Abstract

Penelitian tentang respon cabai rawit terhadap salinitas perlu dilakukan untuk pengembangan varietas yang toleran salinitas. Penelitian ini bertujuan untuk mengetahui respon morfologi dan fisiologi empat genotipe cabai rawit pada kondisi cekaman salinitas. Penelitian dilakukan di kebun percobaan Tajur 2 IPB Bogor pada bulan Oktober 2019-Maret 2020. Rancangan kelompok lengkap teracak dua faktor dengan lima kelompok sebagai ulangan digunakan dalam percobaan ini. Faktor pertama yaitu genotipe cabai rawit (Sigantung, CR10, CSR1, dan CRK1) dan faktor kedua adalah taraf salinitas (0-1, 2-4, 5-7, dan 8-10 mS cm-1). Hasil yang diperoleh menunjukkan bahwa salinitas secara signifikan berpengaruh terhadap peubah morfologi dan fisiologi tanaman cabai rawit. Salinitas 8-10 mS cm-1 pada peubah morfologi menyebabkan penurunan hasil tertinggi pada jumlah bunga (74.65%), jumlah daun (71.23%), bobot buah (58.48%), dan panjang akar (49.81%). Selain itu, pada konsentrasi 8-10 mS cm-1 juga menyebabkan penurunan hasil tertinggi pada peubah fisiologi. Penurunan hasil tertinggi terjadi pada konduktansi stomata (29.37%), laju transpirasi (26.47%), konsentrasi CO2 interseluler (21.83%),dan indeks klorofil (20.19%). Berdasarkan nilai rata-rata indeks sensitivitas cekaman (ISC), genotipe Sigantung (1.2) dan CSR1 (1.0) termasuk katagori peka salinitas (ISC >1.0). Akan tetapi, CR10 dan CRK1 memiliki nilai rata-rata ISC sebesar 0.8, sehingga termasuk ke dalam katagori moderat toleran salinitas (0.5<ISC<0.1). Abstract

A Pineapple (Ananas comosus (L.) Merr.) It is one of the leading commodities in the Indonesian horticultural sub-sector. Based on data from PDSIP in the last 5 years the development of pineapple production has increased but not too high as well as the harvested area. One of the areas that cultivate pineapple plants in Riau Province is Kampar Regency. Its production in 2015 was 8,482 tons, down from 20,046 tons in 2013. However, this amount is not optimal considering the area in Kampar Regency is still large enough for pineapple cultivation. Kampar District has a potential peatland of around 191,363 ha. About half of the area is thin peat, while the rest varies from moderate to deep peat. The success or failure of peatland management for cultivated land is highly dependent on the condition of its characteristics and the mastery and scientific understanding of the character of peat. This shows the need to evaluate the carrying capacity of land-based on its suitability so that it can be used as a guide in wise land-use planning. This study aims to create a fuzzy inference system model with Mamdani method in determining the agroecological suitability of peatlands for pineapple plants, this is due to the target class of land suitability parameters based on FAO provisions, namely S1, S2, S3, and N. Based on the obtained model, decision support systems will be developed for the suitability of peatland agroecology for pineapple plants.

Abstract

Genetic engineering is an alternative technology that can be used to assist breeders for improving the genetic potential of crop variety. Drought tolerant of rice was attempted by applying genetic engineering techniques by introducing gene OsDREB1A using Agrobacterium tumefaciens. The experiment was conducted in 2008-2010 at the laboratory and greenhouse of the Indonesian Center for Agriculture Biotechnology and Genetic Resources and Development, Bogor, using the transformation method. Callus derived from seeds of rice varieties Nipponbare was transformed by inserting the gene OsDREB1A, then regenerated and acclimatized to generate putative transgenic rice plants. The transformation of calli derived from 469 rice seeds of variety Nipponbare produced 103 putative transgenic rice plants. Plant heights of the putative transgenic plants obtained were generally lower than the control plants (88 cm). The numbers of rice panicles of the putative transgenic plants were generally less than the control plants (9 panicle). Production of filled grain of the putative transgenic plants were less than the control plants; three plants did not even produce seeds, namely B14(2)4.1a, D5.1, and D18. The results of the PCR analysis showed that all the putative transgenic rice plants were confirmed to carry the gene OsDREB1A.

Abstract

The aim of this research is to study the effects of synthetic cytokinin (6-benzylaminopurine/BAP) treatments on two groups of pineapple (Ananas comosusL.Merr.) plantlets differentiated by plantlet ages. The research used the Factorial Experiment on Randomized Complete Block Design with two factors, which are BAP concentrations (0 ppm, 25 ppm, 50 ppm, and 75 ppm) and plantlet division by age grouping (52 days and 69 days), with three replications. It was then followed by Tukey’s Honestly Significant Difference Test at error level of 5%. The results show that BAP treatments with the concentration used in this research significantly inhibits the growth of pineapple plantlets observed on four variables (number of leaves, leave length, plantlet height, and plantlet diameter), while age grouping treatments didn’t show any significant effects, except on the plantlet height at 8 and 14 weeks after treatment. There are also treatment-related interactions which significantly affects the number of leaves and plantlet height. It was then suggested that lower concentrations of cytokinin are required if similar research is to be conducted in the future.Keywords : pineapple, post-acclimatization, propagation, 6-benzylaminopurine

Abstract

In supporting the replacement of potato varieties with the varieties that are adaptive to drought, effective and fast methods are needed. One of which is through in vitro screening of drought-tolerant genotypes. The aims of this research were to determine the appropriate concentration of sorbitol as a critical limit in the in vitro screening process of drought-tolerant genotypes, as well as to determine vegetative characteristics that could be used as indicators of in vitro selection and also to obtain in vitro drought-tolerant genotypes. To achieve these objectives, we used single-nodal explants of seven genotypes cultured for six weeks on MS media added with four levels of sorbitol concentration. Of the four treatments, sorbitol level that could distinguish susceptible and tolerant plants was 0.2 M sorbitol and at this concentration, the characteristics that showed significant interactions were plant height and root length, so that both characteristics could be used as selection indicators for the analysis of tolerance tests for the seven genotypes. The results of tolerance test using the analysis of relative decline and stress-susceptibility index (SSI) showed that PKHT4 and PKHT6 were the genotypes that were tolerant in vitro. Abstract

Sebagian besar petani bawang merah di Indonesia menggunakan umbi sebagai bahan perbanyakan meskipun banyak penyakit diketahui ditularkan melalui umbi. Perlakuan air atau udara panas pada organ aktif seperti biji atau umbi adalah metode yang umum digunakan untuk menghasilkan tanaman bebas virus. Penelitian ini dilakukan untuk mendapatkan umbi bawang merah bebas virus melalui perlakuan air panas dikombinasikan dengan kultur jaringan. Deteksi virus sebelum perlakuan menggunakan Dot immuno binding assay (DIBA) untuk memastikan infeksi genus Potyvirus (Onion yellow dwarf virus/OYDV), Carlavirus (Shallot latent virus/SLV dan Garlic common latent virus/GCLV) dari umbi. Penelitian disusun menggunakan rancangan acak lengkap dengan tiga faktor, yaitu kultivar (Sumenep dan Bima Curut), suhu (45 °C, 50 °C, dan suhu kamar), dan waktu pemanasan (15 dan 30 menit). Secara umum, perlakuan air panas yang dikombinasikan dengan kultur jaringan tidak memengaruhi kemampuan tumbuh dan jumlah daun planlet. Berdasarkan deteksi virus menggunakan RT-PCR, suhu 45 °C selama 15 menit mampu mengeliminasi 100% Potyvirus dan Carlavirus pada bawang merah ‘Bima Curut’; sedangkan pada bawang merah ‘Sumenep’ perlakuan air panas pada suhu 50 °C selama 15 menit mampu mengeliminasi 100% Potyvirus dan 33.33% Carlavirus.

Abstract

ABSTRACTCenter for Tropical Horticulture Studies (CTHS) have conducted  melon breeding to meet the need of the expanding melon production  in Indonesia. Four melon genotypes of  IPB Meta 3, IPB Meta 4, IPB Meta 6, IPB Meta 8H exhibit superior performance during selection. Prior to release or to register, these melon genotypes need to be evaluated for their main characteristics. Four potential genotypes  along  with  two  control  varieties  of  Action  434  and Sky  Sweet  were  evaluated  under  a single  factor  Randomized  Complete Block  Design  (RCBD)  with  four  replications.  The  results revelaed  that CTHS  melon  genotypes  exhibited  good  performance.  IPB  Meta  4  has larger  stem diameter and leaf size compared  to  Action 434 and Sky Sweet, subsequently  flesh color  of IPB Meta 3,  IPB  Meta  6,  and  IPB  Meta  8H  are oranges, where  as  both  control  varieties  are  green.  Theseresults indicated that melon genotypes  developed in CTHS have unique characteristics, which could be developed for speciality market.Keywords: fruit quality, melon, morphological characteristics ABSTRAKPusat  Kajian  Hortikultura  Tropika  (PKHT)  telah  melakukan  penelitian pemuliaan  tanaman untuk  mengetahui  kebutuhan  pengembangan  produksi melon  di  Indonesia.  Empat  genotipe  melon yakni  IPB  Meta  3,  IPB  Meta 4, IPB Meta 6, IPB  Meta  8H  menunjukkan  penampilan  baik  pada percobaan sebelumnya. Sebelum  dilepas  atau  didaftarkan,  diperlukan  evaluasi karakteristik  utama dari empat genotipe tersebut. Pengujian keempat genotipe potensial dengan dua varietas pembanding yakni  Action  434  dan  Sky  Sweet disusun  berdasarkan  Rancangan  Kelompok  Lengkap  Teracak (RKLT)  dengan 4  ulangan.  Hasilnya  menunjukkan  bahwa  genotipe  melon  IPB  Meta  4 memiliki diameter batang  dan ukuran daun yang lebih besar dibandingkan Action 434 dan  Sky Sweet. Selain itu,  daging  buah  melon  genotipe  IPB Meta  3,  IPB  Meta  6,  dan  IPB  Meta  8H  berwarna  jingga, dimana kedua varietas pembanding berwarna hijau. Hasil ini menunjukkan bahwa genotipe-genotipe melon yang dikembangkan di PKHT memiliki karakteristik yang unik, dimana dapat berpotensi bagi segmen pasar khusus.Kata kunci: karakteristik morfologi, kualitas buah, melon

Abstract

ABSTRACTHybridization program was started in PKBT IPB Bogor in 2003, entangles of 12 parental cultivars, consisting of five type Smooth Cayenne cultivars and seven type Queen cultivars. The cross yielded 195 genotypes with various different character combinations. The result of cluster analysis based on morphological characters showed that there were 33 groups of hybrid at the degree of genetic similarity of 50%. The result of principal component analysis indicated that, between yield component characters and result most importantly, were fruit weight, fruit diameter and fruit length which were main supporting character of variance in hybrid result of the crosses. Descriptive results of fruit yield and quality characters showed three to five classes with the highest number of individuals around the mean value for each character. Fruit weight, crown weight, fruit length, fruit diameter, flesh thickness, core diameter, total soluble solid (TSS), total acid, vitamin C, pH, plant height, and peduncle length, were characters controlled by nuclear genes.Key words : hybridization, variability, Smooth Cayenne, Queen, genotype

Abstract

ABSTRACTThe objective of the experiment was to investigate the physical and chemical characteristic of eight genotypes of papaya i.e. IPB1, IPB 2A, IPB 3, IPB 3A, IPB 4, IPB 7, IPB 8, and IPB 9 on two stages of ripening period based on percentage of the yellow area of fruit peel (75% yellow and 100% yellow). The fruits were picked at 25% yellow of fruit peel colour. The experiment was conducted in split plot wi th completely randomized block design. The main plot was ripening periods of 75% and 100% ripe, while the genotypes were taken at subplot. There was no significant different on physical and chemical characteristics between papaya at stadium 75% and 100% yellow. Flesh firmness of IPB 9 was better than IPB 1, IPB 4 and IPB 8. Ascorbic acid content of IPB 4 (107.36 mg/100 g) was higher than that of IPB 2A and IPB 3A. Abstract

ABSTRACTIndigenous vegetables have become popular in recent Indonesian diet, but agronomic and economic studies on these crops are limited. The objective of this research was to investigate the cultural technique of indigenous vegetables, their uses and economic importance in West Java, Indonesia. Initial market observation was conducted in Bogor to determine the economic value of indigenous vegetables. In depth observations of the indigenous vegetables and interviews with merchants, farmers and consumers were conducted in three districts, i.e., Bogor, Cianjur and Tasikmalaya, focusing on four indigenous vegetables familiar to local people, i.e., genjer (Limnocharis flava (L.) Buchenau), kenikir (Cosmos caudatus Kunth.), leunca (Solanum americanum Miller) and poh-pohan (Pilea melastomoides (Poir.) Wedd.). This study showed that indigenous vegetables have been produced in extensive and semi-intensive cultivations and are sold in local markets daily, although local people do not consume them frequently. Indigenous vegetables held a market share of less than 5% at local markets, and accounted for less than 10% in household vegetable consumption. The reasons for consumers to choose indigenous vegetables were familiarity to these crops, moderate prices, family members’ preference, availability and ease of preparation. Generally, younger family members (<30 years old) bought indigenous vegetables less frequently than older ones (>30 years old), possibly due to lack of information on its use, unfamiliar flavor and high availability of other commercial vegetables commonly grown worldwide.Keywords: Cosmos caudatus, Limnocharis flava, local knowledge, Pilea melastomoides, Solanum americanumABSTRAKPopularitas sayuran indigenus terus meningkat di Indonesia, namun studi agronomi dan ekonominya masih terbatas. Penelitian bertujuan mengkaji budidaya, pemanfaatan dan nilai ekonomis sayuran indigenus di Jawa Barat, Indonesia. Penelitian pendahuluan dilakukan di pasar di wilayah Bogor. Penelitian mendalam dilakukan dengan mewawancarai pedagang, petani dan konsumen, serta mengamati di lahan petani di tiga kabupaten yakni Bogor, Cianjur dan Tasikmalayauntuk empat sayuran indigenus utama yakni genjer (Limnocharis flava (L.) Buchenau), kenikir (Cosmos caudatus Kunth.), leunca (Solanum americanum Miller) dan poh-pohan (Pilea melastomoides (Poir.) Wedd.). Studi menunjukkan bahwa sayuran indigenus diproduksi secara intensif dan semi intensif, serta dijual di pasar setiap hari, walaupun penduduk setempat tidak mengkonsumsi secara teratur. Sayuran indigenus mengisi celung pasar kurang dari 5% dan memenuhi kurang dari 10% kebutuhan sayuran rumah tangga. Konsumen memilih sayuran indigenus karena sudah terbiasa mengkonsumsi, harga terjangkau, disukai anggota keluarga,ketersediaan dan kemudahan mengolah. Secara umum, anggota keluarga muda (usia < 30 tahun) lebih jarang membeli sayuran indigenus dibanding yang lebih tua (usia > 30 tahun), kemungkinandisebabkan keterbatasan informasi nilai guna, adanya aroma asing, dan ketersediaan sayuran lain.Kata kunci: Cosmos caudatus, Limnocharis flava, pengetahuan lokal, Pilea melastomoides, Solanum americanum
