Abstract

The genetic transformation via pollen-tube pathway in jatropha is the first alternative method that was applied in this plant. The objective of the research was to study fruit set and germination of three genotypes of jatropha following direct transformations via pollen-tube pathways. The research was conducted during April 2014 until January 2015 at jatropa’s experimental field, Pasuruan, and at green house of ICABIOGRAD, Bogor. Three genotypes of jatropha i.e., IP3A, IP3P and JcUMM18 were used. In the first experiment, split plot design was used where 3 levels of DNA plasmid concentration as a main plot and 5 levels of stigma-drip time of DNA plasmid as a subplot. In the second experiment randomized block design was used with single factor consisting of 15 combinations of concentration and stigma-drip time of DNA plasmid as treatments and control. The results demonstrated that interaction between concentration and application time of DNA plasmid did not significantly affect fruit and seeds formation of three Jatropha genotypes. Combination of DNA plasmid concentration with time of stigma-drip had significant effect on seed germination rate of IP3A genotype, but not significant on the other variables. The concentration of 0.05-0.5 µg µL-1 and application time of DNA plasmid at 1-10 hours after pollination could be applied on jatropha genetic transformation via pollen-tube pathways.Keywords: DNA plasmid, Jatropha curcas, pCAMBIA1301, pollen-tube, stigma-drip

Abstract

ABSTRACTThe phenotype is a result of expression from genetic, environment, and their  interaction. Genetic X environment interaction information is important for plant breeders to determine stable genotypes in various environments or stable genotypes in specific environments. The objective of this study was to evaluate morphological and agronomical traits and genetic X environment interaction of IPB sorghum breeding lines. The characterization was conducted at IPB experimental Field, based International Union for The Protection of New Varieties of Plants (UPOV), used 16 IPB sorghum breeding lines and six national varieties. Evaluation of genetic Х environment interaction was conducted in Gowa South Sulawesi and Bogor West Java. This experiment used 16 sorghum lines and two national varieties. The results showed that breeding lines exhibited variation in morphological traits, have medium plant height, small diameter, short panicle length, and medium 1,000 grain weight. There was qualitative genetic X environment interaction on panicle length, days to flowering, 1,000 grain weight, grain filling rate, grain yield, and productivity. UP/N-124-7, UP/N-89-3, UP/N-151-3, UP/N-32-8, UP/N-17-10, UP/N-4-3, UP/N-118-3, and UP/N-118-7 were well adapted in dry soil with dry climate. UP/N-48-2, UP/N-32-8, UP/N-17-10, UP/N-156-8, UP/N-118-3, and UP/N-139-1  were well adapted in dry soil with wet climate.Keywords: characterization, IPB sorgum breeding lines, qualitative genetic X environment interaction

Abstract

The genetic transformation via pollen-tube pathway is an alternative method to overcome the constraints imposed by genotype specificity in transformation and regeneration in jatropha (Jatropha curcas L.) tissue culture. Therefore, it is necessary to establish important parameters for efficient genetic transformation of jatropha via pollen-tube pathway. The objective of the research was to study the efficiency of direct transformation of jatropha via pollen-tube pathway based on histochemical and molecular analysis. Solution of purified pCAMBIA1301 DNA plasmid carrying a hptII marker gene and a gus reporter gene with concentration level of 0.05, 0.25, 0.50 µg µl-1 were applied to stigma of flowers at 1, 2, 4, 7, 10 h after pollination. Seedling of IP3A, IP3P and JcUMM18 jatropha’s genotypes derived from 15 combination treatments of plasmid DNA concentration and application time, also wild type was subjected to histochemical and molecular analyses. Based on those analyses, the efficiency of transformation via pollen-tube pathway of three jatropha genotypes ranged from 1.5-16.7%. PCR analysis showed that a number of positive plants were identified by using specific primers hptII and gus, i.e. 1-3 and 3-7 plants of the 15 combined treatments, respectively. It indicated that the transformation efficiency via the pollen-tube pathway varied in each jatropha genotype.Keywords: Jatropha curcas L., pCAMBIA1301, plasmid DNA, stigma-drip

Abstract

The objective of the research was to identify RAPD markers linked to QTL related to agronomic traits of soybean under low-light intensity condition. The genetic material used in the QTL analysis based on RAPD Markers were Ceneng, Godek, and F6 RILs derived from hybridization between Ceneng (tolerant parent) and Godek (sensitve parent). The results of molecular analysis showed that 9 primers were polymorphic and linked to the tolerant parent. Primers produce 14 RAPD markers which were polymorphic and linked to the tolerant parent. The markers were distributed into a linkage group that containing seven markers. RAPD Markers (OPE15-800, OPM20-800) were linked to two QTL controlling number of productive node and seed weight, respectively. The marker linked to the tolerant parent could be used as a marker assisted selection for high-yielding soybean lines under low-light intensity

Abstract

Water use efficient varieties in rice may have good opportunity in term of economic value and scarcity of water. This study was conducted to determine water consumption among rice genotypes that conventionally cultivated as low land (IR64, IPB97-F-15, Ciherang, Mentik Wangi, and Rokan hybrid), up land (Jatiluhur, Silugonggo), and amphibian type (Way Apo Buru, that could be planted both as lowland and up land). Rice seedlings at 14 days old were transplanted in a plastic container containing 83 kg of air dried soil, 1 plant per whole and 6 plants per container. The experiment was conducted in a vinyl house, using randomized complete block design with three replications. During rice growth, water table was maintained at 2 cm above soil surface, and water was added and recorded weekly. The results showed that varieties reveal production components and yield were different significantly. Water consumptions among varieties were significantly different, ranged from 15.93 L plant-1 for IR64 to 24.13 L plant-1 for Jatiluhur, or equal with 3,639 to 4,827 m3 ha-1. Jatiluhur was the most efficient variety in using water. This finding may guide us to explore benefit of water use efficient variety as sustainable option in water management of rice cultivation.

Abstract

The objective of this research was to determine the trait variance in black soybean lines in terms of yield and yield components. The materials used were F8 lines generated from hybridization of two local varieties, i.e. black seeded Ceneng and greenish yellow seeded Godek, and then selected under single seed descent method, except  SC-39-1, SC-62-2 and GC-74-7 that were generated through bulk selection method. The results showed that the traits between plant genotypes were significantly different, except number of productive node and number of unfilled pod. Total variance was contributed more by number of filled pod, seed weight per plant and 100 seed weight.  Seed weight per plant and 100 seed weight had high heritability with genetic variance and broad-sense heritability which were significantly different from zero. These characters are important for further selection to improve yields of black soybean lines.Keywords: black soybean lines, broad sense heritability, total variance

Abstract

ABSTRACTIndonesia is one of the largest wheat importers. Suitable environmental condition for wheat needs to be studied if wheat is going to be widely cultivated in Indonesia. The adaptability of wheat grown in various climates and altitudes is one of the important aspects. The objective of this experiment was to study the quantitative and morphological character of wheat grown in middle land (540 m asl) in Bogor, West Java. The experiment was arranged in randomized complete block design with three replications. Wheat genotypes used were three national varieties (Nias, Selayar, and Dewata), four new improved varieties (Guri 3 Agritan, Guri 4 Agritan, Guri 5 Agritan, and Guri 6 Unand), and one introduced genotype (SBD). Data were collected for several quantitative variables and seventeen morphological characters based on UPOV descriptor. The result showed that wheat growth was restricted in Bogor. Genotype determined plant height, leaf number, ear length, root length, number of spikelet, harvest time, seed weight, number of tillers, and plant biomass. Based on ear length, grain weight, and plant biomass, Guri 3 Agritan had the highest production than the other genotypes.Keywords: diversity, genetic relationship, high temperature, introduced genotype, phylogenetic

Abstract

The management of P nutrition can be a good solution for aluminum toxicity and P nutrient deficiency in acid soil. This study aimed to determine the role of phosphorus in improving the tolerance of sorghum to Al stress in nutrient culture. This research was conducted at green house of IPB Bogor, tissue culture laboratory in the Department of AGH IPB and Balai Besar Pasca Panen Cimanggu Bogor, from January to November 2014. A completely randomized factorial design was used in three experiments. The results revealed that addition of P improved the tolerance of two sorghum genotypes to Al stress as indicated by the reduction in root length inhibition by Al, where  P was more effective in tolerant genotype Numbu. Addition of P reduced the accumulation of Al in the root tissues as shown by the lighter intensity of hematoxylin staining, especially in Numbu. Aluminum stress increased the secretion of oxalate acid in both sorghum genotypes, where P lowered oxalate acid secretion in both genotypes. These facts showed that the role of P in improving the tolerance of sorghum to Al stress might be associated with the inhibition of Al absorption into the root tissues, although the mechanism is not yet known.Keywords: Al stress, organic acid secretion, role of P, root staining method, sorghum

Abstract

The imbalanced supply-demand of soybean is the main reason of soybean import in Indonesia. One of the efforts to increase of soybean production was expanding the planting area to acid soil with a low pH, high content of Aluminum, and low nutrient availability, through giving ameliorant, such as liming organic materials, and mineral fertilization.This study was aimed to obtain the best combination of fertilizer and amelioran for soybean production in acid soil. The study was conducted in experimental station of Tenjo, Bogor from November 2015 to March 2016. The experiment used a randomized complete block design with two factors. The first factor was combinations of fertilizer and amelioran, i.e., P0 (without fertilization); P1 {25 kg urea + 75 kg SP-36 + 50 kg KCl + dolomite ¼ x Aldd (805 kg) ha-1}; P2 {25 kg urea + 75 kg SP-36 + 50 kg KCl + dolomite ¼ x Aldd (805 kg) + 1,250 kg of manure ha-1}; P3 {50 kg urea + 75 kg SP-36 + 50 kg KCl + dolomite ¼ x Aldd (805 kg) + 2,500 kg of manure ha-1}; P4 {75 kg urea + 150 kg SP-36 + 100 kg KCl + dolomite ¼ x Aldd (805 kg) + 2,500 kg of manure ha-1}; P5 {75 kg urea + 150 kg SP-36 + 100 kg KCl + dolomite ½ x Aldd (1,610 kg) ha-1}; P6 {75 kg urea + 150 kg SP-36 + 100 kg KCl + dolomites 1 x Aldd (3,220 kg) ha-1}. The second factor was soybean varieties of Tanggamus and Anjasmoro. The results showed that application of 75 kg urea + 150 kg SP-36 + 100 kg KCl + dolomite ½ x Aldd (1,610 kg) produced the highest productivity (2.91 ton ha-1), but it was not different from reducing the dose of dolomite into ¼ x Aldd (805 kg) with the addition of 2,500  kg of manure.

Abstract

ABSTRACT Sorghum is one of the potential foods with a higher protein content than rice. Sorghum seeds can be milled and cooked as sorghum rice. This study was aimed to obtain information on the inheritance of morphological and agronomic characters from the population derived from crosses of wide genetic background. The study consisted of the formation of F1 population and evaluation of morphological and agronomic characters in the segregated population (F2) of the three selected F1 genotypes. The location of the research was at IPB Experimental Field, Leuwikopo. The performance of the F1 genotypes were between the two parents, except for the yield components of PI-150-20-A x Numbu and PI-150-20-A x Kawali were higher than the parental lines. The performance of the F2 generation population was also between the parental lines, but with a wider distribution. The sorghum seed color is controlled not only by additive and dominant gene action, but also by epistatic gene action. The character of seed weight per panicle is controlled by epistatic gene action. In the F2 population of the crosses of PI-150-20-A x Numbu and PI-150-20-A x Kawali and PI-10-90-A x Numbu, the seed weight character is inherited with moderate heritability value. Based on genes action dan heritability, bulk or single seed descent method are suitable for yield improvement of the populations.Keywords: gene action, heritability, seed color, segregation, sorghum
