Abstract

Carp is one of the leading freshwater aquaculture commodities in Indonesia. Further improvement of carp strains by the Research Institute for Fish Breeding (RIFB), Sukamandi, Indonesia, has produced a synthetic F2 carp. The strain is assembled from different strains of carps and has shown better growth and health characteristics. Considering that high environmental ammonia (HEA) has affected most carp grow-out systems, this study aimed to determine the performance of the synthetic carp populations in a high ammonia rearing environment. The treatments were rearing media of the synthetic carp seed populations added with and without (control) 200 mg/L NH4Cl arranged in three replicates. A total of 30 fish seeds/aquarium, weighed 10-15 g/fish, was used in the study. Dissolved oxygen levels were maintained above 2 mg/L using aeration. This study shows that higher tolerant carp populations had red blood cells of 232.66 ± 17.24 cells/mL, indicating a direct effect of high ammonia on red blood cell count (p<0.05). Cortisol levels of 80.90 ± 6.35 ng/mL in resistant carp indicate significant differences (p<0.05). The relative expressions of the HSP70 gene in the liver (Log10) ranged between 0.72 and 2.80. The values demonstrate that high ammonia-resistant synthetic carp have a higher relative expression ratio of the HSP70 gene than the less resistant group. This research concluded that the populations of synthetic F2 carp showed a degree of resistance against high-ammonia rearing conditions. When it is ready for aquaculture, this synthetic carp strain could be farmed in high density using intensive systems in HEA-affected artificial lakes and reservoir

Abstract

Indonesian leaffish, Pristolepis grootii (Bleeker, 1852), is an undomesticated freshwater fish species native to the rivers, flooded swamps, and tributaries of Indonesia. The fish is mainly captured for consumption. In order to prevent its extinction and supply its growing demands, the artificial breeding of the fish should be developed. The purpose of this study was to determine the optimum dose of luteinizing hormone-releasing hormone analog (LHRHa) for stimulating the female P. grootii gonadal development at a dosage of 0, 1, 10, and 50 µg kg-1 of fish. Female fish (20.0 ± 0.6 g) were adapted for 30 days in the rearing environment and then separated into 12 aquariums with six fish per aquarium. Fish were then reared for another 21 days and fed with Tubifex sp. The LHRHa injection was conducted twice on day-7 and 14. Fish bodyweight, gonadosomatic index, gonad histology, blood estradiol-17â, and FSH-â and LH-â gene expression were evaluated at day 0, 7, 14, and 21. The results showed that the injection of the LHRHa hormone stimulated the development of fish gonads and was better achieved with a higher concentration of LHRHa. The best treatment was observed by the administration of 50 µg kg-1 of LHRHa that produced the fastest development among all treatments. This study demonstrated that the LHRHa induction could potentially stimulate the gonadal development of the newly domesticated fish. To our knowledge, this is the first study that reported the success of the induction of female gonad development in the Indonesian leaffish P. grooti.KEYWORDS:

Abstract

Koi herpesvirus (KHV) is one of the major pathogen for koi and common carp which cause high mortality and economic losses for the farmer. The purpose of this study was to determine the efficacy of glycoprotein-11 (GP-11) KHV DNA vaccine and compared to GP-25 KHV DNA vaccine. The vaccine in the form of naked DNA plasmidwas delivered by intramuscularly injection to the 3-month-old koi. The fish were divided into six groups, i.e. unvaccinated group (negative control C- and  positive control C+), and vaccinated group (2.5 μg/100 μl of GP-11 (group 1), 7.5 μg/100 μl of GP-11 (group 2), 12.5 μg/100 μl of GP-11 (group 3), and 12.5 μg/100 μl of GP-25 (group 4)). At day 42 post vaccination, all fish of each groups were challenged by injecting KHV titre 10-3 FID50. Number of dead fish was counted everyday after the challenge until 30 days. The results showed that vaccinated fish were had survival rate of 83.33-93.33% (group 2, 3 and 4). It’s show that GP-11 KHV DNA vaccine has high efficacy. As conclusion, GP-11 DNA vaccine could be an alternative DNA vaccine for preventing KHV infection.

Abstract

White spot syndrome virus (WSSV) has caused mass mortality on tiger shrimp (Penaeus monodon) culture and adversely affects prawn industry worldwide including Indonesia. It is well known that the protein structure of WSSV plays an important role in the virus infection and morphogenesis process. A viral protein structure called VP-15 is located in the nucleocapsid of virion virus. The protein structure involves in the life cycle of WSSV in host cells. A gene encoding VP-15 could be involved in constructing the RNA interference (RNAi), so it is needed to isolate and characterize for RNAi technology purpose. The study was aimed to isolate and characterize the VP-15 from the infected WSSV tiger shrimp. The characterization of VP-15 was undertaken through assessment of nucleotide sequence, amino acid deduction, alignment nucleotide/protein searches using Genetyx and BLAST program, and dendrogram construction analysis. The results showed that VP-15 was successfully isolated in form of ORFDNA with a fragment size of 243 bp. The phylogenetic tree analysis revealed three clusters corresponding to the time (year) of isolates collection. The VP-15 consisted of 80 amino acids, two start codons (ATG), one stop codon (TAA), and one Kozak context (AAAATGG). Hydrophilic amino acid was the highest composition (44.2%), followed by neutral (31.2%) and hydrophobic (24.6%) amino acid groups. The VP-15 was rich in amino acid of lysine (21.3%), arginine (22.9%) and serine (24.6%). The successful isolation of VP-15 is a very important step in providing a basic yet suitable material in constructing the dsRNA vaccine to control shrimp diseases in aquaculture.

Abstract

Abstract

Perlakuan Pre-0,5% menunjukkan rata-rata tingkat ekspresi gen tertinggi, namun performa pertumbuhan, dan sintasan tidak berbeda (p>0,05) dengan perlakuan Pre-2%.Fructooligosaccharides (FOS) are natural feed additives with potential application in feed for Pacific white shrimp. This study aimed to evaluate the effects of prebiotic FOS supplementation on the modulation of metabolism-related gene expression and growth performance of Pacific white shrimp. A trial feed consisted of commercial feed supplemented with different doses of FOS: Pre-0% (control), Pre-0.5%, Pre-1%, and Pre-2%; each with triplicate. Pacific white shrimp with an average body weight of 1.58 ± 0.21 g were reared in fiber tanks (d=1.5 m3) with a density of 100 shrimp/tank for each treatment. The shrimp were given the treatment feed five times per day at 6% of the total body mass for 30 days. Shrimp body weight was measured every ten days (n=10). The gene expression level was measured from the hepatopancreas (n=3) by the quantitative-real time PCR (qPCR) method. The results showed that FOS supplementation increased the metabolism-related gene expression levels. FOS supplementation improved the average body weight, average daily growth, and survival higher than control (p<0.05). Pre-0.5% treatment showed the highest average gene expression despite growth performance and survival were not significantly different (p>0.05) compared to Pre-2% treatment. This study concludes that the application of FOS in the feed improves the growth performance of Pacific white shrimp, notably in gained weight, reduced FCR and improved survival rate.
