Abstract

This study aimed to detect the presence of Salmonella sp. in broiler chickens raised in closed house systems using the Polymerase Chain Reaction (PCR) method. Liver and intestinal samples were collected from chickens showing clinical signs such as diarrhea, anorexia, and lethargy, along with pathological lesions observed during necropsy. Bacteriological identification involved isolation using selective media, Gram staining and biochemical tests. Two samples (3A and 3B) showed colony morphology and biochemical characteristics consistent with Salmonella sp. Confirmation using PCR targeting the invA gene (primers 139 and 141, expected amplicon size 284 bp) yielded negative results in both samples, while the positive control successfully amplified the target. The inconsistency between bacteriological and molecular results may be attributed to several factors: absence or mutation of the invA gene in the tested isolates, primer mismatch with local Salmonella strains, or inadequate DNA quality and concentration. These findings suggest that reliance on a single molecular marker may be insufficient for accurate detection and underscore the importance of optimizing PCR conditions. The study highlights the need for locally adapted primers and complementary diagnostic approaches to improve the reliability of Salmonella sp. detection in poultry, particularly in intensive production systems like closed house environments.

Abstract

Lead is a harmful pollutant from engine exhaust that causes free radicals and has detrimental effects on the testicular tissue. This study aimed to determine the effects of tomato juice on the number of Leydig cells and the diameter of the seminiferous tubules of mice exposed to lead acetate. Twenty-five male mice were divided into five groups. Mice in the control (C-) group were given placebos. Meanwhile, mice in C+, T1, T2, and T3 groups were exposed to lead acetate at a dose of 100 mg/kg BW/day for 14 days and given tomato juice respectively at 0, 0.16, 0.32, and 0.64 mL/day from day 8 to day 35. On day 36, all mice were sacrificed, and the testes were collected for histological preparation. The result showed that lead exposure in the C+ group decreased (p <0.05) in the number of Leydig cells and the diameter of the seminiferous tubules compared to the mice in group C-. Administration of tomato juice in groups T1, T2, and T3 increased (p <0.05) the number of Leydig cells and the diameter of seminiferous tubule compared to the mice in the C+ and C- groups. However, tomato juice administration to the T3 group decreased the number of Leydig cells and the diameter of the seminiferous tubules (p <0.05) compared to the T2 group. In conclusion, an effective dose of 0.32 mL/day of tomato juice restored Leydig cell number and seminiferous tubules diameter in mice exposed to lead acetate.

Abstract

Segunung is one of the six Dusun in Carangwulung, Wonosalam, Jombang, East Java where Sapi Perah Jaya and Wanita Tani Perah Jaya groups manage to breed 210 dairy cows. The milk produced is deposited to KUD Anjasmoro before being transferred to the milk processing industry and some is processed by Wanita Tani Perah Jaya into pasteurized milk. The problems faced by dairy farmers, especially after the Foot and Mouth Disease (FMD) outbreak, are the decreasing milk and feed production, and livestock health. The problem faced by Wanita Tani Perah Jaya is a lack of knowledge about dairy product verification and marketing. To solve this problem the researchers administer nano meniran extract containing bioactive compounds such as flavonoids and Saponin flavonoids, as an immunomodulator, stimulate and improve the immune system and the impaired immune function, while Saponins increase the diameter of the villi and the permeability of intestinal cell walls, thereby affecting the absorption of nutrients carried by feed. If the immunity increases and the absorption of feed nutrients is good, it will increase milk production in dairy cows. Through this community service activity, there will be increased empowerment of partners of the Sapi Perah Jaya Group with training in making concentrate with meniran nano extract as well as milk processing at the Wanita Tani Perah Jaya. The solution is to diversify milk in the form of yogurt. This activity was carried out using three methods: 1) Counseling on making beef concentrate with the addition of nano meniran extract, making yogurt and marketing dairy products, 2) Training on making the concentrate and making yogurt 3) Monitoring and evaluation that was carried out every month for 3 months after the counseling and training were carried out. The results obtained from this community service are the increased empowerment of partners and the increased skills of farmers in making concentrates and dairy products in the form of yogurt and marketing them. The successful application of technology through community service can increase the income and welfare of these two groups.

Abstract

The productivity of dairy cows in Sendang Village, Tulungagung Regency, remains below optimal potential, averaging only 4–10 liters per head per day, due to limited access to quality feed and suboptimal livestock health management. This community service program aims to enhance productivity through two feed innovations: nanotechnology-based Phyllanthus niruri extract to improve livestock health and metabolic efficiency, and silage feed to ensure the year-round availability of nutritious feed. The methods employed include socialization, technical training, and distribution of both products to 20 dairy farmers who are members of KPUD Tani Wilis. The application of these two technologies has the potential to increase both the quantity and quality of milk production, while also supporting the achievement of the Sustainable Development Goals (No Poverty, Zero Hunger, Decent Work and Economic Growth, Responsible Consumption and Production, and Climate Action).

Abstract

Broiler farms are generally susceptible to disease attacks caused by viruses, bacteria, parasites, fungi, the environment, and deficiencies in nutritional elements. The use of antibiotics in the livestock industry generally aims to treat livestock so as to reduce the risk of death and restore livestock to a healthy condition. There are three patterns for rearing broiler chickens in Indonesia, especially in West Java, namely modern rearing using a closed house system, semi closed house and conventional rearing using open house system. This study aims to prove the analysis of the cost of antibiotics and their market potential needs to be known so that rational and balanced use of antibiotics can be illustrated with the benefits to the results of raising broiler livestock. The design used in this research was a purposive sampling. The variables in this research are the closed house, semi closed house and open house broiler farming systems. The independent variables in this research are the cost of antibiotics each bird per period. Descriptive analysis is used to obtain stewardship antibiotics based on the existing medication program. The antibiotic cost each bird per period from closed house amount is Rp. 70,49 + 38,65, semi closed house is Rp. 100,20 + 42,28, and open house is Rp. 130,32 + 64,68. The means of antibiotic cost each birds per period amount is Rp. 100,34 + 54,17 use a factor for forecasting market potency of antibiotic. The market potency per year in Subang regency amount Rp. 5.799.005.710 + 3.130.677.091, West Java Province amount Rp. 74.165.707.957 + 40.039.429.938, Indonesia amount Rp. 312.461.530.889 + 168.686.875.905. Apart from that, antibiotic stewardship on a broiler farm scale is also needed to get an idea regarding the appropriate use of antibiotics in broiler farms, especially in the Subang district area of West Java Province.

Abstract

This study aimed to determine the effect of basil (Ocimum basilicum) leaf extract in a Tris-egg yolk extender on the motility, and plasma membrane integrity of Sapudi ram spermatozoa at room temperature. Fresh semen samples were collected from Sapudi ram using an artificial vagina. A completely randomized design was used, consisting of four treatment groups with five replicates each. Data were analyzed using SPPS with factorial analysis of variance (ANOVA) and Duncan’s test. The control group (T0) received Tris-egg yolk extender without basil leaf extract. Treatment groups T1, T2, and T3 received 1%, 2%, and 3% basil leaf extract, respectively, in the extender. Results showed that sperm motility and plasma membrane integrity (Table 4) tended to decrease with increased storage time. Among the treatment groups, the addition of 1% basil leaf extract in the Tris-egg yolk extender (T1) resulted in significantly higher (p <0.05) both sperm motility and plasma membrane integrity compared to the control (T0). However, higher concentrations of basil leaf extract (T2 and T3) did not result in significant differences (p >0.05) in sperm motility after three hours of storage compared to the 1% treatment (T1). In conclusion, the addition of 1% basil leaf extract to a Tris-egg yolk extender was effective in maintaining sperm motility and plasma membrane integrity of Sapudi ram spermatozoa stored at room temperature for up to three hours.
