Abstract

Genotype x environment interaction (GxE) information is needed by plant breeders to assist the identification of superior genotype. Stability analysis can be done if there is a GxE interaction, to show the stability of a genotype when planted in different environments. This study aimed to estimate the effects of genotype x environment interaction on yield and yield components of fruit weight per plant as well as to look at the stability of 14 tomato genotypes at four lowland locations. The study was conducted at four locations, namely Purwakarta, Lombok, Tajur and Leuwikopo. Experiments at each location was arranged in a randomized complete block design with three replications. Stability analysis was performed using the AMMI model. Fruit weight, fruit diameter, number of fruits per plant and total fruit weight per plant characters showed highly significant genotype x environment interactions. Variability due to the effect of GxE interaction based on a AMMI2 contributed by 88.50%. IPBT3, IPBT33, IPBT34, IPBT60 and Intan were stable genotypes under AMMI model.Keywords: AMMI, multilocation trials

Abstract

ABSTRACTThe chili consists of several species, five of which have been cultivated, namely C. annuum, C. chinense, C. frutescens, C. baccatum, and C. pubescens. The classifications of these species are based on: 1) morphological characters, especially floral morphology, 2) crossability between species, and 3) fertile hybrids between species. Species C. baccatum and C. pubescens can be easily identified and distinguished from one another, because there are obvious differences in the two species. However the species C. annuum, C. chinense, and C. frutescens has many common characteristics, so it is difficult to distinguish morphologically. The purpose of this study was to identify capsicum species based on crossability and morphological characters. The crossability experiment was done in housing area TDP 2 Ciampea Bogor from January to December 2013 and the morphological characteristics at Leuwikopo experimental station, IPB. The morphological characters experiment was arranged in randomized complete block design (RCBD) with three replications. Data were collected on successful crosses, seed viability of successful crosses and morphological characters based on descriptor for capsicum IPGRI. The percentage of successful crosses involving IPBC10 and IPBC145 (C. annuum) as female parents and 20 genotypes as male parents ranged between 0-90%; and using IPBC295 (C. frutescens) as female parent was 40%. Genotypes which had successful results equals 40% were alleged as C. frutescens species (IPBC61, IPBC139, IPBC63, IPBC163, IPBC289, IPBC288, IPBC294 and IPBC285). Principal component and of clusters analysis suggests that there are two species groups, i.e. C. annuum and C. frutescens.Keywords: capsicum, cluster analysis, crossing, genotypes, principal component analysis

Abstract

ABSTRACTMolecular genetic analysis of hard bunch phenomenon in oil palm was done in order to elucidate the role of genetic factor underlying hard bunch in oil palm plantation. The aim of this study was to identify the AFLP primer combination that co-segregates with hard bunch phenotype related gene in oil palm. Molecular analysis was done by bulk segregant analysis approach. DNA was isolated from leaves of the normal and hard bunch palm. DNA from ten individual palms from each category were pooled and used as a template. A total of 56 AFLP primer combinations were selected for selection of polymorphic primer, and as a result it was found that 22 AFLP primer combinations (39.28%) were polymorphic. A total of 48 individual of palm DNA containing 24 individual for each group were further genotyped by those 22 polymorphic markers. Of these, one AFLP primer combination (E-ACC/M-CTG) was obtained as a co-segregated marker that distinguished the hard bunch DNA from the normal one. Based on the analysis of the target sequence aligned to the oil palm DNA sequences available in database, we found that our sequence has similarity with Ty-1 copia retrotransposon. This sequence distribute in all 16 linkage group of oil palm genome.Keywords: abnormal fruits, AFLP, oil palm, Ty-1 copia retrotransposon

Abstract

Potassium (K) is required in a large amount for plant growth and production of most fruit crops, including pineapple. However, excess application may decrease its growth and production. Therefore, fertilizer application must be site specifi c, based on soil nutrient status and plant requirements. The aims of the research were (1) to determine the soil K nutritional status of pineapple and (2) to determine the optimum dosage of K fertilization for pineapple grown in Sawah Baru, Darmaga, Bogor. The research was conducted using split plot randomized block design with fi ve soil K status as main plots i.e. 0, 70, 140, 210, and 280 kg K2O ha-1 representing status levels of very low, low, moderate, high, and very high, respectively. The sub plot was dosage of K fertilizer i.e. 0, 200, 400, 600, and 800 kg K2O ha-1. The result of the research showed that plant growth and production of pineapple were improved by soil K nutrient status and dosage of K application. The level of soil K nutrient availability was classifi ed into low (<14 ppm K2O), medium (14-50 ppm K2O), and high (>50 ppm K2O). Potassium fertilizer is recommended for the soil with low soil K status of 634 kg K2O ha-1 .

Abstract

The objective of the research was to identify RAPD markers linked to QTL related to agronomic traits of soybean under low-light intensity condition. The genetic material used in the QTL analysis based on RAPD Markers were Ceneng, Godek, and F6 RILs derived from hybridization between Ceneng (tolerant parent) and Godek (sensitve parent). The results of molecular analysis showed that 9 primers were polymorphic and linked to the tolerant parent. Primers produce 14 RAPD markers which were polymorphic and linked to the tolerant parent. The markers were distributed into a linkage group that containing seven markers. RAPD Markers (OPE15-800, OPM20-800) were linked to two QTL controlling number of productive node and seed weight, respectively. The marker linked to the tolerant parent could be used as a marker assisted selection for high-yielding soybean lines under low-light intensity

Abstract

Characters on hypocotyl dan cotyledon are very potential used for effective and efficient morphology marker for some crop plants. However, the information about the inheritance of qualitative and quantitative characters on hypocotyl and cotyledon of chili pepper was not available. The aim of this research was to determine the inheritance of qualitative and quantitative characters on hypocotyl and cotyledon of chili pepper. This research used purple chili pepper (P1 (IPB C20)), green chili pepper (P2 (IPB C2)), F1, F1R, BCP1, BCP2, and F2 populations. Analysis for qualitative characters used mendelian genetics analysis, whereas model genetic prediction for quantitative characters used join scaling test analysis. The result showed that hypocotyl and cotyledon color of chili pepper controlled by single gene. The gene controlling purple color was dominant than the gene controlling green color on hypocotyl, whereas  the gene controlling green color was dominant than the gene controlling purple color on cotyledon. The additive-dominant genetic model with influence of additive-additive and additive-dominant interaction was suitable for hypocotyl length. The additive-dominant genetic model with influence of additive-dominant and dominant-dominant interaction was suitable for hypocotyls diameter. The additive-dominant genetic model with influence of additive-additive and dominant-dominant interaction was suitable for cotyledon width and cotyledon length.  Keywords: genetic model, morfology marker, natural cross-pollination

Abstract

Leunca (Solanum nigrum L. ) is one of the indigenous vegetables in Indonesia. The fruit of this vegetable is green and has bitter taste because of its tannin content. The objectives of this study were to get lethal dose 50 (LD50), diversity and to obtain candidates of mutant plants with high and low tannin content in young fruit and ripe fruit in leunca. The research was conducted at Pasir Kuda experimental station, Bogor; National Nuclear Energy Agency (BATAN), Jakarta; and postharvest laboratory of the Center for Tropical Horticulture Studies (PKHT IPB), Bogor, from December 2016 to July 2017. Leunca accession of SN 20 and SN 29 were irradiated using doses of 0, 100, 150, 200, and 250 Gy of gamma ray. The LD50 values for SN 20 and SN 29 were 171.944 Gy and 190.949 Gy, respectively. The analysis of tannin content was performed based on permanganate method by titration using potassium permanganate. The results showed that the highest tannin content of green stage fruit was found in SN20D3.5 with 200 Gy dose and the lowest was found in SN29D1.5 with 100 Gy dose. The highest tannin content of ripe fruit was found in SN20D4.6 with dose of 250 Gy and the lowest was found in SN20D3.1 with dose of 200 Gy. Recommended tannin content for leunca is the low tannin content.Keywords: diversity, gamma ray, indigenous vegetable, irradiation, LD50

Abstract

Karakter stay greenness pada sorgum berkaitan dengan kemampuan mempertahankan kehijauan daun dan laju fotosintesis pada kondisi cekaman. Penelitian ini bertujuan untuk memperoleh informasi kendali genetik karakter agronomi, stay greenness serta identifikasi segregan transgresif empat populasi F2 sorgum. Penelitian telah dilaksanakan bulan Februari sampai Mei 2020 di Kebun Percobaan IPB University Leuwikopo, Dramaga, Bogor, dengan ketinggian 250 m di atas permukaan laut (dpl). Bahan genetik yang digunakan adalah populasi Numbu x Samurai 2, Kawali x Pahat, Kawali x B69, Super 2 x PI-150-20-A, dan tujuh genotipe tetuanya. Setiap populasi terdiri atas 300 tanaman F2 dan masing-masing tetua sebanyak 60 tanaman. Pendugaan kendali genetik berdasarkan nilai skewness dan kurtosis. Identifikasi segregan transgresif menggunakan karakter terpilih berdasarkan nilai parameter genetik. Hasil penelitian menunjukkan bahwa karakter bobot biji per tanaman dikendalikan banyak gen aditif pada populasi Numbu x Samurai 2 dan Kawali x Pahat, sedangkan karakter stay greenness pada semua populasi dikendalikan banyak gen aditif. Semua karakter yang diamati memiliki nilai heritabilitas arti luas sedang sampai tinggi. Delapan belas individu dari persilangan Kawali x B69 dan Super 2 x PI-150-20-A diduga sebagai segregan transgresif berdasarkan karakter bobot biji per tanaman dan stay greenness. Segregan yang diperoleh digalurkan dengan menerapkan metode pedigree. Kata kunci: aksi gen, aditif, indeks transgresif, keragaman genetik

Abstract

Pepper Yellow Leaf Curl Virus is a major disease in chili peppers, causing severe damage and yield loss. The use of resistant genotypes is an effective way of controlling the disease. Developing resistance genotypes requires a series of plant breeding processes, starting with the identification of new potential genetic sources. The study aimed to identify the resilience response of 10 cayenne pepper genotypes (C. frustecsens L.) and 4 chili pepper genotypes (C. annuum L.) collection of the Center for Tropical Horticultural Studies (PKHT) to acquire resistant plant candidates. Planting and inoculation were carried out in a greenhouse at Tajur experimental station, and detection of PYLCV was carried out in the molecular laboratory by DNA amplification method using degenerate primers Begomovirus SPG1 and SPG2. The results showed that all plants were infected by PYLCV and it was also confirmed by PCR detection. Based on the disease severity, the CB-EL genotype was categorized as resistant plants with a severity value of 8.89% and the longest incubation period, 21-50 days. Genotypes CB-BA and CB-BJ (13.13%), CB-CA and PKHT-1 (15.56%), CR-SA and PKHT-7 (17.78%) and PKHT-6 (20.00%) were categorized as moderately resistant. Disease severity and disease incidence had a broad-sense heritability values of 0.47 and 0.61, and were categorized as moderate and high heritability, respectively. Abstract

The aim of this research is to study the effects of synthetic cytokinin (6-benzylaminopurine/BAP) treatments on two groups of pineapple (Ananas comosusL.Merr.) plantlets differentiated by plantlet ages. The research used the Factorial Experiment on Randomized Complete Block Design with two factors, which are BAP concentrations (0 ppm, 25 ppm, 50 ppm, and 75 ppm) and plantlet division by age grouping (52 days and 69 days), with three replications. It was then followed by Tukey’s Honestly Significant Difference Test at error level of 5%. The results show that BAP treatments with the concentration used in this research significantly inhibits the growth of pineapple plantlets observed on four variables (number of leaves, leave length, plantlet height, and plantlet diameter), while age grouping treatments didn’t show any significant effects, except on the plantlet height at 8 and 14 weeks after treatment. There are also treatment-related interactions which significantly affects the number of leaves and plantlet height. It was then suggested that lower concentrations of cytokinin are required if similar research is to be conducted in the future.Keywords : pineapple, post-acclimatization, propagation, 6-benzylaminopurine
