Abstract

Tujuan dari penelitian ini adalah untuk mendeteksi adanya jamur pada sarang semi alami penyu lekang di Taman Nasional Meru Betiri Banyuwangi. Sebanyak 1 gram pasir pada 22 sarang penyu hijau digunakan sebagai sampel. Isolasi dan identifikasi dilakukan dengan pengamatan karakteristik makroskopis dari pertumbuhan koloni dan karakteristik mikroskopis dengan pewarnaan lactophenol cotton blue. Berdasarkan hasil penelitian sebanyak 3 sampel (13.6%) negatif dan 19 sampel (83.4%) positif terhadap pertumbuhan jamur. Beberapa jenis jamur yang telah ditemukan antara lain Aspergillus sp., Cunninghamella sp., Fusarium sp. dan Trichoderma sp.

Abstract

Fasciolosis is a disease caused by infection with liver fluke (Fasciola sp). Fasciolosis is a parasitic disease that is important because it can cause high economic losses. The aim of this study was to investigate the effect of gamal (Gliricidia sepium) leaf extract against mortality of Fasciola gigantica in vitro study. In present study was completely randomized design with six treatments and each treatment were divided into four replications. A total of 240 specimens from infected cattles were used in this study. The observation and recording of F. gigantica mortality in all treatments with concentrations of gamal leaf extract P1 (1%), P2 (2%), P3 (3), P4 (4%), K + (Nitroxynil 2%) and K- (physiological NaCl) rate was done in 30, 60, 90, 120, 150, 180, 210 and 240 minutes, respectively. The mortality of F. gigantica was confirmed if there was no movement when triggered by anatomy tweezers then dipped in slightly warm water (50 15í¢„Æ’"> ). The data was analyzed using ANAVA Faktorial and continued with Duncan Multiple Range Test. The results of this study were shown the lowest and the highest mortality rate recorded at minutes 30 and 240 respectively. Moreover, the mean of mortality rate were peaked in nitroxynil (K+) and 1% gamal leaf extract treatment, while the lowest mean of mortality rate occurs in NaCl (K-) treatment. Present study conclude that gamal leaf extract has anthelmintic effect on the mortality rate of Fasciola gigantica. Gamal leaf extract with 1% concentration was claimed to be the most effective.

Abstract

Penelitian ini bertujuan untuk mengetahui seroprevalensi kejadian Infeksius Bronkitis (IB) pada itik dan ayam buras berdasarkan pengujian antibodi. Itik dan ayam buras yang mengandung antibodi dipastikan pernah tertular virus IB karena tidak ada program vaksinasi.Sampling dilakukan di Pasar Mojosari Kabupaten Mojokerto, masing-masing sebesar 120 ekor, yang diambil selama kali empat kali, masing-masing sebanyak 30 ekor. Pengujian antibodi dilakukan dengan teknik indirect-ELISA menggunakan strain Massachusetts sebagai antigen. Hasil penelitian menunjukkan kasus seroprevalensi IB pada itik menunjukkan jumlah yang positif sebesar 30ekor (25%) dari 120 ekor sampel yang diperiksa. Pada ayam buras dari sejumlah 120 ekor yang diperiksa, 87 ekor (72.5%) diantaranya menunjukkan hasil positif. Reaktivitas antiboditerdeteksi ditunjukkan dengan kisaran titer antibodi antara 4.157-12.217 pada itik dan antara 4.173-170.795 pada ayam buras.

Abstract

This study aimed to determine the mutation of amino acid, nucleotide homology, phylogenetic tree, epitope prediction of Hemagglutinin-Neuraminidase protein Newcastle Disease (ND) Virus isolated from traditional market around Surabaya. Samples were from 37 chicken with cloacal swab and one positive samples for control (LaSota). Samples were inoculated on embyonate chicken eggs and identified with HA test confirmed with HI test. Positive samples processed by PCR using forward and reverse primer with 503 bp RNA target. The PCR result then analyzed with sequencing. Result of sequencing analysis showed that theres similiarity between samples amino acid and vaccine isolate its effect the percentage of nucleotide homology and phylogenetic relation between isolate. Epitope NGAANNSGWGAPIHDPDYIGG have high immunogenic value at all of isolate which good as vaccine candidate.

Abstract

This study aimed to determine the in vitro antibacterial activities of essential oil from cinnamon bark (Cinnamomum burmannii) on Methicillin-resistant Staphylococcus aureus (MRSA) isolated from raw milk. Essential oil from cinnamon bark obtained from the steam distillation method and essential oil was made in a series dilution with a concentration of 1%, 2%, 4%, and 8%. The antibacterial activities were tested using the disk diffusion method. Results showed from five isolates of MRSA, one isolate was sensitive of essential oil with a concentration 2% and all of the isolate was sensitive with a concentration 4% and 8%. The antibacterial activity showed by inhibition zones on  MRSA. The results suggest that the activity of the essential oils of cinnamon bark has an antibacterial effect on MRSA and it is developed as phytopharmaca.

Abstract

Infectious Bronchitis (IB) is disease causes great economic impact across nations. Spike protein (protein-S) is one of structural protein of Infectious Bronchitis Virus (IBV) located on enveloped of the virion. Molecular characterization of IBV could be conducted use Sodium Dodecyl Sulphate-Polyacrylamide Gel Electrophoresis (SDS-PAGE) to reveal the protein weight then confirmed through Immuno-Blotting assay. These methods were conducted to know the molecule weight of field isolate compared to seed vaccine. The results of this study indicate the weight of protein-S of field isolate I-147/11 were 168.21 kDa which was same to protein-S of seed vaccine Massachusset.

Abstract

The aim of this study was to identify the antibacterial activity potention of gamal leaf (Gliricidia sepium) ethanol extract against Escherichia coli growth which isolated from Olive Ridley Sea Turtle (Lepidochelys olivacea) semi-natural nest in Boom beach, Banyuwangi. The antibacterial test used disk diffusion method. Oxytetracycline 30 µg (Oxoid CT0041B) was used as the Positive Control (K+). CMC Na 0,5% was used as the Negative Control (K-). The treatment (P1, P2, P3, P4) used various concentrations of gamal leaves ethanol extract, which is 40%, 50%, 60%, and 70%. The study assigned six treatments and four replication. The obtained data were analyzed by ANOVA, then continued with the DUNCAN test if the results showed significant differences (p<0,05). The inhibition result of positive control (K+) was 24,70c ± 7.04; and the negative control was 0,00a ± 0,00. All extract concentration result different inhibition, concentration 1 (P1) was 10,41 b ± 3,10; concentration 2 (P2) 8,14 b ± 0,45; concentration 3 (P3) 8,08 b ± 0,47; and concentration 4 (P4) 8,01b ± 0,29. These results indicated that the gamal leaves ethanol extract has antibacterial activity to inhibit the growth of E. coli isolated from Olive Ridley semi-natural nest in Boom beach, Banyuwangi.

Abstract

Salmonella spp. is one of the major causes of the world's foodborne disease that can spread through meats, eggs, or milk. Salmonella spp. is an enteropathogenic bacteria that can cause gastrointestinal infections. This study aimed to identify the contaminations of Salmonella spp. bacteria on the broiler meats at Banyuwangi and Blambangan traditional markets Banyuwangi district. The method of this study used 24 samples of broiler meat bought from Banyuwangi and Blambangan traditional markets in the Banyuwangi District. The samples were collected using the purposive sampling method. The results found that 5 out of 24 samples (21%) were tested positive for Salmonella spp. contamination and 19 were tested negative. This study was indicated that broiler meats at Banyuwangi and Blambangan traditional markets Banyuwangi district have not suitable with SNI (Indonesia's National Standard) 3924:2009 that requires at least 25 gram of fresh chicken meat to be free of or tested negative for Salmonella.

Abstract

This study aimed to discover the homology of nucleotide sequence, homology percentage, and those relations phylogenetic of protein Fusion (F) gene coding of Newcastle disease in domestic chicken (Gallus gallus domesticus) in Surabaya using some comparison isolate from GenBank. Samples were scoured of digestive organs from native chicken, that was collected from a traditional market in Wonokromo, Surabaya. Samples were tested using RT-PCR with primer forward and reverse with target 976bp, a positive sample which is continued with sequencing then homology and nucleotide analysis which is done and which is translated into amino acid. The result of homology chc/SBY/2018 sample has a similarity of 88% with references sequence, while with Lasota vaccine has a similarity of 87%, and the highest result of homology showed by the comparison with various isolates in Indonesia 90-95%. Translation results from nucleotide alignment into amino acid showed shifts in amino acid structure, which is amino acid shifts could be affected by many things like nutrition, wheater, environment, etc. The conclusion was chc/SBY/2018 sample has a quite high similarity with Indonesian isolates and undergoes mutation on nucleotide structure on amino acid and phylogenetic analysis. This study related to some isolates of vaccine and some isolates in Indonesia.

Abstract

This study analyzed the association between risk factors and Feline panleukopenia at the Ichiyo Veterinary Clinic, Madiun using a retrospective method. A total of 2037 data used was secondary data obtained from cat medical and ambulatory reports from January 2018-March 2021. The risk factors analyzed included age, gender, vaccination status, comorbid diseases, housing system, number of cats, and season. Data was presented descriptively and analyzed using Chi-Square analysis. The results showed that the risk factors for Feline panleukopenia were cats aged ≤6 months, cats that were not vaccinated, and cats with comorbid diseases. It can be concluded that prevention of Feline panleukopenia can focus on age, vaccination and treatment of potential comorbid diseases in cats.
