Abstract

Aging is a complex biological process characterized by progressive functional decline of organs and tissues, largely driven by oxidative stress, chronic inflammation, and cellular senescence. Recent evidence suggests that age-related gut microbiota dysbiosis contributes significantly to inflammaging and oxidative damage, accelerating the aging process. Probiotics have emerged as potential gerobiotics due to their antioxidant, anti-inflammatory, and microbiota-modulating properties. This literature review aims to evaluate the role of probiotics in preventing and delaying the aging process through modulation of gut microbiota, oxidative stress reduction, and regulation of inflammatory. A narrative literature review was conducted using scientific publications from 2012 to 2023 obtained from NCBI (PubMed), ScienceDirect, and Google Scholar databases. Articles were selected based on relevance to probiotics, gut microbiota, oxidative stress, inflammation, and aging mechanisms. A total of 300 articles were identified from PubMed (n = 120), Science Direct (n = 95), and Google Scholar (n = 85). After removing duplicates, 235 articles were screened. 45 full text articles were assessed for eligibility, and 11 studies were included in the quantitative synthesis. The reviewed studies indicate that probiotics, particularly strains from the genera Lactobacillus, exert anti-aging effects by reducing reactive oxygen species (ROS), enhancing endogenous antioxidant enzymes (SOD, CAT, GPx), regulating inflammatory pathways (NF-κB, cytokines), activating Nrf2 signaling, maintaining intestinal barrier integrity, and restoring gut microbiota balance. Several probiotic strains were shown to delay telomere shortening, suppress cellular senescence markers, and improve metabolic. Probiotics play a significant role in preventing aging by modulating gut microbiota composition, reducing oxidative stress, and suppressing chronic inflammation. These findings support the potential application of probiotics as gerobiotics to promote healthy aging and extend health span.

Abstract

Sepsis neonatorum merupakan salah satu penyebab utama morbiditas dan mortalitas bayi baru lahir yang diduga dipengaruhi oleh faktor genetik pada gen-gen yang berperan dalam sistem imun bawaan. Human Defensin β 1 (HDB1) merupakan peptida antimikroba yang berperan penting dalam pertahanan awal tubuh terhadap infeksi. Penelitian ini bertujuan menganalisis hubungan antara polimorfisme gen HDB1 pada lokus rs11362 dan rs1799946 dengan kejadian sepsis neonatorum. Penelitian observasional dengan desain cross-sectional comparative ini menggunakan 60 sampel DNA genomik arsip yang telah tersimpan sebelumnya, terdiri atas 30 sampel neonatus dengan sepsis neonatorum awitan lambat (onset >72 jam dengan kultur darah positif) dan 30 neonatus tanpa sepsis. Genotipe kedua lokus ditentukan melalui amplifikasi PCR dengan sepasang primer yang mengapit kedua lokus (ukuran amplikon 833 bp), dilanjutkan sekuensing Sanger, kemudian dianalisis menggunakan uji chi-square pada derajat kepercayaan 95%. Hasil penelitian menunjukkan hubungan yang bermakna antara polimorfisme HDB1 rs11362 C>T dengan kejadian sepsis neonatorum (p=0,043), dimana dari seluruh subjek yang memiliki genotipe TT rs11362, sebanyak 15 dari 21 subjek (71,4%) merupakan kelompok sepsis, sedangkan 6 dari 21 subjek (28,6%) berasal dari kelompok tanpa sepsis. Sebaliknya, polimorfisme HDB1 rs1799946 C>T tidak menunjukkan hubungan yang bermakna dengan kejadian sepsis neonatorum (p=0,349). Dapat disimpulkan bahwa polimorfisme gen HDB1 rs11362, tetapi tidak rs1799946, berhubungan dengan kerentanan neonatus terhadap sepsis, menunjukkan potensinya sebagai kandidat biomarker genetik untuk diteliti dan dikembangkan lebih lanjut.

Abstract

A phenological study with the aim to elucidate flower and fruit development stages in species of Uncaria gambir was conducted in field trial of agricultural faculty of Andalas University-West Sumatra. Observation during flower and fruit organ development was done in eleven inflorescences. Quantitative and descriptive data were collected during one season of flowering time. Phenology of flower and fruit development could be classified in five stages, namely F0 (flower initiation), F1 (small bud scale), F2 (large bud scale), F3 (anthesis, flower opening) and F4 (fruit development). All these events had average completing time in 112 days, and could be detailed as follows: flower initiation stage (F0) took place in 20 days, small scale bud stage (F1) occurred in 27 days, and large scale bud and anthesis stage (F0 and F3) each took place in 5 days, meanwhile fruit development stage (F4=S0) would complete in 53 days. This result should be useful information especially for creating breeding programme in Uncaria gambir species.Key words: Uncaria gambir, phenology, flower development, fruit development.

The primary objective of this study is to assess genetic and chemical variability among four genotypes of Uncaria gambir cultivated in West Sumatra. Uncaria gambir is native to Indonesia, widely cultivated in West Sumatra, used as an herbal remedy, and become a significant agricultural product for Indonesian export with catechin content as a standard. The phytochemical components of this species have been extensively studied, and its extracts are employed in traditional medicine. To fingerprint genomic DNA, we employed three pairs of Specific Sequence-Related Amplified Polymorphism (SRAP) markers selected from 29 combinations tested. A total of 150 amplified bands were generated, highlighting substantial genetic variability among the tested genotypes. Notably, the highest amplified and polymorphic band was obtained with primer E combination from three selected primers (primer E, primer F, and primer T). In addition, only catechins were identified among the four types of catechins employed as HPLC standards. We detected varying catechin concentrations among the genotypes, with genotype Mancik exhibiting the highest concentration. This finding underscores the potential for genotype-specific bioactive marker identification within U. gambir. In addition, this study shows the efficiency of SRAP markers in genotype identification within U. gambir. Furthermore, the identification of catechin as a prominent bioactive marker emphasizes its significance for further exploration and conservation efforts.

Abstract. Pohan SD, Prima HS, Jamsari, Runifah T. 2025. Molecular screenings of drought tolerance QTLs and agronomic performance in selected rice (Oryza sativa) cultivars. Biodiversitas 26: 3731-3744. This study aimed to identify Quantitative Trait Loci (QTL) associated with drought tolerance (qDTY) and evaluate their agronomic performance under both non-stress and stress conditions. Molecular analysis using eight SSR markers (RM236, RM276, RM511, RM520, RM28099, RM28130, RM28166, and RM1261) was conducted to detect the presence of targeted qDTY loci. A Completely Randomized Design (CRD) with three replications was applied to assess the effects of drought stress on rice plants. Two treatments were implemented: a non-stress control (C1, 100% of field capacity) and a drought stress condition (C2, 50% of field capacity). Agronomic traits such as PH, PTN, PL, NFG, NUFG, DTF, and DTM were measured to evaluate plant performance under both conditions. The SSR markers used for detecting qDTY loci exhibited polymorphism. The number of alleles per marker averaged 8.625, ranging from 1 (RM511) to 12 (RM236). RM236 and RM28099 showed the highest polymorphism with eight alleles each. Phenotypic evaluation found significant differences in trait performance between non-stress and drought-stress conditions. While all traits generally decreased under drought stress (C2), the extent of reduction varied among genotypes. Most genotypes appeared to exhibit a drought escape mechanism by shortening their DTF and DTM under drought stress.
