Abstract

Abstract. Rahmi HA, Syukur M, Sukma D, Aisyah SI. 2024. Diversity of quantitative, qualitative, and vitamin C content characteristics in F5 generation of chili pepper Capsicum baccatum species. Biodiversitas 25: 2878-2886. Chili is a popular vegetable worldwide, and Indonesia is one of the largest chili producers. However, the cultivation of Capsicum baccatum species is still uncommon in Indonesia. This study aimed to assess the diversity and heritability of yield and yield quality of F5 populations of C. baccatum. This study was conducted at the Experimental Field of Perumahan Alam Sinar Sari, Bogor. The experiment followed a randomized complete block design (RCBD) using 12 genotypes with three replications and four sample plants per replication. The F5 populations of the following genotypes were used: 4k-6p, 4k-7p, 7m-4q, 2m-2m, 7m-3q, 7m-3m, 7m-2q, 2m-2q, 7m-8m, 7m-1m, Bishop Crown, and Lemon Drop. The observations included quantitative, qualitative character, vitamin C contents, and soluble solids. The results showed that the fruit weight per plant ranged from 37.73 to 94.24 grams, while the number of fruits per plant ranged from 11.46 to 30.95. Fruit length ranged from 2.65 to 6.08 cm, and fruit diameter ranged from 15.70 to 41.43 mm. The weight per fruit ranged from 3.43 to 14.89 grams. Brightness or L* values ranged from 19.06 to 41.93, a* values ranged from 5.62 to 31.49, and b* values ranged from 22.90 to 57.45. The chroma value ranged from 30.87 to 64.20, and the hue value ranged from 46.54 to 80.23. The analysis of vitamin C content showed a range of 12.20 to 17.74 mg/20 g, and the test results of soluble solids ranged from 8.77% to 13.49%. All quantitative characters showed high heritability, indicating a strong genetic influence on these characters. The qualitative observations were analyzed, and the results showed that the 12 C. baccatum chilies had similarity coefficients ranging from 0.0 to 0.5. The similarity analysis resulted in grouping the 12 genotypes of C. baccatum into four main groups.

Abstract

Abstract. Sari YN, Aisyah SI, Sukma D, Syukur M. 2025. Increasing ploidy level of marigold (Tagetes patula Sudamala Barak) using colchicine with acute and chronic methods. Biodiversitas 26: 2726-2734. Marigold (Tagetes sp.) is a popular and potential ornamental plant in Indonesia, especially in Bali, commonly used in religious ceremonies and industry. A significant challenge in the floriculture industry is producing plants with desirable traits, such as larger flowers. Sudamala Barak has dark red flowers with small size (4,5-5,6 cm), but consumers prefer larger ones (8-10 cm). Colchicine is a chemical mutagen that disrupts spindle fiber formation during mitosis, thereby inducing chromosome doubling and resulting in polyploid cells. Tagetes patula is an allotetraploid species (2n = 48). This study aimed to determine the LC50 value and analyze the morphological, histological, and cytological characteristics of Sudamala Barak mutants. The research was conducted from May 2024 to February 2025 using two application methods: acute (seven concentrations) and chronic (four concentrations) based on LC50 (lethal concentration 50%). The results showed that colchicine significantly affected various plant characteristics. The LC50 value was found to be 0.346%. Overall, the chronic method produced greater diversity than the acute method. Colchicine treatment influenced plant height, canopy width, number of primary branches, stem diameter, flowering time, and flower diameter. It also increased the number of stomata and chloroplasts, potentially improving photosynthetic efficiency. In the acute method, the highest ploidy level was euploid-pentaploid (2n = 60) at concentrations of 0.05%; 0.1%; 0.15%; 0.25%; and 0.3%. In contrast, in the chronic method, the highest ploidy level was euploid-autoalooctaploid (2n = 96) at 10× and 20× LC50 dilution concentrations.

Abstract

The evaluation of citrus resistance to Huanglongbing (HLB) disease is still challenging due to the incapability of the bacteria to be cultured purely in artificial medium, the complexity of inoculation methods, and the long duration required for phenotypic observation. Thus, the use of molecular markers is one of the alternatives to solve this problem. The focuses of this study were to perform in silico analysis of the nucleotide variations in NONEXPRESSOR OF PATHOGENESIS-RELATED GENES 1 (NPR1) gene sequence among several citrus genotypes whose resistance information to HLB have been known previously and to employ phylogenetic analysis among them. The NPR1 gene sequences from 20 genotypes which consisted of 14 citrus and six relative genotypes were collected in silico from Citrus Genome Database and analyzed using multiple sequence alignment program. A total of six interesting SNPs that could distinguish between susceptible and resistant citrus genotypes were detected in this study. As many as five SNPs were non-synonymous, while only one synonymous SNP that did not cause the amino acid change was identified in this study. The phylogenetic analysis also revealed the separation between susceptible and tolerant/resistant citrus genotypes in two main clusters. The SNPs found in this study are expected to be useful for designing new functional markers as a selection tool in future studies. Keywords: in silico, molecular marker, phylogenetic analysis, selection tool, SNP
