Abstract

Antifungi-producing rhizobacteria have been recognized playing an important role in plant disease suppression. In our laboratory, 13 indigenous soybeans' rhizobacteria Pseudomonas sp. that showed strong growth inhibition of root pathogenic fungi, Rhizoctonia solani, Fusarium oxysporum and Sclerotium rolfsii, have been isolated from rhizosphere of soybean plant. For further understanding, the genetic diversity of the antifungi-producing Pseudomonas sp. was investigated using Amplified 16S rDNA Restriction Analysis (ARDRA) and 16S rRNA gene sequences analysis. 16S rDNA were amplified by PCR technique and digested with restriction endonuclease HaeIII, RsaI and AluI. Sequences of 16S rRNA gene were analyzed using the BLAST program for similarity searches on sequence databases. ARDRA based dendrogram analysis was carried out by neighbor-joining of TREECON 1.3b software package. ARDRA indicated the variability of Pseudomonas sp. based on the digestion sites. Dendrogram clustering analysis based on the restriction enzymes profile of the amplified rDNA distinguished Pseudomonas sp. into 7 ribotype groups. The sequences of 16S rRNA gene confirmed that the isolates belonging to Pseudomonas sp. and the phylogenetic tree formed 4 clusters. There was a quite overlap among ARDRA groups and 16S rRNA sequence clusters. This finding suggested that antifungal producing Pseudomonas sp. were present in the rhizosphere of soybean plant and the level of genetic diversity exist within these species. Sequence analysis of the 16S rRNA gene of the Pseudomonas sp. with an identical ARDRA pattern confirmed that members of an ARDRA group were closely related to each other.

Abstract

Fruit rot caused by Botryodiplodia theobromae is one of the most important post harvest disease of mango in Indonesia. Study on biological control on the disease is required to develop environmentally-sound control technology.  The research objectives were to study the potency of yeasts in controlling post harvest mango disease i.e. fruit rot caused by B. theobromae and mechanism involve in the biocontrol. Total yeast isolates used for screening were twenty one, four from collection of Plant Clinic of Institut Pertanian Bogor, and twenty one isolated from healthy mango skin. All of yeast isolates were characterized and identified using BIOLOG. Bioassay on antagonistic activity of yeasts against fruit rot, in-vitro dual culture test and chitinolytic activity were carried out.  Cryptococcus albidus var. aerius WSW1, Pichia guilliermondii K1, and Debaryomyces hansenii K12 were the three most effective antagonistic yeasts against B. theobromae with effectiveness of 70.83%, 45.83%, 37.50% respectively. In vitro bio-assay showed that C. albidus var. aerius WSW1, C. albidus WSW2, C. albidus K6, C. terreus YSW1, Candida edax OSW1, Candida edax K13, and Cryptococcus luteolus K2 had high antibiosis activity.  Biocontrol activity of tested yeasts against fruit rot of mango did not correlate to its antibiosis and chitinolytic activity

Abstract

Identification of plant disease is an important step to decide how it will control. This paper discusses the design and development of an expert system for identification of 12 general diseases of red pepper plant (Capsicum annum L.). Disease identification in this software system was based on the visual symptom of disease(s) in various plant growth stages, similar to the standard rules in plant protection science. The prototype of this software system was implemented by EXSYS Professional for Microsoft Windows (WINEXSYS) version 5.0 and worked under Windows '95 operation system. This software system is user-friendly and equipped by image-visualization that describe disease symptoms.

Abstract

Downy mildew is very detrimental disease of maize production in Indonesia. Adequate information regarding the identification key based on morphological and morphometric characteristic of the causal fungi of maize downy mildew in Indonesia is limited. Study for detection and identification of morphological, morphometric, and moleculer base is urgently required. Artificial sporulation induction method performed to obtain the morphology of the fungus as a whole.The fungi were morphologically identified as symptomatic maize downy mildew collected from 13 provinces in Indonesia.Three species, namely P. maydis, P.sorghi, and P. philippinensis were identified based on the shape,size, and conidial cell wall thickness, size and number of branching coniodiophores, and long sterigmata.Confirmation by PCR succesfully amplified target DNA of P. maydis, P. sorghi, and P. philippinensis. The identification key of third species of Peronosclerospora accurately to be used in identification of Peronosclerospora causes downy mildew of maize in Indonesia.This identification key is recommended as a method of identification of the causal downy mildew of maize in Indonesia.

Abstract

Lasiodiplodia theobromae merupakan cendawan penyebab kanker batang pada tanaman karet (Hevea brasiliensis Muel Arg.), penyakit ini merupakan penyakit baru dan belum pernah dilaporkan sebelumnya di Indonesia.  Banyak cara yang dapat dilakukan untuk mengendalikan penyakit kanker batang. Strategi pengendalian yang terbaik adalah dengan pengelolaan penyakit yang membutuhkan pengetahuan mengenai ekobiologi patogen, inang, dan interaksinya dengan lingkungan biotik dan abiotik. Pengendalian patogen harus didasarkan pada pengetahuan tentang bioekologinya. Diharapkan dengan pengetahuan bioekologi bisa dijadikan dasar untuk bisa menghambat perkembangan atau menurunkan populasi inokulum di lapangan. Penelitian ini bertujuan untuk menentukan pengaruh pH secara in vitro terhadap pertumbuhan patogen kanker batang dan laju penurunan viabilitas tubuh buah akibat penyimpanan.  Kisaran pengaruh pH media meliputi 3, 4, 5, 6 dan 7. Penyimpanan tubuh buah dilakukan dalam wadah tanpa diberi tanah, diberi tanah steril dan tanah tidak steril. Penentuan viabilitas dilakukan pada 30, 60, 90 dan 120 hari masa penyimpanan. Semakin rendah pH menyebabkan pertumbuhan miselia terhambat.

Abstract

Utilization of chitosan to control Bean common mosaic virus (BCMV) on yard long bean. Bean common mosaic virus (BCMV) is one of important viruses infecting yard long bean in Indonesia. One of efforts to control its infection is by utilizing chitosan. Thus, the aim of the conducted research was to test the effectiveness of chitosan in controlling BCMV on yard long bean. Concentration of chitosan tested were 0.1% and 1.0% and it was applied as seed treatment (PB), leaf spraying before (SB) and after (ST) mechanical inoculation of BCMV. Incubation period, disease incidence and severity, peroxidase enzyme activity, and BCMV titer were measured as observation parameters. Incubation period of all treated plants were longer as compared with untreated control plants. Symptoms of treated plants at concentration 0.1% (SB and ST) showed mild to severe symptom and leaf malformation, while at concentration 1.0% showing milder symptoms and some of tested plants remain symptomless. Treated plants showed lower peroxidase enzyme activity and significantly lower BCMV titer as compared with that of untreated control plants. The lowest to highest percentage of virus inhibition exhibited by ST0.1, SB0.1, PB0.1, PB1, SB1 and ST1 treatments with virus relative inhibition level ranged 75.94%-86.07% and severity inhibition level ranged 39.55%-64.63%.

Abstract

The potential of fungi from bamboo rhizosphic soils as endophytic and biocontrol agents of clubroot disease (Plasmodiophora brassicae) on Brocoli. Clubroot, caused by Plasmodiophora brassicae is a serious soilborne disease of plants worldwide, capable of severe infection on broccoli, cabbage and cauliflower. Clubroot is the most destructive diseases on cruciferae in Indonesia. The existing control measures, including biological control do not provide satisfactory result. The objective of the study was to explore fungi from bamboo rhizospheric soils as endophytic that can suppress clubroot disease in broccoli. There were two steps in this study: (1) exploration and identification of fungi from bamboo rhizospheric soils. (2) Effectiveness test of the fungi in suppressing clubroot disease. There were four species of fungi as endophytes in brocoli in this study, e.g Aspergillus sp., Mortierella sp., Paecilomyces sp., and Chaetomium globosum. They had colonized broccoli root endophytically and suppressing clubroot disease. Paecilomyces sp. of endophytic fungi can suppress clubroot diseases incidence for 18.75%.

Abstract

This research was conducted to evaluate the effectiveness of crab shell powder and chitosan to control Fusarium wilt disease on bananas and to analyze the involved control mechanisms. The effectiveness of crab shell powder and chitosan to F. oxysporum f.sp. cubense was examined in a laboratory (in vitro) and greenhouse (in planta). In vitro evaluation showed that chitosan has an antifungal effect while crab shell powder did not. Application of crab shell powder and chitosan suppressed the disease in green house test. The concentrations of crab shell powder and chitosan that most effective to control Fusarium wilt incidences were 0.25% and 0.10% with an efficacy rate of 66.7% and 83.3%, respectively. The highest disease severity reduction was showed by crab shell powder 0.25% and chitosan 0.50% with an efficacy rate of 56.8% and 59.4%, respectively. Suppression of the disease might be due to the fungicidal effect of chitosan and the increase of the total population of bacteria and chitinolytic bacteria in the rhizosphere when banana seedling roots were treated with crab shell powder or chitosan. Experiment results using the split roots technique exhibited the role of crab shell powder and chitosan potentially to induce the resistance of banana to Fusarium wilt.

Abstract

Vascular Streak Dieback (VSD) in cocoa caused by Oncobasidium theobromae. This is one of the important disease of cocoa in the world, including Indonesia. The knowledge on environmental effect and agronomical practices to the epidemics of VSD disease of cocoa, is important to control the diseases. The objectives of this research were to investigate environmental effect and agronomical practices to the epidemic VSD disease of cacao. The study was conducted on February to August 2014 in the three sub districts namely Tinondo (≥ 700 m asl), Mowewe (300-400 m asl) and Lambandia (≤ 100 m asl) East Kolaka Regency, South East Sulawesi. The study was carried out by colleting primary data included agronomical practices, the measurement of VSD disease severity in the field and soil nutrient, as well as secondary data. The results of this research showed that the environment and agronomical practices gave an effect to the epidemics VSD disease. Monoculture plantation had higher severity than mixed garden of cocoa. More condense the canopy of cocoa, more severe the VSD disease. More near the location of cacao plants to the river, more humid the enviroment and more severe the desease. Potassium deficiency in plants will cause susceptible to the disease, the high levels of Mg may limit K and Zn uptake at the soil. Recommendation for reducing VSD disease severity on cocoa plantations are cropping systems mixed with other plants, prunning, reducing the use of insecticides and herbicides, and do not plant the cocoa near to the river.Keywords: cacao, disease, epidemic, Oncobasidium theobromae
