Abstract

The main problem in the culture of clown loach (Chromobotia macracanthus) is the slow growth rate, which takes about six months to reach its market size (two inches total body length). Slow growth eventually cause a long production time and increase the production costs. An alternative solution can be proposed in order to enhance the growth is by using recombinant growth hormone. The aim of this study was to determine the immersion dose of recombinant Epinephelus lanceolatus growth hormone (rElGH) which can generate the highest growth in clown loach. Larvae at seven day after hatching were hyperosmotic treated with NaCl 2.0% for one minute, then immersed for one hour in water containing 0.3% NaCl, 0.01% bovine serum albumin (BSA), and different doses of rElGH, namely: 0.12 (treatment A), 1.2 (B), 12 (C), and 120 mg/L (D). As control, fish were immersed in water without rElGH and NaCl (control-1), water containing 0.3% NaCl and 0.01% BSA (control-2), and 0.3% NaCl water (control-3). Each treatment was replicated three times. The results showed that clown loach juveniles in treatment B, C, and D had longer total body length (P<0.05) than control-1, while fish treatment A was the same as controls. Survival and body weight were similar in all treatments and controls (P>0.05). In addition, the percentage of large size juveniles increased approximately 5% in treatment B, almost the same as in the medium size, while the small size were decrease compared to the control-1. Thus, the best immersion dose of rElGH was 1.2 mg/L water.

Abstract

Indonesian leaffish, Pristolepis grootii (Bleeker, 1852), is an undomesticated freshwater fish species native to the rivers, flooded swamps, and tributaries of Indonesia. The fish is mainly captured for consumption. In order to prevent its extinction and supply its growing demands, the artificial breeding of the fish should be developed. The purpose of this study was to determine the optimum dose of luteinizing hormone-releasing hormone analog (LHRHa) for stimulating the female P. grootii gonadal development at a dosage of 0, 1, 10, and 50 µg kg-1 of fish. Female fish (20.0 ± 0.6 g) were adapted for 30 days in the rearing environment and then separated into 12 aquariums with six fish per aquarium. Fish were then reared for another 21 days and fed with Tubifex sp. The LHRHa injection was conducted twice on day-7 and 14. Fish bodyweight, gonadosomatic index, gonad histology, blood estradiol-17â, and FSH-â and LH-â gene expression were evaluated at day 0, 7, 14, and 21. The results showed that the injection of the LHRHa hormone stimulated the development of fish gonads and was better achieved with a higher concentration of LHRHa. The best treatment was observed by the administration of 50 µg kg-1 of LHRHa that produced the fastest development among all treatments. This study demonstrated that the LHRHa induction could potentially stimulate the gonadal development of the newly domesticated fish. To our knowledge, this is the first study that reported the success of the induction of female gonad development in the Indonesian leaffish P. grooti.KEYWORDS:

Abstract

White spot syndrome virus (WSSV) has caused mass mortality on tiger shrimp (Penaeus monodon) culture and adversely affects prawn industry worldwide including Indonesia. It is well known that the protein structure of WSSV plays an important role in the virus infection and morphogenesis process. A viral protein structure called VP-15 is located in the nucleocapsid of virion virus. The protein structure involves in the life cycle of WSSV in host cells. A gene encoding VP-15 could be involved in constructing the RNA interference (RNAi), so it is needed to isolate and characterize for RNAi technology purpose. The study was aimed to isolate and characterize the VP-15 from the infected WSSV tiger shrimp. The characterization of VP-15 was undertaken through assessment of nucleotide sequence, amino acid deduction, alignment nucleotide/protein searches using Genetyx and BLAST program, and dendrogram construction analysis. The results showed that VP-15 was successfully isolated in form of ORFDNA with a fragment size of 243 bp. The phylogenetic tree analysis revealed three clusters corresponding to the time (year) of isolates collection. The VP-15 consisted of 80 amino acids, two start codons (ATG), one stop codon (TAA), and one Kozak context (AAAATGG). Hydrophilic amino acid was the highest composition (44.2%), followed by neutral (31.2%) and hydrophobic (24.6%) amino acid groups. The VP-15 was rich in amino acid of lysine (21.3%), arginine (22.9%) and serine (24.6%). The successful isolation of VP-15 is a very important step in providing a basic yet suitable material in constructing the dsRNA vaccine to control shrimp diseases in aquaculture.

Abstract

Tanaman obat telah banyak digunakan sebagai bahan pencegah dan pengobatan penyakit pada ikan budidaya. Penelitian ini ditujukan untuk mengetahui status kesehatan ikan lele (C. gariepinus) yang menerima pakan dengan suplemen tanaman obat kombinasi dari daun sirih, jambu biji, dan kipahit melalui pengamatan gambaran darah dan histologi ginjal sebagai organ yang memproduksi darah. Kombinasi satu merupakan kombinasi dari ketiga daun tanaman obat masing-masing sebanyak 33%, kombinasi dua juga terdiri dari daun sirih, jambu biji, dan kipahit masing-masing sebanyak 5%:19%:76%, dan kontrol yaitu pakan tanpa penambahan tanaman obat. Gambaran darah dan histologi ginjal diamati pada minggu ketiga setelah pemberian pakan. Hasil pengamatan gambaran darah menunjukkan bahwa terdapat peningkatan jumlah sel darah merah pada ikan yang menerima pakan perlakuan dibandingkan dengan kontrol (0,4 ± 0,14). Namun tidak terdapat perbedaan nyata antara jumlah sel darah merah dari kelompok perlakuan kombinasi satu dan dua (1,5 ± 0,17 dan 1,4 ± 0,1). Jumlah sel darah putih pada kelompok perlakuan juga meningkat dibandingkan dengan kelompok kontrol (10,5 ± 0,46), namun tidak terdapat perbedaan nyata antara kelompok perlakuan kombinasi satu dan dua (15,1 ± 1,19 dan 17,6 ± 1,14). Hasil pengamatan histologi terlihat jaringan hematopoietik organ ginjal dari kelompok yang menerima perlakuan berproliferasi lebih banyak dibandingkan kelompok kontrol. Namun tidak ada pengaruh pada nilai hemoglobin dan persentase leukosit diferensiasi antara kelompok perlakuan dan kontrol. Penambahan daun tanaman obat dalam pakan ikan mampu meningkakan status kesehatan dari ikan lele.Medicinal herbs have been traditionally used as prophylactic and therapeutic supplement to treat diseases in aquaculture. This study was aimed to improve the health quality of catfish (C. gariepinus) through feeding on diets enriched with a combination of betel, guava, and tithonia as medicine by analyzing hematology and histology of kidney as blood producing organ. Diet-one was feed enrich with 33% of each plant. Diet-two was feed enriched with betel, guava, and tithonia at a proportion of 5%,19%, and 76%, respectively. Control diet was fed without the plants’ supplementation. Hematology and histology of fish kidney were observed after fish received three-week feed treatments. The results showed that there was an increase of erythrocyte levels in the treated fish groups fed with diet-one and diet-two compared with the control (0.4 ± 0.14). However, no significant differences of erythrocyte level were observed between fish groups fed with diet -one and die-two (1.5 ± 0.17 and 1.4 ± 0.1). Leucocyte levels also increased in the treated fish group with diet-one and diet-two compared to the control (10.5 ± 0.46). However, there was no significant difference of leucocyte level between the fish group feed with diet-one and diet-two (15.1 ± 1.19 and 17.6 ± 1.14). Histological observations found that there were more hematopoietic tissues in the fish kidney of proliferated treated group than the control group. However, there was no effect on hemoglobin level and leukocyte percentage differentiation between the treatment and control groups. This study concludes that medicinal herbs as enrichment ingredients in fish diet can increase the health quality of fish.

Abstract

The main problem of this research is the management of zakat, infaq, and alms in alleviating poverty in the pandemic period, study at LAZISNU Makassar City in 2020. The sub-problems are how is the management system of zakat, infaq, and alms at LAZISNU in alleviating poverty in Makassar City, and what are the obstacles or obstacles for LAZISNU in the management of zakat, infaq, and alms. The method used in this study is a qualitative method with a syar'i, social, and juridical normative approach. The results of this study indicate that in compiling the ZIS fund management program carried out by LAZIZNU Makassar City is good enough, to reduce the suffering of the poor, it affects the poverty rate to be reduced. However, there are obstacles that hinder management, one of which is the spread of Covid-19. Keywords: ZIS Management, Poverty, Pandemic Periode.

Abstract

The most common problem in barramundi Lates calcarifer seedling production is the high mortality (> 90%) caused by nervous necrosis virus (NNV) infection. This research aims to evaluate the resistance and gene expression of barramundi challenged by NNV. Two populations were used in this study, i.e., Australian, and Situbondo-originated barramundi populations. The immune-related gene expression levels in the liver, head of kidney, and spleen were observed at 48 and 96 hours after post-infection (hpi). Barramundi’s survival and blood parameters were evaluated post-NNV infection. The results showed that the highest survival was revealed in Situbondo’s barramundi (42.0±4.47%) compared to Australian barramundi (20.0±7.07%) and no mortality was observed in the control without NNV infection. The higher survival rate in barramundi from Situbondo was in line with the blood profile. The number of red blood cell from Situbondo barramundi post-NNV infection (ST) at 96 hpi was higher (P<0.05) than Australian barramundi post-NNV infection (AT). The number of white blood cell of ST at 48 hpi was higher (P<0.05) than AT, but started to decrease at 96 hpi in ST barramundi. The total white blood cell in AT barramundi increased from 48 to 192 hpi. TNFα and IL1-β gene expression levels were significantly higher in the liver, head kidney, and spleen of Situbondo compared to Australian barramundi at 48 hpi, while MHCIIα gene expression in Situbondo’s was significantly higher compared to Australian barramundi at 96 hpi. These results indicate the important roles of all the genes in the barramundi’s immune responses against viral infection. Based on the results of the research, Situbondo’s barramundi has the potential to be used as a candidate for generating broodstock of disease-resistant strain.

Abstract

Abstract The main issue is mediation in the sttlement of inheritance disputes at the Makassar Religious Court Class I A, the subject matter is divided into three sub-problems, namely: 1) How is the ,ediation process in the sttlement of inheritance disputes in PA Makassar Class IA. 2) What is the role of the mediator in the sttlement of inheritance disputes in PA Makassar Class IA. 3) What are the results of mediation in the settlement of inheritance disputes in PA Makassar Class IA. The type of research is descriptive qualitative, namely field research, using a normative juridical and empirical juridical approach with interview and documentation data collection methods. Based on the results of the study, it shows that the mediation process is divided into three stages, namely, the pre-mediation stage, the mediation process stage, and the final stage of implementation. The prsence of he parties is very important for the continuation of the mediation. The mediator must be equipped with psychological knowledge in addition to psychological knowledge of law. The succes in the mediation in the settlement of inheritance disputes at the Makassar Religious Court Class IA is still less effective, as for the causes of the low result obtained, one of which is the complexity of an inheritance case, the obligation to present parties who are not only two or three but can be more than that. The implication of the research is that the mediation process is very important for the mediator to provide a sense of security, peace, and peace to the parties. The mediator must improve the ability to mediate the parties, namely not only from the rules but also the ability to dialogue with the parties. The good faith of the parties is also very influential on the success of the mediation, that is, it is hoped that the partis can regenerate good relations and undergo the mediation process in good faith.

Abstract

Bank Wajib menjaga segala sesuatu yang berhubungan dengan keterangan nasabah penyimpan dan simpanannya, salah satunya ialah data nasabah, di mana anggota direksi, pengurus bank, pegawai bank, dan pihak terafiliasialinnya wajib menjaga hal tersebut. Sistem perbankan di Indonesia menerapkan teori rahasia bank yang bersifat relatif artinya rahasia bank hanya dibuka ketika ada alasan mendesak yang untuk kepentingan masyarakat dan negara. Sehingga menurut Perundang-Undangan pihak bank dan pihak terafiliasi dilarang membuka rahasia bank. Namun bagaimana Indonesia mengatur tentang kewajiban menjaga rahasia bank bagi mantan pegawai bank. Tujuan dari penulisan ini untuk mengetahui peraturan tentang kerahasiaan data nasabah bagi mantan pegawai bank. Penelitian ini menggunakan metode penelitian normatif dengan mengambil kesimpulan bahwa belum adanya pengaturan tentang kewajiban menjaga kerahasiaan data nasabah bagi mantan pegawai bank dalam UU Perbankan di Indonesia sehingga memunculkan celah hukum. Menjaga kerahasiaan data nasabah bagi mantan pegawai bank hanya dapat dibenarkan dengan kode etik sebagai kewajiban moral.

Abstract

Perlindungan terhadap data pribadi menjadi suatu hal yang mendasar sebab data pribadi secara khusus berhubungan dengan privasi right seseorang. Kewajiban untuk melindungi data pribadi tidak hanya sebatas kewajiban suatu negara namun bank sebagai lembaga yang berdiri diatas kepercayaan masyarakat, juga di wajibkan untuk menjaga keberadaan data pribadi yang dikelolanya. Pentingnya bank untuk menjaga data pribadi nasabah tidak hanya sebatas melindungi data pribadi nasabah penyimpan yang sifatnya rasia bank namun termasuk juga kewajiban untuk menjaga menjaga data pribadi nasabah debitur yang tidak termasuk kategori rahasia bank. Tujuan penelitian ini untu menguraikan bagaimana perlindungan hukum terhadap data pribadi nasabah debitur Indonesia. Metode penelitian yang digunakan adalah tipe penelitian normatif, dengan menggunakan pendekatan konseptual. Hasil dari penelitian adalah mengemukakan perlindungan hukum bagi data pribadi nasabah debitur, bentuk-bentuk kejahatan terhadap data pribadi nasabah debitur.

Abstract

Uang panai a significant role in the smooth conduct of marriage. In Bugis culture, giving uang panai is an obligation in marriage. This study utilizes a systematic literature review (SLR) of several articles on uang panai in South Sulawesi. The findings of this research indicate several factors that influence the determination of uang panai, such as noble lineage, education, economy, and women's occupation. The agreement on the value of uang panai should avoid imposing excessive burdens on the groom while respecting the dignity and position of the bride, facilitated by effective communication between both parties.
