Abstract

The research was conducted from February 2016 to April 2017 and was aimed to determine the effect of adding different concentrations of detergent on the growth of bacterial populations in the ocean water column and to see the ratio of bacterial persisted capability in different lengths of dissection. This research used experimental method with Completely Randomized Design. Seawater samples were analyzed at Marine Microbiology Laboratory, Fisheries and Marine Faculty of Riau University. Based on the results of the study, from day 0 untill day 10 after given detergent with different concentrations, bacteria experienced a decreasing trend of growth, in day 15 untill day 20, the trend of bacterial growth contaminated by detergent with different concentration relatively increased to stagnant. Based on the result of this study the addition of different detergent concentration in each treatment affect the growth of heterotrophic bacteria population in the sea water, where the higher concentration of detergent added the lower the ability of bacteria to grow and the ability of heterotrophic bacteria to survive at different concentrations of detergent, it can be concluded that the ability of heterotrophic bacteria to grow from days 0 to 20 has decreased in each of its concentrations.   Keywords: heterotrophic bacteria, Surf detergent.

Abstract

ABSTRACT   Secondary metabolites are a molecule or product of metabolites produced by the metabolic processes of microorganisms where the metabolite products are not a basic necessity of living microorganisms and for growth, but secondary metabolites can also function as emergency nutrients to survive. Marine bacteria including heterotrophic bacteria are capable of producing secondary metabolites. This study aims to determine the ability of secondary metabolite production of heterotrophic bacteria to inhibit the activity of pathogenic bacteria (Vibrio alginolyticus, Aeromonas hydrophila and Pseudomonas sp). In this study using the experimental method. 10 Heterotrophic bacterial isolates obtained from a collection of marine microbiology laboratories were cultured on NB media ± 3 days until the media was cloudy and harvest during stationary times, then extracted secondary metabolites from the media and shells using Etyl Acetate. The secondary metabolite extract will be tested against pathogenic bacteria (Vibrio alginolyticus, Aeromonas hydrophila and Pseudomonas sp). Based on the results of the antagonism test conducted, 10 heterotrophic bacterial isolates were able to inhibit the growth of all three pathogenic bacteria. Based on the antagonism test, the highest inhibitory value was Bacillus cereusspecies (KM489154.1) with an average diameter of about 10.08 mm, while the inhibition power which was classified as the lowest or categorized as moderate was found in the Bacillus safensis species (KY495152.1 ) with an average diameter of around 7.33 mm. The ability to inhibit pathogenic bacteria is suspected because this heterotrophic bacterial species produces antibiotic compounds. Keyword :Secondary metabolites,  Heterotrophic Bacteria, Antagonism test

Abstract

ABSTRACT  Heterotrophic bacteria are bacteria that require organic carbon and inorganic nitrogen as an energy source, Research aims to determine the number of heterotrophic bacteria, knowing the results of its activity test against pathogenic bacteria (Vibrio sp, Aeromonas sp, Pseudomonas sp) and to know the species of heterotrophic bacteria through DNA test isolated from estuari waters of 15 ppt and the industrial estuary at coastal areas Sungai Kayu Ara  village with salinity 27 ppt, Siak District. Distribution of bacteria at station  higher than station 2 with value of nilai 7,04 x 107cfu / ml at station 1 to 6,46 x 107cfu / ml at station 2. Furthermore, through simple linear regression test known that the growth of heterotrophic bacteria on environmental parameters (DO, pH, temperature and salinity) categorized very weakly with the value of r = 0.10 in all parameters. Based on the antagonism test on pathogenic bacteria, it was found that there were 9 bacterial isolates that potentially inhibited the growth of the three pathogenic bacteria, namely: RM3, RM4, RM5, RM6, RM8, RM11, RL15, RL17 and RL 24 with inhibit zone ranges from 0.6 - 18mm, where the RM11 (12-15.3 mm) isolate as the best isolate in its inhibitory properties. By DNA test with 16S rRNA method and BLAST analysis, it was found that Bacillus safensis, Alcaligenes faecalis, Enterobacter sp, Enterococcus sp and Vagococcus sp species were identified as unidentified and possibly non-registered species in Genbank NCBI because it has a homology percentage of only 84%.                                                      Keywords: Heterotrophic Bacteria, Distribution, Antagonism, DNA
