Abstract

Abstract. Harahap F, Siregar ARS, Idramsa, Suriani C, Edi S, Panggabean NH, Daulae AH, Pohan SD, Tanjung AA, Pertiwi SI, Kairani A. 2026. In vitro plant regeneration of Cattleya sp. from Protocorm-Like Bodies (PLBs) using coconut water and activated charcoal. Asian J Agric 10 (1): g100130. https://doi.org/10.13057/asianjagric/g100130. Cattleya sp. is an ornamental orchid whose seeds lack of endosperm, making natural germination and propagation difficult. In vitro culture using Protocorm-Like Bodies (PLBs) offers an alternative for mass propagation. This study aimed to evaluate the effects of coconut water and activated charcoal on the in vitro regeneration of Cattleya sp. PLBs. A completely randomized design was employed with varying concentrations of coconut water (0, 5, and 10%) and activated charcoal (0, 0.1, 0.2, and 0.3 g/L). Morphological parameters observed included time to shoot emergence, number of leaves, shoots, and roots, as well as shoot color. Data were analyzed using two-way ANOVA. The results showed that coconut water significantly accelerated shoot emergence with 10% of coconut water producing the fastest response (12 days after treatment). Activated charcoal enhanced shoot and leaf formation with 0.3 g/L, producing the highest number of shoot and leaf. Root formation was optimal at lower charcoal concentrations. Overall, the combination of coconut water and activated charcoal significantly influenced PLBS growth and regeneration. These findings indicate that appropriate combinations of organic additives can optimize in vitro regeneration of Cattleya sp. and support efficient orchid propagation.

Abstract

This study aims to determine the feasibility of a Project-Based Learning (PjBL)-based Plant Tissue Culture textbook in improving the science process skills of biology students at Universitas Negeri Medan, based on evaluations from material experts and media experts. It also aims to identify the responses of lecturers and students toward the developed textbook and to assess its effectiveness in enhancing students’ science process skills in plant tissue culture material at Universitas Negeri Medan. The study was conducted from August to November 2024 in the Department of Biology, Universitas Negeri Medan. The research employed the 4D development model, which includes the stages of define, design, develop, and disseminate. To measure the effectiveness of the developed textbook on students’ science process skills, the N-Gain test was used. The results showed that the feasibility of the PjBL-based plant tissue culture textbook in improving biology students’ science process skills, based on the assessment of material experts, obtained an average score of 76.66% (feasible), while the media experts gave an average score of 94.04% (highly feasible). The lecturers’ response reached an average percentage of 95% (highly feasible). The students’ responses were also very positive: the individual trial obtained 94.16%, the small group trial obtained 91.75%, and the large group trial obtained 90.92%, all categorized as highly feasible. Based on the N-Gain test results, the developed PjBL-based plant tissue culture textbook was found to be quite effective in improving the science process skills of biology students at Universitas Negeri Medan.

Abstract

This study aims to determine the effect of Liquid Organic Fertilizer (POC) from durian peel (Durio zibethinus Murr.) on the growth of mustard greens (Brassica juncea L.). This study was conducted at the Greenhouse, Medan State University using a non-factorial Completely Randomized Design (CRD) consisting of four treatments with six replications, resulting in 24 experimental units. The parameters observed included plant height, number of leaves, root length, fresh weight, and dry weight of the plant. Based on the results of the study, it was found that the levels of nutrients contained in the liquid organic fertilizer from durian peel waste were 0.93% C-Organic, 0.36% Total Nitrogen, 0.21% P2O5, 0.05% K2O, and 0.02% Total Ca. The application of liquid organic fertilizer from durian peel waste had a significant effect (p <0.05) on all growth and production parameters of mustard greens (Brassica juncea L.). Based on the results of the Duncan's Multiple Range Test (DMRT), the most optimal treatment for mustard greens growth was a concentration of 300 ml of Organic Fertilizer (POC) and 1,000 ml of water. This indicates that the nutrient absorption effectiveness of durian peel POC is sufficient to support maximum vegetative plant growth.

Abstract

This development research was carried out based on the observations of lecturers and students Department of Biology at Universitas Negeri Medan who stated that the tissue culture course was still quite difficult to learn, and the level of scientific literacy in Indonesia according to PISA was still relatively low. So, the purpose of this development research in general is to improve student scientific literacy, one of which is by developing tissue culture teaching materials. The research method for developing tissue culture teaching materials refers to the Dick & Carey model. However, this research has only reached the stage of designing and conducting formative evaluation. Analysis of the data used in this development research using quantitative descriptive data analysis. The result showed that the development of teaching material of in vitro plant breeding based on scientific literacy with a feasibility level based on material expert validation obtained a percentage of 91% in the very worthy category, learning design expert validation obtained a percentage of 95% in the very worthy category, layout design expert validation obtained a percentage of 99,2% in the very worthy category, response from lecturer of tissue culture course obtained a percentage of 95% in the very worthy category, and student responses obtained a percentage of 87% in the worthy category.

Abstract

In the modern society, the internet breaks through the restrictions on time and space and becomes a ubiquitous learning tool. Designing teaching activity for digital learning and flexibly applying technology tools are the key issues for current information technology integrated education. Interactive learning modules could be used in the classroom environment for effective learning. This study explored the effect of instructional modules for providing supplementary instruction in biology concepts. Interactive digital learning module was developed in order to assist students in biology learning. Interactive activities were considered in this study, include teacher – student interaction, student - student interaction, student - content interaction, and student - technology interaction. 180 students of Grade 11 in 6 classes at SMAN 2 Balige were selected as the research subjects. Three groups of students participated in biology learning course, (2 classes) using different modules were interactive digital module, digital module and text module. Goal of this research was to determine the impact of interactive digital learning module on students’ learning activity and autonomy. Quantitative results obtained by using questionnaire to understand the student’s learning activity and autonomy. Results showed student’s activities and autonomy who learned with interactive digital learning modules significantly different with students who learned using text learning modules. The use of interactive features increased the activity and autonomy of learners and created the student-centered learning environment. Students under interactive multimedia instruction performed better than text module class. These findings suggested that learning activity and autonomy of students in biology could be enhanced by multimedia instruction.
