Abstract

The aim of this study was to determine the best combination of cryoprotectant (Ethylene glycol, EG), dimethyl sulfoxide (DMSO) and propanediol (PrOH) and carrier system (hemistraw and cryotop) in improving ewe oocytes viability during cryopreservation. Oocytes with multi layers of compact cumulus cells were colleted from abbatoir and matured in TCM 199 medium supplemented with 10% fetal calf serum for 24-26 h at 38.5 C under 5% CO2 in the air. Matured oocyte was divided into six parts and vitrified in three different vitrification solutions; (i) 17% EG+17% DMSO with hemistraw as carrier system, (ii) 34% EG with hemistraw as carrier system, (iii) 17% EG+17% PrOH in hemistraw (iv), 17% EG+17% DMSO with cryotop as carrier system (v), 34% EG with cryotop as carrier system (vi), and 17% EG+17% PrOH in cryotop. Oocytes were cryopreserved for one week before revived and evaluated for viability. The result showed that oocytes vitrified in media containing EG and DMSO in cryotop had the highest viability (88.16%) compared to media containing EG only or EG and PrOH (70.95% and 68.76%, respectively) (P0.05). Moreover, oocytes viability that vitrified using cryotop and hemistraw as carrier system were not significantly different. The present results indicated that vitrification using combination of EG and DMSO as permeable cryoprotectant and cryotop as carrier system was the best system to maintain oocyte viability after vitrification-warming.

Abstract

AbstractThe use of vaginal sponge in estrous synchronization has been a popular method to uniform estrous cycle of small ruminants. In order to increase the litter size, the use of the sponge is combined with PMSG. Therefore the objective of this study was to evaluate the effect of PMSG in combination with vaginal sponge on estrous response and litter size of Javanese local sheep. The study was conducted in Faculty of Animal Husbandry, Padjadjaran University, Indonesia with 32 non-pregnant local sheep. All these sheep was divided into four groups of treatment, i.e: 1) group of sheep was subjected to vaginal sponge (VS) treatment containing 30 mg Medroxyprogesterone acetat (MAP) inserted in the vagina for 14 days; 2) group of sheep was subjected to the 30 mg MAP in VS plus 300 IU PMSG injection -1 d before VS removal; 3) group of sheep was subjected to the 30 mg MAP in VS plus 300 IU PMSG injection at the day of VS removal; 4) group of sheep was subjected to the 30 mg MAP in VS plus 300 IU PMSG injection +1 d after VS removal. The result showed that all treatment groups had 100% of estrous response. However, the occurrence of estrous time varied among group after VS removal. Group 1 (G1) and G4 tend to have estrous occurrence at day 3 after VS removal by 55% and 75% response, respectively. Whilst, G2 and G3 tend to have estrous occurrence respectively at day 1 (62,5%) and day 2 (50%) after VS removal. A 100% of pregnancy rate occurs in G2 and G4, while G1 and G3 have a 88,9% and 87,5%, respectively. No significant effects of the treatments on the ewes litter size. In conclusion, the use of PMSG administration 24 h prior, after, or at sponge withdrawal increase a tighter synchrony of oestrous and pregnancy rate, but has no effect on the increasing of litter size in Javanese local sheep.Keywords: Vaginal sponge, PMSG, estrous response, pregnancy rate, litter size, sheep

Abstract

Lactic acid bacteria (LAB) are known for their numerous beneficial effects for health and their colonization within intestines is affected by prebiotic consumption. Cogon grass root is an herbal medicine which contain substantial quantities of polyphenols. Recently, many studies suggested polyphenols as a novel group of prebiotics. Therefore, this study aimed to explore prebiotic effect of cogon grass root extract (CGRE) on stimulating growth of LAB in mice intestine. Mice (n =7/group) were divided into three groups; group A (control), group B and C received CGRE orally with daily dosage 90 and 115mg/kg BW, respectively. After 14 days of treatment, all mice were sacrificed. LAB was isolated and counted from ileum. Additional biochemical tests were performed to confirm the bacteria as LAB. Further identification of representatives LAB was performed with API-CHL-50 System. We found increased growth and diversity of LAB in the treatment group with optimum growth stimulation at CGRE dosage 90mg/KgBW daily (group B). Further identification showed Lactococcus lactis was the most frequent LAB found in all groups, with additional Lactobacillus paracasei and Lactobacillus curvatus were found in the treatment group. These findings support additional benefit of CGRE as prebiotics by stimulating LAB growth and diversity in mice intestine.

Abstract

Dairy goat is profitable object since the productivity is relatively brief, easy in maintenance and does not require much investment. Dairy goat become one of the important commodity for farmers in the village Cilengkrang, Cimalaka District of Sumedang. This study used a qualitative approach where 18 farmers who become interviewees. Collected data were observed and compared with the evaluation on the participants’ routine. The majority farmers are depending on individual experiences and conclusions without related guidance and supported references of diseases and procedures in nurturing dairy goats (61.1%), while the rest are consulting the problem to the vet or the group (83,3%). Some of them used the conventional treatment. However, when the condition of sick animal never improved, only 83,3% of farmers who contact veterinarian and the rest would prefer sell the animal in the market. It can be concluded that the majority farmers in the village, has experienced to overcome certain diseases of dairy goats. Nevertheless, the knowledge and sources of supporting references are still limited that affect to the use of inappropriate traditional herbs or regular human medicines from nearby stalls is still relatively high, so the awareness and knowledge of breeders about health management of dairy goat of breeders needs to be improved.

Abstract

ABSTRAK. Penelitian ini bertujuan mengetahui pengaruh keberadaan corpus luteum (CL) pada ovarium domba umur pubertas awal terhadap kualitas oosit hasil koleksi dan tingkat maturasinya secara in vitro (IVM). Sebanyak 279 oosit digunakan, terbagi pada kelompok tanpa CL (CL-) sebanyak 143 dan dengan CL (CL+) sebanyak 136. Oosit dipilih berdasarkan homogenitas sitoplasma dan dikelompokkan sesuai jumlah lapisan sel kumulus yaitu grade 1 (4 lapis), grade 2 (34 lapis) dan grade 3 (02 lapis). Media IVM menggunakan Tissue Culture Media 199 ditambah antibiotik, Follicle Stimulating Hormone, dan 10% Fetal Bovine Serum. Maturasi dilakukan pada inkubator 38,5C, 5% CO2 selama 24 jam. Pasca IVM, tingkat kematangan oosit dievaluasi berdasar kemunculan Polar Body I (PB I). Hasil menunjukkan bahwa keberadaan CL tidak berpengaruh terhadap kualitas oosit yang dikoleksi pada semua grade. Keberadaan CL berpengaruh pada tingkat kematangan oosit pada grade 1 sebesar 48,64% dibandingkan CL- sebesar 47,19% (p0,05). Berdasarkan hasil penelitian disimpulkan bahwa berpengaruh pada tingkat kematangan oosit setelah IVM. Hasil penelitian menggambarkan potensi penggunaan oosit ternak umur pubertas awal untuk digunakan lebih lanjut dalam program produksi embrio secara in vitro.(Effect of the presence of corpus luteum on oocytes quality and in vitro maturation rate of ewes at early puberty)ABSTRACT. This study aims to know effect of presence of corpus luteum (CL) to collected oocytes quality and its maturation rate post in vitro maturation (IVM), on local ewes ovary at early puberty. In total 279 oocytes were collected, 143 without CL (CL-) and 136 with CL present (CL+). Oocytes were selected according to sitoplasma hemogenity, divided into 3 grades according to cumulus cell (CC) layer namely Grade 1, 2 and 3 indicated by 4, 34, and 02 CC layers, respectively. The IVM media was Tissue Culture Media 199 supplemented with antibiotic, Follicel Stimulating Hormone, and 10% Fetal Bovine Serum following culture at 38.5C and 5% CO2. Twenty four hours post IVM, oocytes were evaluated on the presence of Polar Body I. Result showed that oocytes quality were not different among group in all grades. The present of CL gives better maturation rate in grade 1 compared to CL- (48.64 vs 47.19%; p0.05). The present finding show that the presence of CL improves oocytes maturation rate post IVM. Moreover, this study shows the potency of using oocytes from ewes ovary at early puberty for further in vitro embryo production program

Abstract

This study aims to use Google Data Studio to produce an academic data visualization dashboard system to display information, which can manage academic data, namely teacher attendance data and student grades, to provide decision-making support for decision makers in the monitoring process and evaluate school internal processes and monitor student grades at SMPN 8 Pariaman School. The type of research used is the R&D (Research and Development) approach SDLC (Systems Development Life Cycle) method with a prototype model. The stages carried out are communication, planning, modeling, construction and handover (handover). The system testing method studied uses questionnaire guidelines for functional testing to test system functionality to verifiers and usability testing to test system usability and suitability of functions and services to users. Based on the results of the validator functional test study, the results of meeting the functional requirements with both a value of 1 and 89% of the feasibility value of the system usability test are classified as very feasible. Therefore, it can be concluded that the academic data visualization dashboard system using Google Data Studio meets the requirements and is very suitable as a support for SMPN 8 Pariaman School in processing academic data on teacher attendance and student grades.

Abstract

The rapid advancement of information technology has driven significant changes in the management of school libraries, particularly in SMK that require efficient access to learning resources. This study addresses the limitations of manual library management at SMK Kartika 1–2 Padang, which resulted in data redundancy, slow information retrieval, and suboptimal reporting processes. The purpose of this research is to design and implement a web-based library information system that can systematically improve the quality and performance of library services. The study employed a system development research approach using the Waterfall model, encompassing requirements analysis, system design, implementation, and testing phases. Data collection was carried out through observation, interviews, and literature review. The results demonstrate that the developed system successfully integrates the management of book collections, member records, borrowing and returning transactions, and automated reporting within a single platform. The implementation of the system significantly enhances service efficiency by reducing processing time, minimizing data duplication, improving accuracy in transaction recording, and enabling faster access to library information for users. The main contribution of this research lies in providing a practical and applicable model of a web-based library information system that effectively improves operational efficiency and service quality in vocational school libraries.

Abstract

E-commerce applications to sell various products produced by students in higher education can increase income-generating. However, a few universities still use e-commerce to sell student products, especially learning products. This research aims to produce an e-commerce application for web-based learning products in universities. The research method is development research with a research design using the waterfall model. The waterfall model procedure consists of analysis, design, coding, and testing. The system design uses Unified Modeling Language (UML) modeling such as context diagrams, class diagrams, use case diagrams, and activity diagrams. The system's coding uses the PHP programming language, Jquery, and the Boostrap Framework. The testing stage is through black-box analysis carried out by experts in the field of system design, with the results of testing each case being well-integrated and feasible to be applied. The result of the research is a web-based e-commerce application that sells learning products in universities.

Abstract

The current online learning process requires virtual face-to-face media to interact between lecturers and students. However, currently available media requires lecturers to pay a fee to communicate indefinitely. Therefore, we need a media that can facilitate lecturers to do learning easily. This study aims to integrate jitsi with matrix conferences as a real-time online communication medium in learning. This type of research is development research (R&D) with the waterfall method. The steps taken are analysis, design, coding and testing. The analysis is carried out on the system development needs, then the design stage of the Jitsi architecture with a matrix, then coding using PHP and testing through QoS. QoS testing has very good throughput results, poor packet loss, very good delay, and excellent jitter for small-scale applications. It shows that picking video conferencing can be used as a medium of communication in online learning.

Abstract

ABSTRAK Latar Belakang: Kacang gude (Cajanus cajan) memiliki kandungan komponen bioaktif yang tinggi, namun pemanfaatannya dalam produk fermentasi seperti tempe masih terbatas. Tujuan: Penelitian ini bertujuan untuk mengevaluasi pengaruh substitusi kedelai dengan kacang gude terhadap sifat fisik, komponen bioaktif, kandungan gizi, bioaksesibilitas mineral, dan sifat sensori tempe. Metode: Tempe dibuat dengan variasi rasio kacang gude dan kedelai (F1: 60:40, F2: 50:50, F3: 40:60) menggunakan rancangan acak lengkap (RAL) dengan tiga ulangan. Sifat fisik yang dianalisis meliputi pertumbuhan miselium, daya iris, analisis warna dan rendemen. Komponen bioaktif meliputi analisis total kandungan flavonoid, fenolik, serta dengan metode penghambatan DPPH. Kandungan gizi dianalisis yakni kandungan serat total dan kandungan zat besi serta bio aksesibilitasnya. Sifat sensoris tempe dianalisis dengan uji hedonik menggunakan 35 panelis semi terlatih untuk memperoleh formula yang paling banyak disukai. Selanjutnya dilakukan uji deskriptif dengan metode konsensus pada formula terpilih.Hasil:  Semua formula tempe berhasil dikembangkan yang ditunjukkan dengan sifat fisik yang baik, termasuk pertumbuhan miselium, warna putih, dan tekstur yang kompak. Tempe substitusi kacang gude menunjukkan penurunan kecerahan dan  peningkatan daya iris seiring bertambahnya proporsi kacang gude. Selain itu terdapat peningkatan kandungan total flavonoid, fenolik serta antioksidan seiring peningkatan kandungan kacang gude. Analisis kandungan gizi menunjukkan terjadi penurunan signifikan kandungan serat total pada F3 (40:60). Kadar zat besi (Fe) antar formula tidak menunjukkan perbedaan yang signifikan, namun bioaksesibilitas Fe menurun signifikan seiring peningkatan rasio kacang gude. Uji kesukaan menunjukkan penurunan signifikan pada F3 (40:60), sehingga formula F2 (50:50) dipilih untuk uji deskripsi. Tempe substitusi kacang gude segar memiliki aroma fermentasi yang dominan, namun proses penggorengan dapat mengurangi intensitasnya. Kesimpulan: Penelitian ini menunjukkan bahwa substitusi kedelai dengan kacang gude berpotensi meningkatkan nilai fungsional tempe, khususnya potensi antioksidan dengan tetap mempertahankan penerimaan sensori, sehingga berpotensi sebagai bahan alternatif yang dapat dikembangkan pada pembuatan tempe.  KATA KUNCI: antioksidan; fermentasi; kacang gude; kedelai; tempe  ABSTRACTBackground: Pigeon pea (Cajanus cajan) recognized for its high content of bioactive compounds; however, its application in fermented products, such as tempeh, remains underexplored. Objectives: This study aimed to evaluate the impact of substituting soybeans with pigeon peas on the bioactive components, mineral bioaccessibility, and sensory properties of tempeh. Methods: A completely randomized design (CRD) with three replications was employed, using different soybean-to-pigeon pea ratios: F1(60:40), F2(50:50), F3 (40:60).. The physical properties analyzed included mycelial growth, cutting force, color analysis, and yield. Bioactive components evaluated through total phenolic and flavonoid content, and antioxidant activity using the DPPH radical scavenging assay. Nutritional analysis covered total dietary fiber, iron content, and their bioaccessibility. Sensory evaluation involved a hedonic test with 35 semi-trained panelists to determine the most preferred formulation, followed by a descriptive sensory analysis using the consensus method.  Results: All tempeh formulas were successfully developed, exhibiting good physical properties, including mycelium growth, white color, and compact texture. The tempeh samples demonstrated decreased lightness and increased cutting force with the addition of pigeon peas. The results indicated that increasing the proportion of pigeon peas significantly enhanced total antioxidant activity, total phenolic content, and flavonoid levels (p < 0.05). Nutrient analysis showed total fiber content significantly decreased in F3 (40:60). Iron (Fe) levels remained consistent across formulations (8.55 to 8.92 mg/100 g); however, Fe bioaccessibility significantly declined with higher pigeon pea ratios. The acceptance test revealed a notable decrease in acceptability for F3 (40:60), leading to the selection of F2 (50:50) for descriptive analysis.  Fresh tempe substituted pigeon peas exhibited a dominant fermentation aroma, which was subsequently mitigated by the frying process.  Conclusions: These findings suggest that soybean substitution with pigeon peas can enhance the functional value of tempeh, especially its antioxidant potential, without compromising sensory quality, presenting a promising alternative for further tempeh development.  KEYWORD: antioxidants, fermentation, pigeon pea, soybean, tempeh  Article submitted on May 08, 2025; Articles revised on May 26, 2025; Articles received on August 08, 2025; Articles available online on November 28, 2025
