Abstract

Background: Combined oral contraception is one of contraception methods that contains estrogen and progesterone. Both of that hormones have an effect on blood pressure levels because of the using duration of combined oral contraception, Body Mass Index (BMI), and acceptor age. The purpose of this research is to analyze the correlation between the using duration of combined oral contraception and blood pressure levels among fertile women in Jaten area, Karanganyar. Methods: This research used observational cross sectional analytic approach method. The population were fertile women who use combined oral contraception. The total sample were 48 patients who appropriate with inclusion and exclusion criterias. The data were collected by kuesioner, interview, anthropometry measurement, and blood pressure direct measurement. Those variables such as the using duration of combined oral contraception, blood pressure levels (systolic and diastolic), age, and body mass index (BMI) were analyzed using Kolmogorof-Smirnovs test of normality and linear regression analysis using SPSS 16 for Windows. Results: The result of simple statistic test using linear regression showed the correlation significancy between the using duration of combined oral contraception and blood pressure levels, both systolic (p=0,004) and diastolic (p=0,012). Conclusions: There is a correlation between the using duration of combined oral contraception and blood pressure levels among fertile women. Abstract

Background: Systemic Lupus Erythematosus or SLE is a chronic autoimmune disease with varied clinical manifestations. SLE prevalence in Indonesia is being dominated with female, especially in their productive age. Certain etiology of SLE is still unknown until now, but genetic factors and immune system are being estimated as the primary factors. The first line therapy of SLE is cyclophosphamide. Unfortunately if being consumed more than six months, cyclophosphamide has several side effects, like bone marrow depression, infections, alopecia, hemorrhagic cystitis, and infertility. Because of that side effects, herbal medicine is being developed nowday. One of them is Andrographis paniculata Ness. leaf. The aim of this study was quantitative cytotoxic test for Andrographis paniculata Ness. leaf on T lymphocyte culture of Systemic Lupus Erythematosus. Methods: This study was anexperimental study. Subjects in this study were patients with SLE who appropriated SLEs criterias and normal people without allergy and autoimmune diseasess history. This study was done by doing T lymphocyte culture from intravenouss blood sample and observing cells absorbance with ELISA reader. Collected data were used for determining live cells percentage (IC50) and analyzed with Statistical Product and Service Solution (SPSS) 11.00 for Windows. Mean resistance data was tested by linear regression test. Results:The result showed that IC50 dose of Andrographis paniculata Ness. leaf extract treatment on SLE samples was smaller than the normal people. In Andrographis paniculata Ness. leafs extract treatment, IC50 in SLE = 0.00074 and in normal people = 0.19584. Conclusion: Andrographis paniculata Ness.leaf extract had stronger cytotoxic effect on T lymphocyte culture of patients with SLE than the normal people. Abstract

Introduction: Dengue Hemorrhagic Fever (DHF) is an infectious disease caused by dengue virus which commonly found in the tropic and sub-tropic region. Nowadays, the main treatment of DHF is supporting therapy by giving a replacement fluid. However, the number of cases and deaths due to dengue infection in Indonesia still high enough. Another solution to fix this problem is using Indonesians native plants which potentially has antiviral effect for dengue virus, such as guava leaf (Psidium guajava), patikan kerbau leaf (Euphorbia hirta), and betel leaf (Piper betle L.). The aim of this study is to determine the effectiveness of the leaf extract of guava (Psidium guajava), patikan kerbau (Euphorbia hirta), and betel (Piper betle L.) as antidengue in vitro. Methods: This study was laboratory experimental research. The subject of this research was dengue virus serotype 2 strain New Guinea C (DENV2 NGC) which was infected into Huh-7 cell line. The study was held in two places, they were Virology and Biology Molecular Laboratory, Department of Microbiology, Faculty of Medicine, University of Indonesia and Pharmaceutical Laboratory, Faculty of Mathematic and Science, Sebelas Maret University. The study was conducted in 2 parts. First, dengue virus solution was mixed with herbal extract in well which its bottom contained Huh-7 cell monolayer to obtain percent infectivity by FFU assay. Second, Huh-7 cell monolayer was exposed to the herbal extract solution to obtain percent viability by MTT assay. The effectiveness of herbal extract was based on two criterias, there were average percent infectivity 20% and average percent viability > 50%. Results: The leaf extract of Psidium guajava, Euphorbia hirta, and Piper betle L. have average percent infectivity respectively: 7.4%, 65.3%, and 68.4%. While the average percent viability of them were: 88.7%, 95.3%, 96.2%. Conclusion: The leaf extract of Psidium guajava is effective as antidengue in vitro. Abstract

Background: Systemic lupus erythematosus (SLE) is an autoimmune disease that causes chronic inflammation in multiorgan, in which its etiology and pathogenesis are unclear. Curcumin has been found to have a selective cytotoxic effects on various types of cancer cells through induction of apoptosis on the various pathways. This study aimed to demonstrate whether curcumin extract had cytotoxic effects on T lymphocytes autoreactivity of patients with SLE. Methods: This research was an experimental laboratory with post test only controlled group design. Samples were using cell cultures of T lymphocytes from the veins blood of a patient with SLE and normal people. Each of T lymphocytes cell culture consisted of media control (KM), cells control (KS), the positive control with cyclophosphamide (KP1), and a group treated with curcumin extract (KP2). Curcumin extracts and cyclophosphamide were divided into 8 serial doses (50 g/mL, 25 g/mL, 12.5 g/mL, 6.25 g/mL, 3.125 g/mL, 1.5625 g/mL, 0.78125 g/mL, 0.390625 g/mL). Cell inhibition was calculated based on a percentage (%) of cell viability. Then, the data was analyzed using probit analysis to determine the dose of IC50 with SPSS for Window Release 17.0. Results: The result of cell inhibition probit analysis (IC50) on T lymphocytes cell culture of patients with SLE using curcumin extract was active (score 3) and T lymphocytes cell culture of patients with SLE using cyclophosphamide was moderately active (score 2).While, T lymphocytes cell culture of normal people using curcumin extract was active (score 3) and T lymphocytes cell culture of normal people using cyclophosphamide was moderately active (score 2). Conclusions: Curcumin extract had cytotoxic effect on T lymphocytes cell culture of SLE. The cytotoxic effect of curcumin extract on T lymphocytes cell culture of patients with SLE was stronger than cyclophosphamide. Keywords: curcumin extract, cytotoxic, SLE

Abstract

Background: Centella asiatica L.Urban contains antioxidant compounds such as triterpenoid, essential oil and flavonoid. The compounds have significant activities to reduce oxidative stress that can make damage of kidney cells. This study aimed to investigate protection effect of Centella asiatica L.Urban extract on the kidney histological structure of mice (Mus musculus ) induced by toxic doses of paracetamol. Method: This study was an experimental laboratory research. Samples were 30 mice, Swiss webster type, 2-3 weeks old and 20 grams of weight. Samples were divided into 5 groups, each group consisted of 6 mice (Mus musculus). The control group (KK) was given no treatment, just regular feed. Induction group (KI) was induced by paracetamol but did not get any treatment, just got distilled water. KP1, KP2, and KP3 groups induced by paracetamol got treatment with Centella asiatica L.Urban extract at a dose of 5 mg, 10 mg and 20 mg per 20 gram of body weight. Kidney histological damage were assessed by the sum of the cell nucleus pyknosis, karyorrhexis, and karyolysis. Data were analyzed using one-way ANOVA and post hoc multiple comparison test. Result: The ANOVA test for all groups showed that p value was 0.000 (p < 0.05). The post hoc multiple comparisons test showed the significant differences between group KK-KI, KK-KP1, KK-KP2, KK-KP3, KI-KP1, KI-KP2, KI-KP3, KP1-KP2, KP1-KP3, and KP2-KP3. Conclusion: Centella asiatica L.Urban extract has a protection effect to kidney histological structure of mice (Mus musculus) induced by paracetamol. Abstract

Introduction : Broccoli is known to contain antioxidants such as vitaminC, flavonoids and sulforaphane. Antioxidants can protect liver from damage caused by free radicals. This study aims to prove that broccoli extract protect liver cell from damage in mice induced by paracetamol in toxic dose and an increased doses of broccoli extract enhance the effects of protection to liver cell in mice. Methods : This study was an experimental laboratory with post test only controlled group design. Samples (30 mice) were taken by incidental sampling. Samples divided randomly into 5 groups, each group consisted of 6 mice. K1 was given distilled water. K2 was given paracetamol in toxic dose. KP1, KP2 and KP3 were given broccoli extract dose I, II and dose III respectively for 14 days and also were given paracetamol in toxic dose on day 12, 13 and 14. On day 15, the livers of mice were taken for making preparations with HE staining. The liver cell damages were observed by counting the nuclei of cells undergoing pyknosis, karyorrhexis and karyolysis. Data were analyzed by Oneway ANOVA and Post Hoc LSD analysis test (? = 0.05). Result : K2 had the most damaged cells (157.17 7.834 cells) and the least damaged cells were at K1 (1.50 1.049 cells). Oneway ANOVA test result was p = 0.00 (p < 0.05). Result of Post Hoc LSD analysis test showed a significant difference between groups on all pairs with p = 0.00 (p < 0.05). Conclusions : Broccoli extract protect liver cells from damage in mice induced by paracetamol. Increased doses of broccoli extract were proven to increase the protection effect on liver cells. Abstract

Background: Kasus COVID-19 di Indonesia masih terus ada dan mengalami pasang surut, salah satunya di Kabupaten Klaten. Keterbatasan petugas kesehatan dan rendahnya kesadaran masyarakat dalam menerapkan protokol kesehatan menjadi salah satu penyebab masih adanya kasus COVID-19 di Kabupaten Klaten. Kegiatan ini bertujuan untuk meningkatkan peran kader kesehatan dalam membantu meningkatkan kesadaran masyarakat menerapkan protokol kesehatan. Kegiatan ini dilakukan bekerjasama dengan Puskesmas Trucuk 1 dan Puskesmas Trucuk 2. Peserta kegiatan berjumlah 60 orang perwakilan seluruh Desa di Kecamatan Trucuk. Metode: Metode yang digunakan Focus Group Discussion, workshop serta pelatihan yang dilakukan pada bulan November-Desember 2021. Hasil kegiatan yaitu ada peningkatan pemahaman peserta setelah dilakukan kegiatan workshop dan pelatihan. Hasil: Pelatihan dengan praktik simulasi menambah wawasan peserta dalam melakukan edukasi kepada masyarakat. Kesimpulan: Peran kader kesehatan dapat menjadi solusi dalam menyadarkan masyarakat.

Abstract

Sepsis and septic shock are the major health problems worldwide, including in Indonesia. Sepsis and septic shock are medical emergencies that require adequate prompt resuscitation to reverse tissue hypo-perfusion, which will prevent patient mortality. The latest international guideline on septic shock treatment recommended norepinephrine as a first-line vasopressor followed by dopamine and dobutamine as an alternative vasopressor. However, the practices still vary. Therefore, a scoping review is required to scope the existing article, summarize scientific evidence, and give a bearing on future research regarding the effectiveness of norepinephrine-vasopressin combination in reducing mortality in septic shock patients. This scoping review was covered articles published after 2011. A total of 953 articles were collected. Seven articles comprised of 5 systematic reviews, 1 randomized controlled trial, and 1 cohort study with a total 21,670 of patients, were included for qualitative synthesis. From the analysis, there had not been enough scientific evidence to conclusively determine the combination of vasopressors as the best therapeutic outcome of sepsis treatment. In conclusion, based on the existing articles there is inadequate scientific evidence to definitively conclude the effectiveness of the combination of vasopressors and norepinephrine for the treatment of septic shock patients. Further research is needed to explore the effectiveness of the norepinephrine-vasopressin combination in reducing mortality in septic shock patients.

Abstract

Pendahuluan: Diabetes mellitus (DM) merupakan salah satu penyakit metabolik kronis yang masih menjadi perhatian di dunia. Penyakit ini ditandai dengan tingginya kadar glukosa pada sirkulasi darah atau disebut dengan hiperglikemia. Deteksi dini penyakit DM dan pemantauan kadar glukosa darah pada penderita DM sangatlah penting untuk dilakukan. Hal tersebut dikarenakan apabila kadar glukosa darah sangat tinggi atau tidak terkontrol akan menyebabkan terjadinya komplikasi serius diberbagai organ tubuh, sehingga dapat meningkatkan morbiditas dan mortalitas pasien. Saat ini telah tersedia alat pemeriksaan glukosa darah yang mudah digunakan masyarakat yaitu glukometer. Namun, nampaknya masih banyak masyarakat yang belum terampil menggunakannya. Selain itu banyak masyarakat yang belum memahami penyakit DM secara holistik sehingga perlu dilakukan kegiatan pengabdian masyarakat tentang penyakit DM.Metode: Metode pengabdian masyarakat ini adalah edukasi tentang penyakit Diabetes Melitus secara holistik melalui penyebaran video di grup-grup Whatsapp, Instagram, Facebook dan Youtube. Video edukasi tersebut terdiri dari 3 buah video, yaitu video tentang penyakit DM secara holistik, video cara penggunaan glukometer untuk memeriksa glukosa darah, dan video tentang DM di era COVID-19. Setelah disebarkan, masyarakat diminta memberikan respon melalui kuesioner. Hasil dan pembahasan: Sebagian besar responden sangat setuju bahwa video yang diunggah membuat mereka menjadi lebih paham mengenai penyakit diabetes melitus dan cara menggunakan glukometer.Kesimpulan: Kegiatan pengabdian kepada masyarakat dapat meningkatkan pengetahuan dan pemahaman masyarakat mengenai penyakit DM dan cara memeriksa glukosa darah secara mandiri dengan glukometer.

Abstract

ABSTRAKPendahuluan : Awal tahun ini, dunia menghadapi pandemi Covid-19. Berawal dari Wuhan China, dan sampai di Indonesia. Dampak Covid-19 adalah kasus dan kematian yang parah. Di Indonesia, ada 57.000 kasus yang dikonfirmasi dan 2.934 meninggal. Lebih dari 82% kasus orang lanjut usia. Dalam hal komorbiditas, sebagian besar kasus kematian memiliki hipertensi, diabetes melitus, gagal jantung dan gagal ginjal. Kemudian bagaimana Indonesia penduduk rentan ini. Penelitian ini bertujuan untuk menilai respon untuk melawan Covid-19 melalui pengawasan masyarakat, terutama bagi penduduk yang rentan.Metode : Metode yang digunakan adalah responden yang diwawancarai secara online di antara 25 pemimpin rumah tangga sebagai data kuantitatif. Metode pengukuran adalah kualitatif online di kalangan masyarakat, tokoh masyarakat, pimpinan rumah tangga, kader, dan pemerintah desa.Hasil dan diskusi : Hanya ada 80% setuju bahwa infeksi COVID-19 melalui tetesan. Ada 84% pemimpin rumah tangga yang memahami gejala. Mereka memiliki pengetahuan yang baik tentang cuci tangan, masker, jarak sosial, dan isolasi. Pengetahuan tentang pencegahan sangat rendah. Hanya 32% yang tahu bahwa lansia memiliki risiko mati pada COVID-19. Itu termasuk hipertensi dan diabetes melitus. Terkait hasil kualitatif, hanya 2 informasi dari 15 informasi yang memahami jumlah penduduk rentan di daerahnya. Tidak ada kebijakan yang jelas terkait dengan penduduk yang rentan terutama di kabupaten dan di bawah pada perlindungan yang rentan. Tidak ada pengawasan masyarakat di antara penduduk yang rentan.Kesimpulan : Pengawasan masyarakat terkait COVID-19 kesesuaian, tetapi penduduk rentan masih khawatir. Rekomendasinya yaitu mengembangkan model pengawasan masyarakat tentang perlindungan populasi yang rentan terhadap COVID-19.Kata Kunci: Community Surveillance , Covid-19, Era Pandemi
