Abstract

The objectives of research were to obtain basic information on: 1) pesticide use by Tegal farmers, 2) knowledge, attitude and practices of farmers on pesticide, 3) the relation between knowledge, attitude and practices of farmers on pesticide and intoxication level of farmers. Knowledge-attitude- practice (KAP) survey was conducted on 90 farmers (30 chili farmers, 30 shallot farmers, 30 rice farmers) of Tegal District with structured questionnaire. The information collected included private data, type and number of pesticide uses, knowledge on pesticide, practices and attitude in pesticide uses, and symptom rose after pesticide application. The locations of survey were in Sub-district Surodadi for farmers rice, Dukuhturi for shallot farmers and Bumijawa for chili farmers. The data was grouped and calculated for percentage. Further analysis was conducted by cross tabulation and continued by χ2 test for independence of variables. Five most frequently used pesticides by farmers in Tegal were pesticides with active ingredient chlorpyrifos, profenofos, alphamethrin, deltamethrin and propineb, respectively. Knowledge, attitude and practices of most Tegal farmers on pesticide management is still low and not in line with Integrated Pests Management principles. Moreover, KAP level from the best to the worst was chili farmers, paddy farmers and shallot farmers respectively. Intoxication level based on self report, from the most severe to the least was shallot farmers, chili farmers and paddy farmers respectively. There was significant relation of knowledge and practices of farmers and pesticides intoxication level.

Abstract

Pengembangan durian secara intensif dimulai dengan upaya penyediaan bibit berkualitas. Salah satu kendala dalam penyediaan bibit durian berkualitas adalah serangan organisme pengganggu tanaman. Penelitian ini bertujuan mempelajari teknik pembibitan dan menginventarisasi keberadaan organisme pengganggu bibit durian Menoreh Kuning di Kecamatan Kalibawang, Kulon Progo. Wawancara dilakukan untuk mengetahui teknik budidaya bibit durian Menoreh Kuning. Pengamatan hama dan penyakit dilakukan pada bibit durian dengan tujuh umur berbeda (3, 12, 24, 48, 96, 6 double rootstock dan 96 minggu double rootstock). Bibit diperbanyak dengan teknik okulasi menggunakan batang atas durian Menoreh Kuning dengan satu dan dua batang bawah. Hama yang ditemukan adalah Allocaridara sp., Xyleborus sp., Coptotermes sp., Tetranychus sp., Atractomorpha sp., dan Valanga sp., sedangkan penyakit yang ditemukan adalah bercak daun Corynespora sp., hawar daun Rhizoctonia sp., antraknosa Colletotrichum sp., alga Cephaleuros sp., embun hitam Meliola sp., mati pucuk Phytophthora sp. dan layu Phytophthora sp.. Informasi mengenai hama dan penyakit ini dapat digunakan sebagai langkah awal dalam mencegah timbulnya OPT di pembibitan durian.

Abstract

Penyakit layu fusarium yang disebabkan oleh Fusarium oxysporum merupakan salah satu penyakit penting pada bawang merah (Allium ascalonicum). Sebagian besar petani menggunakan umbi sebagai benih dan diketahui bahwa beberapa patogen dapat terbawa oleh benih seperti Fusarium oxysporum. Oleh karena itu diperlukan pengujian kesehatan benih untuk mencegah penyebaran penyakit tersebut. Salah satu metode sederhana dan efektif untuk digunakan di laboratorium adalah blotter test. Namun beberapa strain F. oxysporum terbukti tidak bersifat patogenik, serta tidak dapat dibedakan secara morfologi dengan strain yang bersifat patogenik. Penelitian ini bertujuan menentukan parameter uji dan jumlah sampel minimal dalam deteksi Fusarium oxysporum pada umbi bawang merah dengan metode blotter test. Hasil pengujian menunjukkan bahwa parameter nekrosis pada basal plate umbi bawang merah mempunyai koefisien korelasi (r) sebesar 0,77 terhadap tingkat infeksi pada growing on test (GOT) dan lebih besar dari tingkat infeksi fusarium (0,34) pada blotter test. Dari 195 isolat Fusarium spp. yang diuji, diketahui bahwa sebagian besar isolat bersifat nonpatogenik. Penentuan jumlah minimal umbi dengan plot kurva rerata jumlah nekrosis pada basal plate dan standar deviasi, menunjukkan jumlah sampel umbi minimal untuk blotter test adalah 150 umbi. Perhitungan jumlah sampel dengan formal probability statement yang menunjukkan jumlah umbi minimal untuk blotter test adalah 138 umbi.

Abstract

Antraknosa merupakan penyakit penting pada tanaman cabai yang dapat menyebabkan kerugian ekonomi cukup besar. Khamir merupakan salah satu mikroba yang telah diketahui berpotensi sebagai agens antagonis pada berbagai produk pascapanen. Penelitian ini bertujuan untuk mendapatkan isolat-isolat khamir epifit yang berpotensi sebagai agens antagonis penyakit antraknosa pada cabai. Penelitian dilaksanakan di Laboratorium Mikologi Tumbuhan dan Kebun Percobaan Institut Pertanian Bogor, dari bulan April sampai Desember 2013. Khamir epifit diisolasi dari daun dan buah cabai merah yang diperoleh dari pertanaman cabai di Rancabango dan Panjiwangi (Kabupaten Garut) dan Dramaga (Kabupaten Bogor). Patogen penyebab antraknosa yaitu Colletotrichum acutatum diisolasi dari buah cabai bergejala dari pertanaman cabai di Panjiwangi. Khamir hasil isolasi diuji patogenisitasnya pada benih dan buah cabai. Khamir nonpatogenik diseleksi potensi antagonismenya terhadap  penyebab penyakit antraknosa. Diperoleh 43 isolat khamir epifit, semua isolat bersifat nonpatogenik berdasarkan hasil uji patogenisitas. Seleksi potensi antagonisme isolat khamir epifit menghasilkan 23 isolat yang berpotensi sebagai agens antagonis C. acutatum. Empat belas isolat  khamir epifit menyebabkan penghambatan penyakit antraknosa lebih besar dibandingkan mankozeb.

Abstract

Yellow leaf curl disease of chili pepper caused by Pepper yellow leaf curl virus (PYLCV) has been reported as an important disease in Java and Bali. Disease severity reached 80−100% and it may cause significant yield losses. In order to reduce the negative impact of synthetic insecticides, silica application was evaluated for its potency to suppress the disease. A greenhouse experiment was conducted using randomly block design with 2 factors: PYLCV isolate (Java and Bali) and silica (SiO2) treatment (with and without). Parameters observed were disease symptoms, incubation period, disease incidence and severity, and total silica level. The symptoms of virus infection in Pelita 8 and Seret cultivars were yellow mosaic, leaf curl, green mosaic, dwarf, and cupping upward or downward. A Specific DNA fragment of 912 bp was successfully amplified from 4 samples. Four sequences were obtained and further analysis showed their highest homology, i.e. 96% and 97% with Pepper yellow leaf curl Indonesia virus-Java (PYLCIV-Java) (JX416180) and PYLCIV-KrthAl (LC381274), respectively. Infection by different virus isolates did not affect disease severity significantly. The application of silica was able to delay symptom development and to suppress the severity of the disease in the range of 16.67−30.33%. Silica application on the soil increased the total content of silica in the plants. However, a further experiment is required to understand the mode of action of silica in inducing plant resistance to the pathogen.

Abstract

Epiphytic yeasts have the potency as antagonistic agents against various pathogens of post-harvest products. Anthracnose is a major disease of chilli that causes high economic loss. This research was objected to study the antagonism mechanism of epiphytic yeast isolates that have the antagonistic potency against anthracnose pathogen on chilli (Colletotrichum acutatum). Twenty-two isolates of epiphytic yeasts, isolated from chilli leaves and fruit, were tested. The characterization of the antagonism was carried out by antibiosis, anti-fungus volatile production, and chitinolytic activity tests. The results showed that all tested isolates did not have antibiosis mechanism against C. acutatum. All isolates produced volatile compounds which inhibited the colony growth of C. acutatum. Four isolates showed high relative inhibition rate, i.e. isolates B32DEP (35.68%), B30DEP (37.52%), B23DEP (38.52%), and B29DEP (45.42%). Fourteen isolates showed chitinolytic activities. Three of them had high chitinolytic activities, i.e. B12DEP, B2DEP, and G237DEP.

Abstract

A survey on several cucumber cultivation areas in West Java, Central Java, Yogyakarta, and East Java found many plants showing typical Begomovirus symptoms such as yellow mosaic, cupping, and vein banding. This study was aimed to determine disease frequency, detection and molecular characterization of the causal virus of those symptoms on cucumber in Java. Sampling was conducted by purposive sampling by collecting 50 symptomatic plants from each location in West Java (Indramayu, Subang, and Bogor), Central Java (Brebes and Klaten), Yogyakarta (Kulon Progo), and East Java (Nganjuk, Kediri, and Tulungagung). The detection and disease frequency was determined based on DIBA test using a specific antiserum of Tomato leaf curl New Delhi virus (ToLCNDV) and Squash leaf curl virus (SLCV). The identification of nucleic acid was conducted by PCR using specific primer of ToLCNDV and SLCV, DNA cloning, and sequencing. The results of serological detection showed the disease frequency of ToLCNDV and SLCV ranged from 92.77-100% and 78.33-93.3%, respectively. PCR using specific primer of ToLCNDV successfully amplified the coat protein gene at a size of 600 bp from all samples. Homology nucleotide and amino acid sequences among ToLCNDV Java isolate ranging from 95.6-99.2% and 99.7-100%. ToLCNDV isolates Java had highest nucleotide and amino acid sequences similarity with cucumber isolate from Klaten, Indonesia (AB613825) ranging from 96.1-98.1% and 99.7-100%, and was considered as “Indonesia” strain. SLCV not amplified on all samples by PCR using specific primer, indicating it might not present yet on cucumber in Java.

Abstract

Rhodotorula sp. is widely known as a biocontrol agent and is reported effective in controlling several diseases on crops. Rhodotorula minuta Dmg 16 BEP is an antagonist yeast from Indonesia which was effective in controlling various plant diseases. The objective of this research was to study the in vitro mode of action of R. minuta Dmg 16 BEP against Alternaria solani. The antibiosis ability of R. minuta Dmg 16 BEP to A. solani might be based on the activity of volatile compounds and not from its metabolites products. The growth of A. solani in paper disk assay and dual culture tests were not inhibited nor inhibitory zones  developed, whereas the inverse petridish test showed the growth inhibition of A. solani. The results of the chitinolytic activity test on chitin agar and proteolytic on skim milk agar showed that no clear zone was formed. R. minuta has a strong hyperparasitic ability according to the slide culture test as the yeast cells surrounded A. solani hyphae and caused damage to some parts of hyphae.

Abstract

The observation conducted from 2000 to 2018 found that rice sheath rot has become an emerging disease in several regions. The disease was able to reduce quality and quantity of rice yield. This research objective was to study the history and status of rice sheath rot caused by Sarocladium oryzae. Data was obtained from literature study, farmer interview, and field survey. Data were used to calculate disease incidence followed by visual observation of the symptoms caused by this disease. Identification of sheath rot pathogen was done using microscopic and molecular techniques using specific primers. Rice sheath rot was first reported in Indonesia as a minor disease in 1987 and has only currently become an important disease. This fungus generally causes rice sheath rot in Karawang and Cianjur Regency. Disease incidence of rice sheath rot was 12.56% without considering the varieties at all generative growth stages. Average disease incidence on all rice varieties observed was 12.64%, except for IR 42 (0%). The level of rice plants damage due to rice sheath rot in Cianjur was higher than Karawang with average disease incidence of 11.58% and 9.27%, respectively. Rice sheath rot symptoms have often been found in the last 10 years with average level of damage of 3%‒18% and yield loss of 10%-15%. This disease has proven to be important and has become a challenge in rice cultivation. The fungus that infect rice plants in Karawang and Cianjur had different morphotypes, namely KP, KP2, KP3, PW3, and PW03. The amplification results showed that all fungal isolates were S. oryzae.

Abstract

Vascular Streak Dieback (VSD) in cocoa caused by Oncobasidium theobromae. This is one of the important disease of cocoa in the world, including Indonesia. The knowledge on environmental effect and agronomical practices to the epidemics of VSD disease of cocoa, is important to control the diseases. The objectives of this research were to investigate environmental effect and agronomical practices to the epidemic VSD disease of cacao. The study was conducted on February to August 2014 in the three sub districts namely Tinondo (≥ 700 m asl), Mowewe (300-400 m asl) and Lambandia (≤ 100 m asl) East Kolaka Regency, South East Sulawesi. The study was carried out by colleting primary data included agronomical practices, the measurement of VSD disease severity in the field and soil nutrient, as well as secondary data. The results of this research showed that the environment and agronomical practices gave an effect to the epidemics VSD disease. Monoculture plantation had higher severity than mixed garden of cocoa. More condense the canopy of cocoa, more severe the VSD disease. More near the location of cacao plants to the river, more humid the enviroment and more severe the desease. Potassium deficiency in plants will cause susceptible to the disease, the high levels of Mg may limit K and Zn uptake at the soil. Recommendation for reducing VSD disease severity on cocoa plantations are cropping systems mixed with other plants, prunning, reducing the use of insecticides and herbicides, and do not plant the cocoa near to the river.Keywords: cacao, disease, epidemic, Oncobasidium theobromae
