Abstract

Availability of moleculer marker to identify the important agronomic character of plant is needed to accelerate selection activity of plant. Particularly for the complex quantitatively inherited traits - like e.g. shading tolerance of soybean - the use of such a technique will speed up the process to produce adapted genotypes.The objective of this research is to identify the linkage of molecular marker RAPD with character of shading tolerance. The morphological specific characters wich is corelated to shade tolerance is the number of productive branchs. The intensity 75% of artificial shading is optimal level for doing selection of soybean genotypes. The inheritance of shading tolerance of soybean was controlled by gene with full dominant or by two genes pairs with dominant and ressesive epistasis. There was no maternal effect in the inheritance to shade tolerance. Heritability value (0.45 – 0.54) indicated that the proportion variation caused by the genetic factors was moderate. Molecular analysis by using RAPD technique showed that UBC153, ROTH 480.01, and ROTH 480.03 primer have polymorphic band that can be used for inheritance study and linkage analysis. All polymorphisms segregated independently of each ather. Interval mapping with Mapmaker/QTL indicated that the location of the three QTLs on linkage group were at marker tolerance locus of Roth 480.01-8125 , Roth 480.03-1125, and UBC 153-19125 .

Abstract

The objective of this study were to obtain information about genetic parameters of agronomic characters of sorghum populations derived from Single Seed Descent (SSD) method, and to estimate selection differensial and also to select of F4 population that have a high yield and medium plant height . The experiment was carried out at Leuwikopo Farm, Darmaga, Bogor from January to April 2014. The genetic material were F4 population derived from SSD. The results showed that based on the skewness and kurtosis values, all of the agronomy characters observed in this study were poligenic controlled and influenced by additive gene action. The genetic coefficient of variability values for all parameters varied from intermediate to high in F4 population. Agronomic characters in F4 responded positively to selection because of high broad sense heritability estimates. Correlation analysis showed that plant height, number of leaves, stem diameter, panicle length, panicle weight and 100 seed weight have significant and positive correlation to grain yield/plant. Differential value with selection intensity 10 % based on grain yield/plant will increase grain yield/plant 83.89 % and plant height 8.91 % in the next generation, whereas differensial value based on grain yield/plant and plant height will increase grain yield/plants 68.33 % and plant height 0.26 % in the next generation. It means that selected plant were expected increasing yield 83.89 % or 68.33 % in next generation.Keywords: heritability, cefficient genetic variability, correlation analysis, selection differential

Abstract

The study was aimed to determine the productivity and yield stability of ratoon rice genotypes in three environments and to obtain rice genotypes suistable for ratoonning on specific environment of tidal land. The experiment was designed in a randomized complete block with three replicates. Seedling was planted at 21 days old with spacing of 20 x 20 cm, one seedling per hill. Plants were harvested 30 days after heading by cutting at a height of 10 cm from the soil surface. One day after harvest, the land was irrigated as high as 3 cm and fertilized with Urea 50 kg/ha, TSP 30 kg/ha, and KCl 25 kg/ha. Yield stability was analyzed according to Eberhart and Russel (1966). Ratoons were sensitive to the environmental changes. Genotype IPB97-F-13-1-1, IPB 4S, and IPB 3S produced the main crop + ratoon grain yield of 5.26 t/ha, 5.14 t/ha, and 5.64 t/ha dry milling grain (DMG), respectively. Based on yield of the main crop + ratoon, IPB97-F-13-1-1 and IPB 4S was each considered as adaptable to the suboptimum condition (bi<1). Ratoon crop yield of genotype IPB97-F-13-1-1, IPB 4S, and IPB 3S each was the highest in three locations. The ratoon crop contribution to the rice production was from 31.3% to 61.9% to the main crop.

Abstract

Characterization of mutant wheat (Triticum aestivum L.) lines is a step on the breeding program to determine the beneficial characters for increasing the productivity in tropical lowland. The aim of this research was to obtain information on the variability of morphological, anatomical, and physiological characters that could be used as selection criteria and to obtain adaptive mutant lines of “Alibey” in tropical low altitude land. Research was conducted at the Experimental Farm of SEAMEO-BIOTROP in Bogor 250 m above sea level, from April to December 2013. Mutant lines of “Alibey” consisted of 16 M3 mutants resulted from treatments of EMS. LC50 of “Alibey” at 0.1% EMS for 60 minutes. Results showed that the mutant lines changed their morphological traits significantly, as indicated by the four characters i.e. long stem panicle (8 mutants), grain weight/panicle (1 mutant), weight of 100 seeds (4 mutants) and seed weight/plant (9 mutants). However, the mutant had no significant effect on the nine other characters, including: time of flowering, days to maturing, panicle length, plant height, number of tillers, panicle number, and leaf area. Anatomical characters namely leaf thickness and stomata size showed different values between “Alibey” mutant (AB-0.1.60-1-7-1) and the original Alibey. For the physiological characters there were significant differences among mutants with respect to the amount of proline and glucose levels. Proline level in the control plant was 4.15 ug/g BB, while that in mutant “AB-0.1.60-3-16-1” was 263.47 µg/g BB, and that in “AB-0.1.60-3-3-2” was 235.90 µ/g BB. Likewise, glucose level in control was 132.88 mg/ml, while in mutant “AB-0.1.60-3-16-1” was 181.48 mg/ml, and that in “AB-0.1.60-3-3-2” was 287.41 mg/ml. “Alibey” mutants should be selected based on two characters i.e. stem panicle length and seed weight/plant. Correlation analysis between panicle number and all other characters were not significant. Plant height significantly affected the grain weight/panicle and the grain weight/plant. It is expected that some of the mutants are adaptable to the tropical lowlands, so that the diversity of wheat germplasm in Indonesia is increased.

Abstract

In vitro selection using of salt (NaCl) has been used to select plants that tolerate salinity, so that effective selection process. The purpose of this study was to obtain information about the mechanism of salinity tolerance in rice in vitro conditions. This study consisted of two experiments, they are the selection of salinity tolerance somaclone putative mutants in vitro and study of salinity tolerance mechanism results in in vitro selection. The tolerant plants after being treated NaCl show response to the accumulation of proline is more, the content of K, Mg and Ca which tends to remain and Na content is lower, stomata density is fewer and the length and width ratio stomata is greater.

Abstract

We investigated J. curcas responses to aluminum stress, histochemically and biochemically. Histochemical stainings were observed to analysis aluminum accumulation, lipid peroxidation and the loss of plasma membrane integrity on the surface and tissue of the root apex. Enzymatic analysis was conducted to measure malate content in leaf, root and malate efflux in the medium. We used M. malabathricum as a comparison for Al-tolerance plant. J. curcas root elongation was inhibited by 0.4 mM AlCl3, while M. malabathricum root elongation was inhibited by 0.8 mM AlCl3 treatment. Inhibition of root elongation has high correlation with Al accumulation in the root apex, which caused lipid degradation and cell death. Generally, malate content in J. curcas leaf and root was higher than that in M. malabathricum. In the contrary malate efflux from the root into the medium was lower. J. curcas root has a different pattern compared to M. malabathricum in malate synthesis and malate secretion when treated with a different Al concentration. We categorized J. curcas acc IP3 as more sensitive to aluminum than M. malabathricum.

Abstract

Plant breeding program consists of establishment of the population, selection, and evaluation. The study aimed to observe the variability of yield components, the heritability, and the distribution of the yield component characters in the F3 populations. The experiment was conducted at Muara Experimental Farm Bogor, from April to August 2012 on Latosol soil. The F3 populations derived from crosses of Bintang Ladang x US2, Gampai x IR77674, and Progol x Asahan and their parental were used as plant materials. Twenty one-day-old seedlings from each population were planted in plots of 2 x 12 m, with planting space of 20 x 20 cm and 3-5 seedlings per hole. Panicle length and weight were observed on 300 randomly selected plant samples from each population. The results showed that there was a variation of agronomic characters among genotypes. The heritability of characters, the panicle length and weight was low to high. Panicle length and weight were controlled by many genes with additive gene action in the Gampai x IR77674 derived population, while panicle weight was controlled by few genes with complementary epistatic additive gene action in both Bintang Ladang x US2 and Progol x Asahan derived populations. The more genes controlling a character, the more distribution classes formed and the greater variance among genotypes.

Abstract

The research on adaptation selection for utilization in phytoremediation of soil contaminated by crude oil using four type plants, such as Helianthus annuus, Paspalum conjugatum, Sorghum bicolor, and Tagetes erecta were conducted. The adaptability of four types of plants on crude oil contaminated soil at total petroleum hydrocarbon (TPH) level at 0, 3, and 6% were observed and evaluated to their morphological and anatomical responses. Parameters observed were vegetative growth of plants including growth precentage, plant height, number of leaves, root length, root dry weight, shoot dry weight, root/shoot ratio, total dry weight, and stomatal density for 9 weeks cultivation in screen house. The results show that increasing in TPH level caused in significant reductions on morphological of four plants, such as percentage of plant growth, plant height, number of leaves, root length, root dry weight, shoot dry weight, and total dry weight. In contrast, the increasing in TPH level caused to increasing in root/shoot ratio. The four types of plants studied were effective to be used as plants for phytoremediation of petroleum contaminated soil. The plants of P. conjugatum and S. bicolor are recommended as phytoremediators for further studies.

Abstract

This study aimed to find out the lead tolerance of sengon (Paraserianthes falcataria) seedling based on growth performance, tolerance index, and ecretion and accumulation of organic acids content. Seedlings were exposed to lead (Pb) with the concentration of 0, 0.5, 1, 1.5, 5, and 10 mM in liquid nutrient culture for 4 days in order to investigate secretion and accumulation of oxalic, malic, and citric content, and for 15 days to examine growth performance and olerance index. The result showed that tolerance index and growth performance of engon seedling were insignificant (p > 0.05) to the rising of Pb concentration up to 1.5 mM with tolerance index at least 95%, and even caused an increase of fresh weight. However, the tolerance index and growth of sengon decreased significantly due to Pb exposure of 5 and 10 mM. Among the three organic acids, citrate was most dominant as compared to malate and oxalate. Secretion of citrate increased significantly (p < 0.05) with the rising concentration of Pb 0.5, 1 and 1.5 mM, reaching to 0.464, 0.540, and 0.587 µg mℓ-1, respectively, or rising according linear line (r = 0.9, p < 0.5). Citrate accumulation showed inconsistent pattern with the rising Pb exposure. The result suggested that sengon seedling have a slightly tolerance to lead by secretion of organic acid especially citric acid.

Abstract

ABSTRACTThe yellow sap is produced naturally in mangosteen organ except in the root. The yellow sap contaminated the aryl and rind if the epithelial cell walls rupture due to deficiency of calcium (Ca). Calcium is one of structural component of cell walls, whereas naphthaleneacetic acid (NAA) has its role in improving cell division and cell elongation. Interaction of Ca and NAA can improve sink strength and capacity because the newly formed cells need Ca to construct wall structure. This experiment aimed at finding out the effect of Ca and NAA applications in reducing the contamination of yellow sap in mangosteen. The experiment was conducted by using factorial random block design consisting of 2 factors and 3 replications. The first factor was Ca dosage (0 and 4.8 kg/tree), and the second factor was NAA concentration (0, 200, 400 and 600 ppm). The results showed that application of 4.8 Ca/tree and 200 ppm NAA as much as 5 ml / fruit effectively improve the content of Ca pectate in pericarp, reduced the percentage of yellow sap contamination on the fruit segment, aryl and rind to 0% and 12.3% respectively compared to control (17.8% on fruit segment, 36.8% on aryl and 56.1% on rind).Key words: aryl, Ca pectate, cell wall, middle lamela.ABSTRAKGetah kuning adalah getah yang dihasilkan secara alami pada setiap organ manggis, kecuali pada akar. Getah kuning akan keluar dan mencemari aril serta kulit jika dinding sel epitel pecah karena kekurangan kalsium (Ca). Kalsium adalah komponen dinding sel, berperan dalam struktur dan permeabilitas membran sedangkan asam naftalenasetat (NAA) berperan penting dalam meningkatkan pembelahan dan pembesaran sel. Interaksi keduanya dapat meningkatkan kapasitas sink buah karena sel yang baru terbentuk membutuhkan Ca dalam menyusun struktur dinding sel. Percobaan bertujuan untuk mengetahui pengaruh aplikasi Ca dan NAA dalam menurunkan cemaran getah kuning manggis. Percobaan menggunakan rancangan acak kelompok faktorial 2 faktor dengan 3 ulangan. Faktor ke-1 yaitu dosis Ca (0 dan 4.8 kg Ca/pohon) dan faktor ke-2 yaitu konsentrasi NAA (0, 200, 400 dan 600 ppm) dengan volume semprot 5 ml perbuah. Hasil percobaan menunjukkan bahwa aplikasi 4.8 kg Ca/pohon dengan NAA 200 ppm sebanyak 5 ml/buah efektif meningkatkan kandungan Ca pektat perikarp dan menurunkan persentase buah tercemar getah kuning menjadi 0% pada juring dan aril serta 12.3% pada kulit dibandingkan dengan perlakuan kontrol (17.8% pada juring, 36.8% pada aril dan 56.1% pada kulit buah).Kata kunci: aril, Ca pektat, dinding sel, lamela tengah
