Abstract

Produksi tomat di Indonesia banyak menghadapi gangguan penyakit, baik biotik maupun abiotik, diantaranya penyakit bercak daun yang disebabkan Alternaria solani dan blossom end rot yang disebabkan defisiensi kalsium. Penelitian ini bertujuan untuk menguji kombinasi teknik perlakuan konsorsium plant growth promoting rhizobacteria (PGPR) dengan Trichoderma hamatum THSW13 dalam mengendalikan penyakit bercak daun dan blossom end rot serta pengaruhnya terhadap pertumbuhan dan produktivitas tanaman tomat di lapangan. PGPR yang diuji adalah Pseudomonas fluorescens PF1 dan Bacillus polymixa strain BG25.Percobaan telah dilakukan di pertanaman tomat Kabupaten Tegal, Jawa Tengah dengan perlakuan: (A) perlakuan benih dan di persemaian, (B) perlakuan benih, perlakuan di persemaian dan perlakuan pada tanaman di lapangan, (C) perlakuan fungisida kimia sintetik sebagai kontrol. Percobaan dilaksanakan dengan metode eksperimental dan disusun dalam rancangan acak kelompok dengan pengamatan terhadap penyakit tanaman dan agronomis. Hasil uji menunjukkan bahwa perlakuan teknik aplikasi konsorsium PGPR dan T. hamatum THSW13 dengan perlakuan benih, di persemaian dan pada tanaman di lapangan menunjukkan kemampuan lebih baik dalam menekan tingkat keparahan penyakit bercak coklat dan persentase kejadian blossom end rot serta meningkatkan tinggi tanaman, diameter batang dan produktivitas per satuan luas dibandingkan perlakuan hanya pada benih dan di persemaian.

Abstract

Mangga merupakan salah satu komoditas hortikultura yang sudah mampumenembus pangsa pasar internasional dengan negara tujuan Timur Tengah, Hongkong,Singapura, Malaysia, dan Brunei Darussalam. Selama proses pemasaran, beberapaeksportir mengalami beberapa permasalahan diantaranya rendahnya mutu visual buahmangga yang dipasarkan jika dibandingkan dengan negara maju. Tujuan penelitian iniadalah untuk melihat pengaruh pencucian dan penyimpanan terhadap kandunganpadatan terlarut (PTT), asam tertitrasi total (ATT), rasa dan aroma buah mangga Gedong.Rancangan penelitian yang digunakan adalah rancangan acak lengkap dengan pola splitplot, terdiri dari 2 faktor perlakuan yaitu aplikasi bahan pencuci, [Tanpa dicuci; BahanPencuci (Detergen 1% + CaO 0.5%); Bahan Pencuci + Fungisida 0.025%; Bahan Pencuci+ Khamir] dan suhu penyimpanan (12 °C, 15 °C, 18 °C dan suhu ruang). Hasil penelitianmenunjukkan bahwa suhu berpengaruh terhadap peningkatan jumlah padatan terlaruttotal, penurunan asam tertitrasi total serta penerimaan konsumen terhadap rasa danaroma.Kata kunci: pascapanen, pencucian, penyimpanan

Abstract

Endophytic fungi are fungi that live in healthy plant tissues without causing disease symptoms. Endophytic fungi in chili have been tested as both biocontrol agents and growth promoters, but their colonization has not been reported. Therefore, this study aimed to determine the level of colonization of endophytic fungi and its effect on the growth of chili seedlings. A total of 8 endophytic fungi were prepared at a density of 2.8 × 106 CFU mL-1. Then the endophytic fungus was inoculated 2 times, first by soaking the seeds, and secondly by watering the endophytic fungus suspension on chili seedlings aged 3 weeks after sowing. Endophytic fungi were re-isolated on chili seedlings that were 4 weeks old after sowing on the roots and stems to determine their colonization ability. Chili seeds were then maintained for up to 4 weeks after transplanting to observe their growth. The results showed that the endophytic fungal colonization ranged from 26-60% on the chili root, while at the base of the stem it was 20-40% with a different pattern of colonization distribution. In addition, endophytic fungus colonization was also able to increase the shoot height and root length of chili seedlings.

Abstract

Teknologi mikroba intensif merupakan paket integrasi berbagai agens hayati dalam budidaya cabai yang diketahui efektif dalam menekan berbagai penyakit cabai.  Pengaruh teknik tersebut terhadap dua hama utama cabai yaitu lalat buah (Bactrocera sp) dan thrips (Thrips sp) belum diketahui. Tujuan penelitian ini adalah untuk meneliti pengaruh teknik mikroba intensif terhadap tingkat serangan lalat buah dan thrips pada cabai.  Penelitian terdiri dari empat perlakuan yaitu mikroba intensif penuh, mikroba intensif di pesemaian, konvensional dan tanpa pengendalian.  Penelitian dilakukan di tiga lokasi dengan ketinggian tempat yang berbeda. Pengamatan tingkat serangan lalat buah dilakukan pada umur 10 dan 11 minggu setelah tanam, sementara pengamatan terhadap kerusakan thrips dilakukan seminggu sekali mulai 2 hingga 10 minggu setelah tanam.  Perlakuan mikroba intensif secara nyata mengurangi tingkat serangan lalat buah.  Perlakuan mikroba intensif mengurangi serangan thrips pada dua lokasi tanam yaitu Margasari dan Bojong, namun tidak berpengaruh nyata pada satu lokasi lainnya yaitu Bumijawa.

Abstract

Dieback disease caused by Lasiodiplodia theobromae is an important disease on the nutmeg tree in Aceh Selatan. The disease has been widespread in the district. One alternative to control the disease is the use of antagonistic fungi from the nutmeg tree to inhibit the development of the cause of the disease. In vitro experiments were carried out by evaluating 4 types of antagonistic fungi from the nutmeg tree to L. theobromae. The aim of the study was to determine the potential of in situ soil fungus as an antagonistic agents against L. theobromae, the cause of dieback disease on the nutmeg tree in Aceh Selatan. Microbial isolation using serial dilution method. The fungus test which was successfully isolated as an antagonist candidate was carried out through the dual culture method. The results showed that four fungi namely; Trichoderma harzianum, T. virens, T. asperellum, and Talaromyces pinophilus have the potential as antagonistic agents against pathogens cause dieback disease on the nutmeg tree in Aceh Selatan, with inhibitory power ranging from 66-100% in vitro.

Abstract

Application of biocontrol agents on pre- and post-harvest commodities is a promising disease control strategy. This research was objected to study the potencies and antagonism mechanisms of eight isolates of yeast-like fungus Pseudozyma, i.e. P. hubeiensis Dmg 18 BEP, Dmg 20 DEP, Dmg 23 DEP, Dmg 27 BEP, and Dmg 32 DEP, P. shanxiensis Dmg 28 DEP, dan P. aphidis SG 25 BE and SG 53 BE in controlling Colletotrichum acutatum, the cause of anthracnose on chili. Those isolates of Pseudozyma were obtain from chili leaf and fruit surfaces and tissues. To study their potencies in controlling anthracnose on chili caused by C. acutatum, the eight Pseudozyma isolates were applied on chili fruit. While the study of the antagonism mechanisms was performed by antibiosis, volatile compound formation, chitinolitic activities, hyperparasitism, and ACC deaminase production tests. The results showed that all isolates of the Pseudozyma had the potencies to control anthracnose on chili caused by C. acutatum with more than 60% control levels. The mechanisms were production of volatile compounds, chitinolitic activities, and hyperparasitism. Those Pseudozyma isolates did not produce ACC deaminase.

Abstract

Cucumber mosaic virus strain soybean (CMV-S) is one of economically important virus infecting soybean in Indonesia. However, it is very few information related with the CMV-S Indonesia isolates. Thus, the aim of present work is to detect and identify CMV-S isolates from different origin. Leaf samples collected from six different soybean fields in Java and North Sumatera. Molecular detection was carried out by reverse transcription polymerase chain reaction (RT-PCR) using specific primer of the coat protein (CP) gene and DNA sequencing. RT-PCR were successfully amplified the entire CP gene size ~657 bp from all samples and encoded 218 amino acids. Analysis of nucleotide and amino acid sequences showed high homology among isolates ranging from 99.5% and 100%. However isolate from Sindang Laut either nucleotides or amino acids showed lower homology to that of five isolates ranging from 89.4%-89.6% and 91.7%, respectively. The differences between CMV-S with other non legume strain found in six amino acid positions in the CP gene, suggesting the differences might related with host specificity. Phylogenetic analysis of amino acid of six isolates to other CMV strain showed that the five CMV-S isolates were in the same cluster, while Sindang Laut isolate was in different cluster together with the non-legume CMV from India. It is indicating the present of genetic variability among CMV-S isolates in Java.

Abstract

Climate change affects the agricultural sector, which has an impact on droughts or floods. Ecological agriculture is expected to be an adaptation method for reducing losses due to climate change. This study aims to describe the characteristics of farmers, ecological rice cultivation practices, and the relationship between the two parameters. The research in Kendayakan Village, Terisi District, Indramayu Regency, involved 60 farmers surveyed using accidental sampling techniques in February 2022. The data collected include farmer characteristics (age, education, land area, land ownership status), length of farming, sources of farming knowledge, participation in groups, and rice cultivation practices. Data on the application level of ecological agricultural practices were analyzed descriptively and tested usin the Spearman Rank correlation between farmer characteristic variables and rice cultivation practices. The results showed that farmers were of productive age, with a low level of formal education but experienced and active in local organizations. The level of application of farmers' ecological agriculture is moderate. The return of straw, tillage, and control of plant pests are the components of cultivation that are least following the principles of ecological agriculture. Internal factors of farmers that correlate with the components of ecological rice cultivation are age, education, length of farming, source of farming knowledge, and land ownership status. Abstract

Study on resistance mechanism to vascular-streak dieback (VSD) disease in cacao (Theobroma cacao L.) is limited due to the lack of fungal spores for artificial inoculation. This research was conducted to study the production of secondary metabolites that appear to be evidence of defense signaling in resistant clone of Sca 6 and susceptible clone of TSH 858 to Ceratobasidium theobromae natural infection. A fungal staining method was employed to detect C. theobromae hyphae at early infection stages, before VSD symptoms appear. Metabolite profiling was analyzed using pyrolysis gas chromatography mass spectrometry (Py-GCMS) at pre-, early and late stages of C. theobromae infection. Histochemical and anatomical characteristics of both healthy and infected leaves were also observed to identify the accumulation sites of secondary metabolites on and in cocoa leaf tissues. The results confirmed that fungal staining using trypan blue can detect early stages of C. theobromae infection; at the 14th week (on susceptible seedlings) and the 18th week (on resistant clones), following placement of the seedlings under infected cacao plants. Phenylpropanoid biosynthesis, terpenoid biosynthesis, environmental information processing signal transduction pathways, and aromatic biodegradation were detected as important metabolite pathways during defense mechanism. I-limonene (terpenoid), p-ethylguaiacol (phenols) and 2.3 dihidrobenzofuran (heterocyclic compounds) were proposed as an active defense produced by the host after infected by pathogen mainly on late infection of C. theobromae. Terpenoid and phenol compounds were accumulated on the glandular trichomes, idioblast of upper and bottom epidermis, phloem vessel and cortex idioblast of cacao leaves. Epidermis thickness of resistant clone was significantly greater than that of susceptible clone on both surfaces. Leaf epidermis tissue and the accumulated compounds in epidermis idioblast may act as the physical and biochemical markers of cocoa resistance to VSD.

Abstract

This study aimed to study the profile of starch content in cocoa leaf and phytoalexin production based on GC-MS analysis at several stages of VSD pathogen infection. Research was conducted on January – October 2015 at Kaliwining Experimental Field, Indonesian Coffee and Cocoa Research Institute, Jember, East Java. The research was designed based on a Completely Randomized Block Design with two factors with three replications. The first factor was clone, i.e. the resistant clone (Scavina 6) and susceptible (TSH 858) to VSD infection. The second factor was the level of O. theobromae infection, i.e. pre-infection, early infection, and late infection. Starch catabolism revealed during Vascular Streak Dieback infections in Cacao. Starch content in Sca 6 (resistant clone) in late infection decreased 24,33 % than healthy condition (no infection), however, starch content in TSH 858 (succeptible clone) in late infection decreased only 9,63 % than healthy condition (no infection). This indicated that starch catabolism rate on resistant clone was higher than susceptible clone. Some secondary metabolites releases under Vascular Streak Dieback i.e. I-limonene, eugenol and coumaran. Scavina 6 (resistant clone) had higher concentration of eugenol and coumaran than TSH 858 (susceptible clone). I-limonene compound, TSH 858 (Susceptible clone) had higher concentration than Scavina 6. I-Limonene concentration increased in lined with the severity of pathogen infection. There were an negative correlation between starch content with contentration of I-limonene (R= - 0,74), concentration of Eugenol (R= - 0,44), and contentration of Coumaran.
