Abstract

SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2) is a virus responsible for the infectious disease of COVID-19 (Coronavirus Disease 2019), whose development is still being monitored. One way to deal with the virus’s development is by searching for natural-based medicines that prevent and treat SARS-CoV-2 infection. The abundant biodiversity can be used as a source of treatment. Among many natural resources, seaweed is one of the natural resources rich in bioactive components. Sulfated polysaccharide is one of the potential bioactive compounds in seaweed because it has antiviral effects and the potential to treat SARS-CoV-2. This research aims at increasing the potential of Karimunjawa green seaweed sulfate polysaccharides for antiviral capabilities in SARS-CoV-2 through molecular docking. This research began with identifying the morphology of Karimunjawa seaweed. Identify the components of sulfated polysaccharide compounds based on literature studies according to the species that have been identified. Analysis of the antiviral ability of SARS-CoV-2 based on its binding ability to the SARS-CoV-2 target protein through a molecular docking computational program and testing drug compounds using the Lipinski rule. Based on the research results, it is known that the seaweed obtained from Karimunjawa based on morphology, belongs to the genus Kappaphycus. Molecular docking with a ligand and spike glycoprotein (6LZG) resulted in the hexadecanoic acid compound having a binding free energy of -5.3 kcal.mol-1, which was the compound with the lowest yield compared to other test compounds. The prediction of the physicochemical properties of all test compounds fulfills Lipinski's five rules and has the potential to be used as medicinal compounds.

Abstract

Pemalang Regency has a mangrove area of around 2,839.44 but has been damaged reaching 453.38 Ha or about 16% of the existing area. This causes ecological functions in resisting abrasion, erosion, sedimentation, and carbon capture to decrease. Government efforts to restore the function of mangroves on the North Coast of Central Java carried out by rehabilitation require input from a water productivity approach. This study aims to determine the content and distribution of chlorophyll-a found in Pemalang Waters, Central Java. This was done by in situ observations in Pemalang Waters to see the fertility level of the waters, by purposive sampling at 20 sampling site, on April 30, 2024. The results obtained will be interpolated with ex situ data, including the Indonesian Earth Map (RBI), tidal data obtained from the SRGI website, flow data obtained from the Batnas website, and wind data obtained from the BMKG website. The next geospatial approach is ODV 4, Ms.Excel, and WR Plot application mapping; current analysis will be performed using ArcGIS 10.8 and Mike 21 applications; while the tidal analysis is carried out by the Admiralty method. The results of research conducted in Mojo Waters, Pemalang showed that the range of chlorophyll-a values was in the range of 1,858 - 41,287 mg / m3. The results were analyzed in sampling areas including rivers, estuaries, lagoons, eastern drift currents, western drift currents, waters still affected by estuaries, and the high seas. The distribution of chlorophyll values in these waters is the impact of nutrient supply from land because it is closely related to human activities, freshwater and seawater resuspension, drift currents, breaking waves, tidal currents. This causes the distribution pattern to occur from northeast to southwest.

Abstract

In the lagoon area of the Mangrove Information Center (PIM), Pekalongan, annual flooding leads to inundation in some land areas, particularly pond areas, lagoons, and residential areas. The area of submergence due to tidal floods in 2020 reached 783.99 hectares. The construction of embankments to the south of the lagoon as protection against flooding has transformed the northern area into a lagoon. Our research, crucial for understanding and managing these environmental changes, aims to determine and analyze the content and distribution of nitrate in the Lagoon Area of Mangrove Information Center Waters, Pekalongan. This was achieved through a thorough process of in situ observations based on purposive sampling at six sampling sites on June 22, 2023. The nitrate distribution data was then interpolated with a 2D current hydrodynamic model, ensuring the most accurate representation of the data. The value of nitrate content in the Mangrove Information Center lagoon ranges between 0.0065-0.1072 mg/L. This distribution pattern, influenced by deposits of mangrove litter, the depth of the lagoon, and water circulation, reflects the accumulation of physical and chemical influences in the waters.

Abstract

Rajungan (Portunus sp) merupakan salah satu komoditas utama di Kabupaten Lamongan, Jawa Timur. Penangkapan rajungan di Daerah Kabupaten Lamongan khususnya di Desa Sedayulawas menggunakan Gillnet satu lapis dan dua lapis karena lebih mudah dan optimal digunakan. Pada penelitian ini menggunakan dua jaring yakni Gillnet satu lapis dengan mata jaring 4 inch dan dua lapis dengan mata jaring 4 inch dan 9 inch. Penelitian ini bertujuan mengetahui deskripsi Gillnet satu lapis dan dua lapis dan perbedaan komposisi tangkapan hasil tangkapannya di Desa Sedayulawas, Kabupaten Lamongan. Metode penelitian ini dilakukan dengan 10 kali pengulangan dengan lama perendaman jaring 8 jam, kemudian data yang didapatkan dianalisa secara deskriptif kuantitatif. Gillnet satu lapis dan dua lapis memiliki bagian yang sama seperti tali ris atas, tali ris bawah, timah pemberat, namun terdapat perbedaan pada bagian pelampung jaring dan lembar jaring. Dari total tangkapan keseluruhan, Gillnet satu lapis mendapatkan tangkapan rajungan 34%, hasil tangkapan sampingan ekonomis 3%, dan hasil tangkapan sampingan non ekonomis 18%. Sedangkan Gillnet dua lapis mendapatkan rajungan 10%, hasil tangkapan sampingan ekonomis 7%, dan hasil tangkapan sampingan non ekonomis 29%. Dengan demikian dapat disimpulkan penggunaan Gillnet satu lapis dan dua lapis terdapat perbedaan terhadap jumlah individu tangkapan.

Abstract

Permasalahan efisiensi pakan di dunia perikanan adalah harga bahan dasar pakan yang semakin tinggi dan sukar diperoleh. Pemanfaatan probiotik komersil ini masih belum dapat meningkatkan nilai produktivitas dan angka produksi dari hasil kegiatan budidaya ikan yang diharapakan, salah satunya pada komoditas ikan lele (Clarias gariepinus). Oleh karena itu, inovasi baru sangat diperlukan untuk mengatasi masalah tersebut dengan mengedepankan prinsip ramah lingkungan yang dapat diterapkan kepada masyarakat. Perlunya penelitian terkait aplikasi formulasi probiotik ikan menjadi hal yang menarik untuk dikaji. Tujuan dari penelitian ini adalah mengetahui pembuatan probiotik ikan, parameter kualitas air, dan pertumbuhan (panjang dan bobot) mutlak ikan lele

Abstract

Soft-shell crabs are a type of crab harvested after molting or shedding their old shells. Soft-shell crabs are known for their high market value, both in domestic and international markets. Technological advancements in crab farming include the mangrove crab mutilation system to produce soft-shell crabs (soka). However, many communities still lack an understanding of this technology. The mutilation technology is known to provide benefits, so we conducted a community outreach program to impart knowledge about the molting process in crabs and provide training on how to mutilate crabs in Kamal Village. Additionally, in the crab farming process, there are issues related to water quality management and feeding. The outreach program also aims to provide solutions to these problems. The method employed is the Adult Learning Approach, which is carried out through participatory lectures and interactive discussions. Meanwhile, the crab mutilation training is conducted using the Demonstration and Example Method, where participants practice the crab mutilation process after receiving demonstrations from the facilitators. The molting process and the sustainability of the outreach program are continued with ongoing mentoring until desired outcomes are achieved.

Abstract

Pupuk pro analitik (PA) biasanya digunakan budidaya Chlorella sp., tetapi mahalnya harga pupuk PA sehingga diperlukan alternatif pupuk yang bisa penuhi kebutuhan nutrisi Chlorella sp. Inovasi pupuk alternatif ini memiliki prospek dalam pengganti pupuk PA yang mahal dipasaran dengan pemanfaatan limbah ikan yang diperoleh dari pasar hamadi, Jayapura. Pupuk yang digunakan merupakan pupuk organik yang diambil dari limbah ikan cakalang. Umumnya penjualan ikan dipasar bermacam-macam ikan, namun mayoritas ikan yang dijual di pasar hamadi adalah ikan cakalang. Tujuan dari riset ini merupakan menganalisis pengaruh pupuk organik cair dari limbah cakalang terhadap laju perkembangan, biomassa, serta klorofil a Chlorella sp. Tata cara yang digunakan dalam riset ini merupakan tata cara Rancangan Acak Kelompok (RAK) dengan memakai 2 perlakuan dengan masing masing 3 ulangan. Perlakuan yang diberikan pada riset ini merupakan pupuk organik cakalang dengan dosis 1 ml/ l serta kontrol (1 ml/ l Guillard). Parameter utama yang diamati merupakan pertumbuhan Chlorella sp., serta klorofil a. Bersumber pada hasil pertumbuhan dari aplikasi pupuk organik cair dari limbah ikan cakalang dengan pupuk kontrol (Guillard) diperoleh hasil tertinggi pada control dan pupuk ikan sebesar 12 x 108 dan 11.15 x 108 sel/mL. Sedangkan pada hasil klorofil diperoleh bahwa untuk control sebesar 1.85 mg/L serta hasil pupuk ikan 1.62 mg/L Hasil menunjukkan bahwa limbah pupuk ikan cakalang berpotensi dijadikan pupuk komersial guna meningkatkan pertumbuhan mikroalga Chlorella sp.

Abstract

Cadmium (Cd) heavy metal pollution not only affects aquatic ecosystems but also has toxic effects on human health. Bioremediation using microalgae Nannochloropsis sp. is considered more economical and sustainable for overcoming heavy metal pollution. This study aims to molecular characterization of Nannochloropsis sp. with tufA gene markers, determine the effect of different Cd concentrations on the growth and morphology of Nannochloropsis sp. and the ability of Nannochloropsis sp. to absorb Cd concentrations. The methods used include DNA isolation, quantitative and qualitative DNA analysis, amplification of tufA gene, sequencing result analysis, phylogenetic tree analysis, and bioremediation test. The results of molecular characterization showed that Nannochloropsis sp. has similarities with N. oceanica strain BR2 plastid (CP044614.1) with 60% bootstrap value. Nannochloropsis sp. grew at different Cd concentration (0, 2, 4, and 6 ppm) but optimal at 0 ppm Cd concentration. Morphology of Nannochloropsis sp. showed morphological differences in the cell structure. The cell was broken and the color turned increasingly clear. Different concentrations of Cd metal on Nannochloropsis sp. growth significantly affected the decline in Nannochloropsis sp. growth. Nannochloropsis sp. has the potential to absorb heavy metal Cd with an efficiency that increases as the concentration reaching 62.6% at 6 ppm. It can be concluded that molecular characterization using the tufA marker was effective in showing that Nannochloropsis sp. had closed relation with N. oceanica strain BR2 plastids. Cadmium (Cd) exposure significantly reduced growth and caused cell damage, while Nannochloropsis sp. effectively adsorbed Cd and lowering Cd levels by 3.76 ppm.

Abstract

Mangrove litter is a critical component of the nutrient cycle, reflecting soil fertility and water quality within the mangrove ecosystem. This study investigates the dynamics of litter productivity in the Segara Anakan mangrove forest in Cilacap. The study area is divided into two locations with three points, with five litter traps installed at each point. Sonneratia caseolaris dominates vegetation characteristics at Station 1, while Station 2 has more diverse vegetation, including Sonneratia caseolaris, Aegiceras corniculatum, Acanthus ilicifolius, Derris trifoliata, Rhizophora mucronata, Avicennia alba, and Avicennia marina. Sampling follows seasonal changes in Indonesia, covering June 2023 to March 2024. The primary data for this study include aboveground litterfall production, estimated using litter traps, and the litter layer on the forest floor collected through soil transects. Secondary data comprise weather measurements, including air temperature, rainfall, wind speed, and humidity. Litterfall and litter layer were collected four times: June (dry season), September (transition from dry to rainy season), December (rainy season), and March (transition from rainy to dry season). The results indicate a total litter productivity of 18.99 tonnes ha-1 yr-1. The lowest litter production was observed in September, while the highest was in March. Leaf litter contributed the most, accounting for 71.49%, followed by twigs at 19.19%, flowers/fruits at 8.31%, stipules at 0.62%, and the lowest portion from other unidentified materials at 0.39%. Litter productivity shows a strong correlation (0.99) with rainfall. The main factors influencing litter productivity include environmental factors, primarily rainfall, and physiological factors, such as flowering and fruiting phenology.

Abstract

Heavy metal lead (Pb) pollution in aquatic environments poses a serious threat to aquatic ecosystems and human health. The microalga Thalassiosira  sp. has potential as a bioremediation agent for heavy metals; however, further studies are needed to maximize its capability. This study aims to molecularly characterize Thalassiosira  sp. using the tufA gene, evaluate the effects of Pb on growth and cell morphology, and assess its Pb uptake efficiency. Methods included DNA isolation, phylogenetic analysis, growth measurement using a hemocytometer, Pb uptake testing via ICP-OES, and cell morphology analysis. Results showed that the tufA gene could molecularly characterize Thalassiosira  sp., with a phylogenetic similarity of 99.53% to Conticriba weissflogii (MH571875.1). Pb significantly inhibited growth at concentrations of 0.03 ppm and 0.05 ppm and caused cell aggregation and pigment degradation at higher concentrations (2 ppm). This microalga exhibited the highest Pb uptake efficiency at lower concentrations (0.03–0.05 ppm), reaching 100%, while at 2 ppm, the efficiency decreased to 40%. This study highlights the potential of Thalassiosira  sp. in remediating Pb, providing a foundation for the development of microalgae-based biotechnological applications.
