Abstract

Streptococcus agalactiae is a bacterium that causes streptococcosis, which attacks tilapia. This research was conducted from March to June 2022 at the Laboratory of Parasites and Fish Diseases, Faculty of Fisheries and Marine, University of Riau. This study aims to understand the best dose of fermented Kaempferia galanga and Curcuma xanthorrhiza, which are added to the feed to increase Oreochromis niloticus immunity. The method used was experimental with a completely randomized design (CRD) with five treatment levels and three replications. The treatments applied were Kn: Negative control (no treatment used), Kp: positive control (no treatment used and infected with S.agalactiae), P1 (100 mL/kg feed), P2 (125 mL/kg feed), P3 (100 mL/kg feed) and infected with S. agalactiae. The fish were contaminated on the 32nd day at a density of bacteria 108 CFU/mL of 0.1 mL/fish intramuscularly, and then the fish were reared up to the 46th day. The results showed that the fermented K.galanga and C.xanthorrhiza affected the haematology of O.niloticus. The best dose was in P3 with a dose of 150 ml/kg of feed which showed total erythrocytes of 2.01 x106 cells/mm3, hematocrit value of 27.33%, a hemoglobin level of 7.93 g/dL, total leukocytes of 6.59 x104 cells/mm3, lymphocytes 77.33%, monocytes 14.00%, neutrophils 8.67%, growth with a weight of 34.06 g, a length of 7.5 cm and a survival rate of 93.33%. Based on the data obtained, it can be concluded that fermented K.galanga and C.xanthorrhiza can increase the immunity of O.niloticus against Streptococcosis disease

Abstract

This research was conducted 30 days from June to July 2022 at the Aquaculture Laboratory, Faculty of Fisheries and Marine Sciences, Universitas Riau, Pekanbaru. This study aimed to analyze the osmoregulation response of the Asian redtail catfish to different salinities and to obtain optimal salinity for good physiological performance for the Asian redtail catfish. The method used was an experimental method using a completely randomized design (CRD) with five treatments and four replications, namely, three ppt salinity treatment, six ppt salinity treatment, nine ppt salinity treatment, 12 ppt salinity treatment, and 15 ppt salinity treatment. The test fish used were Asian redtail catfish with an average size of 7.17±0.04 cm. Asian redtail catfish seeds were obtained from the Tibun Fish Seed Center, Kampar. The container used is an aquarium with a size of 60x40x40 cm. Research data shows that nine ppt salinity is the best for Asian redtail catfish osmoregulation of 0.5 mOsm/L H2O. The best cortisol hormone content is found at three ppt salinity, the lowest oxygen consumption is at three ppt salinity of 0.9 mgO2/g/hour, the best survival was found at three ppt salinity of 100%, and the growth in weight and length was in the three ppt treatment of 8.90 g and 2.04 cm. Water quality parameters include temperature 25-30oC and dissolved oxygen 5.04-8.04 ppm.

Abstract

Herbs Fermented consist of Kaempferia galanga L, Curcuma xanthorrhiza Roxb, and Curcuma domestica Val, which are environmentally friendly disease prevention alternatives that can be added to feed because they have several advantages, such as increasing appetite, digestion, growth, disease resistance, and reducing stress levels in fish. This research was carried out from August to December 2021 at the Marine Science Biotechnology Laboratory and Fish Parasites and Diseases Laboratory, Faculty of Fisheries and Marine, Universitas Riau. This study aimed to obtain the best dose from adding fermented herbs to increase the immune system of Oreochromis niloticus and prevent diseases caused by Aeromonas hydrophila bacteria in terms of profile erythrocytes. The method used was an experimental method using a Completely Randomized Design (CRD) with one factor, five levels of treatment, and three replications so that 15 experimental units were obtained. The challenge test was carried out on the 32nd day using A.hydrophila with a density of 108 CFU/mL. The best dose was found in P3 with a dose of 200 mL/kg of feed, which showed a total erythrocyte of 2.09x106 cells/mm3, hemoglobin level of 7.70 g/dL, hematocrit value of 37.70%, and a normal erythrocyte morphology. Water quality ranged from temperature 27-29oC, pH 6.9-7.2, DO 3.1-4.4 mg/L, and Ammonia 0.016-0.024 mg/L. Based on the data obtained, it can be concluded that fermented herbs can increase the immunity of O. niloticus to diseases caused by A.hydrophila.

Abstract

Multi-cell probiotic containing bacteria Nitrosomonas sp., Nitrobacter sp., and Bacillus sp. This research was conducted from March to May 2021 at the Fish Disease and Parasite Laboratory, Department of Aquaculture, Faculty of Fisheries and Marine, Universitas Riau. This study aimed to analyze the hematology of Asian redtail catfish reared in media that was added with probiotics periodically and to obtain the best time interval for giving probiotics to the rearing medium. The method used in this study is an experimental method by applying a Completely Randomized Design (CRD) with five treatment levels and three replications. The treatments used were the administration of probiotics at a dose of 0,01 mL/L of water with different time intervals, P0 (without treatment), P1 (5 days once), P2 (10 days once), P3: (15 days once), and P4 (20 days once). Fish were reared for 50 days. The results show that the Asian redtail catfish reared with the addition of probiotics in the rearing medium had a significant effect (P<0.05) with the best time interval being once every 5 days in the P1 treatment, seen from the total erythrocytes 2.43 x 106 cells/mm3, hematocrit level 28.67%, hemoglobin level 7.40 g/dL, total leukocytes 9.92 x 104 cells/mm3, leukocrit level 1.83%, and glucose 49.67 g /dL. Water quality during the study was still in the normal range that could support the growth of Asian redtail Catfish, namely temperature 26-27.1 C, pH 5.8-6.5, DO 6.0-7.4 mg/L, and ammonia 0.0011-6.5 0.0047 mg/L.

Abstract

Aeromonas hydrophila bacteria often attack internal organs such as the liver and intestines. This research was conducted in June – August 2021 at the Laboratory of Fish Parasites and Diseases, Faculty of Fisheries and Marine, Universitas Riau. Histology preparations were made at the Bukittinggi Veterinary Center. This research aimed to obtain the best dose addition of papaya leaf fermented solution which could reduce intestinal and liver tissue damage of striped catfish (P.hypophthalmus) which was challenged with A.hydrophila. The method used is an experimental method with a completely randomized design (CRD) with 4 treatment levels and 3 replications; Kp (Feed not given papaya leaf fermented solution + infected with A.hydrophila), P1 (16 ml/kg feed of papaya leaf fermented solution + infected with A.hydrophila), P2 (16 ml/kg feed of papaya leaf fermented solution) + infected with A.hydrophila), P3 (16 ml/kg of papaya leaf fermented solution feed + infected with A. hydrophila). The fish used was 8-10 cm with a density of 1 fish/3 L totaling 10 fish with a container measuring 40x30x30 cm. Infection by injection in the intramuscular section as much as 0.1 mL/head with a bacterial density of 108 CFU/mL. The parameters observed were clinical symptoms for 72 hours and histopathology of the intestine and liver of striped catfish. The results showed that the intestine saw the presence of hemorrhage, necrosis, and edema. While the liver shows abnormalities, such as hemorrhage, necrosis, hypertrophy, and fatty degeneration.

Abstract

The demand for ornamental fish is currently increasing while the availability in nature is decreasing due to overfishing. Clownfish (Amphiprion ocellaris) is a type of marine ornamental fish that is widely kept in aquariums, but the problem is the rapid decline in water quality due to the activity of these fish. The recirculation system is one way that can be used in water quality management. The study aimed to analyze the type of filter used to stabilize seawater quality in clownfish (rearing containers. This research was conducted from January to April 2021 at the Research Centre of Sibolga Fisheries College. The method used was an experimental method using a one-factor Completely Randomized Design (CRD) with four treatment levels and three replications. The treatments in this study were P0 (unfiltered), P1 (cotton filter), P2 (fiber filter), and P3 (activated charcoal filter). The fish used was 2 – 3 cm in size, kept in an aquarium measuring 60x40x40cm with a stocking density of 2 fish/L of water and fed tubifex 2 times a day, at 08.00 and 18.00 WIB as much as 5% of their body weight. Parameters measured were temperature, pH, DO, salinity, ammonia, and phosphate, abundance of bacteria, phytoplankton, and survival of clownfish. Water quality measurements are carried out every 7 days. The results obtained from the improvement and analysis of water quality for the cultivation of clownfish in the recirculation system provides an influence on water quality in the rearing container. The results obtained were temperature 27.9–28.0 0C, pH 8.3–8.4, DO 7.5–8.0 mg/L, salinity 33-34 ppt, ammonia 0.04–0.06 mg/L, phosphate 0.47–0.48 mg/L and bacterial abundance 2.35x104 CFU/mL with a survival rate of 95.55%.

Abstract

The aim of this study was to analyze the effects of different salinities on osmotic performance rate, growth performance, and oxygen consumption rate Pangasianodon hypophthalmus. The method used is an experimental method by applying a completely randomized design (CRD) with four treatments, salinity 0 ppt, salinity 5 ppt, salinity 7 ppt, and salinity 9 ppt. The fish specimen used is 8 cm in length, weighing 4 g, were raised in a 54 L tank containing 40 L of water at a density of 1 fish 2 L-1 and kept for 45 days. Feed with commercial pellet 3 times a day in at satiation. The treatment with 5 ppt salinity showed the best effects, yielding an osmotic performance rate of 21,66 mOsm/I H2O, absolute weight growth of 9,63 g, the absolute length of 3.54 cm, the specific growth rate of 2,99 %, the oxygen consumption rate of 0,410 mgO2/g body weight/hour and survival rate 100 %. The water quality is at a level suitable for the growth of P. hypophthalmus. It was concluded that 5 ppt salinity can minimize the value of the osmotic performance rate, increase growth rates, minimize the level of oxygen consumption and increase.

Abstract

This research was conducted in April – June 2021in the Laboratory of Parasites and Fish Diseases, Faculty of Fisheries and Marine, Universitas Riau. Histological preparations were carried out at the Bukittinggi Veterinary Center (BVet). The aim of this study was to analyze the kidney structure of striped catfish (P.hypophthalmus) infected with Edwardsiella tarda with different bacterial densities. The benefit of this research is to provide information about changes in the structure of kidney tissue due to infection with E.tarda, so that later treatment solutions can be sought. The method used was an experimental method with a completely randomized design (CRD) consisting of 4 treatment levels and 3 replications. The treatments applied were P0 (fish not infected with E.tarda), P1 (fish infected with E.tarda at a density of 106 CFU/mL), P2 (107 CFU/mL), and P3 (108 CFU/mL). Striped catfish (P.hypophthalmus) used are 8-12 cm in size with a density of 1 fish/3 L. Infection is done by injection in the intraperitoneal as much as 0.1 mL/fish. The results of the study found that clinical symptoms caused by dull and protruding eyes, dull body surface color, flaky fins, body surface wounds, and bleeding. In histopathological observation, the kidneys with different bacterial densities experienced the highest damage at P3 (108 CFU/mL), namely necrosis, hemorrhage, and hypertrophy.

Abstract

Lactic acid bacteria can improve digestibility because they can be absorbed directly by the body. The purpose of this research was to obtain lactic acid bacteria types from the intestines of striped catfish (Pangasionodon hypophthalmus) that had been fed fermented herbal feed. This research was conducted from June 2019 to April 2020. The treatment used in this research was with different doses and stocking densities. The test was carried out using the disc diffusion method or the Kirby-Bauer method, which was done by measuring the inhibition zone around the paper disc. Identification of LAB was carried out by phenotypic and genotypic. The results showed that as many as four of the selected isolates were able to inhibit the growth activity of the pathogenic bacteria Aeromonas hydrophila. Isolates with code K had an inhibition zone of 6.7 mm against A.hydrophila, an inhibition zone of 7.5 mm for P1D2 isolates, P2D1 isolates had an inhibition zone of 7.0 mm, P2D2 isolates had an inhibition zone of 7.8 mm. Abstract

Metode Bioflok adalah salah satu dari sekian banyak strategi pemeliharaan yang diterapkan untuk meningkatkan produktivitas ikan lele. Teknologi Bioflok pada kegiatan ini berupaya untuk menekan konsumsi air yang dibutuhkan selama budidaya, menekan kuantitas limbah yang dihasilkan, serta meningkatkan produktivitas ikan lele yang dihasilkan. Kegiatan ini bertujuan untuk memberikan wawasan yang mendalam terkait budidaya ikan. Pemahaman yang lebih baik mengenai budidaya ikan lele serta meningkatkan kemampuan produksi. Metode kegiatan ini dimulai dengan perencanaan, pelaksanaan, refleksi, laporan serta pendampingan dan evaluasi kegiatan. Hasil yang diperoleh dari kegiatan ini adalah budidaya ikan lele dengan sistem bioflok dapat mengembangkan skill yang berkaitan dengan kewirausahaan berbasis perikanan seperti budidaya ikan, pengolahan produk perikanan, pengemasan produk serta manajemen pemasaran produk perikanan tersebut. Budidaya Lele sistem bioflok merupakan inovasi baru yang memiliki keunggulan dapat diterapkan di lahan yang sempit serta dapat mengolah air limbah secara biologis.
