Abstract

Bacterial phosphate solvent is bacteria that have role in enrichment of soil. Origin material and formation ofpeat soil have correlation with the maturity and the level of fertility of peat soil. The recent research aimed to find out genus and density of bacterial phosphate solvent at the level of maturity of different peat soil in protected forest areas of Ambawang Mountain in District of Teluk Pakedai, Kubu Raya Regency. The isolation of bacterial was conducted by using the media of pikovskaya and pour plate method, whereas bacterial density calculation was done by total plate count method. The research finding showed that there were five genera of bacterial phosphate solvent, namely; Acinetobacter, Chromobacterium, Flavobacterium, Micrococcus, and Stapylococcus. There was no different of the total density of bacterial colonies phosphatesolvent in fibric, hemic, and sapric.

Abstract

Canal A Kuala Dua is one of the water source used in daily activities by the society lives along that river banks. The occurence of markets and inhabitans activities such as bathing and washing can affect the water quality of Canal A Kuala Dua due to the disposals entering the water body. One of the bioindicator used to detect water pollution is coliform bacteria. The purpose of this research was to find out the dispersion pattern of coliform bacteria when ebb and flow occurs in Canal A Kuala Dua. The Research was done in November 2011 through March 2012. Sampling was done at 9 stations along Canal A Kuala Duan when the ebb and flow occurred. The density of coliform bacteria was calculated by MPN method and Escherichia coli qualitative test was done by IMVIC test. Statistics sofwares such as Microsoft Excel, SPSS 15 (t test), and Arcgis 10.1 were used in data anylisis. Based on its use, Canal A Kuala Dua is categorized into class C. Average values of coliform bacteria densities in Canal A Kuala Dua are 74,249 x 103 MPN/100 mL when the uptide occurs and 59,259 x 103 MPN/100 mL when the downtide occurs.

Abstract

Nitrifying bacteria has an important role in oxidizing ammoniac to be nitrit which is required by plants. The purpose of this study is to investigate character and density of nitrifying bacteria at maturity level of peat soil. The sampel was taken at wildlife sanctuary of Ambawang mountain Kubu Raya district. The proses of isolation was conducted by pour plate method that applying spesific media of nitrification and total plate account was used to calculate the density of bacteria. Observation and identification was implemented macroscopically, microscopically and biochemically. The finding indicated that 5 genus of nitrifying bacteria had different level of maturity in peat soil. Nitrosomonas, Nitrobacter and Nitrosococcus were some types ofbacteria that were pound in fibrik meanwhile in hemik, Nitrobacter and Nitrosocystis were identified. In addition, in saprik, Nitrobacter, Nitrospina and Nitrosococcus were discovered. The total density of nitrifying bacteria colonies in maturity level of peat soil in fibrik, hemik and saprik was 160 x 105 CFU/g, 107,5 x 105 CFU/g, and 19 x 105 CFU/g.

Abstract

Liquid wastes from palm oil containing lipid which, if not managed properly could potentially contaminate the environment. One of effective waste treatments that relatively cheap and environmentally friendly is by using bacteria as agents of biodegradation. The purpose of this study was to acquire lipid degrading bacteria and verify the isolate ability of lipid degrading bacteria which derived from sewage treatment plants PPKS PTPN XIII Ngabang, Landak District. The research was conducted from September 2011 to March 2012 at the laboratory of Microbiology and Chemistry laboratory of the Faculty of Mathematics and Natural Sciences, Tanjungpura University. The method used for isolation of bacteria was pour plate and streak plate. The results obtained 9 isolates of lipid degrading bacterium. LSWJ1 bacterium isolate were capable to degrade lipids better since they could grow in liquid media oily and had the ability to reduce lipid levels by up to 25% for 14 days. Identification showed that isolate belonged to the genus Bacillus.

Abstract

Blas caused by Pyricularia grisea Sacc. is one of disease on rice (Oryza sativa L.). The disease is controlled using bio-control agents which are antagonist to pathogenic fungi. The objective of this research was identify the fungi from rizosfer on rice and to the test their ability to suppress the growth of Pyricularia grisea Sacc. The research was carried out for July to September 2012 in the rice field in Kalimue Village, Keranji Paidang Village, Sengah Temila Sub-district, Landak Regency. Isolating Pyricularia grisea Sacc. was conducted based on direct plantation method. Dilution method and antagonism test using paired method are used to isolate rizosfer fungi. Antagonistic fungi like Aspergillus flavus, Aspergillus fumigatus, Aspergillus niger, Curvularia sp., and Trichoderma harzianum were successfully isolated. The result showed the growth of Pyricularia grisea Sacc. was inhibited. The highest suppression shown by Trichoderma harzianum isolation was 88.63%.

Abstract

Lalat rumah (Musca domestica) merupakan serangga yang memiliki bulu dan cairan lengket pada kakinya dan hidup di tempat yang kotor, sehingga dapat membawa berbagai macam mikroorganisme. Jamur adalah salah satu mikroorganisme yang dibawa oleh M. domestica. Tujuan dari penelitian ini adalah mendapatkan genus jamur dari seluruh tubuh lalat rumah (M. domestica). Penelitian ini dilakukan dari bulan April sampai Juni 2012. Tempat pengambilan sampel M. domestica yaitu warung makan, tempat pembuangan sampah sementara, peternakan ayam dan penjualan ikan. Isolasi dilakukan pada seluruh permukaan tubuh dan usus M. domestica dengan metode tanam langsung, pengenceran dan goresan. Hasil penelitian ini didapatkan 7 genus jamur dari tubuh dan usus M. domestica. Genus jamur tersebut adalah Acremonium, Aspergillus (2 spesies), Debaryomyces, Hanseniaspora, Fusarium, Penicillium, dan Geotrichum

Abstract

Selulosa merupakan salah satu bahan organik yang sukar untuk dirombak pada tanah gambut.Bakteri pendegradasi selulosa memiliki peranan dalam proses dekomposisi selulosa secara enzimatik dengan memutuskan ikatan rantai 1,4-?-glukosida. Penelitian ini bertujuan untuk mengetahui bakteri yang dapat mendegradasi selulosa dan mengetahui kepadatan bakteri pendegradasi selulosa pada tanah gambut. Sampel diambil di Desa Parit Banjar Kecamatan Mempawah Hilir Kabupaten Pontianak. Isolasi dilakukan dengan metode pengenceran cawan tuang dan cawan gores menggunakan media Carboxymethylcellulose (CMC) dan kepadatan bakteri dihitung dengan metode Total Plate Count. Pengamatan dan identifikasi dilakukan secara makroskopis, mikroskopis dan melalui rangkaian uji biokimia. Hasil isolasi diperoleh enam genera bakteri yaitu Acetobacter, Acinetobacter, Azotobacter, Acidomonas, Pseudomonas dan Cellvibrio. Kepadatan bakteri pendegradasi selulosa yang ada di lokasi penelitian berkisar antara 23,126x107 sampai 24,942x107 CFU/gr.

Abstract

Cellulose is the main carbohydrate synthesized by plants and it occupies almost 60% of the components of the structure of plants. Natural process of decomposition of cellulose requires the help of microorganisms that secrete the enzyme cellulase. Bagasse contains 32-34% cellulose. This study aims to determine the cellulose degrading bacteria of yellow bagasse and know the ability of the bacteria to degrade cellulose bagasse. The samples were taken from ice-cane seller in the District of South Pontianak. To isolate of cellulose degrading bacteria, the dilution method of Carboxymethylcellulose (CMC) was used and HC test was performed on solid CMC. Dry weight of test was performed using a fresh bagasse as a source of cellulose. The results showed that there are fifteen pure isolates of cellulose degrading bacteria consisting of eight genera, namely Psedomomas, Bacillus, Nocardia, Kurthia, Vibrio, Flavobacterium, Neissera, and Micrococcus. AT3BPS2 isolates (Nocardia) has the largest ratio of 2.67 HC. Isolates that have the lowest HC constellation is AT1BPS1 isolates (Pseudomonas) 1.06. Isolates that has the highest cellulose degradation ability is AT1BPS2 (Pseudomonas) at 19.28% and isolates with the lowest degradation ability is AT3BPS2 (Nocardia) at 7.21%.

Abstract

Jamur makroskopis memiliki peranan yang penting dalam kehidupan manusia. Penelitian inventarisasi jamur makroskopis ini bertujuan untuk mengetahui jenis-jenis jamur makroskopis dan pemanfaatannya oleh masyarakat Desa Teluk Bakung, Kecamatan Sungai Ambawang, Kabupaten Kubu Raya. Penelitian ini dilaksanakan selama 4 (empat) bulan dari bulan Agustus 2011 sampai November 2011 dengan menggunakan metode jelajah (Cruise Method) (Rugayah et al., 2004). Hasil penelitian diperoleh 20 spesies jamur makroskopis dari kelas Basidiomycetes yang terdiri dari 4 ordo, 9 famili dan 15 genera. Jamur dari ordo Aphylloporales paling banyak ditemukan di hutan rawa gambut Desa Teluk Bakung Kecamatan Sungai Ambawang Kabupaten Kubu Raya. Dari 20 spesies yang ditemukan, 10 spesies jamur dimanfaatkan sebagai makanan, 3 spesies sebagai obat-obatan, 3 spesies bersifat racun dan 6 spesies tidak diketahui pemanfaatannya

Abstract

Fosfat di dalam tanah merupakan unsur hara yang berperan penting bagi proses pertumbuhan tanaman. Ketersediaan unsur fosfat dibantu oleh bakteri pelarut fosfat yang banyak dijumpai di daerah rizosfer. Tujuan penelitian ini untuk mengetahui kepadatan total dan karakteristik bakteri pelarut fosfat yang di isolasi dari rizosfer tanaman pisang nipah (Musa paradisiaca var. nipah) pada jenis tanah aluvial, gambut dan podsolik merah kuning (PMK) di Kecamatan Singkawang Tengah. Isolasi bakteri dilakukan dengan menggunakan media Pikovskaya dan metode agar tuang (pour plate method), sedangkan penghitungan kepadatan bakteri dengan metode cawan hitung (total plate count). Hasil penelitian ini menunjukkan bahwa diperoleh 4 genus bakteri pelarut fosfat pada tanah aluvial yaitu Acetobacter, Bacillus, Flavobacterium dan Micrococcus. Tanah gambut ditemukan 5 genus yaitu Azotobacter, Bacillus, Micrococcus, Pseudomonas, Staphylococcus. Tanah PMK ditemukan 6 genus yaitu Acetobacter, Azotobacter, Bacillus, Escherichia, Flavobacterium, Paracoccus. Kepadatan total koloni bakteri pelarut fosfat pada tanah aluvial, gambut dan PMK berturut-turut adalah 7,9x108 CFU/gr, 7,3x108, 7,1 x108 CFU/gr.
