Abstract

ABSTRAK. Penelitian ini bertujuan untuk isolasi, identifikasi dan uji sensitivitas antibiotik eritromisin dan penisilin G terhadap bakteri Streptococcus sp. penyebab endometritis subklinis pada sapi aceh. Penelitian ini dilakukan dengan cara swab dibagian vagina sapi  sebagai sampel, dan dimasukkan ke dalam tabung reaksi berisi Nutrien Broth (NB), kemudian diisolasi menggunakan media Manitol Salt Agar (MSA) dan diidentifikasi koloni bakteri menggunakan pewarnaan Gram, serta dilakukan uji katalase. Hasil isolasi bakteri tersebut diuji sensitivitas antibiotik dengan cara menempelkan disk antibiotik eritromisin dan disk antibiotik penisilin G pada permukaan media Mueller Hinton Agar (MHA) dan setelah inkubasi pada suhu 37 ­­­0C selama 24 jam diamati zona bening yang terbentuk disekitar disk antibiotik tersebut. Hasil penelitian menunjukkan adanya pertumbuhan bakteri pada media NB dan terdapat koloni terpisah pada media MSA yang menunjukkan Gram positif berbentuk kokus dan tidak menimbulkan gas pada uji katalase sebagai reaksi spesifik dari bakteri Streptococcus sp. Hasil isolasi bakteri dari vagina sapi aceh tersebut, secara uji sensitivitas menunjukkan adanya diameter zona bening seluas 23,86 mm pada antibiotik eritromisin dan 8,86 mm pada antibiotik penisilin G. Hasil penelitian dapat disimpulkan bahwa pada vagina sapi aceh ditemukan adanya bakteri Streptococcus sp. yang berpotensi menyebabkan endometritis subklinis dan sensitif terhadap antibiotik eritromisin serta resisten terhadap penisilin G.Kata kunci: antibiotik, endometritis subklinis, sapi aceh, Streptococcus sp.Isolation, Identification and Sensitivity Test to Antibiotics of Bacteria Streptococcus sp. Causes of Subclinical Endometritis in Aceh CattleABSTRACT. This study aims to isolate, identify, and test the sensitivity of erythromycin and penicillin G antibiotics against Streptococcus sp. bacteria causing subclinical endometritis in Aceh cattle. The study was conducted by swabbing the vaginal area of the cattle to collect samples, which were then placed in test tubes containing Nutrient Broth (NB). The bacteria were isolated using Manitol Salt Agar (MSA) media and identified by Gram staining, along with a catalase test. The isolated bacteria were tested for antibiotic sensitivity by applying erythromycin and penicillin G antibiotic disks onto the surface of Mueller Hinton Agar (MHA) media. After incubation at 37°C for 24 hours, the clear zones formed around the antibiotic disks were observed. The results showed bacterial growth on NB media and separate colonies on MSA media, which were Gram-positive cocci bacteria and did not produce gas in the catalase test, indicating a specific reaction of Streptococcus sp. The sensitivity test results showed a clear zone diameter of 23.86 mm for erythromycin and 8.86 mm for penicillin G. This study concluded that Streptococcus sp. bacteria found in the vagina of Aceh cattle have the potential to cause subclinical endometritis and are sensitive to erythromycin but resistant to penicillin G.

Abstract

Essential oils have gained interest in wound management, with prior studies exploring combinations of hemp seed oil (Cannabis sativa) and other oils. However, single-oil strategies may offer simpler formulation, reducing the risk of interactions while preserving therapeutic benefits. The aim of this study was to explore the effect of hemp seed oil on accelerating wound healing, focusing on wound size reduction, epithelialization, granulation tissue formation, and vascularization in murine models. An in vivo with a post-test-only control group was conducted using 36 male Mus musculus mice (3−4 months, 150–250 grams) which were divided into three groups: negative control (NC), positive control (PC, treated with chloramphenicol ointment twice daily), and treatment group (TG, treated with hemp seed oil 400,000 mg/mL twice daily). Mice were euthanized on day 3, 7, 14, and 21 for wound healing assessment, including macroscopic evaluation (visual observation, wound size, and wound healing rate) and microscopic evaluation (epithelialization, granulation tissue formation, and vascularization). The present study found that the TG group demonstrated smaller wound sizes on day 14 (p<0.001) and day 21 (p<0.001). This group also enhanced wound healing rates observed on day 14 (p<0.001) and day 21 (p=0.001) compared to PC and NC groups. Epithelialization was significantly higher in the TG group compared to PC and NC groups on day 14 (p=0.007), while granulation tissue formation showed significant improvement on day 3 (p=0.045), day 14 (p=0.028), and day 21 (p=0.003). Additionally, TG group showed significantly greater new blood vessel formation on day 21 (p=0.001) compared to the PC and NC groups. In conclusion, hemp seed oil demonstrated significant potential in accelerating wound healing processes suggesting a superior effect compared to chloramphenicol ointment. Therefore, hemp seed oil may serve as a promising natural and cost-effective adjunct for wound management.

Abstract

ABSTRAK. Penelitian ini merupakan penelitian observasional yang dengan rancangan acak lengkap (RAL) dengan 3 kelompok perlakuan jenis semen beku sapi aceh (S1), semen beku sapi bali (S2) dan semen beku sapi simmental (S3). Masing-masing kelompok diulang sebanyak 10 kali. Data kualitas spermatozoa dan angka kebuntingan yang diperoleh dianalisis dengan analisis of variance (ANOVA) dan dilanjutkan dengan uji Duncan dan hubungan kualitas spermatozoa semen beku dengan tingkat kebuntingan diuji dengan regresi berganda. Persentase motilitas, spermatozoas hidup dan TAU sapi aceh dan sapi bali tidak memperlihatkan perbedaan yang nyata (P0,05), namun keduanya berbeda secara nyata (P0,05) dengan sapi simmental. Hasil uji regresi menunjukan ada hubungan yang nyata (P0,05) antara kualitas spermatozoa (motilitas, spermatozoa hidup dan TAU) semen beku dengan tingkat kebuntingan, dengan persamaan regresi adalah Y= - 2,586 + 0,017 X1 - 0,001 X2 + 0,043 X3, nilai koefisien korelasi r = 0,695 dan nilai koefisien determinasi (r2) sebesar 0,483. Keutuhan TAU memiliki hubungan yang lebih kuat (r = 0,695) dibanding dengan motilitas spermatozoa (r = 0,505) dan spermatozoa hidup (r = 0,195) terhadap angka kebuntingan. Disimpulkan kualitas semen beku sapi unggul berpengaruh terhadap tingkat kebuntingan setelah inseminasi pada induk aseptor sapi aceh betina.(Comparison of frozen semen quality of aceh cattle, bali cattle, brahman cattle and simmental cattle and the relationship with the success level of artificial insemination in female aceh cattle)ABSTRACT. This study is an observational study with a completely randomized design (CRD) with 3 groups of frozen semen aceh cattle (S1), bali cattle frozen semen (S2) and simmental cattle frozen semen (S3). Each treatments groups was repeated 10 times. Spermatozoa quality data (motility, live spermatozoa, and intact acrosome) of frozen semen and pregnancy rates obtained were analyzed by analysis of variance (ANOVA) and continued with Duncant test. Percentage motility, live sperm and intact acrosome of aceh cattle, bali cattle and simmental cattle showed no significant differences (P 0.05), but they differ significantly (P 0.05) compared with brahman cattle. Results of regression analysis showed no significant relationship (P 0.05) between spermatozoa quality (motility, live sperm and intact acrosome) with a pregnancy rate of frozen semen, the regression equation is Y = - 2,586 + 0,017 X1 - 0,001 X2 + 0.043 X3, Correlation coefficient value r = 0,695 and coefficient of determination (r2) equal to 0,483. intact acrosome of spermatozoa have a stronger relationship (r = 0.695) compared with sperm motility (r = 0.505) and live spermatozoa (r = 0.195) on the pregnancy rate. It was concluded that the quality of frozen semen had an effect on the pregnancy rate after insemination on female aceh cow.

Abstract

ABSTRAK. Penelitian ini bertujuan untuk menentukan karakteristik dan komposisi plasma semen sapi aceh. Sampel semen dikoleksi dari 2 ekor sapi aceh jantan sehat berumur 3,0-3,5 tahun menggunakan vagina buatan. Koleksi semen dilakukan satu kali dalam seminggu selama enam minggu. Evaluasi karakteristik semen meliputi volume, warna, konsistensi, pH, konsentrasi spermatozoa, motilitas, viabilitas, abnormalitas spermatozoa dan komposisi kimia plasma semen. Data kualitas dan komposisi kimia plasma semen yang diperoleh dianalisa secara diskriptif. Hasil penelitian menunjukkan semen segar sapi aceh memiliki volume 3,82 0,47 ml, warna krem keputihan, konsistensi kental, pH 6,84 0,17, konsentrasi 1194 4,80 106sp/ml, persentase motilitas 77,28 3,17 % dan abnormalitas 5,98 1,77%. Plasma seminalis semen segar sapi aceh mengandung protein total sebanyak 1,51 0,21 g/100ml, Kalium 75,80 22,63 mg/100 ml dan Magnesium 32,30 17,64 mg/100 ml lebih tinggi, tetapi Natrium 170,00 81,60 mg/100 ml; kalsium 40,00 12,76 mg/100 ml, Fosfor 16,70 6,17 mg/100 ml, dan Mangan 7,08 2,48 mg/100 ml lebih rendah dibandingkan dengan sapi potong. Secara umum kuantitas dan komposisi plasma seminalis semen sapi aceh yang dihasilkan cukup baik, dan memenuhi syarat untuk diproses sebagai semen beku.(The characteristics of semen and chemical composition of the seminal plasma of aceh cattle maintaned in BIBD Saree Aceh Besar)ABSTRACT. This study aim were to determine characteristics and composition of semen plasma Aceh cattle. Semen samples were collected from two healthy Aceh bulls that aged from 3.0 to 3.5 year. Samples were yielded by using an artificial vagina. Samples collecting were conducted once a week for six consecutive weeks. The evaluation of semen characteristics includes volume, color, consistency, pH, sperm concentration, motility, viability, abnormal spermatozoa and seminal plasma chemical composition. Data of semen quality and chemical composition of seminal plasma were analyzed descriptively. The evaluation showed that Aceh Cattle semen had 3.82 0.47 ml volume, beige whitish color, thick consistency, 6.84 0.17 pH, 1194 4.80106 sp/ml sperm concentration, 77.28 3.17% motility and 5.98 1.77% abnormal spermatozoa. Plasma seminalis fresh semen of Aceh Cattle contained 1,51 0,21 g/100ml total protein; 75,80 22,63 mg/100ml potassium and 32,30 17,64 mg/100ml magnesium higher than the heifer, but 170,00 81,60 mg/100ml sodium; 40,00 12,76 mg/100ml calcium, 16,70 6,17 mg/100ml fosfor, and 7,08 2,48 mg/100ml mangan lower. It was concluded that quality and composition of seminal plasma Aceh cattle semen could be made as frozen semen.

Abstract

This study aims to determine the effect of administering Chromolaena odorata L. leaf extract gel on increasing angiogenesis in the healing of open incisional wounds in rats (Rattus norvegicus). In this study, 27 healthy male rats, aged 3-4 months with a body weight of 150-200 g were used. The rats were anesthetized and incised in the back area with a size of 2x2 cm (WxL) and a depth reaching subcutaneous. The rats were divided into three groups (n= 9), the group only given gel (P0), the group of rats that were given 10% Chromolaena odorata leaf extract gel (P1), and the group of rats that were given 20% Chromolaena odorata leaf extract gel (P2). Collection of wound skin tissue samples for histopathological preparations was carried out on days 3, 7, and 14 after treatment, 3 individuals per group for each collection. The results showed that application of Chromolaena odorata leaf extract gel on P1 and P2 could increase angiogenesis significantly (P0.05) compared to P0 on days 3 and 7, but was not significantly different (P0.05) on day 14. The average angiogenesis in P1 and P2 on day 3 was similar, while on day 7 the average angiogenesis on P1 was significantly different compared to P2. It was concluded that administration of Chromolaena odorata leaf extract gel could increase angiogenesis in rats during phase II of the open incision wound healing process.

Abstract

This study aimed to determine the concentrations of Follicle Stimulating Hormone (FSH) and Luteinizing Hormone (LH) in New Zealand White (NZW) rabbits after superovulation induction using FSH. Six female NZW rabbits and one male NZW rabbit were used in this study. Criteria for female NZW rabbits used include having previously given birth, not being pregnant, and having a body weight of 2-3 kg. The rabbits were divided into two treatment groups; the first group was injected with physiological NaCl (NZW1, n= 3), while the second group was injected with FSH (NZW2, n= 3). Injections of physiological NaCl or FSH were administered five times at 12-hour intervals and first injection was given at 20:00 p.m. In NZW1, each injection was given physiological NaCl with volumes of 0.2:0.2+0.2;0.4+0.4 mL, respectively whereas in NZW2, each injection was given FSH with doses of 4:4+4:8+8 mg, respectively. Twelve hours after the last FSH injection, NZW2 rabbits were injected with 100 IU hCG and mated with male rabbit, while NZW1 rabbits were mated without hCG administration. On days 1, 3, and 5 after mating, blood samples were collected for FSH and LH hormone examination using the Enzyme-Linked Immunosorbent Assay (ELISA) method. The FSH concentrations on days 1, 3, and 5 between NZW1 vs NZW2 were 34.781.46 vs 39.753.18; 41.434.90 vs 41.425.14; and 44.471.51 vs 30.130.19 mIU/mL, respectively (P0.05). The LH concentrations on days 1, 3, and 5 between NZW1 vs NZW2 were 33.784.01 vs 40.1812.36; 29.342.24 vs 31.3110.13; and 42.0911.38 vs 30.578.87 mIU/mL, respectively (P0.05). It is concluded that FSH administration does not affect the concentrations of FSH and LH in NZW rabbits. The FSH concentration five days after mating tends to fluctuate, while the LH concentration tends to decrease as the days increased.

Abstract

The objective of this study was to determine the effect of ethanol extract of neem (Azadirachta indica) leaves on the number of spermatids and androgen receptor expression in the testicular tissue of New Zealand White (NZW) rabbits. Twelve adults male NZW rabbits with body weight (BW) 2-2.5 kg were divided into three treatment groups (n = 4), namely P1 (control group) given 0 mg/kg BW, P2 and P3 were given 200 mg/kg BW and 300 mg/kg BW of neem leaves ethanol extract respectively. The extract was administered orally using a gastric tube for 52 days. At the end of the treatment, the rabbits were terminated and the testes were collected to be processed into histological preparations and stained with Periodic Acid Schiff (PAS) staining to detect the number of spermatid and Immunohistochemical (IHC) staining to detect androgen receptor expression. The results showed that neem leaves extract administered at P3 rabbits significantly decreased the number of spermatids and androgen receptor expression in myoid peritubular cells and connective tissue P0.05), but not significantly different in the Leydig cells and blood vessels (P0.05) of testicular tissue. The conclusion of this study is that the ethanol extract of neem leaves at a dose of 300 mg/kg BW can be used as a contraceptive candidate compared to a dose of 200 mg/kg BW.

Abstract

The purpose of this study was to analyze the potential of tomato extract (Lycopersicum esculentum L.) in increasing the Sertoli and spermatogenic cells count of the contralateral testis in white rats (Rattus novergicus) with unilateral torsion. A total of 24 male Wistar white rats, aged 3-4 months old with a weight of 180-200 g were used in this study. The rat samples were randomly divided into 4 treatment groups: Group 1 as a negative control (NC) which consisted of rats without testicular torsion and given tomato extract; Group 2 as a positive control (PC) which consisted of rats with unilateral testicular torsion 720 for 6 hours without given tomato extract; Group 3 as treatment 1 (T1) consisted of rats that were subjected to unilateral testicular torsion 720o for 6 hours and were given tomato extract at a dose of 100 mg/kg BW/day; and Group 4 as treatment 2 (T2) consisted of rats that were subjected to unilateral testicular torsion 720 for 6 hours and were given tomato extract at a dose of 200 mg/kg BW/day. Each group consisted of six rats. Tomato extract was administered orally for 30 days. The observations of the number of Sertoli cells and spermatogenic cells (spermatogonia cells, spermatocytes, and spermatids) were carried out histologically with Hematoxylin-Eosin (HE) staining and the slides were examined with a light microscope. The data obtained was analyzed by ANOVA and continued with Duncan's test. The results showed that the administration of tomato extract significantly increased (P0.05) the number of Sertoli cells and spermatogenic cells of the contralateral testicular seminiferous tubule. Giving tomato extract 100 mg/kg BW/day resulted in an increase in the number of Sertoli cells and better spermatogenic cells than tomato extract 200 mg/kg BW/day. In conclusion the administration of tomato extract after detorsion can increase the number of Sertoli cells and spermatogenic cells of the contralateral testicular seminiferous tubules in white rats subjected to unilateral torsion of 720 for 6 hours

Abstract

The aim of this study was to further understand the histomorphometry of seminiferous tubules from local goat testicles that were fixed with different fixatives. Nine testicles were randomly chosen, each fixed with 10% formalin, 10% neutral buffered formalin (NBF), and 4% paraformaldehyde (PFA) solutions. The sample was then proceeded to histology preparation and stained with haematoxylin eosin (HE). Observation of the tissue was carried out using microscope and the data obtained was analyzed statistically. The results showed that there was a significant difference (P0.05) on each fixative in tubular diameter and membrane, and very significant difference (P0.01) on germinal cell and staining absorbability. Duncan test result on tubular diameter showed that the use of PFA fixative was significantly different (P0.05) compared to formalin fixative while the use of NBF fixative had no significant difference (P0.05) compared to PFA fixative. In addition, the measurement of interstitial membrane of seminiferous tubules showed no significant difference (P0.05) among three fixatives. To conclude, formalin, NBF, and PFA fixatives affect seminiferous tubules, basal membrane structure, and germinal cell appearance inside tubular lumen. Type of tissues and fixative must be considered in selecting the suitable fixative.

Abstract

This study aimed to evaluate the genetic potential of local meat chickens (LMC) to get the candidate of parent stock through the development of the selection method applied, as well as economical and practical approach and adjusment of nutritional requirement, especially protein and energy balance. Research was divided into 2 groups, where the female and male parent was reared for 2 months and the offspring (F1) was reared for 4 months and selected. The parent was mated by the method of artificial insemination (AI). Method of selection was used on the orientation of the meat chicken production parameters with a positive correlation with the gene expression Diet treatments were formulated by optimal nutrition standards in order to drive maximum primary production of local meat chicken, contained 17, 19, and 21% of protein. The result showed that feeding diet with 19% protein can stimulate optimum growth and food conversion of ALPU male, whereas female growth LMC in line with the increasing availability of protein in the diet, wherein feeding 21% of protein result in optimal growth until age 4 months.
