Abstract

Estrogen plays important roles in breast cancer as it binds its receptor in breast tissue. The most studied variants in estrogen receptor α encoded by ESR1 gene are the ESR1 PvuII and XbaI polymorphisms, which were associated with lower sensitivity to estrogen. We determined the proportion of ESR1 XbaI and PvuII polymorphisms in Javanese woman in Yogyakarta, Indonesia and analyzed the correlation between genetic variations with mammogram density. ESR1 XbaI and PvuII polymorphisms of 50 cases and 58 controls were identified using PCR-RFLP. Breast density was assessed based on digitizer mammograms. Quantitative analysis was performed using an interactive program based on cumulus of two thresholds. Mean of density and frequencies of SNPs were compared between cases and controls to identify the association between SNPs and cancer susceptibility. Mammographic density was significantly higher in cases (52%) than controls (0.41%) (p < 0.05). Women with one or two copies of the PvuII T allele and XbaI A allele had higher mammographic density compared with women with C and G alleles, respectively. The proportion between PP and TT genotype was not statistically significant (p > 0.05), while the proportion between AA and GG was significantly different (p < 0.05). Haplotype 2 (CG/PX) was associated with lower sensitivity to estrogen and reflects a decrease of mammographic density. These findings were consistent with other studies that showed that ESR1 polymorphisms may affect breast cancer risk through differences in breast density.

Abstract

Epilepsy and migraine are common neurological diseases in many populations. Mutation of the voltage gated natrium channel Nav1.1 (SCN1A) are important causes of different genetic epilepsies and can also cause familial hemiplegic migraine (FHM-III). This study aimed to identify SCN1A gene variation in patients with epilepsy and common migraine. Gene variation analysis of exon 26 of the SCN1A gene was carried out in 33 patients with epilepsy, 33 patients with migraine and 30 control individuals from Neurology Polyclinic at Dr. Zainoel Abidin General Hospital, Banda Aceh. The PCR and direct sequencing methods were performed in this study. SCN1A gene variations were identified in two epilepsy patients. These gene variations located at exon 26 were four silent mutations in patient E27 at position A4440T (Leu1480Leu), T4443C (Leu1481Leu), A5046G (Leu1682Leu) and C5121T (Asp1707Asp). One silent mutation in patient E30 at position G5505A (Glu1835Glu). None of these gene variations were identified in controls and patients with common migraine in this study. This study has identified 5 genetic variations of SCN1A in patients with epilepsy but not in common migraine. The mechanism and relationship between these variants and epilepsy need to be clarified.

Abstract

Rifampin is one of first-line drugs for the treatment of tuberculosis. In Indonesia nearly alltuberculosis patients show lower rifampin plasma concentrations possibly due to genetics.Rifampin is a substrate of the organic anion-transporting polypeptide 1B1 (OATP 1B1)encoded by the solute carrier organic anion transporter family member 1B1 (SLCO1B1).This study aimed to identify haplotype polymorphisms of tuberculosis drug transporterswith an impact on clinical outcome in tuberculosis patients. Thirty-six patients from AbdulWahab Sjahranie General Hospital, Samarinda, East Kalimantan were involved in thestudy. Buffy coat from patient blood samples were tested for SLCO1B1 and SLCO1B3polymorphisms by RFLP and ARMS PCR, whereas the clinical outcome was examinedbased on the sputum conversion. The frequency of patients with SLCO1B1*15 haplotypewas 63.9%. The SLCO1B1*15 haplotype was associated with susceptibility to failureof clinical outcome (p=0.005; RR=4.52; 95% CI: 1.22-16.64). The OATP1B1*15haplotype revealed that the failure of clinical outcome was markedly increased comparedto the three other haplotypes. These results suggest that the SLCO1B1*15 haplotypeis an important predisposing factor for lower clinical outcome. Our data indicate thatindividualized treatment should be considered for Indonesian tuberculosis patients basedon genetics characteristics of patients.

Abstract

Polyherbal formulation (PHF) containing extracts of Sauropus androgynous, Trigonella foenum-graceum and Moringa oleifera has been proven can induce milk production in animal model. However, its molecular of action has not been elucidated, yet. This study aimed to investigate the effect of the PHF on the mRNA expressions of α-actin smooth muscle (ACTA2) and cytokeratin 14 (CK14) on the myoepithelial cells of the lactating rats mammary glands. Thirty female Wistar rats were divided into five groups with six of each. Group I was orally administered aquadest. Group II, III, and IV rats were orally administered the PHF at dose level of 26.25, 52.5, and 105 mg/kg once a day, for 15 days, respectively. Group V was orally administered 2.7 mg domperidone. On 16th day, rats were sacrificed. Mammary glands were isolated and processed for mRNA expression analysis using real-time polymerase chain reaction (qRT-PCR). The results demonstrated that the mRNA expression of ACTA2 and CK14 increased in dose-dependent manner in the groups of PHF. Significantly different between the Group III, IV, and V compared to Group I was observed (p < 0.05). However, there was no significantly different between Group IV and Group V (p>0.05). In conclusion, the PHF increases the mRNA expression of ACTA2 and CK14 on myoepithelial cells of the mammary glands on lactating rats.

Abstract

The prevalence of metabolic syndrome (MetS) is high worldwide which it can increase the risk of some diseases such as cardiovascular disease, type 2 diabetes mellitus even mortality. The prevalence pattern and determinants of MetS risk factors might differ among ethnics in Indonesia. This study aimed to determine the anthropometry and metabolic measurements determinants to predict the MetS prevalence of the Javanese population in Yogyakarta. It was a case control study conducted from December 2018 to March 2019 involving 214 Javanese subjects aged 20-74 years resided in Yogyakarta Special Region, Indonesia. NCEP ATP III criteria were used to identify MetS as case and not diagnosed with MetS as control. The results showed that BMI, WC, BP, total cholesterol and HDL-C were significantly different between MetS and non MetS patients (p<0.005). In MetS subjects, prevalence of obesity was 75.3%, visceral fat was 75.3%, WC 92.95%, WHtR 97.64% and total cholesterol/HDL-C ratio 553%, which independently increased the risk of MetS 7.30, 5.32, 13.37, 20.75, and 7.16 times, respectively. Result of logistic regression analysis showed central obesity based on WC increased the risk of Met-S by 17.62 time and the ratio of total cholesterol/HDL-C>5 by 9.54 time. In conclusion, WHtR is a better marker for MetS prediction independently. However, the WHtR in combination with WC and total cholesterol/HDL-C ratio are better for MetS prediction in the Javanese population.

Abstract

Plumbum bersifat toksik terhadap manusia. Teh hijau digunakan untuk mengobati penyakit karena mengandung catechin. Tujuan penelitian untuk mengetahui efek catechin ekstrak teh hijau sebagai kelator Pb. Sebanyak 36 ekor tikus Wistar digunakan dalam penelitian ini. Tikus dibagi 6 kelompok, 6 ekor per kelompok. Kelompok I sebagai kontrol negatif. Kelompok II sampai VI diberi Pb asetat selama penelitian (66hari). Kelompok II sebagai kontrol positif tidak diberi perlakuan. Kelompok III, IV, dan V pada hari ke 35 diberi ekstrak teh hijau masing- masing 0,75 g/kgbb, 1,5 g/kgbb dan 3 g/kgbb. Kelompok VI diberi dimerkaprol dosis 3 g/kgbb sebagai pembanding. Ada tidaknya beda nyata antar perlakuan dianalisis dengan ANOVA diikuti dengan uji Duncan untuk mengetahui letak perbedaan. Parameter yang diukur adalah malondialdehid, aktivitas glutation peroksidase dan nitrit oksida, diukur dengan komersial kit. Hasil penelitian menunjukkan semua dosis ekstrak teh hijau dapat menurunkan MDA, Pb darah dan meningkatkan GPx serum serta NO. Dosis 3 g/kgbb optimum dalam menurunkan kadar MDA dari 2,75–0,07 µmol/L dan Pb darah dari 2,35–0,02 ppm. GPx serum meningkat dari 58,5–177 µmol/L dan NO serum dari 1,27,95 µmol/L. Simpulan hasil penelitian, ekstrak teh hijau dapat digunakan sebagai kelator Pb.

Abstract

Diabetes mellitus type 2 is the most common type of diabetes. This study was conducted to determine the effect of Mesenchymal Stem Cell-Conditioned Medium (MSC-CM) in Homeostatic Model Assessment of β-cell function (HOMA-β) value, normal Langerhans cells, and Transcription Factor 7-Like 2 (TCF7L2) gene expression in type 2 diabetic rats model. As many as 27 male Sprague Dawley rats were divided into 3 study research groups: normal control (9 normal rats), diabetic control (9 type 2 diabetic rats, induced by 60 mg/kg BW Streptozotocin and 120 mg/kg BW Nicotinamide i.p.), and treatment (9 type 2 diabetic rats treatment with 0.1 ml/200g BW MSC-CM i.p.). On day 30 after therapy, the expression of TCF7L2 gene was performed with real time-quantitative PCR (RT-qPCR). The HOMA-β value were calculated based on Fasting Insulins (FINs) levels and Fasting Blood Glucose (FBG) levels data from other research team members. Based on results, MSC-CM increases the HOMA-β value and amount of normal Langerhans cells of treatment group that indicates amelioration effect of MSC-CM, but there was no significant difference in TCF7L2 gene expression level between diabetic control and treatment group.

Abstract

Objectives: To ascertain the nitric oxide (NO) level of the follicular fluid of endometriosis sufferers compared with that of the non-endometriosis patients.Materials and Methods: Follicular fluid was taken from 64 women, but only 54 continued with further laboratory test. Women with endometriosis had their follicular fluid taken during laparoscopy, while the follicular fluid from non-endometriosis women was taken during tubectomy. Nitric Oxide level was analyzed using the PCR-RFLP method. Data were analyzed using software STATA Intercooled Version 11.Results: NO level in endometriosis is higher than that in non-endometriosis and has a significant relationship with all the variables (age, parity, dysmenorrhea, dyspareunia, types of endometriosis, and grade of endometriosis).Conclusion: The NO level in endometriosis is higher than that in non-endometriosis. The distribution of the polymorphism of allele GG is greater than that in non-endometriosis. The existence of polymorphism GG caused a higher eNOS, increasing NO level, causing a decrease in fertility which derived from the degenera-tive oocyte.

Abstract

Objective: To ascertain the IL-1b level in the follicular liquid of endometriosis sufferers compared with that of non-endometriosis sufferers.Materials and Methods: The ELISA method, for detecting the IL-1b, is taken from follicular fluid of endometriosis as well as non-endometriosis patients that diagnosed according to the ASRM criteria.Results: The 54 subjects were dominantly endometriosis <34 year old (88.9%).  The IL-1b levels of endometriosis patients and non-endometriosis patients were 82.86 ± 30.15 and 56.05 ± 23.27 respectively (p=0.01). The ROC value of IL-1b was found at a cut-off point of 26 (with the IL-1b cut-off point of 65.16). The discriminating value of AUC (area under curve) was 59.3%.Conclusion: It was concluded that the IL-1b level in endometriosis is higher than that in non- endometriosis.

Abstract

Background: Diabetes is a result of oxidative stress which causes the impaired function of pancreatic beta-cells. Fermented tempeh aerobic anaerobic (FETAA) containing gamma-aminobutyric acid and isoflavones can reduce oxidative stress in diabetes. Objective: The aim of this study is to evaluate FETAA in improving pancreatic β-cell function in diabetic mice. Methods: Twenty streptozotocin-induced diabetic mice, divided into four groups (n = 5 each group): DM, DM + FETAA 10 mg/100 g BW, DM + FETAA 20 mg/100 g BW, DM + FETAA 40 mg/100 g BW as well as normal group (n = 5). DM mice were treated with FETAA for 21 days. Fasting glucose was determined using the GOD-PAP method, while insulin level was determined by ELISA. The homeostasis model assessment of β-cell function (HOMA-β) was calculated using the HOMA2 calculator, and the Pdx1 mRNA level was determined by Real Time-PCR. Results: The DM mice group treated with FETAA had lower glucose levels than the DM mice group. FETAA dosage of 40 mg/100 g BW was able to reduce the highest blood glucose levels (p<0.05). DM mice group treated with FETAA had higher levels of insulin and HOMA-β than the DM mice group (p <0.05). Treatment of FETAA 10 mg/100 g BW produced the highest insulin content of 57.44 ± 8.132 pmol/L, while treatment of FETAA 40 mg/100 g BW had a HOMA-β value of 72.86 ± 21.85%. Pdx1 mRNA expression in group FETAA-treated DM mice was higher than the DM mice group, although it was not statistically significant (p> 0.05). Conclusion: FETAA could improve HOMA-β, blood glucose levels, but did not affect Pdx1 mRNA expression.
