Abstract

Hyperlipidemia is a major risk factor for cardiovascular disease, with a continuously increasing prevalence worldwide. Turmeric leaves (Curcuma longa L.) contain various bioactive compounds with potential antihyperlipidemic properties through modulation of metabolic biomarkers such as acyl ghrelin, growth hormone (GH), and AMP-activated protein kinase (AMPK). To analyze the effects of ethanolic turmeric leaf extract on acyl ghrelin, GH, and AMPK levels in hyperlipidemic rats induced by a high-fat diet (HFD) and propylthiouracil (PTU) using combined in silico and in vivo approaches. This laboratory experimental study used Federer's formula to determine the sample size, resulting in 36 male Wistar rats randomly divided into six groups: normal control (K1), hyperlipidemia control receiving HFD+PTU (K2), positive control receiving HFD+PTU+simvastatin 0.9 mg/kg body weight (BW) (K3), and three treatment groups receiving HFD+PTU plus ethanolic turmeric leaf extract at doses of 300 mg/kg BW (K4), 600 mg/kg BW (K5), and 1200 mg/kg BW (K6). In silico molecular docking was conducted to evaluate interactions between active compounds and target proteins. Measured parameters included lipid profiles, aortic histopathology, and levels of acyl ghrelin, GH, and AMPK assessed using ELISA.  Phytochemical screening confirmed the presence of five classes of bioactive compounds in the extract. In silico analysis identified rutin as the principal compound, exhibiting strong binding affinities to acyl ghrelin (−10.6 kcal/mol), growth hormone (−8.0 kcal/mol), and AMPK (−9.3 kcal/mol). The 300 mg/kg BW dose demonstrated the most pronounced lipid-lowering effects, with significant reductions in total cholesterol (210.50±2.74 to 181.67±6.98 mg/dL), triglycerides (246.83±8.47 to 179.17±8.80 mg/dL), and LDL (96.33±5.68 to 70.17±8.11 mg/dL), alongside an increase in HDL (65.00±3.74 to 75.67±1.97 mg/dL) (p<0.001). Histopathological analysis revealed reduced aortic wall thickness (192.59±6.99 μm) and foam cell count (24.17±4.31) in this group. Acyl ghrelin levels differed significantly among groups (p<0.001), with the hyperlipidemic control showing the lowest levels. The 600 mg/kg BW dose optimally increased AMPK (10.33±0.95 ng/mL) and GH (1.96±0.15 ng/mL) compared with the hyperlipidemic control (p<0.05). Ethanolic turmeric leaf extract exerts significant antihyperlipidemic effects by improving lipid profiles, attenuating atherosclerosis, and modulating metabolic biomarkers including acyl ghrelin, GH, and AMPK. A dose of 300 mg/kg BW was most effective for lipid profile improvement and atherosclerosis reduction, while 600 mg/kg BW was optimal for hormonal biomarker modulation.

Abstract

Hyperlipidemia is a major cardiovascular risk factor with increasing global prevalence. To analyze the effects of ethanolic turmeric leaf extract on leptin, HMG-CoA levels, and blood vessel histopathology in hyperlipidemic rats using in silico and in vivo approaches. Thirty-six male Wistar rats were divided into six groups: normal control (K1), hyperlipidemia control (K2), simvastatin 0.9 mg/kg BW (K3), and turmeric extract at 300 (K4), 600 (K5), and 1200 mg/kg BW (K6). Hyperlipidemia was induced with high-fat diet and propylthiouracil. In silico molecular docking evaluated compound-protein interactions. Phytochemical screening confirmed five bioactive compound classes. In silico analysis showed rutin with strong binding affinities to leptin (−10.2 kcal/mol) and HMG-CoA reductase (−9.8 kcal/mol). The 300 mg/kg dose significantly improved lipid profiles, reduced aortic wall thickness (192.59±6.99 μm), foam cells (24.17±4.31), and HMG-CoA levels (3.45±0.42 ng/mL) (p<0.001). The 600 mg/kg dose optimally restored leptin (485.67±62.34 pg/mL). Ethanolic turmeric leaf extract exerts significant antihyperlipidemic effects by improving lipid profiles, attenuating atherosclerosis, and modulating leptin and HMG-CoA reductase.

Abstract

Background: Diabetes mellitus is a chronic disease in the form of metabolic disorders, which is characterized by blood glucose levels (BGL) that exceed normal limits and are accompanied by disorders of carbohydrate, lipid, and protein metabolism, as a result of abnormalities in insulin secretion. Diabetics are known to have slow wound healing. Moringa oleifera leaves contain flavonoid compounds, which can reduce glucose absorption. Objective: This study aims to determine the effect of Moringa leaf infusion on blood sugar levels, glucose tolerance, cholesterol levels, body weight, and speed of wound healing in mice model. Method  : A total of 25 male mice aged 2-3 months weighing 25-30 grams were divided into 5 experimental groups namely: negative control, positive control, 10% Moringa infusion, 20% infusion, 30% infusion. The negative control was not given any treatment. Positive controls were induced by high fat diet and PTU and given sucrose for 3 weeks. The infusion group was the same as the positive control but at the end of each week, 10%, 20%, and 30% Moringa leaf infusion were given successively. Moringa leaf intervention was given for 3 weeks after feed induction and observations were made at that range. The wound was made after feeding induction on the back 1 cm long. Result  : Moringa leaf infusion had a significant effect on reducing the levels of each variable, namely a dose of 10% on body weight, a dose of 30% on KGD, 20% on KGD with glucose loading, 20% on cholesterol, and 20% on wound closure, which proved based on the results of statistical tests. Conclusion: Moringa leaf infusion can reduce blood sugar levels, blood sugar levels by loading glucose, cholesterol levels, body weight, and accelerate wound healing in mice induced by sucrose, high fat diet, and PTU.

Abstract

Background: Diabetes mellitus (DM) sufferers in the world and even inIndonesia are expected to continue to increase, especially type 2 DM(DMT2). DMT2 often occurs in people who are obese, the cause of obesityis often consuming foods that contain a lot of fat. Hyperlipidemia plays arole in the pathogenesis of pancreatic β-cell dysfunction.Research method: This study is an experimental laboratory study with apretest-posttest only controlled group design, using male white rats(Rattus norvegicus) with a sample size of 30 divided into 6 groups, namelythe normal group, the negative control group, the positive control group,the group given a high fatty diet (HFD). The independent variable is HFD,while the dependent variable is blood sugar levels. Data were analyzedusing the Anova test.Results: Giving HFD of 4 ml/day for 4 weeks has not been able to increaseKGD to hyperglycemic, this is thought to be because the HFD was notgiven for long enough so that insulin resistance had not occurred in thetest animals.Conclusion: Based on the results of the study, it was concluded that givingHFD for 4 weeks still could not increase KGD in male white mice tobecome hyperglycemic

Abstract

Diabetes Mellitus (DM) is a disease with metabolic disorders characterized by chronichyperglycemia and disorders of carbohydrate, fat, and protein metabolism due to insulinsecretion disorders. In general, DM patients require pharmacotherapy such as injectedinsulin or oral antidiabetic drugs. Pharmacological treatment is relatively expensive andneeds to consider its efficacy and safety. Therefore, this study explores natural sources suchas medicinal plants. The aim of this study was to determine the antihyperglycemic effect of acombination of boiled moringa leaves, lemongrass leaves and stevia leaves on mice (Musmusculus) induced by alloxan. In this study, 25 mice were divided into 5 groups. The micewill undergo adaptation for 7 days and then given HFD for 30 days. After 30 days, the micewill be given alloxan 170 mg/kgBW for 3 days to induce DM. After DM was successfullyinduced in mice, polyherbal tea combination of moringa leaves, lemongrass leaves and stevialeaves was made and given, then continued with evaluation blood glucose, blood glucosepercentage, body weight, body weight percentage, index lee and liver organ index. The groupgiven herbal tea combination of moringa leaves (Moringa oleifera), lemongrass leaves(Cymbopogon citratus) and stevia leaves (Stevia rebaudiana) with a concentration of 3.6mg/mL showed a better decrease in KGD and %KGD compared to other treatment groups.The decrease in BB, %BB and lee index was better in the group given a combination ofMoringa leaves (Moringa oleifera), Lemongrass leaves (Cymbopogon citratus) and Stevialeaves (Stevia rebaudiana) with a concentration of 7.2 mg/mL. The decrease in the liverorgan index was also better seen in the group given a combination of Moringa leaves(Moringa oleifera), Lemongrass leaves (Cymbopogon citratus) and Stevia leaves (Steviarebaudiana) with a concentration of 7.2 mg/mL. The group given herbal tea combination ofMoringa leaves (Moringa oleifera), Lemongrass leaves (Cymbopogon citratus) and Stevialeaves (Stevia rebaudiana) with a concentration of 3.6 mg/mL reduced BG and BGpercentage, while the group given herbal tea combination of Moringa leaves (Moringaoleifera), Lemongrass leaves (Cymbopogon citratus) and Stevia leaves (Stevia rebaudiana)with a concentration of 7.2 mg/mL reduced BW, BW percentage, Lee index, and hait organindex.

Abstract

ABSTRACT Currently, modern life, especially in urban areas, promotes an instant consumption lifestyle, particularly among teenagers. With technological advancements, teenagers can easily order fast food through delivery apps, making fast food a common choice. In addition to fast food, school snacks are also highly favored by teenagers. This excessive consumption pattern, if sustained over time, can lead to fat accumulation that affects body mass index (BMI) and waist circumference. According to the 2019 health profile, 35.4% of teenagers experience obesity, with the highest prevalence in urban areas at 25.1%. Excess fat accumulation puts pressure on cardiorespiratory function, reducing lung capacity as reflected in VO2Max or cardiorespiratory endurance. This decline in cardiorespiratory endurance significantly affects a person's quality of life, causing them to feel fatigued easily and limiting their ability to engage in prolonged physical activities.Conclusion: There is a significant relationship between body mass index and waist circumference on cardiorespiratory endurance among students of Methodist 1 High School, Medan, North Sumatra..

Abstract

Background: Tuberculosis (TB) is still a major public health problem in most developing countries and its incidence is rising in many developed countries. This study aimed to examine factors associated with tuberculosis in Deli Serdang, North SumateraSubjects and Method: This was a cross-sectional study conducted in Deli Serdang, North Sumatera. A sample of 190 study subjects was selected for this study comprising of 95 TB patients and 95 non-TB patients. The dependent variable was tuberculosis. The independent variables were sex, education, BCG immunization, smoking, nutritional status, and dwelling density. Data on TB status were taken from medical record. The other variables were measured by questionnaire. The data were analyzed by a multiple logistic regression.Results: The risk of TB decreased with female sex (OR= 0.33; 95% CI= 0.11 to 0.98; p= 0.046). The risk of TB increased with low education (OR= 8.47; 95% CI= 3.01 to 23.80; p&lt;0.001), had no BCG immunization (OR= 8.86; 95% CI= 3.28 to 23.94; p&lt;0.001), smoking (OR 6.69; 95% CI= 2.20 to 20.38; p=0.001), high dwelling density (OR= 10.35; 95% CI= 4.06 to 26.41; p&lt;0.001), and poor nutritional status (OR= 3.44; 95% CI= 0.85 to 14.03; p= 0.085).Conclusion: The risk of TB decreases with female sex, but increases with low education, had no BCG immunization, smoking, high dwelling density, and poor nutritional status.Keywords: Tuberculosis, sex, BCG immunization, dwelling density, nutritional statusCorrespondence:
