Abstract

This research was conducted to identify some leaf characters related to light capture efficiency. It used split plot design with sub plot nested at the main plot. The main plot was light intensity consisted of two levels: I100 = light intensity 100% and I50 = light intensity 50%, while the sub plot was soybean genotype consisted of G1 = Pangrango, G2 = Ceneng, G3 = Godek, and G4 = Slamet.  Leaf characters measured were : leaf total area, specific leaf area, trifoliate leaf area, leaf hairs (trichome) density, leaf thickness, length of  palisade, chlorophylls content (chlorophyll a, b, and ratio a/b), and the greenness intensity.  The result of research indicated that Ceneng has lower total leaf area, higher specific and trifoliate leaf area, less leaf hair density, thinner leaf, shorter palisade, higher greenness intensity, higher chlorophyll content (a and b), and also lower ratio chlorophyll a/b.   Key words:  Light capture efficiency, light intensity, tolerant genotype, sensitive genotype, soybean

Abstract

Aluminum (Al) toxicity is one of the most important yield-limiting factors for upland rice grown on acid soils. Since many small farmers may have difficulty in soil liming, the genotypes tolerant to soil acidity and aluminum toxicity should be developed. Anther culture can substantially speed up new variety development through recombination of parental characters in early generations and immediately homozygous lines were upon chromosome doubling. The Doubled haploid (DH) rice lines were screened under both nutrient solution containing either 0 or 45 ppm Al and acid soils containing either low or high-Al saturation. The relative root length (RRL) was determined at 14-day-old stage to characterize genotypes for Al-tolerance in nutrient solution. The relative grain weight (RGW) was determined to characterize genotypes for Al-tolerance in soils conditions. The results of this study indicated that Al reduced root elongation. The differential tolerance for Al among genotypes was found to be highly significant for RRL. Of the 120 genotypes tested, 16, 77 and 27 genotypes were found to be Al-tolerance, moderate and sensitive in term of RRL respectively. KRGM4, JTGR13, JTGR17, JTGR18, JTKR1, JTKR5, GRGM4, GRGM6, GRGM9, GRGM14, GRGM25, GRJT11 and SGJT27 lines were consitently Al-tolerance under both nutrient solution and acid soils. The RRL of doubled haploid upland rice lines in nutrient solutions were strongly correlated with RGW in acid soils.   Key words: Rice, doubled haploid, aluminum tolerance, relative root length, relative grain weight

Abstract

The main objective of this study was to identify the leaf morpho-physiological characters as markers for adaptation of soybean to low light intensity. The field experiment was conducted using split plot design with 3 replications. First factor was soybean genotype consisted of shade tolerant genotypes (Ceneng, Pangrango) and sensitive genotypes (Godek, Slamet). Second factor was low light intensity consisted of: L0 = control (under full sun light), L1 = 5 days under 50% shading (exposed after 21 days after planting/DAP), L2 = 5 days under dark condition (exposed after 21 DAP), L3 = 3 days 50% shading + 5 days sun light (exposed after 18 DAP), dan L4 = 3 days 50% shading + 3 days sun light + 5 day dark conditions (exposed after 15 DAP). The 50% shading was made artificially using black plastic paranet with 50% light transmitted. Results of this study showed that under the conditions of low light intensity, leaf morpho-physiological characters of soybean that could be used as markers for adaptation were leaf area, specific leaf weight, and chlorophyll content. Leaves of shade tolerant genotypes were wider and thinner and also contained higher chlorophyll especially for chlorophyll b than that of shade sensitive genotypes under low light intensity.   Key words :  Low light intensity, tolerant genotype, leaf area, chlorophyll, specific leaf weight

Abstract

The objective of this study was to analyze the inheritance pattern of soybean adaptation to shade stress based on leaf morpho-physiological characters. Genetic materials used in this study consisted of 22 plants of low irradiance (LI)-tolerant genotype (Ceneng) and 22 plants of LI-sensitive genotype (Godek); 21 plants of F1 (Ceneng x Godek); and 114 plants of F2 populations (derived from F1). These populations were planted under shading of paranet 50%. The population was arranged in a Randomized Complete Block Design with two replicates. Analysis of inheritance of soybean adaptation involved estimation of heritability (broad sense) and gene action. Results of this study showed that: adaptation of soybean to low light stress based on characters of yield per plant was highly heritable (68% of broad sense) with partial dominant mode of action. Characters of leaf area and specific leaf weight were highly (68% of broad sense) and moderately (48% of broad sense) heritable, respectively, with additive mode of action. Soybean adaptation based on leaf physiological characters (chlorophyll contents) was highly heritable (70% - 86% of broad sense) in epistatic mode of action.   Key words:  Soybean, leaf morpho-physiological character, heritability, gene action

Abstract

Twenty four upland rice cultivars from East Kalimantan and control cultivar were evaluated for tolerance to aluminium using nutrient culture with two treatments of 0 ppm and 45 ppm AI. Result showed that eight cultivars (Pimping, Ketan Hitam, Mayas, Padi Bulu, Popot, Siam Kuning, Ketan Merah, Serai) have PAR > 0.5 and were categorized as tolerant cultivar, the other 16 cultivars were categorized as suceptable. The study was conducted in the greenhouse of the University Farm, Bogor Agricultural University from August to October 2009. The experiment was carried out as a Factorial experiment in a Completely Randomized Design with three replications. The fi rst factor was sorghum genotypes consisted of Numbu (tolerant) and B-75 (sensitive), and the second factor was combination of lime and phosphorous fertilization consisted of: no lime-no P (R1), no lime-low P (R2), no lime-suffi cient P (R3), lime-no P (R4), lime-low P (R5) and lime-suffi cient P (R6). The results showed that all variables were signifi cantly affected by the interaction of media and genotype, except for length of shoot and total P uptake. Under low P and Al toxicity, sorghum variety Numbu showed a higher nutrient effi ciency than B-75 with ability of forming greater dry mass. The sensitive genotypes showed a higher nutrient uptake but a lower P nutrient use efficiency under Al toxicity and P defi ciency. Total P uptake and P nutrient use effi ciency were highly correlated with root growth and dry material accumulation. Abstract

ABSTRAKPenelitian ini bertujuan untuk menganalisis daya gabung tujuh galur jagung manis koleksi PT. BISI International,Tbk di dua lokasi menggunakan persilangan dialel lengkap berdasarkan Griffing metode 1. Penelitian ini menggunakanrancangan kelompok lengkap teracak (RKLT) tiga ulangan, dengan lokasi pengujian di Malang, Jawa Timur dan Magelang,Jawa Tengah. Pengaruh resiprok tidak nyata pada semua karakter, memberikan indikasi bahwa semua karakter dipengaruhioleh gen-gen yang terdapat di inti. Interaksi DGUxL berpengaruh nyata pada karakter hasil panen dan diameter tongkol,memberikan indikasi bahwa galur yang memiliki kemampuan bergabung yang baik di satu lokasi belum tentu memberikanhasil yang sama di lokasi yang lain. Interaksi DGKxL tidak berpengaruh nyata pada semua karakter, sehingga seleksicukup dilakukan di satu lokasi. Pengaruh aksi gen non-aditif lebih besar terhadap pewarisan sifat di semua karakter. GalurDMSG781 (D) merupakan penggabung umum yang baik untuk karakter hasil panen, galur DMSE711 (E) untuk karakterdiameter tongkol serta galur DMST531 (B) dan DMSF11 (G) untuk karakter panjang tongkol. Nilai DGK yang tinggi padaumumnya dihasilkan dari tetua dengan DGU tinggi x DGU rendah kecuali untuk karakter panjang tongkol. Kombinasipersilangan DMST531 (B)xDMSK5 (C) memiliki DGK terbaik untuk karakter hasil panen, DMSC499 (A)xDMSS491 (F)untuk panjang tongkol dan DMSS491 (F)x DMSF11 (G) untuk diameter tongkol.Kata kunci : daya gabung, jagung manis, persilangan dialel

Abstract

The efectiveness of selection depends on the gene action cotrolling the character.  The presence of epistatic gene action will delay the genetic gain in self pollinated crops like soybean. The objective of this research was to obtain information on genetic control of soybean tolerance to aluminum toxicity on nutrient culture. This research was conducted at green house of  the University Farm IPB Cikabayan, Bogor, from April to June 2013. This research used 120 seedlings of F2 population  generated from  crossing between Argomulyo (Al-sensitive) with Tanggamus (Al-tolerant). Seedlings were grown in nutrient culture for 14 days with Al concentration of 1.5 mM and pH 4.0, to observe the vegetative growth under aluminum stress condition. The result showed that mean of roots length, shoot length, shoot to root ratio, wet and dry weight of root and shoot exceeded  both parents mean value with moderate to high heritability. All observed characters had continue distribution pattern, and controlled by many genes. Duplicate epistasis additive gene action controlled only root length while other characters were controlled by complementary epistasis gene action.Keywords: gene action, heritability, nutrient culture, selection, tocixity

Abstract

Ratooning can be used as an alternative cultivation technique for production of sorghum. This study was aimed to evaluate the growth and production of ratoon following the main crop. The study was conducted during August 2013 until November 2014, at Cikabayan Experimental Station, Bogor Agricultural University. The research was arranged in a randomized complete block design with four replications. The results showed that genotypes was significantly affected the vegetative and reproductive characters, physiologycal characters of the main crop, ratooning ability, and vegetative characters of ratoon crop. Plant height, stem diameter, fresh and dry weight, harvest time, and harvest index of main crops had significant correlation with ratooning ability. Plant height, total chlorophyll, fresh and dry weight, panicle weight, grain weight per panicle, 1,000 grain weight, harvest index, and transpiration rate of main crops had significant correlation with ratoon growth. Based on path analysis, the main crop growth directly affected ratoon height, and vegetative characters of ratoon directly affected the yield. The ratoon yielded 48.9-65.7% of the main crop yield. Abstract

The genetic transformation via pollen-tube pathway in jatropha is the first alternative method that was applied in this plant. The objective of the research was to study fruit set and germination of three genotypes of jatropha following direct transformations via pollen-tube pathways. The research was conducted during April 2014 until January 2015 at jatropa’s experimental field, Pasuruan, and at green house of ICABIOGRAD, Bogor. Three genotypes of jatropha i.e., IP3A, IP3P and JcUMM18 were used. In the first experiment, split plot design was used where 3 levels of DNA plasmid concentration as a main plot and 5 levels of stigma-drip time of DNA plasmid as a subplot. In the second experiment randomized block design was used with single factor consisting of 15 combinations of concentration and stigma-drip time of DNA plasmid as treatments and control. The results demonstrated that interaction between concentration and application time of DNA plasmid did not significantly affect fruit and seeds formation of three Jatropha genotypes. Combination of DNA plasmid concentration with time of stigma-drip had significant effect on seed germination rate of IP3A genotype, but not significant on the other variables. The concentration of 0.05-0.5 µg µL-1 and application time of DNA plasmid at 1-10 hours after pollination could be applied on jatropha genetic transformation via pollen-tube pathways.Keywords: DNA plasmid, Jatropha curcas, pCAMBIA1301, pollen-tube, stigma-drip
