Abstract

Ikan nila merah (Oreochromis niloticus) merupakan salah satu komoditas perikanan yang memiliki potensi ekonomi tinggi. Penelitian ini dilaksanakan pada bulan Juni sampai dengan November, 2023 di Laboratorium Parasit dan Penyakit Ikan, Fakultas Perikanan dan Kelautan Universitas Riau, Pekanbaru. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian pakan mengandung probiotik terhadap hematologi ikan nila merah yang dipelihara dalam sistem bioflok. Metode yang digunakan Rancangan Acak Lengkap (RAL), dengan 4 taraf perlakuan dan 3 ulangan, yaitu P0 (kontrol), P1, P2 dan P3 adalah penambahan probiotik dengan dosis 10, 15 dan 20 mL/ kg Pakan. Hasil penelitian didapatkan bahwa perlakuan terbaik pada penambahan probiotik sebanyak 15 mL/ kg Pakan dengan nilai total eritrosit 1,83 x 106 sel/mm3, nilai hematokrit 27,67%, kadar hemoglobin 12,67 g/dL, total leukosit 3,6 x 104 sel/mm3, kadar leukokrit 3,53%, limfosit 78,67% monosit 2,33% neutrofil 1,67% trombosit 17,33% dan tingkat kelulushidupan 96,67%. Kualitas air berkisar suhu 27-27,5 oC, pH 6,5-6,9, DO 5,2-5,4 mg/L, ammonia 0,004-0,005 mg/L. Berdasarkan data yang diperoleh dapat disimpulkan bahwa pelet yang diperkaya probiotik mampu meningkatkan derajat kesehatan ikan nila merah

Abstract

Bay leaf (Syzygium polyantha) has antibacterial compounds like flavonoid, tannin, alkaloid, essential oil, vitamin C, and minerals. The purpose of this study was to obtain the best dose of bay leaf solution to cure catfish infected by Aeromonas hydrophila and seen from total erythrocytes, hemoglobin levels, hematocrit levels, and morphology of erythrocytes. The method used is an experimental method with a Completely Randomized Design (CRD), one factor with five treatments. The treatment applied is Kn (without immersed with bay leaf solution and not infected by A.hydrophila), Kp (without immersed with bay leaf solution and infected by A.hydrophila), P1 (100 ppm), P2 (1100 ppm), P3 (1200 ppm). The curing is done by immersed with bay leaf solution for 5 minutes, carried out three times with an interval of 24 hours. The results show that bay leaf solution able to cure catfish infected by A.hydrophila. The best dose is 1200 ppm with total erythrocytes was 244 x 106 cell/mm3, hemoglobin level was 11,67 g/dL, hematocrit level was 34,33% and the survival rate was 96,67%. Water quality during the study is temperature 27,3-28,0°C, pH 6,5-6,8, DO 3,4-4,2 mg/L and ammonia 0,016-0,019 mg/L.

Abstract

Lactic Acid Bacteria (LAB) are probiotic-forming bacteria that have characteristics, like be able to ferment sugar or carbohydrates and produce lactic acid as the final product of fermentation. The purpose of this study was to obtain isolates of lactic acid bacteria from the digestive tract of Asian Swamp Eel (Monopterus albus) and to determine the inhibition produced by these lactic acid bacteria in inhibiting pathogenic bacteria such as Aeromonas hydrophila and Pseudomonas sp. The method was used in this study is experimental method, by taking the digestive tract of Asian Swamp Eel and then identifying the lactic acid bacteria and characterized by morphological and biochemical characteristics and tested of the antagonist against the pathogenic bacteria A.hydrophila and Pseudomonas sp. using the Kirby Bauer disc method (disc method). From the research results obtained 5 types of candidate lactic acid bacteria with each code BL121, BL263, BL142, BL242 and BL342. The five isolates of lactic acid bacteria were able to inhibit A.hydrophila and Pseudomonas sp. with a Inhibition range of 8-16 mm. Based on the results of the study, there were three isolates that had inhibitory power against bacteria A. hydrophila (12-16 mm) and Pseudomonas sp (11-13 mm) with codes BL121, BL263 and BL142 and were classified as strong. The identification results obtained are bacteria from the genus Bacillus sp.

Abstract

Streptococcus agalactiae is a pathogen that causes significant losses in tilapia (Oreochromis niloticus) aquaculture. One alternative control is the use of natural ingredients such as fermented herbs. This study aims to analyze changes in the histopathological structure of the gills and liver of tilapia fish fed with feed containing fermented herbs after being infected with S. agalactiae. The research was conducted from March to December 2023 at the Laboratory of Parasites and Fish Diseases, Faculty of Fisheries and Marine Sciences, Universitas Riau and Bukittinggi Veterinary Center. The method used was a one-factor, completely randomized design (CRD) with five treatments and three replicates. The treatments consisted of negative control (Kn), positive control (Kp), and three feed treatments with fermented herbal medicine doses of 100 (P1), 125 (P2), and 150 mL/kg feed (P3). S. agalactiae infection was carried out on day 31 at a dose of 10⁸ CFU/mL. The results showed that the P3 treatment gave the best results, characterized by minimal damage to gill tissue (only hypertrophy) and liver (mild vacuolar degeneration) and the highest survival rate of 83.33%. In conclusion, adding fermented herbal medicine at a dose of 150 mL/kg feed can increase the immunity of tilapia against S. agalactiae infection, as shown through the improvement of organ tissue structure and improved survival

Abstract

Penelitian ini bertujuan untuk mengetahui gejala klinis ikan komet yang terinfeksi bakteri Aeromonas hydrophila dan pascapengobatan dengan larutan propolis. Metode yang digunakan adalah metode eksperimen dengan menerapkan rancangan acak lengkap (RAL) dengan satu faktor lima taraf perlakuan dan tiga kali ulangan perlakuan. Perlakuan tersebut adalah adalah Kn (tidak terinfeksi A.hydrophila dan tidak diobati propolis), Kp (terinfeksi A. hydrophila  tetapi tidak diobati propolis), sedangkan ikan terinfeksi A.hydrophila diobati dengan propolis dosis P1 (700 ppm), P2 (800 ppm), dan P3 (900 ppm). Pengobatan dilakukan dengan cara penyuntikan propolis pada ikan yang terinfeksi A.hydrophila di bagian intramuscular sebanyak 0,1 ml. Bahan uji yang digunakan adalah ikan komet (Carassius auratus) ukuran 8-10 cm sebanyak 150 ekor. Hasil penelitian menunjukkan bahwa ikan yang terinfeksi bakteri A.hydrophila menunjukkan gejala klinis sirip ekor gripis, bercak merah, mata menonjol (exopthalmia), perut mengembung (dropsy), sisik tubuh lepas. Setelah 14 hari pascapengobatan dengan larutan propolis perlakuan terbaik adalah perlakuan P2 dosis (800 ppm) mengalami kondisi pemulihan mendekati gejala klinis kondisi ikan normal terlihat dari warna tubuh cerah, produksi lendir normal, sirip ekor utuh, tidak terdapat mata menonjol (exopthalmia), dan perut tidak mengembung (dropsy).

Abstract

This study aims to determine the immunity response of  Carassius auratus infected with Aeromonas hydrophila bacteria and post-treatment with propolis solution by measuring total leukocytes and phagocytosis index. The method used is experimental by applying a completely randomized design (CRD) with five treatments and three replications. The treatments were Kn (not infected with A. hydrophila and not treated with propolis), Kp (infected with A. hydrophila but not treated with propolis), while fish infected with A. hydrophila were treated with propolis doses P1 (700 ppm), P2 (800 ppm), and P3 (900 ppm). Treatment was done by injecting 0.1 ml of propolis into fish infected with A. hydrophila intramuscularly. The test material was 150 fish of 8-10 cm in size. The results showed that propolis significantly gave an immune response to the C. auratus infected with A. hydrophila (p<0.05). The 800 ppm propolis dose was the most effective, as indicated by a total leukocyte count of 3.70×104 cells/mm3, a phagocytosis index value of 28.33%, and a survival rate of 83.33%. This study highlights that propolis has potential as a natural immunostimulatory agent in enhancing non-specific defense mechanisms in C. auratus infected with pathogenic bacteria

Abstract

Fermented herbs made from turmeric, ginger, and kencur have the potential to be antimicrobials and immunostimulants that can increase appetite and reduce fish stress due to changes in water quality. The research was conducted from June 2024 to August 2024 at the Fish Disease and Parasite Laboratory, Faculty of Fisheries and Marine, Universitas Riau. This study aims to determine the best frequency of fermented herbs enriched on the health of striped catfish (Pangasianodon hypophthalmus) hematology after challenge with Aeromonas hydrophila. The research method used a completely randomized design (CRD) with one factor, five treatments, and three replicates. Treatments consisted of Kn (negative control, no challenge test), Kp (positive control and challenge test), P1 (herbs enriched every day and challenge test), P2 (giving every 3 days and test challenge), and P3 (giving every 5 days and a test challenge). Fish were kept for 46 days and challenged with A. hydrophila (10⁸ CFU/mL) on day 32. Clinical symptoms were observed for 14 days post-challenge. The best results were obtained in treatment P1 with the following hematological parameters: erythrocytes 2.37×10⁶ cells/mm³, hematocrit 36.00%, hemoglobin 9.60 g/dL, leukocytes 11.14×10⁴ cells/mm³, glucose 69.33 mg/dL, and normal leukocyte differentiation. The survival rate reached 100%. Feeding fermented herbs effectively improved the health and body endurance of striped catfish

Abstract

Motile Aeromonas septicemia (MAS) merupakan penyakit yang disebabkan oleh Aeromonas hydrophila dan sering menyerang ikan jambal siam (Pangasius hypophthalmus). Kayu manis merupakan tanaman yang memiliki senyawa aktif, seperti minyak atsiri yang mengandung cinnamaldehyde dan berperan sebagai antibakteri. Tujuan penelitian adalah untuk mengetahui efektifitas penambahan larutan kayu manis dalam mencegah infeksi A. hydrophila penyebab penyakit MAS pada ikan jambal siam. Metode yang digunakan adalah metode eksperimen dengan rancangan acak lengkap (RAL), lima perlakuan dan tiga ulangan. Perlakuan yang diterapkan adalah sebagai berikut: Kn (kontrol negatif), Kp (kontrol positif), dan (P1; P2; dan P3 penambahan larutan kulit kayu manis dengan dosis 15; 30; dan 45 mL kg-1 pakan). Ikan (10 ± 2 cm) dipelihara dalam akuarium berukuran 40 x 30 x 30 cm3 dengan kepadatan 10 ekor per akuarium. Pada hari ke-32 pemeliharaan, ikan diuji tantang dengan menginfeksikan A. hydrophila sebanyak 0,1 mL per ekor dengan kepadatan 108 CFU mL-1 yang dilakukan secara intramuskular. Ikan dipelihara selama 14 hari hingga hari ke-46. Parameter yang diamati adalah kadar kalsium, magnesium, dan fosfor dalam serum darah, aktivitas lisozim, tingkat kelulushidupan, dan relative percent survival 91,07 ±7,78%. Hasil penelitian menunjukkan bahwa penambahan larutan kayu manis yang terbaik adalah dosis 30 mL kg-1 pakan yang ditandai dengan angka kelulushidupan pascauji tantang (93,33%) dan profil biokimia darah (kadar kalsium 9,33 mg dL-1, magnesium 3,10 mg dL-1, dan fosfor 3,93 mg dL-1) serta aktivitas lisozim (285 unit mL-1) tertinggi. Hal ini menunjukkan bahwa penambahan larutan kulit kayu manis pada pakan efektif untuk mencegah penyakit MAS pada ikan jambal siam.Motile Aeromonas septicemia (MAS) is a disease caused by Aeromonas hydrophila and often attacks Siamese catfish (Pangasius hypophthalmus). Cinnamon is a plant that has active compounds, such as essential oil which contains cinnamaldehyde and acts as an antibacterial. The aim of this study was to determine the effectiveness of cinnamon solution addition in preventing A. hydrophila infection which causes MAS disease in Siamese catfish. The study used an experimental trial consisting of five treatments with three replicates arranged in a completely randomized design (CRD), five treatments and three replications. The treatments applied were as follows: Nc (negative control), Pc (positive control), and (P1; P2; and P3 cinnamon bark solution addition at doses of 15; 30; and 45 mL kg-1 feed). Fish (10 ± 2 cm) were reared in aquariums with a dimension of 40 x 30 x 30 cm3 at a density of 10 fish per aquarium. On the 32nd day of rearing, each fish were challenged with A. hydrophila through the injection of 0.1 mL solution containing A. hydrophila at a density of 108 CFU mL-1 through intramuscular route. The fish were reared for 14 days until the 46th day. The parameters observed were levels of calcium, magnesium and phosphorus in blood serum, lysozyme activity, survival rate, and relative percent survival 91.07 ± 7.78%. The results of the study showed that the best addition of cinnamon solution was a dose of 30 mL kg-1 feed, which was characterized by better post-challenge survival rate (93.33%) and blood biochemical profile (calcium level 9.33 mg dL-1, magnesium 3.10 mg dL-1, and phosphorus 3.93 mg dL-1) and the highest lysozyme activity (285 units mL-1). These findings showed that the addition of cinnamon bark solution to feed was effective in preventing MAS disease in Siamese catfish.

Abstract

Ikan jambal siam (Pangasianodon hypophthalmus) merupakan komoditas ikan air tawar yang permintaan pasarnya terus meningkat, sehingga pembudidaya menerapkan sistem budidaya intensif. Namun, jika tidak dikelola dengan baik dapat menurunkan kualitas air, menimbulkan stres, dan meningkatkan risiko penyakit, seperti penyakit Motile Aeromonas Septicemia (MAS). Salah satu alternatif pencegah penyakit adalah dengan memanfaatkan bahan alami seperti ekstrak biji pepaya yang mengandung antibakteri. Penelitian ini bertujuan untuk mengetahui dosis terbaik penambahan ekstrak biji pepaya pada pakan ikan jambal siam (P. hypophthalmus) yang diujitantang dengan Aeromonas hydrophila. Penelitian ini dilaksanakan pada Juli hingga Desember 2024 di Laboratorium Parasit dan Penyakit Ikan, Fakultas Perikanan dan Kelautan, Universitas Riau. Metode eksperimen dengan Rancangan Acak Lengkap (RAL) satu faktor dengan lima perlakuan dan tiga pengulangan. Perlakuan dalam penelitian ini adalah Kn: pakan tidak ditambahkan dengan ekstrak biji pepaya dan ikan tidak diujitantang dengan A. hydrophila, Kp: pakan tidak ditambahkan dengan ekstrak biji pepaya dan ikan diujitantang dengan A. hydrophila, P1: (1 g/kg), P2: (2 g/kg), P3: (3 g/kg) dan ikan diujitantang dengan A. hydrophila. Didapatkan hasil yaitu dosis terbaik penambahan ekstrak biji pepaya pada pakan adalah P3 (3 g/kg) dengan total eritrosit 2,27 x 10⁶ sel/mm³, nilai hematokrit 37,33%, kadar hemoglobin 11,80 g/dL, total leukosit 10,37 x 10⁴ sel/mm³, aktivitas fagositosis 26,00%, dan kelulushidupan 86,67%, dapat disimpulkan ada pengaruh penambahan ekstrak biji pepaya dengan dosis berbeda pada pakan terhadap gambaran darah ikan jambal siam sebelum dan setelah diujitantang dengan A. hydrophila dan dapat meningkatkan imunitas ikan jambal siam

Abstract

Asam Jungga (Citrus jambhiri L.) berpotensi sebagai bahan tambahan pakan alami karena mengandung alkaloid, saponin, steroid, triterpenoid, tanin, flavonoid, dan vitamin C. Tujuan dari penelitian ini adalah untuk memperoleh dosis terbaik larutan asam jungga yang ditambahkan ke dalam pakan guna meningkatkan sistem imun non-spesifik ikan jambal siam dengan melihat jumlah eritrosit total, kadar hematokrit, kadar hemoglobin, jumlah leukosit total, aktivitas fagositosis, dan sintasan. Penelitian ini dilaksanakan pada bulan Agustus hingga Desember 2024. Metode yang digunakan adalah metode eksperimental dengan rancangan acak lengkap (RAL) satu faktor dengan lima perlakuan, yaitu: Kn (kontrol, tanpa asam jungga dan tanpa uji tantang), Kp (tanpa asam jungga, diuji tantang dengan Aeromonas hydrophila), serta tiga perlakuan dengan penambahan larutan jeruk jungga dalam pakan masing-masing dengan dosis 6,0 mL/kg (P1), 6,5 mL/kg (P2), dan 7,0 mL/kg (P3), yang semuanya diuji tantang dengan A. hydrophila. Masa pemeliharaan berlangsung selama 46 hari dalam wadah bervolume 35 L. Pengambilan sampel darah dilakukan sebanyak tiga kali, dosis terbaik diperoleh pada perlakuan P3 (7 mL/kg) dengan jumlah eritrosit total 294,08 x10⁴ sel/mm³, kadar hematokrit 37,67%, kadar hemoglobin 9,87 g/dL, jumlah total leukosit 10,67 x10⁴ sel/mm³, aktivitas fagositik 32,67%, dan tingkat kelangsungan hidup 93,33%. Parameter kualitas air selama penelitian meliputi suhu berkisar antara 26–29°C, pH 6–7,3, dan DO (oksigen terlarut) 4,6–5,2 ppm. Hasil penelitian menunjukkan bahwa suplementasi dengan asam jungga secara signifikan meningkatkan parameter imun dan kelangsungan hidup pada ikan jambal siam
