Abstract

The pletekan plant (Ruellia tuberosa L.) is a family of Acanthaceae Ruellia tuberosa L.) one of the plants used as a medicinal plant, especially the leaves, have been used empirically for therapy as a diuretic, antidiabetic, antihypertensive. Also used in the treatment of syphilis, kidney stones, cancer, heart disease, colds, hypertension and digestive problems. The purpose of this study was to determine the toxicity by determining the LC50 in the ethanol extract of pletekan leaves (Ruellia tuberosa L). The analysis carried out was a toxicity test on the ethanol extract of pletekan leaves against Artemia Salina Leach shrimp larvae using the BSLT method with various concentrations, namely 10, 15, 20, 25, 30, 35, and 40 ppm and determining the LC50 value with probit analysis. Based on the results of the toxicity test, the LC50 value was 96.4595 µg/Ml. These results show that the ethanol extract of pletekan leaves (Ruellia tuberosa L) has a toxic effect on Artemia Salina Leach shrimp larvae.

Abstract

Penggunaan tumbuhan sebagai ramuan obat sangat berkaitan dengan kandungan kimia yang terdapat dalam tumbuhan tersebut terutama zat aktif biologisnya. Senyawa bioaktif yang terdapat dalam tumbuhan merupakan senyawa metabolit sekunder seperti steroid, flavonoid, alkaloid, saponin, terpenoid dan tanin. Salah satu tanaman yang mengandung senyawa metabolit sekunder yaitu daun pala (Myristica fragrans Houtt). Tujuan penelitian ini adalah untuk mengetahui senyawa kimia yang terdapat didalam ekstrak etanol dan ekstrak etil asetat dan untuk mengetahui nilai flavonoid total ekstrak etanol dan ekstrak etil asetat daun pala. Tahapan penelitian ini meliputi pengolahan bahan tumbuhan, pembuatan ekstrak etanol dan ekstrak etil asetat, pemeriksaan karakterisasi, skrining fitokimia dan penetapan kadar flavonoid total ekstrak etanol dan ekstrak etil asetat daun pala dengan metode spektrofotometri Visible. Ekstrak daun pala dibuat dengan metode maserasi dengan menggunakan etanol 70% dan etil asetat ekstrak yang diperoleh dipekatkan dengan rotary evaporator, Selanjutnya dilakukan penetapan kadar flavonoid total dengan metode spektrofotometri Visible. Hasil skrining fitokimia pada ekstrak etanol dan ekstrak etil asetat daun pala terdapat kandungan golongan senyawa kimia seperti alkaloid, flavonoid, saponin, tanin, dan steroid/triterpenoid. Hasil penentuan kadar flavonoid total pada ekstrak etanol sebesar 39,7252 ± 1,9596 mg QE/g dan  ekstrak etil asetat sebesar 31,1395  ± 0,3983 mg QE/g.

Abstract

Rhodamin B dalam kosmetika tidak diperbolehkan sesuai No.445/MenKes/Per/V/1998. Penggunaan Rhodamin B dapat mengiritasi saluran pernapasan, penumpukannya dapat menyebabkan gangguan fungsi hati. Penelitian ini bertujuan untuk mengetahui adanya kandungan Rhodamin B serta kadar Rhodamin B pada lipstik, untuk mengetahui cara analisis kualitatif dan kuantitatif Rhodamin B pada lipstik secara KLT Densitometri. Penelitian ini meliputi cara pengumpulan dan pembuatan larutan lipstik, pembuatan larutan Rhodamin B, pengujian Rhodamin B dan penetapan kadar Rhodamin B. Penelitian ini menggunakan 5  lipstik. Analisis kualitatif secara KLT menggunakan fase diam silika gel GF254 dan fase gerak etil asetat : metanol : ammonia (15:3:3). Analisis kuantitatif menggunakan metode Densitometri. Hasil analisis kualitatif secara KLT didapat, 2 dari 5 sampel lipstik positif Rhodamin B ditandai dengan noda berwarna orange dan berfluoresensi kuning pada lampu UV 254 nm dan 366 nm, bewarna merah muda secara visual. Nilai Rf lipstik S1 (0,68), Rf lipstik S2 (0,45), Rf lipstik S3 (0,87), Rf lipstik S4 (0,87) dan Rf lipstik S5 (0,56) dengan Rf Rhodamin B (0,87). Lipstik positif Rhodamin B dilakukan penetapan kadar dengan metode Densitometri. Kurva Kalibrasi diperoleh persamaan y = 0,221x + 1,088. Kadar Rhodamin B pada lipstik S3 sebesar 1,505 mg/g dan lipstik S4 sebesar 3,118 mg/g. Kesimpulannya lipstik kode S3 dan S4 tidak memenuhi uji persyaratan.

Abstract

Uji sitotoksik adalah pemeriksaan yang menentukan tingkat kerusakan zat terhadap sel. Penyakit yang dikenal sebagai kanker dicirikan oleh pembelahan sel yang tidak terkendali dan kemampuan sel-sel tersebut untuk menyerang jaringan biologis lainnya. Senyawa aktif tanaman herbal dapat berupa ekstrak tanaman atau ekstrak tanaman. Daun tahi ayam adalah salah satu tumbuhan yang mengandung banyak manfaat. Untuk mengetahui apakah larva udang Artemia salina Leach memiliki keidentikan dengan sel kanker manusia, uji pendahuluan dilakukan dengan metode BSLT.Penelitian ini bertujuan untuk mengetahui golongan senyawa metabolit sekunder yang memiliki kemampuan efek sitotoksik pada ekstrak etanol daun tahi ayam. Dalam penelitian ini, ekstrak etanol daun tahi ayam dikarakterisasi, di skrining fitokimia, dan diuji untuk sitotoksisitas dengan menggunakan metode BSLT terhadap larva udang Artemia salina Leach, yang ditunjukkan dengan nilai LC50. Dalam berbagai konsentrasi, seperti 100, 200, 300, 400, 500, 600, 700, 800, 900, dan 1000 ppm, uji sitotoksisitas dilakukan. Hasil penelitian ini menunjukkan bahwa ekstrak etanol daun tahi ayam mengandung senyawa metabolit sekunder seperti alkaloid, flavonoid, tanin, saponin, dan steroid, dan uji sitotoksisitas dengan probit menunjukkan nilai LC50 319,6686 g/ml. Oleh karena itu, penelitian ini menunjukkan bahwa ekstrak etanol daun tahi ayam bersifat toksik dan mungkin berfungsi sebagai antikanker.

Abstract

Raru bark (Cotylelobium melanoxylon (Hook.f) Pierre) is a plant with antidiabetic properties. Raru bark plants contain flavonoids, saponins, and tannins. The purpose of this study was to determine the class of secondary metabolites contained in the sample, determine flavonoid levels, and determine antibacterial activity against Staphylococcus aureus bacteria. This research includes processing samples made simplisia, then extracted and thickened using rotary evaporatory. Simplisia was tested for phytochemical screening to determine the content of metabolite compounds, in addition to testing its characteristics. The next test was the determination of total flavonoid content using UV-Vis spectrophotometry. Followed by testing antibacterial activity against Staphylococcus aureus. The results of phytochemical screening obtained flavonoids, tannins, and saponins. Flavonoid levels were 42.4308 ± 2.4739 mgQE/g; 38.1813 ± 1.5799mgQE/g; and 23.8153 ± 3.049276 mgQE/g, respectively. In testing antibacterial activity using concentrations of 10%, 40%, 70%, and 100%, and the highest concentration is found at a concentration of 100% which is 19.83mm.

Abstract

The use of decorative cosmetics such as loose powder and blush is increasingly widespread, raising concerns regarding the safety of the coloring agents used, particularly Rhodamine B. This study aimed to identify the presence and determine the levels of Rhodamine B in loose powder and blush cosmetic products using Thin Layer Chromatography (TLC) for qualitative analysis and High-Performance Liquid Chromatography (HPLC) for quantitative analysis. The HPLC method applied in this study was validated and showed good linearity, precision, and sensitivity in accordance with ICH guidelines. A total of 16 cosmetic samples were analyzed, and 3 samples were confirmed to contain Rhodamine B. The highest Rhodamine B level was found in sample X at 66.54 µg/g, followed by sample Z at 21.03 µg/g, and sample Y at 0.83 µg/g. These findings indicate that the presence of Rhodamine B in cosmetic products remains a concern. Therefore, it is necessary to strengthen regulatory supervision and increase public awareness to select safe cosmetic products that are officially registered by BPOM.

Abstract

The bajakah plant is one of the biological plants used in traditional medicine. Bajakah is also a medicinal herbal plant that can be used in all its parts. This plant can be found in the depths of Kalimantan Province, but it has not spread to other regions. There has been no effort to cultivate bajakah because of the lack of understanding of the local population about the use of this plant. This research includes the stages of processing plant materials, making ethanol extracts, characterization examinations, phytochemical screening, and determining total flavonoid levels of ethanol extract of Bajakah wood using the visible spectrophotometry method and antibacterial activity. Bajakah wood extract was made by maceration method using ethanol concentrations of 96%, 70%, and 50%, and the extract obtained was concentrated with a rotary evaporator, then the total flavonoid level was determined using the visible spectrophotometry method and antibacterial activity tests were carried out against Staphylococcus aureus As a positive control using chloramphenicol and negative control using dimethyl sulfoxide. The results were obtained by determining the maximum wavelength of quercetin and operating time, measuring the quercetin calibration curve, and calculating the total flavonoid level using the visible spectrophotometry method. The results obtained in the determination of total flavonoid levels in Bajakah wood ethanol extract at a concentration of 96% were 349.888 ± 4.5022 mg QE/g, at a 70% concentration of 307.137 ± 6.7626 mg QE/g and a 50% concentration of 6.15173 ± 2.9148 mg QE/g.

Abstract

Radiasi ultraviolet (UV) merupakan salah satu ancaman utama bagi kesehatan kulit yang berkontribusi pada terjadinya penuaan dini, kerusakan DNA, dan peningkatan risiko kanker kulit. Senyawa antioksidan, seperti flavonoid yang banyak ditemukan pada tumbuhan, memiliki kemampuan untuk menetralkan radikal bebas yang dihasilkan akibat paparan sinar UV. Penelitian ini bertujuan untuk menghasilkan potensi ekstrak kulit jeruk bali, yang kaya akan flavonoid, sebagai tabir surya alami. Untuk meningkatkan stabilitas dan homogenitas formulasi, ekstrak kulit jeruk bali dimodifikasi menjadi nanopartikel, yang diharapkan dapat meningkatkan efektivitasnya dalam sediaan tabir surya bubuk. Formulasi sediaan dilakukan dalam empat konsentrasi, yaitu formulasi blanko, F1 (5%), F2 (10%), dan F3 (15%). Evaluasi mutu fisik sediaan meliputi uji organoleptis, homogenitas, derajat halus, daya lekat, derajat keasaman (pH), hedonik/kesukaan, dan Sun Protection Factor (SPF). Hasil penelitian menunjukkan bahwa uji mutu fisik sediaan tabir surya bubuk berbasis ekstrak nanopartikel kulit jeruk bali memenuhi kriteria yang ditetapkan. Ekstrak nanopartikel yang dihasilkan memiliki ukuran 205,2 nm, indeks polidispersitas 0,359, dan zeta potensial -16,0 mV. Nilai SPF dari formulasi tabir surya bubuk berturut-turut adalah 1,617 (minimal), 7,946 (ekstra), 16,554 (ultra), dan 19,072 (ultra), menunjukkan efektivitas perlindungan yang signifikan terhadap sinar UV. Penelitian ini memberikan pengetahuan baru mengenai potensi penggunaan ekstrak tumbuhan sebagai alternatif dalam pengembangan produk tabir surya alami.

Abstract

Cytotoxic activity is a process that can induce cell death. One of the plants that has cytotoxic properties is bandotan (Ageratum conyzoides L.). The purpose of this study was to identify the class of secondary metabolites present in the ethanol extract of Bandotan herb and to evaluate its cytotoxicity by determining the LC50 value using the Brine Shrimp Lethality Test (BSLT) method. This research includes phytochemical screening and cytotoxicity testing of an ethanol extract of Herba bandotan using the BSLT method. BSLT is the first method for cytotoxicity testing using Artemia salina Leach larvae at the nauplii stage. Used in the BSLT method because it has similar responses with mammals, namely DNA-dependent RNA polymerase in Artemia salina Leach, similar to DNA-dependent RNA polymerase in mammals. By phytochemical screening, the secondary metabolites in Herba Bandotan (Ageratum conyzoides L.) are alkaloids, flavonoids, saponins, steroids, and tannins. Based on the results of the cytotoxicity test using the BSLT method, it shows that the ethanol extract of Herba bandotan (Ageratum conyzoides L.) has the potential to be anti-cancer because it has an LC50 value of 255.1915 g / mL, which is included in the toxic category and has the potential to be anti-cancer. Potential as an anticancer agent in the BSLT test if it has an LC50 value < 1000 µg / mL.

Abstract

The Chinese petai plant (Leucaena leucocephala (Lam.) de Wit) has been used in various systems of traditional medicine for the treatment of various diseases in humans. The Chinese petai plant (Leucaena leucocephala (Lam.) de Wit) contains antioxidant secondary metabolites that can inhibit oxidation reactions by binding to free radicals and highly reactive molecules. The purpose of this study was to determine the class of chemical compounds contained in the ethanol extract of Chinese petai peel and the antioxidant activity of the ethanol extract of Chinese petai peel (EEKPC). The stages of this research included: collection and processing of simplicia; preparation of the extract by maceration with 70% ethanol; characterization test of simplicia powder; phytochemical screening of ethanol extract of petai cina; antioxidant activity test of EEKPC using the DPPH (1,1-diphenyl-2-pikrilhidrazyl) free radical scavenging method using UV-Vis spectrophotometry at a wavelength of 516 nm. The results of the phytochemical screening examination of Chinese Petai Peel Ethanol Extract (EEKPC) contained alkaloids, flavonoids, saponins, tannins, and triterpenoids. The results of measuring the antioxidant activity of EEKPC showed strength in the "strong" category with an IC50 value of 91.0189 μg/ml.
