Abstract

DNA optimization procedures can be carried out on the type of buffer used during extraction or physical handling techniques in separating genomic DNA from other compounds. The research using Three types of buffer: 1. CTAB, 2. Detergents containing Alkyl Benzene Sulfonate (ABS) surfactants and Three Detergents containing Sodium Laureth Sulphate (SLS) surfactants. This study aims to obtain an optimal DNA isolation method to produce genomic DNA of good quality and sufficient quantity so that it can be used for genetic diversity analysis in forest trees and to determine the optimal alternative buffer for CTAB to facilitate DNA isolation in remote areas, which generally hard to get CTAB. The parameters measured in this study were the presence of DNA and DNA concentration. The results showed that DNA isolation of 12 species of forest trees was successfully carried out using CTAB buffer and Detergent containing ABS surfactants by visualizing the genomic DNA bands from the results of the electrophoresis and Nanodrop spectrophotometer meanwhile Detergent containing SLS surfactants buffer was not successful in DNA isolation. The highest DNA quantity (DNA concentration) was found in 19 samples using CTAB buffer and the detergent containing ABS surfactants buffer with a concentration of 1403,8 - 3412,7 ng/μl. The conclusion of this study was CTAB buffer and the Detergent containing ABS surfactants can also be used as an alternative to a simple buffer for DNA isolation experiments.

Abstract

Independence in learning is very necessary for the application of online learning. Students as learners must have Self Directed Learning. This study aims to develop independent learning resources to facilitate student learning independence. Development research using the ADDIE model is carried out until the development stage. Needs analysis is conducted on 49 Biology Education Students with data collection instruments using the SDL test consisting of 5 question indicators: 1) Identifying learning needs; 2) Set up study routines and discover concept mappings; 3) Using concept/outline mapping as a useful understanding various information; 4) Self-assessment, honing potential; 5) Can explore self-ability. The test results show that in indicator 1 there are 9 students (18.37%) and indicator 5 as many as 11 students (22.45%) who have SDL abilities. The results of the needs analysis become the basis for the development of interactive electronic books through features such as material summaries, images, animations, videos, references (links and barcodes), interactive exercises, keywords, interest facts, practice questions, formative tests, and reflections. It is hoped that this interactive e-book can increase the independence of undergraduate and PPG students in studying biotechnology material as a source of independent learning.Abstrak Kemandirian belajar sangat diperlukan dalam penerapan pembelajaran daring. Mahasiswa sebagai pembelajar harus memiliki kemandirian belajar (Self Directed Learning/SDL). Penelitian ini bertujuan untuk mengembangkan sumber belajar mandiri untuk memfasilitasi kemandirian belajar mahasiswa. Penelitian pengembangan menggunakan model ADDIE penelitian ini dilakukan sampai tahap development. Analisis kebutuhan, dilakukan pada 49 orang Mahasiswa Pendidikan Biologi dengan instrumen pengumpulan data menggunakan tes SDL terdiri dari 5 indikator pertanyaan: 1) Mengidentifikasi kebutuhan belajar; 2) Mengatur rutinitas belajar dan menemukan pemetaan konsep; 3) Menggunakan pemetaan konsep/garis besar sebagai metode yang berguna untuk memahami berbagai informasi; 4) Menilai sendiri, merefleksikan dan mengasah potensi; 5) Bisa mengeksplor kemampuan diri. Hasil tes menunjukkan pada indikator 1 sebanyak 9 orang (18,37%) dan 5 sebanyak  11 orang  (22,45%) yang memiliki kemampuan SDL. Hasil analisis kebutuhan menjadi dasar untuk pengembangan buku elektronik interaktif melalui fitur-fitur barupa ringkasan materi, gambar, animasi, video, referensi (link dan barcode), latihan interaktif, key word, interesring fact, latihan soal, tes formatif dan refleksi. Diharapkan e-book interaktif ini dapat meningkatkan kemandirian mahasiswa S1 dan PPG dalam mempelajari materi bioteknologi sebagai sumber belajar mandiri.

Abstract

Terasi memiliki warna asli coklat kehitaman yang berpengaruh terhadap daya tarik konsumen. Secara umum bahan pewarna yang sering digunakan terbagi atas pewarna alami dan sintetis (buatan). Pewarna sintetis lebih mudah ditemukan dipasaran dengan harga yang relatif lebih murah dibandingkan pewarna alami. Namun banyak sekali produsen yang menyalahgunakan pemakaian pewarna. Oleh karena itu dilakukan penelitian ini untuk mengetahui pengaruh limbah kulit buah manggis, kulit buah naga dan karmoisin terhadap kualitas terasi udang rebon (Acetes erythraeus) serta menghasilkan rancangan modul sebagai bahan ajar pada pembelajaran biologi kelas XII SMA. Penelitian ini terdiri dari dua tahap yaitu eksperimen dan perancangan modul pada bulan April-Mei 2021. Eksperimen dengan desain Rancangan Acak Lengkap (RAL) dan menggunakan 4 perlakuan dengan 3 kali ulangan. Perlakuan yang diberikan berupa perbedaan pewarna, pewarna alami (kulit buah manggis dan kulit buah naga) dan pewarna sintetis (karmoisin). Adapun perlakuan: P0(perlakuan tanpa pewarna), P1 (ekstrak pewarna kulit buah manggis), P2 (ekstrak pewarna kulit buah naga), dan P3 (pewarna karmoisin). Parameter yang diukur meliputi kadar protein, kadar lemak, kadar pH, kadar air, dan uji organoleptik. Hasil penelitian menunjukkan bahwa limbah kulit buah berpengaruh sangat nyata terhadap kualitas terasi udang rebon. Perlakuan yang terbaik adalah P1 (ekstrak pewarna kulit buah manggis) dengan kadar protein sebesar 20,15%, kadar lemak 0,08%, kadar pH 6,8, kadar air 23,97%, nilai uji kenampakan 8,9, nilai uji bau 8,6,  nilai uji rasa 8,6, nilai uji tekstur 8,5 dan nilai hedonik 8,6. Hasil eksperimen digunakan untuk modul sebagai bahan ajar pada materi bioteknologi konvensional pada pembelajaran Biologi di SMA kelas XII.

Abstract

White Oyster Mushrooms (Pleurotus ostreatus) is one foodstuffs favored by the people. This mushrooms demand higher beucase it is tasty and seful to health. The research was conducted to determine the effect of the addition NPK complex fertilizers to the growth and protein content white oyster mushrooms (pleurotus ostreatus). This research using non factorial completely randomized design that consist 5 treatments and 3 replicatesand totally obtain 15 experimental design. It would further DMRT test if effect. The observed parameters include growth and protein content white oyster mushrooms. Based on the results revealed that the addition of NPK effect on all parameters observed where the best dosage is treatment P2 with the addition of NPK 10gr / 1 kg of growing media. The P2 treatment effect on time appears pinhead, pinhead number, stem length and wet weight white oyster mushrooms

Abstract

Calamansi fruits is one horticulture crops to make juice and flavor foods. This research aimed to determine the effect of Kinetin and NAA on the growth of explants Musk lime (Citrus microcarpa). The results of this research showed of Kinetin and NAA significantly affect the growth of explants Musk Lime. The percentage of explants grown 100% all treatments, except treatment K5A0, The earliest of root rise in treatment K0A0,5 HST, The earliest of bud rise in treatment K5A0 HST, Highest amount of buds in treatment K3A2is 2.4 buds, Highest length of shoot in treatment K3A0is 7.1 buds, and the amount of roots the best treatment of K0A0,5is 8,1 roots.

Abstract

The aim of this study was to produce compost from rice straw and maize straw in according to SNI 19-7030-2004 and also to know the best bioactivator which can be applicated to those wastes. This study held on February until June 2014. This reasearch used True Experimental Reasearch and Completely Random Used Design (Factorial RAL) from 4 factors and 3 reiterations. The subject was rize straw and maize straw to be composted by using bioactivator EM-4, Acticomp and Orgadec and also cow faeces. This prosedure of reasearch was preparing tools and materials, execution the step of composting, observing the process of composting and finished step. The perception variables are texture, composting time, C/N, temperature, humidity and pH. Data was analyzed in quantitative and qualitative. The result showed that the shrinkage of compost weight was 50%, the texture of compost including to small criteria and showed C/N ratio lower than before. All perception variables showed that compost was according to SNI 19-7030-2004, including temperature, humidity and pH as secondary variables. That result also indicated that EM-4 was the best bioactivator in composting rice straw and maize straw.

Abstract

The purpose of this study was to determine the effectiveness of using macromedia flash in the distance lectures for students of the Biology Education Study Program, FKIP University of Riau in 2021. This media application was applied to learning low-level plant systematics (STR) courses. The research was conducted at the Biology Education Study Program, FKIP Riau University, in the even semester of 2021. The object of the research was 35 students of Class A lectures on low-level plant systematics. The research was conducted using the classroom action method, namely applying macromedia flash learning media. The lecture period is divided into 2 rounds where the first round is learning without using macromedia flash media and the second round using macromedia flash. The indicators set in the form of student learning outcomes during lectures and student responses to the use of macromedia flash. The conclusion of the study is that the use of macromedia flash can improve student learning outcomes with an effectiveness ratio of 83% and is considered effective in terms of learning outcomes. Student responses to the application of macromedia flash media are classified as good with a percentage of 79.91%. Key words: Macromedia flash, Low Plant Systematics, Effectiveness   Abstrak. Tujuan dari penelitian ini adalah untuk mengetahui efektivitas Penggunaan macromedia flash pada perkuliahan jarak jauh mahasiswa Program Studi Pendidikan biologi FKIP Univeristas Riau tahun 2021. Aplikasi media ini diterapkan pada pembelajaran mata kuliah sistematika tumbuhan tingkat rendah (STR). Penelitian dilakukan di program studi pendidikan biologi FKIP Universitas Riau pada semester genap tahun 2021. Objek penelitian adalah mahasiswa Kelas A oerkuliahan sistematika tumbuhan tingkat rendah sebanyak 35 orang. Penelitian dilakukan dengan metode tindakan kelas yaitu menerapkan media pembelajaran macromedia flash. Masa perkuliahan dibagi dalam 2 putaran dimana putaran pertama adalah pembelajaran tanpa penggunaan media macromedia flash dan putaran kedua dengan menggunakan macromedia flash. Indikator yang ditetapkan berupa hasil belajar mahasiswa selama perkuliahan serta respon mahasiswa terhadap penggunaan macromedia flash.. Kesimpulan dari Penelitian diketahui  bahwa penggunaan macromedia flash dapat meningkatkan hasil belajar mahasiswa dengan rasio efektivitas 83% dan tergolong efektif dari segi hasil pembelajaran. Respon mahasiswa terhadap penerapan media macromedia flash tergolong baik dengan presentase 79.91%. Kata kunci: Macromedia flash, Sistematika Tumbuhan Rendah, Efektivitas

Abstract

Abstrak: Kurikulum merdeka memiliki berbagai perbedaan dengan kurikulum pendahulunya. Salah satu aspek pembeda adalah asesmen yang menekankan pada perkembangan karakter peserta didik dan mengutamakan asesmen formatif. Hasil wawancara pada sekolah mitra, diketahui masih banyak guru yang belum memahami penyusunan asesmen pada kurikulum merdeka. Kegiatan pengabdian ini bertujuan untuk meningkatkan kompetensi guru dalam menyusun asesmen diagnostik, formatif, dan sumatif. Metode pengabdian ini yaitu sosialisasi dan praktik penyusunan asesmen oleh guru dengan didampingi oleh tim pengabdi. Terdapat tiga tahap yang dilaksanakan yaitu persiapan, pelaksanaan, dan evaluasi yang terdiri dari 10 soal pretest dan postest. Peserta pengabdian terdiri dari guru-guru yang tergabung dalam MGMP Fisika, Kimia, dan Biologi SMA berjumlah 18 orang. Tingkat ketercapaian kegiatan didasarkan pada peningkatan nilai pretest-posttest dan angket kepuasan peserta yang dibagikan melalui googleform. Hasil kegiatan diperoleh bahwa (1) pengetahuan peserta tentang penyusunan asesmen meningkat sebesar 58%; (2) hasil angket kepuasan mendapatkan nilai 94%; dan (3) adanya peningkatan kompetensi guru dalam menyusun asesmen kurikulum merdeka.Abstract: The independent curriculum has various differences from its predecessor curriculum. One of the differentiating aspects is assessment which emphasizes the development of students' character and prioritizes formative assessments. As a result of interviews at partner schools, it is known that there are still many teachers who do not understand the preparation of assessments in the independent curriculum. This service activity aims to increase teacher competence in preparing diagnostic, formative and summative assessments. This service method is socialization and practice of preparing assessments by teachers accompanied by a service team. There are three stages carried out, namely preparation, implementation and evaluation which consists of 10 questionspretest andposttestThe service participants consisted of 18 teachers who were members of the MGMP Physics, Chemistry and Biology High School. The level of activity achievement is based on increasing valuepretest-posttest and participant satisfaction questionnaires distributed viagoogleform. The results of the activity showed that (1) participants' knowledge about preparing assessments increased by 58%; (2) the results of the satisfaction questionnaire got a score of 94%; and (3) there was an increase in teacher competence in preparing independent curriculum assessments.

Abstract

This study aims to develop inquiry-based student worksheets to improve students’ critical thinking skills and learning motivation. This development research uses the ADDIE (analysis, design, development, implementation, and evaluations) method. The development of inquiry-based student worksheets is developed on ecosystem materials. The results of the development of inquiry-based Student Worksheets with validity tests obtained an average score of 3.58 with very valid categories, so they have excellent quality and are worthy of use. The results of the development of student worksheets and the teacher response practicality test obtained an average score of 92.83 in the very practical category. From the results of the practicality test obtained, inquiry-based Student Worksheets have very practical qualities. The results of the development of inquiry-based student worksheets have a significant influence on improving students’ critical thinking skills and learning motivation with a test significance value of t 0.000 <0.005. The results of the t-test on improving students' critical thinking skills showed a mean statistics percentage of 83.12 very critical categories. The results of the t-test on increasing student learning motivation showed a mean statistics percentage of 82.8 categories of highly motivated.

Abstract

Bakau api-api (Aivicenniai mairinai Forsk. Vierh) merupakan tumbuhain pionir paida lahan pantai yang terlindungi. Bakau api-api (Aivicenniai mairinai Forsk. Vierh) ini bergunai untuk menekan laju abrasi paintaii, oleh sebab itu mangrove api-api ini perlu dilestarikan. Degradasi hutan mangrove yang signifikan di Indonesia memerlukan upaya rehabilitasi yang efektif dan efisien. Upaya penyediaan benih unggul dalam waktu singkat dan dalam jumlah banyak, salah satunya melalui kultur jaringan. Pemanfaatan ilmu bioteknologi dalam pembudidayaan bakau dengan cara kultur jaringan. Tujuan penelitian adalah untuk dapat memperoleh bibit bakau api-api dalam waktu yang singkat dan cepat. Penelitian ini menggunakan metode eksperimen dengain Rancangan Acak Lengkap faiktoriail 4×4 dengain perlakuan diulang 3 kali. Pada penelitian ini menggunakain hormon NAA dain BAP untuk mempercepat perkecambahan mangrove api-api, dan menggunakan eksplain biji. Analisis data menggunakan Analysis of Variances dan uji lainjut Duncan Multiple Range Test (DMRT) taraf 5%. Hasil penelitian menunjukkan adanya interaksi dengan faktor waktu tumbuh tanaman, laju pertumbuhan tanaman terbaik dicapai paidai perlaikuain N2B2 dengain  hormon 2mg L-1 NAiAi + 2 mg L-1 BAiP merupaikain perlaikuain yaing terbaik dalam mempercepait percambahan eksplain bii bakau api-api yaitu  waiktu muncul eksplain yang tercepat yaitu 21 hairi setelaih kultur  dengain persentase tumbuh eksplain 100%, jumlaih daun plantlet 4 helai  dan rata-rata  tinggi per plantlet 4 cm.
