Abstract

An experiment to compare age of Macaca fascicularis (Mf) as pheripheral blood (PB) mesenchymal stem cell (MSC) isolate sources and the impact of its concentration on the pheriperal blood mononucleous cells (PBMC) development has been conducted. Twelve male Mf were used in this experiment. Three different age groups (infant (A1), juvenil (A2) and adult (A3)) of the Mfs were compared as treatments. Isolate of pheriperal blood MSC were created by taking 1 ml, 5 ml or 10 ml the Mfs pheriperal blood, processed them into PBMC, counted, isolated, cultured, subcultured, pelleted, extracted for their messenger Ribonucleic Acid (mRNA). Reverse transcriptase - polymerase chain reaction (RT-PCR) were conducted to obtain complentary Deoxyribonucleic Acid (cDNA). PCR amplification were performed to look cluster differentiation (CD) of the MSC gene expression. Incomplete block design was used and the data were analysed using descriptive statistic and T-Test. The results showed that PBMC counted from infant, juvenil and adult were 6.78 – 7.28, 6.18 – 7.30, and 6.01 – 7.34 log cell, respectively. The subculture and pelleting cells were only obtained from A3 with positive 73, 90, 105 and negative 34, 45 CD markers. It is concluded that pheriperal blood of adult Mf can be utilized as MSC source.

Abstract

Tujuan dari penelitian ini adalah mempelajari karakteristik gambaran sel-sel ephitelial vagina dan gejala klinis estrus pada banteng. Sebanyak 2 ekor banteng betina yang sehat, sudah dewasa kelamin dan pernah beranak diobservasi secara klinis selama siklus estrus normal dan dilakukan pengambilan sampel ulas vagina. Observasi gejala klinis estrus dan ulas vagina dilakukan tiga kali dalam seminggu dan 5 hari berturut-turut menjelang sampai sesudah estrus. Pemeriksaan fisik ini terdiri atas pemeriksaan simptomatis dengan melihat gejala-gejala yang timbul dari luar seperti gelisah, adanya kebengkakkan, kemerahan, kebasahan pada vulva, naik menaiki, melenguh (meskipun pakan cukup tersedia), keluar lendir jernih transparan. Tingkah laku ketika banteng sedang estrus memperlihatkan perilaku saling naik menaiki sesama banteng dan diam ketika dinaiki, sedangkan tanda klinis lainnya seperti kemerahan, kebengkakan, dan lendir serviks tidak terlihat dengan jelas. Sel-sel ephitel yang diperoleh adalah parabasal, intermediet, superfisial dan kornifikasi. Sel superfisial mendominasi (50,20%) pada saat estrus diikuti sel kornifikasi (38,51%), sel intermediet (26,82%) dan sel parabasal 13,44% selama 3 siklus estrus. Hasil penelitian ini dapat disimpulkan bahwa naik-menaiki sesama banteng pada saat estrus merupakan tanda klinis yang paling jelas terlihat dan teknik sitologi vagina dapat digunakan dalam penentuan siklus estrus banteng.

Abstract

Satwa primata merupakan salah satu hewan model yang banyak digunakan dalam penelitian biomedis karena secara anatomi dan fisiologis memiliki kesamaan dengan manusia. Darah merupakan salah satu cairan tubuh ekstraseluler yang berfungsi sebagai alat angkut atau media transport dan pertahanan tubuh. Gangguan keseimbangan yang terdapat pada setiap variabel kuantitatif dan kualitatif dari jenis sel-sel darah dapat menyebabkan validitas suatu penelitian menurun. Penelitian ini bertujuan untuk mendapatkan data dasar hematologi Macaca siberu dan untuk mendapatkan informasi kekerabatan melalui data sekunder hematologi secara kuantitatif dan kualitatif dari Macaca leonina, dan Macaca nemestrina. Sampel diambil dari 8 ekor Macaca siberu (5 jantan dan 3 betina) dengan umur yang bervariasi dan tergolong sehat serta telah dipuasakan. Analisis sampel yang dilakukan yaitu berupa pemeriksaan diferensial leukosit, pemeriksaan kuantitatif sel-sel darah, dan indeks eritrosit yang dianalisis secara deskriptif. Profil hematologi pada Macaca siberu secara keseluruhan menunjukkan hasil yang tidak jauh berbeda dengan Macaca nemestrina dan Macaca leonina. Variasi nilai yang ditemukan dapat dipengaruhi oleh faktor intrinsik seperti jenis kelamin, nutrisi, aktivitas dan stres, serta faktor ekstrinsik seperti suhu, ketinggian, dan letak geografis.

Abstract

Pengukuran estron terkonjugasi (E1C) dan pregnanediol glucuronat (PdG) di feses dilakukan pada tiga ekor Owa Jawa (Hylobates moloch AUDEBERT 1797) betina untuk mengevaluasi fungsi ovarium ketiga owa tersebut yang masing-masing dipelihara di kandang individu di Pusat Studi Satwa Primata IPB dan di Taman Margasatwa Ragunan, Jakarta.. Contoh feses dikoleksi secara regular pada jam 07.00-09.00, 5-7 kali seminggu selama 4-9 bulan. Setelah melalui proses pengeringbekuan, contoh feses diekstraksi menggunakan metanol dan asai E1C dan PdG dilakukan dengan enzyme immunoassay (EIA). Profil hormon yang diperoleh menggambarkan adanya satu betina yang bersiklus dengan panjang siklus 21-25 hari. Lama fase folikular yang diperoleh pada betina tersebut bervariasi antara 11-18 hari, dan fase luteal yang relative konstan dengan lama 8-12 hari. Akan halnya dua betina yang lain, tidak diperoleh adanya gambaran pola yang menunjukkan siklus ovarium yang regular. Pembengkakan organ kelamin luar yang berfluktuasi juga hanya teramati pada betina yang bersiklus dengan lama pembengkakan 3-5 hari, dan perdarahan menstruasi terjadi selama 2-3 hari. Hasil yang diperoleh menunjukkan bahwa pengukuran steroid di feses merupakan suatu metoda nonivasif yang dapat diterapkan untuk mengevaluasi fungsi ovarium Owa Jawa. Keadaan fisiologis dari individual betina mungkin menjadi bahan pertimbangan lain dibandingkan tipe perkandangan sebagai faktor yang mempengaruhi profil hormonal.Kata kunci: Owa Jawa, steroid di feses, pembengkakan organ kelamin luar

Abstract

Carbohydrate is a diverse compound in the body and can be  as glycoconjugate which bound to protein in proteoglycan and glycoprotein shape, and bound to lipid in glycolipid shape as well. The study was conducted to get information image of pattern distribution of the glycoprotein at stomach gland from various age levels at Macaca fascicularis (Mf) after and before bearing. Gastric specimen was collected processed through microtechnique procedure, followed by histochemistry Lectin staining to detect glycoprotein from stomach. Observation result of given score subjectively into four categories: (-) none/negatif, (+) low, (++) medium,and (+++) height. The results obtained were tabulated and analyzed diskriptively. We found that glycoproteins complex exist in the glandullar stomach of long tailed macaques are mannose, glucose, galactose, N-acetil-D-glocosamine and N-acetil-D-galactose all of these glycoproteins complex was observed at age foetus 70 days. Abstract

Curcuminoids of Curcuma manggo believed to have antioxidant properties that can prevent the occurrence of atherosclerosis.  However, the mechanism at the molecular level is not clearly known. Increase in intracellular adhesion molecule-1 (ICAM-1) will result in endothelial cell dysfunction that initiates athersclerosis. The expression of ICAM-1 can be triggered by oxidized LDL molecules. The purpose of this study is to elucidate the effects of curcuminoids temumangga (Curcuma mango) on the expression of ICAM-1 induction in endothelial cell culture. Prior to the induction of the expression of ICAM-1, endothelial cell culture was pre-incubated with curcuminoids extract for 48 hours with concentration of 2 ppm and 8 ppm, then further incubated with oxidized LDL for 24 hours. The treated cells were compared to 5 µM Cu2+ induced endothelial cell culture with or without LDL  from Macaca fascicularis.  ICAM-1 expression was determined by immunohistochemistry using anti-ICAM-1 antibodies and visualized with staining by 3.3-diaminobenzidin tetrahidrochloride (DAB). Results showed that surface-induced endothelial cell culture with ion Cu2+ stained well by DAB as brown yellow, which indicates ICAM-1 expression. Curcuminoids extract of  8 ppm reduced induction of ICAM-1 response. Abstract

Abstrak: Lima belas monyet ekor panjang obes (Macaca fascicularis) dengan resiko aterogenesis digunakan untuk memperoleh informasi tingkat low density lipoprotein (LDL) dan ekspresi protein uncoupling-1 (UCP-1) setelah tiga bulan intervensi nikotin dosis rendah per oral. Kelompok hewan dibagi menjadi tiga dengan komponen diet berbeda, yaitu diet A (lemak), diet tipe B (lemak dan kuning telur), dan diet tipe C (komersial diet-monyet chow), dan setiap kelompok terdiri dari lima monyet. Ketiga diet tersebut dicampur nikotin dan diberikan ke dalam makanan sehari dengan dosis 0,50-0,75 mg/kg/12 jam. Abstract

Abstrak: Diet berenergi tinggi dengan sumber dari lemak dapat mendorong perkembangan obesitas yang biasanya disertai dengan perubahan profil lipid darah. Tujuan dari penelitian ini adalah untuk mengevaluasi pengaruh diet energi tinggi dengan sumber lemak hewani dan kuning telur sebagai diet obesitas terhadap profil lipid darah monyet ekor panjang (Macaca fascicularis) yang diberikan selama empat bulan. Lima belas ekor monyet jantan dewasa dibagi menjadi tiga kelompok diet secara acak menjadi kelompok 1) lemak tinggi dari lemak hewani tanpa kuning telur, atau 2) lemak tinggi dengan kuning telur, (lemak 19,62% pada kedua diet), atau 3) diet standar monkey chow sebagai kontrol (lemak 5,55%). Pengukuran dilakukan tiap bulan untuk berat badan, profil lipid darah seperti kolesterol, trigliserida, kolesterol lipoprotein densitas tinggi (HDL-C) dan kolesterol lipoprotein densitas rendah (LDL-C), dan pada akhir studi, konsumsi lemak dan kecernaan dievaluasi. Abstract

Banteng is considered as endangered species by the International Union for Conservation of Nature (IUCN). Banteng is currently distributed in Java, in Kalimantan [Indonesian Borneo], Sabah [part of Malaysian Borneo], Myanmar, Thailand, Cambodia and, probably Lao PDR and Vietnam (1). The populations in the Asian mainland have decreased by about 80% in the last decades. The total number of wild banteng is estimated to about 5,000-8,000 animals. No population has more than 500 animals, only a few have more than 50. Reasons for the population decline are reduction of habitat, poaching, hybridisation with domesticated cattle, and infections with cattle diseases (4).Since the wild population has been declining continously and the numbers of individual are getting smaller, therefore captive breeding is urgently needed in order to support the conservation of Banteng sustainably. Although breeding program of Banteng is important, but information about the reproductive biology of Banteng is very limited. Breeding programs in Banteng can be done by natural mating or using assisted reproductive technology such as artificial insemination (AI). The aplication of AI in Banteng can support Banteng conservation program in order to increase heterozygosity, prevent inbreeding and possible to use of the Banteng bulls from different facilities for AI of females in all breeding facilities of Banteng.AI applications require information on the estrous cycle and estrous signs to determine the precise AI time. Clinical signs of estrous in Banteng is difficult to observe, therefore determining the time of AI must be done through rectal palpation and ultrasonography. The objectives of  this research is to assess estrous cycle to determine time of insemination.

Abstract

Banteng (Bos javanicus) is listed as endangered species in IUCN Redlist. Only few thousand wild banteng survive and their numbers are decreasing as a result of hunting and the loss of habitat due to deforestation and conversion to agricultural land. One of the conservation measures is to preserve this species in ex-situ breeding center. The breeding centre with the largest number of individuals is Ragunan Zoo in Jakarta, with 21 animals. Despite those numbers of individuals available, the sex ratio was far from ideal at 16 males and five females. While in Taman Safari Prigen, there were with 16 individuals and a sex ratio of five males and 11 females [1] Since number of males are usually less than females, and also to avoid inbreeding in order to keep high heterozygosity population, therefore, assisted reproductive technology such as artificial insemination is considered to be necessary.For artificial insemination, semen collection sufficient quantitiy and quality of semen from males are needed. Thus, best semen collection method should be considered. Though transrectal massage procedure does not require the male to be anesthetized  prior to semen collection, semen collected by this technique contains urine contamination. Besides, volume of semen sample obtained by transrectal massage is lesser compared to electroejaculation. Electroejaculation is also suitable for wild animal because in this particular semen collection procedure, males are not required to be trained.The information about semen characteristics of banteng bulls collected by electroejaculation is less known. To date, study done by Johnston et al. (2002) is the only available data. That study only provided data from one semen sample from one 4-year-old banteng bull located at Western Plains Zoo [2]. Moreover such volume parameter and consistency was not reported from that study. Thus, this study is aimed to provide more reliable data from larger number of semen samples  collected by electroejaculation and to add information to previous data that are availabe.
