Abstract

Pencarian obat antikanker terus dilakukan salah satunya pencarian inhibitor Check point kinase 1 yang mampu meningkatkan level apoptosis sel kanker. Telah dilakukan skrining virtual inhibitor Check point kinase 1 dari derivat 1,3-dibenzoylthiourea dengan menggunakan perangkat lunak PLANTS 1.1 (Protein-Ligand ANT System). Dari 23 senyawa yang di-docking pada Check point kinase 1 didapat 5 senyawa dengan energi ikatan paling stabil yaitu, 1,3 – bis ( { [ 3-dimethylamino ) phenyl ] carbonyl } ) thiourea, 1,3-bis ( { [ 4- (dimethylamino)-3-methylphenyl] carbonyl}) thiourea, 1,3-bis ( { [ 3-(trifluoromethyl) phenyl] carbonyl } ) thiourea, 1,3-bis ( { [ 4-( dimethylamino ) -3- methylphenyl ]carbonyl})thiourea dan 1,3-bis ( {[4-chloro-3- (trifluoromethyl)phenyl] carbonyl})thiourea    dengan  energi  ikatan  berturut  -  turut    -95,6248,  -92.9495,  -92,5560, -92,5509 sedangkan untuk (UCM) native ligand adalah         -95.3636 . Terdapat kesamaan interaksi antara UCM ( native ligand ) yaitu dengan senyawa 1,3 – bis ( { [ 3-dimethylamino ) phenyl ] carbonyl } ) thiourea dimana keduanya berinteraksi dengan 7 residu yaitu Lys 132, Ile 131, Ile 146, Asn 135, His 128,  Leu136 dan Asp 130. Hal tersebut menunjukan bahwa 1,3 – bis ( { [ 3-dimethylamino ) phenyl ] carbonyl } ) thiourea diprediksi memiliki aktivitas inhibitor Check point kinase 1 yang memiliki energi ikatan paling stabil menurut metode docking. Key words: Check point kinase 1, Thiourea, Docking

Abstract

3-benzoil-1-feniltiourea telah disintesis melalui reaksi asilasi antara senyawa benzoil klorida dengan senyawa 1-feniltiourea menggunakan pelarut tetrahidrofuran dan katalis trietilamin dengan cara refluks dan pengadukan selama 6 jam. Persentase hasil sintesis yang diperoleh adalah 89 %. Kemurnian hasil sintesis ditunjukkan dengan adanya noda tunggal pada KLT dan jarak lebur yang sempit. Hasil identifikasi struktur dengan spektrofotometri UV, spektroskopi infra merah, spektroskopi 1HNMR dan spektroskopi massa menunjukkan struktur senyawa hasil sintesis sesuai dengan yang diperkirakan. Studi interaksi secara in silico menggunakan program Autodock Vina dan MMV menunjukkan senyawa 3-benzoil-1-feniltiourea memiliki interaksi yang baik terhadap reseptor kanker paru-paru (2ITO) melalui ikatan hidrogen dengan residu-residu asam amino yaitu Ala 839, Asn 842 dan Lys 852 dan dengan nilai energi afinitas -6,5 kkal/mol.

Abstract

Senyawa N’-(3,5-dinitrobenzoyl)-isonicotinohydrazide disintesis melalui reaksi asilasi antara isoniazid dengan 3,5-dinitrobenzoil klorida dalam pelarut tetrahidrofuran dengan cara refluks selama 8 jam. Persentase hasil senyawa sintesis yang diperoleh adalah 11,88%. Kemurnian hasil sintesis telah diuji dengan menggunakan Kromatografi Lapis Tipis (KLT) menggunakan beberapa eluen yang berbeda menghasilkan noda tunggal dengan nilai Rf yang berbeda dari isoniazid dan 3,5-dinitrobenzoyl klorida dan uji jarak lebur menunjukkan rentang yang sempit. Identifikasi struktur dilakukan dengan menggunakan metode secara spektroskopi yaitu spektrofotometri UV – VIS, IR dan MS menunjukkan bahwa senyawa hasil sintesis merupakan senyawa N’-(3,5-dinitrobenzoyl)-isonicotinohydrazide sesuai dengan hipotesis. Studi interaksi secara In Silico dilakukan menggunakan software ArgusLab yang menunjukkan energi ikatan senyawa N’-(3,5-dinitrobenzoyl)-isonicotinohydrazide lebih baik daripada isoniazid dengan parameter nilai bindding afinity lebih kecil daripada senyawa isoniazid, nilai binding affinity terbaik senyawa N’-(3,5-dinitrobenzoyl)-isonicotinohydrazide adalah -8,466699 kcal/mol pada reseptor tuberkulosis dengan kode PDB 2X23. Hasilnya divisualisasi menggunakan bantuan sofware Molegro Molecular Viewer (MMV) menunjukkan bahwa senyawa N’-(3,5-dinitrobenzoyl)-isonicotinohydrazide dapat berinteraksi dengan residu asam amino melalui interaksi ikatan hidrogen. Analisis Drugs Scan menunjukkan bahwa senyawa N’-(3,5-dinitrobenzoyl)-isonicotinohydrazide memenuhi syarat sebagai obat dengan mengikuti aturan Lipinski’s rule of five serta di uji toksisitasnya menggunakan ECOSAR (Ecological Structure Activity Relationships) dan hasilnya menunjukkan bahwa senyawa N’-(3,5-dinitrobenzoyl)-isonicotinohydrazide memiliki nilai LC50 dan EC50 >100 mg/L sehingga termasuk dalam kategori rendah karena >100mg/L. Kata Kunci: Modifikasi, Isoniazid, Mycobacterium tuberculosis.

Abstract

The Bis-(1-(2,4-dichlorobenzoyl)-3-methylthiourea) iron (III) complex has been synthesized from the reaction between 1-(2,4-dichlorobenzoyl)-3-methylthiourea and Fe (III) metal ion by reflux method with ethanol solvent at a temperature of 75°C for 7 hours. It was characterized by a hot stage microscope (HSM), UV-Vis, FT-IR, and mass spectroscopy. The % yield of the synthesis result was 97.58%. From the docking study on the ribonucleotide reductase enzyme, the binding affinity value was -7.76 kcal/mol, and the inhibition constant was 2.11 mM. The Bis-(1-(2,4-dichlorobenzoyl)-3 methylthiourea) iron (III) complex compounds can be synthesized and predicted as anticancer candidates.

Abstract

Diabetes Mellitus (DM) is a chronic disease that occurs when the pancreas does not produce enough insulin, or the body cannot use insulin effectively. Type 2 DM treatment can be done using antidiabetic drugs, but the continuous use of synthetic drugs will cause side effects. Empirically, the people of Nias Indonesia use palm juice (Arenga pinnata Merr.) as an antidiabetic, which can reduce blood glucose levels. This study aimed to find the active compounds in palm juice that can potentially be an antidiabetic type 2 using an in silico approach. The methods used were toxicity screening, profile pharmacokinetics, drug scanning, docking, and molecular dynamics simulation. Screening, molecular docking, and molecular dynamics of 30 compounds generated from pyrazine and furan revealed that two compounds, PF 16 and PF 30, can bind to receptors and produce lower ∆G values than metformin HCl. Molecular dynamics simulation results using the MM-GBSA calculation method showed that the PF 16 compound was more selective than the 2PDY (aldose reductase) with a value of -39.23 kcal/mol, while compound PF 30 was more selective to 1Z89 (aldose reductase) with a value of -7.36 kcal/mol. It can be concluded that the level of affinity of the PF 30 compound to the 1Z89 receptor and the PF 16 compound to the 2PDY were predicted to have the potential as antidiabetic (DM type 2).

Abstract

In December 2019, the first COVID-19 cases were in Wuhan, China. This case is a global concern and a threat to public health. Based on previous research using molecular docking methods, it was found that flavonoids exhibit strong inhibitory activity in SARS-CoV-2 main proteases. The study aims to determine the flavonoid compound Syzygium cumini (L.) Skeels can interact with the SARS-CoV-2 main protease receptor and can be used as a candidate for COVID-19 therapy with virtual screening. Myricetin 4”-O-acetyl-2-O-gallate has the lowest Gibbs free energy (ΔG) of -9.82 kcal/mol. The molecular dynamics of the best compound, Myrcetin 4-O-acetyl-2-O-gallate, RMSD, and RSMF values are quite stable. As a result of pharmacokinetic prediction and toxicity, the best compounds have a relatively good pharmacokinetic profile and are non-toxic. Thus, it can be concluded that the compound Myricetin 4”-O-acetyl-2-o-gallate in the Syzygium cumini (L.) Skeels is predicted to interact with the SARS-CoV-2 main protease receptor (7C6S) as a potential drug candidate for COVID-19 therapy.

Abstract

Cancer is a disease that starts from the uncontrolled growth of abnormal cells in the organs or tissues of the body, which is the second leading cause of death in the world. One of the targets in discovering and developing anticancer drugs is Sirtuin-1. SIRT1 can act as a tumor suppressor or tumor promoter depending on its target in a particular signalling pathway or on particular cancer. This study aimed to study the interaction of a thiourea derivative with SIRT1 (PDB ID:4I5I) through its inhibition of histone deacetylase. Research has been carried out in silico with molecular docking (MGLTools.1.5.6) and molecular dynamics (Desmond 2019) of three thiourea derivatives to the receptor. In addition, pharmacokinetic parameters, toxicity, and selection of Lipinski's Rule of Five were also tested. Molecular docking results showed that compound b ([2-(methylcarbamothioylcarbamoyl)phenyl]benzoate) had the lowest ∆G value of −9.29 kcal/mol with a KI value of 0.156 µM compared to other thiourea derivatives and was proven by molecular dynamics tests for 30 ns and amino acids that play an active role in the interaction include the residue PheA:297. In terms of pharmacokinetics and toxicity, compound b is better than natural ligands. Compound b is predicted to be used as an anticancer candidate through further research.

Abstract

This study examines the influence of training, leadership effectiveness, and work environment on employee performance in the Goods and Services Procurement Section of the Regional Secretariat of North Konawe Regency. The method used is a quantitative approach with multiple linear regression. The population of this study includes all 31 employees of the Procurement Section, who also served as the sample for this research. Data was collected through a questionnaire using a Likert scale, then analyzed with statistical tests. The results show that training, leadership effectiveness, and the work environment collectively have a positive and significant impact on employee performance. Individually, each variable was also proven to positively and significantly affect performance. This study emphasizes that improving the quality of training provided, leadership effectiveness, and a comfortable work environment play a significant role in enhancing employee performance. These findings can serve as a reference for government agencies in formulating more effective employee development strategies.

Abstract

Hasan Langgulung as one of the leading Islamic education figures. Hasan Langulung's concept of thought, emphasizes that education is a process to transfer the cultural values ??of society inherited from the older generation to the younger generation so that its cultural identity is maintained as a continuation of the life of society and education as well as education as a process of developing the potential of each individual. This research is a library research. The primary data source here is data or information obtained from the written works of Islamic education figures, namely Hasan Langgalung which are discussed in writing this thesis, namely the Islamic education method according to Hasan Langgalung and Islamic Education according to Hasan Langgalung. The data collection techniques used in this study are literature and documentation. The results of the study concluded that the Islamic education method proposed by Hassan Langgulung is a more practical method, namely the method used must be related to the objectives of Islamic Education, the education method does not force something that is contrary to human nature, the method should talk about motivation and discipline or in terms of the Qur'an, rewards and punishments. The advantages of Hasan Langgulung's Islamic education method are an increase in spiritual, moral, intellectual, and social processes. Hasan Langgulung's Islamic education methods guide humans and give them ideal values, principles, and role models in life aimed at preparing for worldly and afterlife life. ABSTRAKHasan Langgulung sebagai salah satu tokoh pendidikan Islam yang terkemuka. Konsep pemikiran Hasan Langulung, menegaskan bahwa pendidikan adalah proses untuk memindahkan nilai-nilai budaya masyarakat yang diwariskan dari generasi tua ke generasi muda agar identitas budayanya tetap terjaga sebagi kelanjutan hidup masyarkat dan pendidikan juga pendidikan sebgai proses pengembangan potensi-potensi setiap individu. Penelitian ini merupakan penelitian kepustakaan (library research). Sumber data primer disini adalah data atau informasi yang diperoleh dari karya tulis dari tokoh pendidikan islam yaitu Hasan Langgalung yang dibahas dalam penulisan skripsi ini, adalah sebagai Metode pendidikan Islam menurut Hasan Langgalung dan Pendidikan Islam menurut Hasan Langgalung. Teknik pengumpulan data yang digunakan dalam penelitian ini adalah kepustakaan dan dokumentasi. Hasil penelitian menyimpulkan bahwa Metode pendidikan Islam yang dikemukakan oleh Hassan Langgulung yaitu metode lebih bersifat praktikal yaitu metode yang dilakukan harus berkaitan dengan tujuan Pendidikan Islam, metode pendidikan tidak memaksakan sesuatu hal yang bertentangan dengan fitrah manusianya, metode hendaknya membicarakan tentang pengerakan (motivtion) dan disiplin atau dalam istilah Al-Qur’an ganajaran dan hukuman. Keunggulan metode pendidikan Islam Hasan Langgulung ialah suatu meningkatkan proses spritual, akhlak, intelektual, dan sosial. Metode metode pendidikan Islam Hasan Langgulung membimbing manusia dan memberinya nilai-nilai, prinsip, dan keteladanan yang ideal dalam kehidupan bertujuan mempersiapkan kehidupan dunia dan akhirat.

Abstract

In this study, the researcher used a type of field research or also called Qualitative with Descriptive research nature, with data collection tools used, namely observation, interviews and documentation. The researcher used primary data sources and secondary data sources, the data collection method used was non-participant observation and unstructured interviews, while the data analysis technique used was inductive and holistic, with several stages, namely Data reduction (Data Reduction), Data display (data presentation), Conclusion Drawing/ verification, Based on the results of this study, it shows that the implementation of the bahtsul masa'il method in motivating learning fiqh at the Ahsanul Ibad Purbolinggo East Lampung Islamic High School includes three stages, namely with teachers making a learning implementation plan using the bahtsul masa'il method, then after planning, namely by implementing the fiqh learning process in accordance with the learning implementation plan using the bahtsul masa'il method, the last stage is evaluating the fiqh learning process using the bahtsul masa'il method. ABSTRAKPada penelitian ini peneliti menggunakan jenis penelitian lapangan (field research) atau disebut juga Kualitatif dengan sifat penelitian Deskriptif, dengan alat pengumpulan data yang digunakan yaitu observasi, wawancara dan dokumentasi. Peneliti menggunakan sumber data primer dan sumber data sekunder, Metode pengumpulan data yang digunakan aalah observasi  non-Partisipan dan Wawancara tidak terstruktur, sedangkan teknik analisis data yang digunakan bersifat induktif dan holistic, dengan beberapa tahapan yaitu Data reduction (Reduksi Data), Data display (penyajian data), Conclusion Drawing/ verification, Berdasarkan hasil penelitian ini menunjukan bahwa implementasi metode bahtsul masa’il dalam memotivasi belajar fiqih di madrasah aliyah ahsanul ibad purbolinggo lampung timur mencakup tiga tahapan yaitu dengan guru membuat rancangan pelaksanaan pembelajaran dengan menggunakan metode bahtsul masa’il, kemudian setelah merencanakan yaitu dengan melaksanaan proses pembelajaran fiqih sesuai dengan rancangan pelaksanaan pembelajaran dengan menggunakan metode bahtsul masa’il, tahap yang terakhir adalah mengevaluasi proses pembelajaran fiqih dengan menggunakan metode bahtsul masa’il.
