improving the number of sertoli cells.

Abstract

Anti-Müllerian hormone (AMH) has been identified as a potential biomarker for assessing ovarian reserve and reproductive capacity in cattle, with high heritability and relatively stable expression. However, studies detecting AMH mRNA in bovine serum remain scarce due to the matrix’s inherently low RNA yield, susceptibility to degradation, and potential presence of PCR inhibitors. This study provides an exploratory validation of AMH mRNA detection in serum — a challenging matrix compared with ovarian tissue — using EvaGreen-based reverse transcriptase–quantitative PCR (RT–qPCR). Specific primers for the AMH and β-actin genes were designed in silico and validated through melting curve analysis and linearity testing. The results showed high amplification efficiency (AMH: 100.2%, R² = 0.994; β-actin: 109.1%, R² = 0.996), with specific amplification of both targets. AMH detection in serum samples was successful in some samples, while the β-actin gene was consistently amplified as a reference gene. Despite the low RNA quality from serum and the presence of organic contaminants, the method demonstrated its feasibility for detecting AMH transcripts in a minimally invasive manner. Physiologically, AMH levels positively correlate with antral follicle count, superovulation success, and embryo quality, and are sensitive to heat stress and other environmental factors. These findings provide a foundational basis for developing molecular diagnostic approaches based on AMH gene expression in cattle reproductive management programs and support the future development of efficient, accurate, and context-specific biomolecular-assisted selection technologies for tropical livestock systems.
