Abstract

Peer teaching adalah sebuah model pembelajaran yang dapat mengembangkan aspek afektif seperti kerjasama, komunikasi dan mengembangkan rasa percaya diri sehingga Jumlah Waktu Aktif Belajar siswa dapat meningkat. penelitian ini bertujuan untuk mengidentifikasi peningkatan Jumlah Waktu Aktif Belajar (JWAB) dalam pembelajaran bolatangan dengan menerapkan medel pembelajaran Peer Teaching. Metode menggunakan Penelitian Tindakan Kelas (PTK), di SMA AL-Musyawarah Lembang, dengan sampel 33 orang dan terdiri dari 14 siswa putra dan 19 siswa putri. Hasil penelitian secara umum menunjukan siswa bergerak secara aktif sehingga meningkatkan Jumlah Waktu Aktif Belajar (JWAB) yang ditunjukan dari motivasi tinggi pada saat terlibat dalam pembelajaran permainan bolatangan. Tutor dan siswa dapat meningkatkan kemampuan berkomunikasi dan berinteraksi dengan orang lain. Kesimpulan penelitian bahwa model peer Teaching dapat gunakan pada pembelajaran bolatangan, karena secara bertahap dapat meningkatkan nilai kerjasama, komunikasi, menyampaikan informasi dan mengembangkan rasa percaya diri sehingga Jumlah Waktu Aktif Belajar (JWAB) sebagai indikator bahwa siswa terlibat secara aktif selama pembelajaran juga meningkat.

Abstract

Penelitian ini merupakan penelitian eksperimen yang bertujuan untuk; (1) Mengetahui perbedaan kemampuan representasi matematis antara mahasiswa yang pembelajarannya menggunakan model pembelajaran learning cycle 7E dan pembelajaran konvensional. (2) Mengetahui peningkatan kemampuan representasi matematis pada mahasiswa yang pembelajarannya menggunakan model pembelajaran learning cycle 7E. Uji coba penelitian ini dilakukan di FKIP Unswagati Cirebon, sampel penelitian kelas D dan G mahasiswa semester II Program Studi Pendidikan Ilmu Ekonomi tahun akademik 2013/2014. Hasil penelitian diperoleh (1) Terdapat perbedaan rata-rata kemampuan representasi matematis antara mahasiswa yang pembelajarannya dengan menggunakan learning cycle 7E dengan pembelajaran secara konvensional.(2) Adanya peningkatan kemampuan representasi matematis yang signifikan pada mahasiswa yang pembelajarannya menggunakan learning cycle 7E. Abstract

ABSTIIAKPcnelitian ini bertujuan untuk mengetahui pembelajaran dengan menggunakan model pembelajnran koeperaiif tipe TOT dapat meningkatkan kcmampuan pcmahaman matemaus siswa. Penelitian ini dilaksanakan di SMP Negeri 1 Lemahabang Kabuparcn Cirebon. Populasi pada penelitian ini adalah seluruh slswa kelas VIII SMPNegeri I Lemahebang. Kelas yang djJttdikan sampel dalam penelitien Ini sebanyak 2 kelas dari 8 kelas yang ada dengan mengambil sampel secara purposif. Masing-masing kelas sebaayak 33 siswa, kelas VIII B sebagai kclas eksperimen dan kelas VIII C sebagai kelas kontrol. Hasil pcnelitian menunjukkan bahwa pembelajaran dengan meuggunakan model pembelajaran kooperatif tipe TGT dapat meningkarkan kemampuan pemahaman matemaiis secara signifikan berdasarkan hasil uji hipotesis dengan menggunakan uji t yang nilainya 0.047 < 0,05. Secara deskriptif statistik juga dapat dilihal rata-rata peningkatan pemahaman pada kelas eksperimen sebesar 0,45 dan peningkalan pemahaman pada kelas komrol sebesar 0,35.

Abstract

Gold nanoparticles have good surface plasmon properties in the presence of free electrons on their surface. The presence of free electrons on the surface of the plasmon causes gold nanoparticles to be able to bind to various kinds of biomolecules and proteins, including single-chain variable fragments (scFv). In this study, gold nanoparticle conjugates with Chikungunya antiviral scFv have been successfully prepared with the aim of developing a colorimetric method for rapid detection of envelope protein Chikungunya virus (E2 CHIKV). The manufacture of gold nanoparticles was carried out using malva nut gum and trisodium citrate as reducing agents. The results showed the formation of the AuNP-scFv conjugate which was characterized by a shift in the maximum wavelength for gold nanoparticles synthesized using malva nut gum as a reducing agent from 532.89 nm to 536.62 nm. Similarly, for gold nanoparticles synthesized using trisodium citrate as a reducing agent, the maximum wavelength shifted from 528.99 nm to 531.95 nm. The detection result of E2 CHIKV protein using the AuNP-scFv conjugate has not shown a positive result

Abstract

Prethrombin-2 (PT2) is a thrombin precursor, which plays a role in the conversion of fibrinogen into fibrin during blood clotting process. Previous study reported that the expression of human prothrombin-2 (rhPT2) in Escherichia coli formed inclusion bodies. The aim of this study was to establish a strategy to express a soluble rhPT2 in E. coli. This study was animed to design and codon optimize human prethrombin-2 gene as well as to optimize the expression condition using four strains of E. coli. The codon adaptation index (CAI) of the unoptimized hpt2 gene was 0.336, with 56.8% GC content. After optimization, the CAI of optimized hpt2 became 1.000 with 53.1% GC content. The optimized gene was successfully cloned into pTWIN1 expression vector. Expression analysis indicated that only E. coli ArcticExpress strain could successfully express a soluble recombinant rhPT2 protein, with only part of rhPT2 being expressed in insoluble form. However, the rest of the E. coli strains used in the experiments failed to express the rhPT2 in soluble form. We are deducing that the success in achieving soluble expression was not only due to the availability of chaperonins Cpn60/Cpn10, which played a crucial role in the protein folding in E. coli ArcticExpress strain, but also due to the codon optimization of hpt2 gene.

Abstract

α-Amylase from Saccharomycopsis fibuligera R64 is a non-adsorbing raw-starch degrading enzyme, a unique characteristic. This character is difficult to explain in the absence of its three-dimensional structure. Here we discuss the expression of a-amylase from Saccharomycopsis fibuligera in Pichia pastoris and the effect of site directed mutagenesis on its activity. A model based on the structure of its homologs suggested mutation of codon of Tyr401 into that of a Trp residue. An activity study using whole cells P. pastoris showed similar substrate degradation rates by cells carrying either the native or mutant amylase encoding gene. However, the purified enzyme of the mutant strain showed faster starch hydrolysis.

Abstract

Prethrombin-2 is a thrombin precursor that has important role in blood coagulation. It is the smallest precursor which is activated into thrombin by FXa prior to coagulation process. However, as a commercial theurapetic protein in fibrin sealant component, prethrombin-2 must be activated by ecarin before used. Thus, the production process of this protein needs further purification. In order to eliminate ecarin activation step and to increase production efficiency, we designed, cloned and expressed the recombinant autoactivated human prethrombin-2 in Pichia pastoris SMD1168. The variant was designed with 4 mutations, E40A, D47A, G48P, and E52A, following the result of a previous study. The synthetic variant gene was first optimized to conform with P. pastoris codon preference. The optimized synthetic gene was cloned in pD912 plasmid using XhoI and SacII restriction enzymes. The transformed P. pastoris was selected on agar plate supplemented with 1,000 µg.mL-1 Zeocin as a selection marker. This study showed that autoactivated prethrombin-2 was succesfully expressed extracellularly by P. pastoris SMD1168. The activity of recombinant autoactivated prethrombin-2 using a chromogenic substrate S-2238 was 0.540 unit/mg. Taken together, these results demonstrated that autoactivated human prethrombin-2 was successfully produced extracellularly in P. pastoris.

Abstract

The symptoms of chikungunya disease are generally similar to the symptoms of other viral diseases, except that chikungunya disease can cause joint disorders and even paralysis. Incorrect initial diagnosis will result in inappropriate treatment of the disease Thus it is necessary to study the identified antibodies that can neutralize and protect the body from alphavirus infections, including chikungunya. There are several antibodies that can neutralize chikungunya including CHK152 which neutralizes virus by inhibiting fusion with stabilizing the surface area of the virus, hindering the exposure of the fusion-loop, likely neutralizing infection by blocking fusion. Otherwise CHK265 inhibits the life cycle of the virus in a way neutralize infection by blocking entry at a post-attachment pre-fusion step. The low energy of binding between antigen-antibodies will stabilize the interaction of both. Therefore, this aims to find the best affinity energy for antibodies to be used in the rapid diagnostic component of anti-chikungunya virus (CHIKV). The binding energies were calculated using FireDock an efficient method for the refinement and rescoring of rigid-body docking solutions, through rearrangement of the interface sidechains and adjustment of the relative orientation of the molecules.The results of this study  obtained energy scores from CKH152 and CHK265 are -125.78 and -29.94, indicate that CHK265 antibody affinity energy is higher than CHK152. It can be concluded that the best affinity energy for CHIKV diagnostic component is CHK152.Keywords: energy affinity; CHK152; CHK265; E1E2 CHIKV; molecular docking

Abstract

The association of type-2 diabetes mellitus (T2DM) and cataract with mtDNA mutation has been reported before. Despite the high prevalence of DM and cataract in Indonesia, a study of the mtDNA variants in Indonesia in correlation with the two diseases is still limited. MT.3243A>G is one of the hotspots mutations for mitochondrial diseases, but the explanation for its occurrence in patients with pure cataract is still elusive. Therefore, the objective of this study was to analyze the mitochondrial genome variants from T2DM and cataract patients in Indonesia using the direct sequencing method. The homology analysis of the genome to the Cambridge reference sequence resulted in 86 variants, including 20 variants that cause amino acid substitutions. Based on the Mitomap data, 17 of the 20 variants were novel. Upon comparison with the 12 normal variant genomes, 11 of 17 variants were suggested to be associated with T2DM and cataract diseases since they code the protein in complex-I (ND4L, ND5, and ND6), complex-III (cytb), and complex-V (ATP6) of the respiratory complex. Interestingly, MT.3316G>A, for the first time, is shown in a pure cataract patient. In addition, the novel phenotype of MT.5460G>A and MT.10398A>G were revealed, which are T2DM and cataract in one patient. Based on our study, these diseases might be related to the disruption of the ATP metabolism due to the structure and function changes of proteins involved in the respiratory complex. This discovery is expected to offer an understanding of the origins of gene-level clinical differences, particularly in Indonesia.

Abstract

Teachers' enthusiasm for taking educator certification is still high. This enthusiasm grew because of the recognition of positions as professional educators after passing certification. This article is based on the results of observations of implementing the teacher professional development competency model through PPG Jabatan. This research aims to obtain an overview of the implications of PPG in the Department for teachers' degrees in mastering professional knowledge competencies, professional learning practices, and professional development. This study employs qualitative research methodologies, utilizing the library as the primary information source and additional literature relevant to the paper's thematic focus. Research findings show that PPG demands participants' and managers' ability, sincerity, readiness, and ability. By analyzing the findings of this research, it can be concluded that the Teacher Professional Education (PPG) program can significantly increase teacher professionalism through an in-depth qualitative approach. PPG participants increase their pedagogical knowledge and skills and experience significant growth in self-understanding and reflection abilities. Conclusion PPG increases the mastery of professional knowledge, professional learning practices, and professional development ranging from advanced, decent, and capable to advanced degrees.
