Abstract

Abstract. Fachrial E, Putri RRJS, Lister INE, Anggraini S, Harmileni, Nugroho TT, Saryono. 2020. Molecular identification of cellulase and protease producing Bacillus tequilensis UTMSA14 isolated from the geothermal hot spring in Lau Sidebuk Debuk, North Sumatra, Indonesia. Biodiversitas 21: 4719-4725. The industrial need for stable microbial enzymes tends to increase every year. The aim of this study was to isolate and identify the protease and cellulase producing thermophilic bacteria isolated from the geothermal spring of Lau Sidebuk Debuk, North Sumatra, Indonesia. The protease activity was determined based on the formation of halo zone on Skim Milk Agar, while cellulase was confirmed using CMC plate agar method. Nine bacterial isolates were successfully obtained from the sediments and water, and both activities were observed in only one isolate, known as UTMSA14. These isolates were characterized biochemically and morphologically. The molecular identification of UTMSA14 was done by 16S rRNA sequencing based on BLAST and phylogenetic analysis using MEGA X. Phylogenetic analysis showed that UTMSA14 clustered together with Bacillus tequilensis strain 10b (accession number NR_104919.1), which ascertained the isolate as Bacillus tequilensis strain 10b (accession number NR_104919.1). This is the first report of a thermophilic bacteria isolated from Lau Sidebuk Debuk hot springs, with the ability to produce cellulase and protease. further research is needed to purify and characterize the resulting enzymes, with a potential for industrial applications.

Abstract

Abstract. Fachrial E, Krisdianilo V, Harmileni, Lister INE, Nugroho TT, Saryono. 2021. Isolation, characterization, activity test and molecular identification of thermophilic bacteria producing proteases from Dolok Tinggi Raja Natural Hot Springs, North Sumatra, Indonesia. Biodiversitas 22: 1725-1732. Proteases are complex enzymes that can be applied in both commercial products and physiological fields. This enzyme accelerates the breakdown of peptide bonds in polypeptides and proteins using hydrolysis reactions, transforming them into simpler bonds. In recent years, the demand for protease enzymes in Indonesia is still dependent on imported products, and there has also been an increase in their use. This is because there are many advantages in terms of efficiency, environmental friendliness, and its ability to catalyze a reaction without by-products. Proteases dominate the enzyme market with a total of 70% sales. Furthermore, they are manufactured from thermophilic bacteria as a solution to the problem of enzyme use in industries, where enzymes are easily damaged at high temperatures. The thermophilic protease enzyme can be found in thermophilic microorganisms living in extreme temperatures such as hot springs. There are lots of mountainous areas in North Sumatra Province, Indonesia with many hot springs. For example, the Dolok Tinggi Raja Natural Hot Springs of Simalungun District which has a water temperature of 50oC with a pH of about 6, the potential of which still needs to be explored. This study aims to isolate and determine the activity and molecular structure of thermophilic bacteria producing protease in Dolok Tinggi Raja Natural Hot Springs. Protease activity was determined based on the formation of clear zones in Skim Milk Agar medium. In addition, a total of 20 isolates were successfully obtained from sediment and water. Each was tested biochemically and morphologically, where 19 isolates had the activity of proteases, with the highest in isolate UTMTR VK S9 of 23.67 U/mL. Molecular identification was performed by the amplification of 16SrRNA gene based on BLAST and phylogenic analysis with MEGA X. Phylogenic results showed that isolate UTMTR VK S9 possessed similarities to Bacillus paralicheniformis strain A30103. Meanwhile, more research is required to purify enzymes for application in the industrial world.

Abstract

Continuous exposure to ultraviolet radiation triggers the formation of Reactive Oxygen Species (ROS), leading to oxidative stress and damage to cellular biological components. This condition is closely associated with increased activity of the tyrosinase enzyme, a key enzyme in the melanin biosynthesis process that can cause hyperpigmentation. This study aims to evaluate the antioxidant activity and tyrosinase enzyme inhibition potential of the ethanolic extract of pandan leaves (Pandanus amaryllifolius) in vitro. The research method began with the extraction of pandan leaf powder using 70% ethanol as a solvent. Subsequently, qualitative  phytochemical screening was conducted to identify secondary metabolites. The antioxidant activity was evaluated using the DPPH method across various concentrations, with Vitamin C as the standard comparison. The tyrosinase enzyme inhibition assay was performed spectrophotometrically using L-DOPA as a substrate at a wavelength of 492 nm. Phytochemical screening revealed that the extract contains secondary metabolites, including flavonoids, alkaloids, saponins, and tannins. The antioxidant test using the DPPH method showed that at a concentration of 500 ppm, the extract achieved an inhibition percentage of 97.73%, which is comparable to the effectiveness of Vitamin C (99.36%). In the tyrosinase enzyme assay using L-DOPA as a substrate, the blank absorbance was 0.794, while the sample absorbance was 0.907. Although the sample absorbance was higher due to the extract's intrinsic color pigments, the calculation indicated a very strong enzyme inhibition effectiveness of 90.7%. In conclusion, the ethanolic extract of pandan leaves (Pandanus amaryllifolius) holds significant scientific potential as a natural therapeutic agent to counteract oxidative stress and inhibit melanogenesis in skin pigmentation disorders.

Abstract

Introduction: Gentamicin, an aminoglycoside antibiotic, is widely used for severe Gram-negative bacterial infections but poses a significant risk of nephrotoxicity, characterized by elevated serum creatinine levels. Muntingia calabura L. (cherry fruit) contains bioactive compounds with potential nephroprotective properties. Objective: This study aimed to evaluate the nephroprotective effect of cherry fruit extract on serum creatinine levels in gentamicin-induced white rats. Methods: An experimental post-test only control group design was employed using 25 male Wistar rats divided into five groups: normal control (K1), gentamicin control (K2), and treatment groups receiving gentamicin plus cherry fruit extract at doses of 100 (K3), 200 (K4), and 400 (K5) mg/kgBW. Gentamicin (100 mg/kgBW/day) was administered intraperitoneally for 7 days, while the extract was given orally for the same period. Serum creatinine levels were measured using the Jaffe method and analyzed using One-way ANOVA. Results: Serum creatinine levels in the gentamicin control group were 0.64 ± 0.09 mg/dL, while treatment groups K3, K4, and K5 showed levels of 0.62 ± 0.09, 0.61 ± 0.06, and 0.59 ± 0.10 mg/dL, respectively. Statistical analysis revealed no significant differences among groups (p > 0.05). A dose-dependent decreasing trend was observed, with the 400 mg/kgBW dose showing the lowest creatinine level, approaching the normal control value (0.55 ± 0.05 mg/dL). Conclusion: Cherry fruit extract demonstrated a tendency toward nephroprotective effects, indicated by dose-dependent reduction in serum creatinine levels; however, the effect was not statistically significant. Further research incorporating additional parameters, including BUN, renal histopathology, and oxidative stress biomarkers, is necessary to confirm its nephroprotective potential.

Abstract

The primary function of skin is to protect the inside of the body from physical and mechanical stresses. Skin is perceptive to its surroundings. Thus, a variety of skin issues may develop, including wounds. The standard wound treatment is chemical therapy using antiseptics. Antiseptics contain antimicrobial qualities that can eradicate bacteria. However, antiseptics can also destroy fibroblast tissue, which creates new skin tissue, and leukocytes, which are responsible for eliminating harmful bacteria. Alternative medicines, such as traditional medicine, are thus required to aid in wound healing. The purpose of this research is to determine the effectiveness of Allium chinense G. Don extract on the healing of cut wounds in white rats. This research is experimental research using a post-test control group design. Data analysis used One way ANOVA. The research found that Allium chinense G. Don extract contained alkaloids, flavonoids, saponins, and triterpenoids/steroids, but no tannin compounds. The antibacterial properties of Allium chinense G. Don are effective against Pseudomonas aeruginosa and Staphylococcus aureus bacteria. Collagen density and fibroblast count increased in Wistar rat wound tissue after being treated with the extract. Allium chinense G. Don is effective in healing cut wounds in white rats (p <0,001).

Abstract

Introduction: Diabetes mellitus is a chronic metabolic disorder with a high global prevalence and serious complications. The prediabetes stage is a critical target for prevention, often managed with α-glucosidase inhibitors. However, synthetic drugs such as acarbose are frequently associated with gastrointestinal side effects, highlighting the need for safer alternatives. Endophytic microbes from medicinal plants, including areca nut (Areca catechu), have the potential to produce bioactive compounds with antioxidant and antidiabetic activities. Objective: This study aimed to explore endophytic bacteria from fresh areca nut seeds as a source of metabolites with antioxidant and α-glucosidase inhibitory activity. Methods: Fresh areca nut seeds were sterilized using the surface sterilization method, followed by endophytic bacterial isolation and Gram staining. Cultures were fermented in Nutrient Broth, and supernatants were obtained by centrifugation. Antioxidant activity was assessed using the DPPH method, while α-glucosidase inhibition was tested in vitro with p-nitrophenyl α-D-glucopyranoside as substrate. Acarbose served as the positive control. Results: Seven Gram-negative, rod-shaped endophytic isolates (BP1-BP7) were obtained. Isolate BP1 exhibited the highest antioxidant activity with 45.7% inhibition. Furthermore, the BP1 supernatant demonstrated potent α-glucosidase inhibitory activity up to 93,6%, comparable to that of acarbose (91%). Conclusion: Endophytic bacteria from areca nut seeds possess dual bioactivity as antioxidants and α-glucosidase inhibitors, suggesting their potential as sustainable alternative antidiabetic agents worthy of further investigation.

Abstract

Cardiotoxicity is a common side effect of cisplatin (CIS) chemotherapy, often accompanied by gastrointestinal, renal, and hematological toxicities. This study investigated cardioprotective potential of hydroalcoholic basil leaf extract against CIS-induced toxicity in rats, evaluated through Troponin-T, high-sensitivity C-reactive protein (Hs-CRP), superoxide dismutase (SOD), malondialdehyde (MDA) levels, and cardiac histopathology. A quasi-experimental design was employed with eight groups of rats (n = 6 per group): normal, negative control, positive control (vitamin C), and treatment groups receiving basil extract (400 and 800 mg/kg body weight). CIS administration significantly elevated Troponin-T, Hs-CRP, and MDA levels while reducing SOD activity. Treatment with basil extract, particularly at 800 mg/kg, markedly attenuated these changes, restoring biochemical markers toward normal values. Histopathological analysis further confirmed the cardioprotective effect. In conclusion, basil leaf extract demonstrates promising potential as a cardioprotective agent against CIS-induced cardiotoxicity.
