Abstract

Isolation of bacterial cellulose from fruits in traditional market of Makassar have been done. This objective of this research was to determine optimum temperature of isolate bacteria that produce cellulose from Dragon fruit (Hylocereus polyrhizus) using dilution agar method. Isolation of bacteria produced cellulose using Hestrin-Schramm agar (HSA) obtain 5 (five) isolate NB02, NB03, NS01 and NS02. The result of screening activity showed that isolate NB04 has a potential as bacteria produce cellulose. The temperature optimize of NB04 has been done at 25oC and 40oC. Based on research results showed that optimum temperature for NB04 was at 25oC and has highest cellulose content at 1,29 g.

Abstract

Infectious diseases are one of the public health problems caused by the transmission and multiplication of microorganisms. The research on the antibacterial activity of painted nettle leaves (Coleus scutellarioides L. Benth) extracts was carriedout to examine the activity and chemical components that provide inhibition against gastrointestinal infection bacteria. The extraction of painted nettles leaves performed maceration methd using a solvent of 96%. The extraction results were examined to determine the antibacterial activity at 1% concentration of the test bacteria: Escherichia coli, Salmonella thypi, and Shigella dysenteriae. The results showed that 96% ethanol extract of painted nettle leaves inhibited Escherichia coli, Salmonella thypi, and Shigella dysenteriae. The research conducted using agar diffusion indicated that painted nettleleaf extract showed activity against bacteria and had highly strong potential with diameters of 6-20 mm. The results of Thin Layer Chromatography (TLC) Bioautography test of ethanol extract of painted nettle leaves using chloroform:methanol eluent (3:1) showed inhibition of Escherichia coli, Shigella dysenteriae, and Salmonella thypi at Rf values of 0.90, 0.80, 0.56. Abstract

Kopasanda  (Chromolaena odorata L.) leaves are one of the plants (herbs) containing various types of secondary metabolites, including saponins, tannins, alkaloids, and flavonoids, which function as antibacterial agents used as wound medicine for the community. This study aims to determine the antibacterial activity of the ethanol extract of Kopasanda (Chromolaena odorate L.) leaves using the TLC-bioautography and diffusion methods. The results of the antibacterial activity screening test obtained the active extract at a concentration of 0.1% against Escherichia coli and Staphylococcus aureus. The results of the antibacterial activity test using the TLC-Bioautography method with the eluent N-Hexane:Ethyl Acetate (4:1) showed results from Staphylococcus aureus bacteria with Rf values = 0.90, 0.78, 0.67, 0.52, 0.38, 0.27, 0.14, 0.05, and Escherichia coli bacteria with Rf values = 0.90, 0.52, 0.38, 0.27, 0.14, 0.05. Based on the results of testing the antibacterial activity with the agar diffusion method, the largest diameter of the inhibition zone at a concentration of 10% was obtained for Escherichia coli bacteria with an inhibition zone diameter of 13.59 mm, and Staphylococcus aureus with an inhibition zone diameter of 15.52 mm.

Abstract

Endophytic fungi that live on plant tissue has the potential to produce secondary metabolites, same as its host. This study aims to isolation endophyte fungi and antibacterial activity of isolate endophyte fungi of Jatropha multifida L. stem to Escheriacia coli and Staphylococcus aureus bacteria. The preliminary research was performed by isolation endophyte fungi of Jatropha multifida L. stem use PDAC medium and the result of isolation endophyte fungi Jatropha multifida L. stem obtained 6 isolates. Isolates endophyte fungi done examination morphology by microskopic method obtained that islolates have different carasteristic. Based on examination of antibacterial activity by agar diffusion method showed that isolates endophyte fungi that potential as antibacterial is isolate IFJT1 code where isolate IFJT1 code active to Escheriacia coli bacteria obtained inhibitory zone diameter of 17,7 mm and to Staphylococcus aureus bacteria.obtained inhibitory zone diameter of 16,7 mm

Abstract

White weed (Ageratum conyzoides L.) is known to have antibacterial elements containing chemical compound such as saponin and flavonoids. This study aimed to examine the antibiotic activity of endophytic fungi isolates of white weed leaves by the TLC-Bioautography method. The results of endophytic fungi isolation of white weed leaves obtained 10 isolates consisting of IFDP 1, IFDP 2, IFDP 3, IFDP 4, IFDP 5, IFDP 6, IFDP 7, IFDP 8, IFDP 9, and IFDP 10. The results of the macroscopic examination of the ten isolates of endophytic fungi found different characteristics. The results of the screening using 9 samples obtained isolates that found activity showing high inhibitory power, such as IFDP 1, IFDP 2, and IFDP 4 isolates. Isolates of IFDP 1, IFDP 2 and IFDP 4 were fermented on MYB medium for 14 days. Then filtering and evaporation were carried out to produce an extract. Isolate fermentate extracts were identified using Thin Layer Chromatography with chloroform eluent: methanol (4:2). The antibiotic activity test was done using TLC-Bioautography method, and obtained an Rf value in isolate 1, at Rf 0.74 on Vibrio cholerae. Isolate 2 resulted rf value of 0.67 on Salmonella thypi, Vibrio cholerae, Eschericia coli, disentriae, Staphylococcus epidermis. Isolate 4 generated an Rf value of 0.74 on Pseudomonas aeruginosa and Vibrio cholerae. While the group of active chemical components contained in the endophytic fungi isolates of white weed leaves was lavonoids by using AlCl3 reagents, AICI3 reagent alkaloids, and Sulfuric acid for saponin identification

Abstract

Averrhoa bilimbi L. is one of the traditional medicines that people empirically use as medicine to treat various diseases such as cough, diabetes, rheumatism, mumps, canker sores, toothache, bleeding gums, acne, diarrhea to high blood pressure. The research aimed to determine the antibacterial activity of Averrhoa bilimbi L. against gram-positive bacteria. This research applied a literature study method in which the references were obtained from 3 data bases, namely google scholar, pubmed and mdpi, then selected journals that metthe criteria in the literature study.  From the journal exploration, 7 journals were obtained. Then the antibacterial activity of Averrhoa bilimbi L. against gram-positive bacteria was observed. The results confirmed that the fruit and leaves of Averrhoa bilimbi L. had an antibacterial activity against Staphylococcus aureus, Streptococcus mutans, Staphylococcus epidermidis, and Propionibacterium acnes due to the presence of secondary metabolites, namely saponins, tannins, alkaloids and flavonoid.

Abstract

Pantai minanga merupakan slah satu objek wisata yang berada di provinsi Gorontalo kabupaten Gorontalo utara kecamatan atinggola kelurahan kotajin utara. Latar belakang dari perancangan Kawasan ekowisata pantai minanga yakni merujuk pada Kawasan yang strategis namun tidak terkelola dengan baik Adapun salah satu alasan di desain adalah untuk melestarikan Kembali hutan mangrup yang terdapat pada area pantai minanga dengan pendekatan arsitektur eko wisata. Persoalan desainnya adalah Bagaimana merancang kawasan wisata pantai minanga dengan menghidupkan kembali keberadaan konservasi mangrove di pantai minanga.tujuan dari perancangan ini adalah Untuk merancang kawasan wisata pantai minanga dengan menghidupkan kembali keberadaan konservasi mangrove di pantai minanga dan menggunakan pendekata desain arsitektur eko-wisata.  Metode yang digunakan dengan melakukan obserpasi lapangan yaitu dengan meninjau langsung kondisi lapangan dan mengumpulkan data dengancara mewawancarai pihak terkait selain itu dilakukan studi literatur dan setudi banding objek yang menyangkut faktor - faktor teknis dan persaratan bangunan yang mempengaruhi pada desain bangunan kemudian mengidentivikasi aspek - aspek yang dapat di terapkan dalam perancangan teknik dan desain yang sesuai dengan keriteria perancangan yang akan dicapai. Dari permasalahan yang di peroleh selanjutnya di analisis sehingga menghasilkan konsep perancagan dengan tema Arsitektur Eco Wisata.  Hasil yang diperoleh, yaitu desain Kawasan wisata pantai minanga dengan pendekatan arsitektur eko wisata yang di aplikasikan pada desain Kawasan yang memiliki kawasan konservasi mangrove, Kawasan edukasi serta Kawasan yang dapat di manfaatkan oleh warga sekitar untuk menumbuhkan perekonomian

Abstract

The aim of this research is to determine the feasibility of developing a brain-based learning mathematics module using a crossword puzzle game as seen from the level of validity, practicality and effectiveness. The method used is research and development with the ADDIE research design. This research was conducted at Yapis Ranting 2 Middle School with test subjects of 27 class VIII students. The validity results show that media validation reached a percentage of 83.33% and material validation was 89.98% in the "very valid" category. The module developed is also practical to use with a percentage of teacher response questionnaires of 70.90 with "practical" criteria and student response questionnaires of 81.61% with "very practical" criteria. Furthermore, the effectiveness seen from the results of the posttest with 21 students completing and 6 students not completing received a percentage of 80.76% for the "very effective" criteria. Based on these results, it was concluded that the module developed was suitable for use in learning.

Abstract

Vocational high school graduates are prepared to go straight into the workforce but there is a problem with high unemployment. The purpose of this study was to examine the effect of counseling teacher social support and gender on career optimism of vocational high school students. This study was conducted in March 2024, involved 348 students of Vocational High School. Using multiple regression analysis with the results showed that counseling teacher social support and gender had a significant effect on career optimism with a sig of 0.000 (sig<0.05). The social support of counseling teachers was also tested for each aspect, only instrumental support had an effect on students' career optimism and men have higher career optimism. The findings reveal that there is a need for social support from counseling teachers in order to influence students' career optimism to face the world of work after graduating from school.

Abstract

Gaharu (Aquilaria malaccensis) leaves contain alkaloids, terpenoids, flavonoids, saponins, and tannins that can help cure infections. This study aims to determine the antibacterial activity of n-hexane and ethyl acetate fractions of Gaharu leaves against skin infection bacteria such as Propionibacterium acnes, Pseudomonas aeruginosa, Staphylococcus aureus, and Staphylococcus epidermidis by Bioautography-TLC. Gaharu leaves symplisia was extracted using the maceration method using ethanol 96% solvent and fractionation with solid-liquid method to obtain n-hexane and ethyl acetate fractions. The result of the screening antibacterial activity of n-hexane and ethyl acetate fractions of Gaharu leaves with concentrations of 0,5% active to 4 bacteria test. The results of antibacterial activity n-hexane and acetaet ethyl fraction by Bioautography-TLC using n-hexane:ethyl acetate (1:4) eluent showed Rf 0.67 and Rf 0,16 value in the n-hexane fraction and an Rf 0.78 value in the ethyl acetate fraction active to 4 bacteria test.  The identification results of the chemical compound of the n-hexane fraction contain flavonoids and tannins (AlCl3 and FeCl3 reagents.), the ethyl acetate fraction contains flavonoids ( AlCl3 reagents).
