Abstract

Tanjung Emas Port is the entry and exit point for trade commodities, both regional and international filled with many ships. This condition makes biofouling a very massive process in that place by various types of marine bacteria. The initial formation of a biofilm is relevant to bacterial diversity, colonization and adhesion. The objective of the study was assessing bacterial diversity in relation to with biofouling within Tanjung Mas Port Semarang, by using 16S rRNA eDNA metabarcoding Next-Generation Sequencing (NGS). Seawater samples from aquatic sites of Tanjung Mas harbor was used for DNA extraction and amplification of the 16S rRNA V-3-V4 hypervariable region, followed by sequencing and library construction of eDNA Metabarcoding. Sequence processing and analysis was performed in QIIME 2 and RStudio using DADA2 for advanced sequencing processing and Phyloseq. The results of this research showed that bacteria is the predominant taxon constituting 100% of the community. The taxon consists of Proteobacteria (49.38%), Bacteroidota (8.67%), and Firmicutes (8.88%). Alphaproteobacteria (20.92%) and Gammaproteobacteria (12.39%) dominate at the Class level, emphasizing their versatility and ecological influence. At the Order and Family levels reveals the prevalence of Rhodobacterales (10.04%), Chitinophagales (2.53%), Rhizobiales (3.61%), Rhodobacteraceae (5.67%), Saprospiraceae (3.63%), and Rhizobiaceae (2.15%). It was found that the unculturable taxa dominance in Tanjung Mas Port was 44.66%. These taxonomic entities contribute significantly to the taxonomic and functional diversity of the microbial community, influencing nutrient cycling, organic matter degradation, ecosystem stability and biofilm formation.

Abstract

Produktivitas kapal penangkapan ikan merupakan tingkat kemampuan kapal penangkap ikan untuk memperoleh hasil tangakapan ikan per tahun. Jaring Tarik Berkantong merupakan alat tangkap berkantong besar berbentuk seperti kerucut dan merupakan alat tangkap yang mendominasi di Pelabuhan Branta Pesisir. Tujuan dari penelitian ini yaitu untuk mengkaji penangkapan ikan menggunakan Jaring Tarik Berkantong dan produktivitas hasil tangkapannya. Penelitian ini dilaksanakan pada April-Mei 2022 di Pelabuhan Branta Pesisir, Tlanakan, Pamekasan, Madura. Metode yang digunakan pada penelitian ini adalah metode observasi secara langsung dengan jenis penelitian studi kasus. Metode analisis data yang digunakan adalah status pemanfaatan, produktivitas berdasarkan jumlah hasil tangkapan, produktivitas per ABK, dan produktivitas per trip. Pengoperasian Jaring Tarik Berkantong terbagi menjadi 3 tahapan yaitu persiapan, setting, dan hauling. Hasil analisis status pemanfaatan diperoleh nilai 3.492,84 ton/tahun yang diperoleh dari 80% nilai MSY. Hasil penelitian menunjukkan produktivitas berdasarkan jumlah hasil tangkapan, per ABK dan per trip diperoleh nilai sebesar 5.658 kg/unit atau 5,658 ton/unit, 0.566 ABK/trip, dan 1.885 ton/trip. Nilai produktivitas Jaring Tarik Berkantong di Pelabuhan Branta Pesisir menunjukkan bahwa nilai produktivitas pada penelitian ini telah melebihi peraturan yang telah ditetapkan.The productivity of fishing vessels is the level of ability of fishing vessels to obtain fish catches per year (KEPMEN-KP 2021). Cantrang is a fishing gear that is like a large bag shaped like a cone and is the dominant fishing gear in the Coastal Branta Harbor. The purpose of this study was to determine the process of catching fish using cantrang fishing gear and to determine the productivity value of cantrang catches. This research was conducted in April-May 2022 at the Port of Branta Pesisir, Tlanakan, Pamekasan, Madura. The method used in this research is the observation method with the type of case study research. The data analysis methods used are utilization status analysis, productivity analysis based on the number of catches, productivity analysis per crew, and productivity analysis per trip. Cantrang operation is divided into 4 stages, namely preparation, setting, and hauling. The results of the utilization status analysis obtained a value of 3,492.84 tons/year which was obtained from 80% of the MSY value. The results of the productivity analysis based on the number of catches, per crew and per trip obtained values of 5,658 kg/unit or 5,658 tons/unit, 0.566 crew/trip, and 1,885 tons/trip. The productivity value of cantrang at the Coastal Branta Port shows that the productivity value in this study has exceeded the established regulations.

Abstract

Desa Durbuk merupakan salah satu desa yang terletak di Kecamatan Pademawu, Kabupaten Pamekasan, Madura. Pelaku budidaya ikan pada desa ini tidak banyak karena letak desa yang kurang strategis serta minat maupun harga pakan menjadi kendala untuk sektor perikanan budidaya. Peningkatan minat dan penyelesaian masalah dilakukan oleh tim pengabdian kepada masyarakat dari Univesitas Trunojoyo Madura dengan cara melakukan inisiasi pelatihan budidaya ikan lele untuk menyelesaikan permasalahan pada sektor perikanan di desa tersebut. Kegiatan pelatihan budidaya ikan lele yang bertujuan untuk meningkatkan dan mengembangkan minat masyarakat untuk bergerak kembali di sektor perikanan dilaksanakan dengan menggunakan metode Pendekatan Orang Dewasa (POD) dimana kegiatan dilakukan secara partisipatif dengan diskusi interaktif. Pada saat ini permasalahan pakan bisa dicoba menggunakan campuran probiotik yang dapat membantu laju pertumbuhan dan kekebalan tubuh ikan sehingga bisa mengefisiensikan penggunaan pakan. Manfaat yang dapat diberikan adalah ilmu ataupun tahapan proses budidaya dengan benar, sehingga para pembudidaya dan pemuda atau masyarakat setempat lebih mendalami dan memahami dengan benar. Perlu adanya pendampingan secara intensif demi keberlanjutan kegiatan ini supaya masyarakat bisa benar-benar memahami dan nantinya kegiatan ini memperoleh hasil yang maksimal untuk saat ini dan masa yang akan datang.

Abstract

Perairan Teluk Rembang merupakan salah satu sumber penghidupan bagi penduduk bermata pencaharian nelayan. Penelitian ini menggunakan citra Sentinel-3 OLCI dan SLSTR memiliki resolusi sebesar 300 m x 300m, dengan resolusi spasial tersebut citra yang dihasilkan oleh Sentinel-3 OLCI dan SLSTR dapat dikatakan cukup baik untuk diteliti lebih lanjut. Penelitian ini menggunakan data citra Sentinel-3 OLCI dan SLSTR serta data angin reanalysis ERA-5 dari (Climate Copernicus) yang diolah menggunakan bantuan program SNAP dengan metode C2RCC. Berdasarkan hasil penelitian didapatkan bahwa nilai variabilitas bulanan klorofil-a memiliki nilai yang bervariasi setiap bulannya. Nilai konsentrasi klorofil-a tertinggi terjadi pada bulan Juli sebesar 6,72 mg/l dan terendah pada bulan November sebesar 0,87 mg/l. Nilai konsentrasi Klorofil-a memiliki keterkaitan dengan suhu permukaan laut dan angin. Nilai konsentrasi klorofil-a berbanding lurus dengan kecepatan angin yang nilai korelasinya sebesar -0,54, sedangkan nilai konsentrasi klorofil-a berbanding terbalik dengan suhu permukaan laut yang nilai korelasinya sebesar 0,42. Tingginya nilai konsentrasi klorofil-a terjadi pada saat kecepatan angin meningkat, dan suhu permukaan laut rendah terjadi ketika kecepatan angin tinggi. Begitupun sebaliknya, konsentrasi klorofil-a menurun dan suhu permukaan laut meningkat terjadi pada saat kecepatan angin menurun. Kata kunci: Klorofil-a, Suhu Permukaan Laut, Sentinel-3, Perairan Teluk Rembang Abstract Rembang Bay has been one of the sources of livelihood for residents who work as fishermen. This study uses Sentinel-3 OLCI and SLSTR images with a resolution of 300 m x 300m, with this spatial resolution the images produced by Sentinel-3 OLCI and SLSTR can be said to be good enough to be investigated further. This research uses Sentinel-3 OLCI and SLSTR image data as well as ERA-5 wind reanalysis data from (Climate Copernicus) which was processed using the SNAP program with the C2RCC method. Based on the results of the study, it was found that the monthly variable value of chlorophyll-a has a value that varies every month. The highest value of chlorophyll-a concentration occurred in July of 6.72 mg/l and the lowest was in November of 0.87 mg/l. Chlorophyll-a concentration value have a relationship with sea surface temperature and wind. The value of chlorophyll-a concentration is directly proportional to wind speed with a correlation value of -0.54, while the value of chlorophyll-a concentration is inversely proportional to sea surface temperature with a correlation value of 0.42. High concentration values of chlorophyll-a occur when wind speed increases, and low sea surface temperatures occur when wind speeds are high. Abstract

Pulau Kemujan memiliki kelimpahan sumberdaya ekosistem seperti ekosistem karang, mangrove dan lamun. Perairan bagian barat memiliki bentuk yang unik, karena ada sebagian perairannya berada pada lokasi yang tertutup oleh daratan dan tumbuh subur pohon mangrove. Keberadaan ekosistem mangrove dan bentuk perairan yang semi tertutup akan berpengaruh terhadap distribusi ukuran butirnya. Kajian sebaran sedimen pada Pulau Kemujan ditujukan untuk mendapatkan gambaran mengenai kondisi  sedimen  dan keterkaitannya  dengan  proses  hidrooseanografi  yang  terjadi.  Daerah penelitian mencakup perairan tertutup dan perairan terbuka sebanyak 30 stasiun. Sampel sedimen diambil menggunakan Vanveen grab. Analisis ukuran butir sedimen menggunakan metode dry sieing (pengayakan) dan wet sieving (pemipetan).  Hasil penelitian menunjukkan bahwa pola sebaran sedimen lebih didominansi oleh pasir dan jenis sedimen halus ditemukan di perairan yang lebih dalam ≥7 meter serta lokasi stasiun jauh dari sumbernya (daratan). Pola distribusi sedimen kasar berkaitan dengan kondisi arus yang lebih lemah dan ditemukan pada perairan lebih dangkal. Kata kunci: Ukuran Butir, Sedimen, Arus, Karimunjawa Abstract Kemujan Island has ecosystem resources such as coral, mangrove and seagrass ecosystems. The western waters have a unique shape, because some of the waters are in a location that is covered by land and mangrove trees thrive. the existence of mangrove ecosystems and the shape of the waters that are semi-enclosed will affect the grain size distribution. The study of the distribution of sediments on Kemujan Island is aimed at obtaining an overview of the sediment conditions and their relationship to the hydrooceanographic process. The research area of closed waters and open waters is 30 stations. Sediment samples were taken using a vanveen grab. Sediment grain size analysis used dry sieving and wet sieving (pipepeting) methods. The results obtained show that the distribution of sediment patterns is more dominated by sand and the type of fine sediment is found in deeper waters ≥ 7 meters and the location of the station is far from the source (land). The distribution pattern of coarse sediment is associated with weaker currents and shallower waters. Keywords: Grain Size, Sediments, Current, Karimunjawa

Abstract

Permasalahan efisiensi pakan di dunia perikanan adalah harga bahan dasar pakan yang semakin tinggi dan sukar diperoleh. Pemanfaatan probiotik komersil ini masih belum dapat meningkatkan nilai produktivitas dan angka produksi dari hasil kegiatan budidaya ikan yang diharapakan, salah satunya pada komoditas ikan lele (Clarias gariepinus). Oleh karena itu, inovasi baru sangat diperlukan untuk mengatasi masalah tersebut dengan mengedepankan prinsip ramah lingkungan yang dapat diterapkan kepada masyarakat. Perlunya penelitian terkait aplikasi formulasi probiotik ikan menjadi hal yang menarik untuk dikaji. Abstract

Segara Anakan Mangrove Forest in Cilacap is one of the most extensive mangrove forests on the southern coast of Java Island. This study aims to determine the level of herbivory and environmental conditions. The sampling technique used was purposive random sampling, which considered the different characteristics of the vegetation. Sonneratia caseolaris dominated Station 1, while Station 2 was more diverse, including Avicennia marina, Avicennia alba, Rhizophora mucronata, Sonneratia caseolaris, and Aegiceras corniculatum. Each station is spread over three points, each with five sampling plots, representing the distribution and zonal homogeneity of carrying capacity. The research was conducted in May, August, and November 2023, based on the different temporal seasons in Indonesia. The results showed biotic factors from herbivory activity and abiotic factors such as the hydrological cycle caused mangrove leaf damage. The class of leaf damage caused by herbivory at all observation stations was mainly in class I (<2.5%). The most leaf damage was found in old leaves, which is related to the optimal photosynthesis process producing carbohydrates as a food source for herbivores. Additionally, tannin content in the leaves plays a role in protection from excessive UV radiation during the dry season. This condition indicates that Segara Anakan mangrove forest is still in good condition. PCA analysis results show that mangrove leaf damage is closely related to the age of the leaves.

Abstract

The density of community activities around the Ujing Piring Bangkalan estuary has the potential to produce waste containing ammonia which is very dangerous for the environment and causes a decrease in water quality and threatens the existence of aquatic organisms including phytoplankton. The purpose of this study was to determine the relationship between ammonia levels and the abundance of phytoplankton in the morning and evening in the Ujung Piring Bangkalan estuary. The sampling time was carried out in the morning (08.00-09.30 WIB) and afternoon (15.00-16.30 WIB) three times in three weeks using 10 micron plankton nets in the Ujung Piring Bangkalan estuary waters. The sampling method used a purposive sampling method with phytoplankton analysis using the census method and continued with statistical tests using the linear regression method. The relationship between ammonia levels and phytoplankton abundance in the morning and evening were 0.1537 with a percentage of 15% and 0.0593 with a percentage of 0.05%, respectively. This value states that ammonia levels have a negative correlation with the abundance of phytoplankton in the morning and evening in the Ujung Piring Bangkalan estuary

Abstract

SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2) is a virus responsible for the infectious disease of COVID-19 (Coronavirus Disease 2019), whose development is still being monitored. One way to deal with the virus’s development is by searching for natural-based medicines that prevent and treat SARS-CoV-2 infection. The abundant biodiversity can be used as a source of treatment. Among many natural resources, seaweed is one of the natural resources rich in bioactive components. Sulfated polysaccharide is one of the potential bioactive compounds in seaweed because it has antiviral effects and the potential to treat SARS-CoV-2. This research aims at increasing the potential of Karimunjawa green seaweed sulfate polysaccharides for antiviral capabilities in SARS-CoV-2 through molecular docking. This research began with identifying the morphology of Karimunjawa seaweed. Identify the components of sulfated polysaccharide compounds based on literature studies according to the species that have been identified. Analysis of the antiviral ability of SARS-CoV-2 based on its binding ability to the SARS-CoV-2 target protein through a molecular docking computational program and testing drug compounds using the Lipinski rule. Based on the research results, it is known that the seaweed obtained from Karimunjawa based on morphology, belongs to the genus Kappaphycus. Molecular docking with a ligand and spike glycoprotein (6LZG) resulted in the hexadecanoic acid compound having a binding free energy of -5.3 kcal.mol-1, which was the compound with the lowest yield compared to other test compounds. The prediction of the physicochemical properties of all test compounds fulfills Lipinski's five rules and has the potential to be used as medicinal compounds.

Abstract

Pemalang Regency has a mangrove area of around 2,839.44 but has been damaged reaching 453.38 Ha or about 16% of the existing area. This causes ecological functions in resisting abrasion, erosion, sedimentation, and carbon capture to decrease. Government efforts to restore the function of mangroves on the North Coast of Central Java carried out by rehabilitation require input from a water productivity approach. This study aims to determine the content and distribution of chlorophyll-a found in Pemalang Waters, Central Java. This was done by in situ observations in Pemalang Waters to see the fertility level of the waters, by purposive sampling at 20 sampling site, on April 30, 2024. The results obtained will be interpolated with ex situ data, including the Indonesian Earth Map (RBI), tidal data obtained from the SRGI website, flow data obtained from the Batnas website, and wind data obtained from the BMKG website. The next geospatial approach is ODV 4, Ms.Excel, and WR Plot application mapping; current analysis will be performed using ArcGIS 10.8 and Mike 21 applications; while the tidal analysis is carried out by the Admiralty method. The results of research conducted in Mojo Waters, Pemalang showed that the range of chlorophyll-a values was in the range of 1,858 - 41,287 mg / m3. The results were analyzed in sampling areas including rivers, estuaries, lagoons, eastern drift currents, western drift currents, waters still affected by estuaries, and the high seas. The distribution of chlorophyll values in these waters is the impact of nutrient supply from land because it is closely related to human activities, freshwater and seawater resuspension, drift currents, breaking waves, tidal currents. This causes the distribution pattern to occur from northeast to southwest.
